Low temperature modulates the carbon allocation in different metabolic pathways to improve the tolerance of Arctic Chlorella to high light stress

Tong Wang,Meng Gao,Hong Song,Changhai Wang,Meilin He

Polar microalgae, renowned for their exceptional adaptability in extreme environments, are vital for carbon fixation and nutrient cycling. To understand their metabolic resilience under low temperature and high light stress, we conducted an extensive biochemical and full-length transcriptome analysis of Arctic Chlorella ( Chlorella- Arc) and temperate Chlorella ( Chlorella- Temp). This research aimed to comprehensively assess Chlorella 's adaptability to the interactive effects of temperature and light stress. Under low-temperature stress, intense light inhibited Chlorella -Temp growth by 14.8 %, while it significantly promoted Chlorella -Arc growth by 88.8 %. The cold-adapted Chlorella -Arc demonstrates higher tolerance to high light stress. Elevated temperature (+0.80) and light (+0.47) directly increased cell density, revealing direct and indirect effects of temperature and light on Chlorella- Arc bioproducts via structural equation modeling. We then analyzed key molecules involved in lipid metabolism using orthogonal projections to latent structures discriminant analysis, confirmed that Chlorella -Arc maintained a relatively constant fatty acids composition under low temperature and high light stress. Compared to its temperate counterpart, Chlorella -Arc exhibits unique adaptability by modulating polysaccharide composition, downregulating carbon fixation, reducing nitrogen absorption, and maintaining stable unsaturated fatty acid levels with a higher unsaturated to saturated fatty acid ratio. Chlorella- Arc, a prime example of polar microalgae adaptability, exhibits unique metabolic responses. Compared to the low-temperature normal-light group, the high-light Chlorella demonstrates precise regulation of nitrogen uptake and carbon allocation, while activating multiple antioxidant pathways to mitigate the increase in ROS levels under light stress. Acclimated Chlorella- Arc tends to increase triacylglycerol accumulation, suggesting a shift in carbon flow towards lipid production. Our analyses reveal specialized adjustments, enabling survival in harsh conditions of low temperature and intense light. This research enhances our understanding of microalgal resilience in extreme environments, offers valuable insights into the utilization of polar microalgal bioproduct resources.

Starch accumulation dynamics and transcriptome analysis of Chlorella sorokiniana during transition of sulfur nutritional status

Xu Haiqing,Yang Jinzhi,Wang Xu,Peng Qing,Han Yanxia,Liu Xudong,Liu Kexin,Dou Shijuan,Li Liyun,Liu Guozhen,Yang Ming

Microalgae can effectively accumulate starch by using nutritional limitation methods in the context of bioalcohol fuel production. However, relatively few studies have focused on starch accumulation in microalgae and its molecular basis, especially under sulfur limitation conditions. In this study, the starch accumulation dynamics and physiological responses of Chlorella sorokiniana under sulfur starvation (SS) and sulfur replenishment (SR) conditions were investigated, and the genes involved in the transcriptional regulation were explored using RNA-seq. The starch content in C. sorokiniana cells significantly increased from 1.6% to 55.0% of dry weight within 24 h under SS conditions, and then, it decreased to 3.4% within 12 h after transition to SR conditions. However, cell growth was inhibited, and pigment content decreased under SS conditions. Using RNA-seq analysis, a total of 9720 differentially expressed genes (DEGs) induced by sulfur status were obtained. These genes were narrowed down to 454 starvation and replenishment cross-validated (SRV)-DEGs, among which 283 SRV-DEGs were significantly up-regulated and 171 SRV-DEGs were down-regulated under SS conditions, and returned to their previous state under SR conditions. The SRV-DEGs enriched in the sulfate metabolism pathway were all up-regulated under SS conditions after 6 h to speed up the sulfur metabolic cycle, and the transcriptional abundance of a sulfate transporter (SULTR4), cysteine synthase[O-acetylserine(thiol)-lyase] (OASTL), serine acetyltransferase (SAT), and methanethiol oxidase (SELENBP1) increased 8.6-fold, 12.6-fold, 8.7-fold, and 12.4-fold, respectively. Protein synthesis was correspondingly inhibited, which resulted in the reallocation of carbon and elevated the starch synthesis pathway, in which the expressions of glycogen branching enzyme (GBE) and starch synthase (SS) were up-regulated 12.0- and 3.0-fold, respectively. The fatty acid desaturase (FAD) and phosphatidic acid phosphatase (PAP) in the lipid synthesis pathway were strongly up-regulated 8.8- and 16.2-fold, respectively, indicating the competitive synthesis of lipids. The down-regulation of SRV-DEGs associated with carbon fixation, such as those in the Calvin cycle, possibly affected cell growth. The time-resolved transcriptional analysis identified the SRV-DEGs, revealing the underlying starch accumulation mechanism, as well as the relationship with cell growth and lipid synthesis.

A sucrose transporter from Gongronella butleri w5 mediates plant-fungus-bacteria interaction

Junnan Fang,Xiaojie Wang,Li Li,Juanjuan Liu,Ling Fang,Yazhong Xiao,Yan Wang,Zemin Fang

Beneficial fungi in the plant rhizosphere connect plants and soil bacteria by releasing carbon sources as a bridge. Sucrose, a direct product of plant photosynthesis, plays a crucial role in this process. Here, we report that the sucrose transporter (SUT) protein GspSUT1 worked as a key regulator in the plant-beneficial fungus Gongronella butleri w5 (w5) and promoted plant growth by modulating carbon allocation during plant-fungus-bacteria interactions. The GspSUT1 expression was upregulated during the w5-plant interaction, accompanied by a decrease in sucrose in the root of Actinidia chinensis var. Chinensis “Hongyang.” Knocking down the expression level of GspSUT1 through host-induced gene silencing (HIGS) led to the accumulation of sucrose in the plant root and a reduction in the release of monosaccharides into the environment, which in turn decreased the abundance of soil nitrogen-fixing bacteria and lowered the available nitrogen content in the soil, thereby weakening the ability of w5 to promote plant growth. Our research suggested that GspSUT1 from w5 played a crucial role in promoting plant growth and proved that the SUTs in beneficial fungi can influence soil microbial composition by regulating carbon allocation during plant-fungus-bacteria interactions, thereby improving soil nutrient availability and promoting plant growth.

Precise control of chromatin loop extrusion enhances sustainable green revolution yield in rice

Liu Qian,Liu Hui,Sun Lili,Wang Dekai,Sun Xueyuan,Wu Kun,Ye Yafeng,Wang Yun,Wang Shuoxun,Zhong Nan,Wu Xiaokang,Li Yu,Deng Min,Wang Meiyue,Wu Chenchen,Liu Xueying,Zhang Yijing,Xiao Jun,Wu Yuejin,Fu Xiangdong

Continuous and excessive use of inorganic fertilizers underlies current global crop production; therefore, reducing fertilizer use while increasing crop productivity is critical for ensuring agricultural sustainability and food security. Here we show that the natural variant of RCN2 , a rice TERMINAL FLOWER 1/CENTRORADIALIS homolog, enhances photosynthesis, nitrogen assimilation and grain yield by restricting chromatin loop extrusion. RCN2 competitively inhibits the interaction between growth-repressing DELLA proteins and SQUAMOSA PROMOTER BINDING PROTEIN-LIKE transcription factors, breaking the green revolution trade-off between plant growth and metabolism of carbon and nitrogen. We demonstrate that targeting CCCTC-containing insulator elements at the RCN2 locus confers not only decoupling tillering and panicle branching without affecting beneficial semi-dwarfism, but also improves source-to-sink carbon allocation and nitrogen-use efficiency, consequently increasing harvest index and rice yield at low nitrogen fertilization levels. Precise modulation of loop extrusion thus enables new breeding strategies to reduce nitrogen fertilizer use in high-yield cereal crops.
