Biocatalytic kinetics of the reaction between CO2 and tertiary amine using carbonic anhydrase

Meng-Meng Du,Yuan-Cheng Wang,Bao-Chang Sun,Yong Luo,Liang-Liang Zhang,Guang-Wen Chu,Hai-Kui Zou

Carbonic anhydrase (CA) is a high-efficiency biocatalyst that significantly improves the absorption of CO 2 by tertiary amine. This work aims to investigate kinetics behaviors from the perspective of enzymatic reaction mechanism. The influences of the CA concentration, type of tertiary amines, pH, and temperature on the reaction rate between CO 2 and tertiary amine ( ν ) and catalytic activity of CA ( φ ) were first investigated in a stopped-flow device. Adding 50 g∙m⁻³ CA enhanced ν in tertiary amine solutions by a factor ranging from 22 to 42 at 298 K and pH=9.5, demonstrating its excellent catalytic performance. The ν increased with increasing CA concentration, pH, temperature, and tertiary amine's pK a. φ increased with the increase of CA concentration, as well as the decrease of temperature, pH, and tertiary amine's pK a. Proteomics analysis further revealed that conformational changes of the CA's secondary structure induced by high pH and temperature altered the expressions of the local active-site region and deactivated CA, ultimately leading to a decrease in φ . Additionally, the CA-catalysis kinetics equation accorded with the Michaelis-Menten model, with catalytic second-order rate constants on the magnitude of 10 7 . Overall, this work provides a guideline for its industrial application in the CO 2 capture process.

Cost-Effective Preparation of Carbonic Anhydrase with Superior Performance for Assisting Amine and Amino Acid Ionic Liquid Blends in CO2 Absorption and Desorption

Lei Mao,Yaxin Chen,Ruifang Zhang,Chun Yang,Yanhong Zhou,Guangya Zhang

The utilization of carbonic anhydrases (CAs) for CO2 capture aligns with the principles of environmental sustainability. However, there is an urgent need to minimize the cost of CA purification while improving their performance in harsh environments. A novel strategy was proposed to solve the above problems by employing the ferritin-tagged CA variant (DvCA8.0-F). The purification is achieved by low-speed centrifugation, yielding 92% activity recovery and 95% purity. After 14 weeks of incubation in a 50% MDEA solution at 50 °C, DvCA8.0-F maintained activity nearly equivalent to the initial activity, which showed remarkable stability compared to the free ones. Encouragingly, DvCA8.0-F had a yield of up to 800 mg/L, allowing for cost-effective CA-assisted CO2 absorption and desorption in MDEA solutions. The results show that DvCA8.0-F reduced the absorption time from 70 to 50 min at 40 °C and desorption time from 40 to 25 min at 96 °C in 25%MDEA+1%[N1111][Gly] solution with a final CO2 load and a stripped amount of CO2 at the same time. DvCA8.0-F exhibits the maximum reaction rate at 96 °C; the CO2 regeneration efficiency improved from 60% at 87 °C to 72%. When the concentration of DvCA8.0-F was 1.5 g/L, the absorption rate of CO2 reached about 90% of that of the 25% MEA solution. These results demonstrate the simplicity of preparation and the outstanding properties of DvCA8.0-F, which paves a feasible way for CA-assisted CO2 capture. This exciting strategy may also have excellent potential in enzymes for innovative chemical synthesis to achieve the sustainable development of green chemistry.

One-pot synthesis of efficient carbonic anhydrase-zeolitic imidazolate framework-8 composite for enhancing CO2 absorption

Mine Du,Han Chen,Jiexu Ye,Shihan Zhang,Jianmeng Chen,Lidong Wang

Carbonic anhydrase (CA) enzyme-based absorption process is under development for CO 2 capture. How to maintain the activity and stability of CA enzyme is the important issue related to this emerging process. The immobilization technologies are regarding as the promising method to stabilize CA in harsh environments. This work encapsulated CA into zeolitic imidazolate framework-8 (ZIF-8) with different particle sizes according to several preparation protocols and investigated their activity and stability. The experimental results revealed that the preliminary crystallization time, enzyme dosing amount, and particle sizes of ZIF-8 had a significant influence on the catalytic performance of immobilized CA. The remaining activity of the CA after immobilization ranged from 52 to 93 % depending on the employed preparation protocol. In addition, the immobilized CA with particle size of ∼400 nm exhibited better activity compared to those with particle sizes of ∼100 nm and 1000 nm. Moreover, with the increase of the preliminary crystallization time, the stability of immobilized CA descended.

Bimetal based inorganic-carbonic anhydrase hybrid hydrogel membrane for CO2 capture

Huan Wen,Lei Zhang,Yingjie Du,Ziyuan Wang,Yunhong Jiang,Hongjie Bian,Jiandong Cui,Shiru Jia

In this study, we synthesized for the first time a bimetal-based inorganic-carbonic anhydrase (CA) hybrid nanoflower to immobilize CA using Cu 2+ and Zn 2+ instead of single metal ion. Subsequently, the synthesized bimetallic hybrid nanoflowers (CANF) were embedded into the poly(vinyl alcohol) (PVA)-chitosan (CS) hydrogel networks to obtain PVA/CS@CANF hydrogel membrane. The CANF exhibited a significantly higher activity recovery of 70 % compared with 35 % with CA/Zn 3 (PO 4 ) 2 hybrid nanoflowers and 10 % with CA/Cu 3 (PO 4 ) 2 hybrid nanoflowers. The PVA/CS@CANF hydrogel membrane possessed excellent mechanical strength, high catalytic activity, and were easy to flow out without centrifugation or filtration. At the same time, the PVA/CS@CANF displayed higher thermostability, storage stability, and pH stability than free CA and CANF , and superior reusability and CO 2 capture capacity. The hydrogel membrane maintained more than 75 % of its original activity after 8 cycles. However, CANF only maintained 12 % of its original activity. Furthermore, the amount of CaCO 3 produced by PVA/CS@CANF membrane was 9.0-fold and 2.0-fold compared with free CA and CANF, respectively. Therefore, This approach to synthesizing bimetallic-based protein hybrid hydrogel membrane could have a bright future in CO 2 capture.

Carbonic anhydrase encapsulation using bamboo cellulose scaffolds for efficient CO2 capture and conversion

Xiaoqiang Wang,Menghan Li,Zhiyuan Liu,Zhuang Shi,Daoyong Yu,Baosheng Ge,Fang Huang

Utilizing carbonic anhydrase (CA) to catalyze CO 2 hydration offers a sustainable and potent approach for carbon capture and utilization. To enhance CA's reusability and stability for successful industrial applications, enzyme immobilization is essential. In this study, delignified bamboo cellulose served as a renewable porous scaffold for immobilizing CA through oxidation-induced cellulose aldehydation followed by Schiff base linkage. The catalytic performance of the resulting immobilized CA was evaluated using both p -NPA hydrolysis and CO 2 hydration models. Compared to free CA, immobilization onto the bamboo scaffold increased CA's optimal temperature and pH to approximately 45 °C and 9.0, respectively. Post-immobilization, CA activity demonstrated effective retention (>60 %), with larger scaffold sizes (i.e., 8 mm diameter and 5 mm height) positively impacting this aspect, even surpassing the activity of free CA. Furthermore, immobilized CA exhibited sustained reusability and high stability under thermal treatment and pH fluctuation, retaining >80 % activity even after 5 catalytic cycles. When introduced to microalgae culture, the immobilized CA improved biomass production by ∼16 %, accompanied by enhanced synthesis of essential biomolecules in microalgae. Collectively, the facile and green construction of immobilized CA onto bamboo cellulose block demonstrates great potential for the development of various CA-catalyzed CO 2 conversion and utilization technologies.

Carbonic Anhydrase Immobilized by ZnO Nanoparticles for Catalytic CO2 Conversion

Liu Lian,Wang Xin,Gao Zixin,Zhan Yue,Yao Mengqin,Bao Jia

Carbon capture, utilization, and storage (CCUS) has emerged as a critical strategy in combating climate change and global warming. Carbonic anhydrase (CA) exhibits exceptional potential as a biocatalyst for CCUS processes due to its remarkable ability to accelerate CO 2 hydration. In this study, CA was immobilized on ZnO nanocarriers of varying morphologies. The optimal immobilization conditions and enzymatic properties of the immobilized CA were investigated, followed by catalytic CO 2 uptake and mineralization experiments. The immobilization process resulted in several improvements: enhanced overall thermal stability, an elevated optimal reaction pH of 8.50 (compared to 8.00 for free CA), retention of 84.98% original activity after 30 days of storage at 4 °C, and preservation of 85.36% initial activity after five recycling cycles. While the affinity between immobilized CA and its substrate increased, a decrease in activity was observed. The Michaelis constant (Km) for immobilized and free CA were determined to be 17.22 mM and 34.92 mM, respectively, with corresponding catalytic efficiency (Kcat/Km) of 20.49 M −1 -S −1 and 148.91 M −1 -S −1 . The catalytic performance of immobilized CA was investigated by carrying out the CO 2 adsorption assay. Notably, the catalytic efficiency of immobilized CA in CO 2 absorption and mineralization under various conditions was significantly enhanced. Graphical

Comparison of amino and epoxy functionalized SBA-15 used for carbonic anhydrase immobilization

Xiaoyao Fei,Shaoyun Chen,Dai Liu,Chunjie Huang,Yongchun Zhang

Two functionalized SBA-15 [amine-functionalized SBA-15 (AFS) and epoxy-functionalized SBA-15 (GFS)] with different types of functional groups were synthesized by a hydrothermal process and post functionalized with 3-aminopropyltriethoxysilane (APTES) and 3-glycidyloxypropyltrimethoxysilane (GPTMS), respectively. They were used for the immobilization of carbonic anhydrase (CA). The physicochemical properties of the functionalized SBA-15 were characterized by X-ray powder diffraction (XRD), N 2 adsorption–desorption, 13 C, 29 Si solid-state nuclear magnetic resonance (NMR) spectroscopy, and scanning electron microscopy (SEM). Before and after CA was immobilized on AFS and GFS, the effects of temperature and pH value on the enzyme activity , storage stability, and reusability were investigated using para -nitrophenyl acetate ( p -NPA) assay. CA/GFS showed a better performance with respect to storage stability and reusability than CA/AFS. Moreover, the amount of CaCO 3 precipitated over CA/AFS was less than that precipitated over CA/GFS, which was almost equal to that precipitated over the free CA. The results indicate that the epoxy group is a more suitable functional group for covalent bonding with CA than the amino group, and GFS is a promising support for CA immobilization.

Preparation of defective UiO-67 for CA immobilization to improve the ability of CO2 capture and conversion

Rui Zhao,Wenhui Yang,Pengyan Yang,Yunhan Cao,Fenghuan Wang

Carbonic anhydrase (CA) has been suggested as a biocatalyst for carbon dioxide capture since it could effectively catalyze the hydration of carbon dioxide. Metal-organic frameworks (MOFs) have attracted significant attention in the development of immobilized enzyme carriers due to their pore tunability and excellent stability. In this study, UiO-67 materials with varying defect degrees (DUiO-67) were prepared using the organic linker missing method to enhance the catalytic performance. The results indicated that both the defect degree and the pore size of DUiO-67 materials decreased with an increase in acetic acid equivalent. After CA immobilization, all DUiO-67 materials exhibited excellent thermal, pH and recycling stability. CA@DUiO-67–25 showed higher enzyme catalytic activity and conversion ability than the other materials. After 10 cycles, CA@DUiO-67–25 retained 57 % of its initial conversion capacity. The construction of defective MOFs provides a promising new strategy for CA immobilization to achieve the catalytic conversion of CO 2 .

Structure–Activity Relationship of Carbonic Anhydrase Enzyme Immobilized on Various Silica-Based Mesoporous Molecular Sieves for CO2 Absorption into a Potassium Carbonate Solution

Peijing Shao,Han Chen,Qin Ying,Shihan Zhang

In order to fulfill the application of carbonic anhydrase (CA) in the carbon dioxide capture process, immobilization technology will be used to enhance the stability of the enzyme. The structure of the support for the enzyme immobilization is important to determine the enzyme activity and stability. In this work, mesoporous molecular sieves with different dimensions and pore sizes were used as supports for the immobilization of CA. Characterization analysis indicated that the carrier material retained its integrity after enzyme immobilization. The half-life (t1/2) of CA/KIT-6, CA/SBA-15, and CA/MCM-41 increased to 3.0, 2.8, and 2.0 times compared to the free CA enzyme. Additionally, the experimental results showed that when using SBA-15 with a two-dimensional structure and a large pore size as the carrier, the CA enzyme loading was high and retained 78% relative activity and 98% initial activity after 6 days at 40 °C. Comprehensively, CA/SBA-15 was more suitable for capturing CO2 in flue gas than CA/MCM-41 and CA/KIT-6.

Carbonic Anhydrase Enzyme-MOFs Composite with a Superior Catalytic Performance to Promote CO2 Absorption into Tertiary Amine Solution

Shihan Zhang,Mine Du,Peijing Shao,Lidong Wang,Jiexu Ye,Jun Chen,Jianmeng Chen

Carbonic anhydrase (CA) enzyme-based absorption technology for CO2 capture has been intensively investigated. The main issue related to this novel technology is the activity and stability of the CA enzyme under the typical flue gas conditions. To address this issue, CA enzymes were embedded into zeolitic imidazolate framework (ZIF-L) nanoparticles to synthesize a novel CA/ZIF-L-1 composite. The composite exhibited a superior apparent catalytic activity (1.5 times higher) for CO2 absorption compared with their free counterparts, which was due to the synergistic enhancement of CO2 adsorption by support ZIF-L and enzymatic catalysis. The analyses of Fourier transform infrared spectroscopy and circular dichroism revealed that the CA enzyme’s secondary structure was not significantly varied during the CA/ZIF-L-1 preparation, resulting in a high enzyme activity retention. Moreover, the CA/ZIF-L-1 possessed a high thermal stability and reusability due to the structural rigidity and confinement of ZIF-L scaffolds. Compared with the free enzyme, its thermal stability was improved by approximately 100% at 40 °C. After six cycles of reuse, CA/ZIF-L-1 still retained a relative activity of 134%. Therefore, the CA/ZIF-L-1 can be a good candidate to promote the CO2 capture in industrial application.

Core-shell magnetic ZIF-8@Fe3O4-carbonic anhydrase biocatalyst for promoting CO2 absorption into MDEA solution

Qin Ying,Han Chen,Peijing Shao,Xiaowei Zhou,Xiaojia He,Jiexu Ye,Shihan Zhang,Jianmeng Chen,Lidong Wang

Tertiary amine-based absorption process possesses high absorption capacity and low heat duty for CO 2 capture. However, its slow CO 2 absorption rate retards its industrial application. This work developed a core-shell magnetic ZIF-8@Fe 3 O 4 -carbonic anhydrase composite to promote CO 2 absorption into tertiary amine, e.g., diethanolamine (MDEA) due to its easy recovery from the solution. The catalytic performance of CA@ZIF-8@Fe 3 O 4 that had three particle sizes were investigated. Experimental results revealed that the obtained CA@ZIF-8@Fe 3 O 4 can achieve the relative activity as high as 95.2 % compared with the free counterparts. Large particle size of ZIF-8@Fe 3 O 4 was beneficial to activity but adverse to loading. Magnetic analysis shows that the developed CA@ZIF-8@Fe 3 O 4 possessed remarkable magnetic response which made recycling more convenient.

The Kinetics Investigation of CO2 Absorption into TEA and DEEA Amine Solutions Containing Carbonic Anhydrase

Bin Liu,Zhe Cui,Wende Tian

Tertiary amines have been used as alternative absorbents for traditional primary and secondary amines in the process of carbon capture. However, the carbon dioxide (CO2) absorption rates in these kinds of amine are relatively slow, which implies greater investment and construction costs and limits the large-scale application of carbon capture. Carbonic anhydrase (CA) is considered to be an ideal homogeneous catalyst for accelerating the rate of CO2into aqueous amine solution. In this work, CO2absorption combining CA with two single aqueous tertiary amines, namely triethanolamine (TEA) and 2-(diethylamino)ethanol (DEEA), was studied by use of the stopped-flow apparatus over temperature ranging from 293 to 313 K. The concentrations of selected aqueous amine solution and CA used in the experiment were ranging among 0.1–0.5 kmol/m3and 0–50 g/m3, respectively. Compared to the solution without the addition of CA, the pseudo first-order reaction rate in the presence of CA (k0,withCA) is significantly increased. The values ofk0,withCAhave been calculated by a new kinetics model. The results of experimental and calculatedk0,amineandk0,withCAin CO2-amine-H2O solutions were also investigated, respectively. Keywords:carbon capture;CO2absorption;carbonic anhydrase;reaction kinetics
