Magnet-Guided Bionic System with LIFU Responsiveness and Natural Thrombus Tropism for Enhanced Thrombus-Targeting Ability

Ni Fang,Jia Liu,Jingxin Hou,Yixin Zhong,Ying Luo,Liu Hu,Wenli Zhang,Junrui Wang,Jie Xu,Jun Zhou,Yu Zhang,Haitao Ran,Dajing Guo

Background Arterial thrombosis is a serious threat to human health. Recently, many thrombus-targeted nanoparticles (NPs) have been constructed for detecting thrombi or monitoring thrombolysis, but their thrombus-targeting performance is limited. Considering this drawback, we designed a specific bionic system with enhanced thrombus-targeting ability.Materials and Methods In the bionic system, gelatin was chosen as a carrier, and Fe3O4 served as a magnetic navigation medium and a magnetic resonance (MR) imaging agent. The CREKA peptide, which targets fibrin, was conjugated to the surface of gelatin to prepare targeted NPs (TNPs), which were then engulfed by macrophages to construct the bionic system. At the targeted site, the bionic system released its interior TNPs under low-intensity focused ultrasound (LIFU) irradiation. Moreover, the targeting performance was further improved by the conjugated CREKA peptide. Results In this study, we successfully constructed a bionic system and demonstrated its targeting ability in vitro and in vivo. The results indicated that most TNPs were released from macrophages under LIFU irradiation at 2 W/cm2 for 10 min in vitro. Additionally, the enhanced thrombus-targeting ability, based on the natural tropism of macrophages toward inflammatory thrombi, magnetic navigation and the CREKA peptide, was verified ex vivo and in vivo. Moreover, compared with the bionic system group, the group treated with TNPs had significantly decreased liver and spleen signals in MR images and significantly enhanced liver and spleen signals in fluorescence images, indicating that the bionic system is less likely to be cleared by the reticuloendothelial system (RES) than TNPs, which may promote the accumulation of the bionic system at the site of the thrombus. Conclusion These results suggest that the magnet-guided bionic system with LIFU responsiveness is an excellent candidate for targeting thrombi and holds promise as an innovative drug delivery system for thrombolytic therapy.

Fabrication of Ag-armed iron oxide magnetic Janus nanoparticles for enhanced elimination of robust biofilm under magnetic field

Wang Xi,Wu Juan,Sun Haibin,Lu Bingyan,Huang Jingjing,Liu Qiaoling,Li Tingting

Biofilms pose a significant challenge to public health, fostering persistent infections that are notoriously difficult to eradicate. Their resilience stems partly from the inadequate penetration of conventional antibacterial agents into these structured microbial communities. To address this issue, we have developed a novel magnetite iron oxide Janus nanoparticles (JNPs), harnessing the magnetic field assistance for targeted biofilm elimination. In which, natural tannic acid (TA) and chitosan (CS) were coated on IONPs for improved antibacterial property and biocompatibility. Subsequently, the surfaces were functionalized with epoxy chloropropane and butanediamine, creating anchor points for the deposition of silver nanoparticles (AgNPs). Subsequently, AgNPs were embedded via an in situ reduction process, utilizing polyvinyl pyrrolidone (PVP) to precisely control the morphology of the Ag-armed magnetite JNPs. Remarkably, IONPs@BCS/Ag exhibited dual-responsive release characteristics, sensitively adjusting their Ag + release in response to both oxygen levels and pH variations within the biofilms microenvironment. This system leveraged both stimulus-responsiveness and magnetic guidance to deliver exceptional antimicrobial performance against both planktonic bacteria and established biofilms. Critically, the superparamagnetic nature of IONPs@BCS/Ag enabled enhanced penetration into biofilms under an external magnetic field, facilitating the efficient delivery of AgNPs. Furthermore, the sustained release of Ag + triggered by low pH and oxygen levels within the biofilm triggered the generation of reactive oxygen species (ROS), which not only accelerated the decomposition of the biofilm matrix but also disrupted the integrity of microbial communities. In summary, the IONPs@BCS/Ag magnetite JNPs, augmented by an external magnetic field, demonstrated enhanced anti-biofilm capabilities, offering immense potential as a potent weapon against biofilm-associated infections. Magnetic guidance shape memory PLA/TBC/Fe3O4 microspheres for dentin tubule sealing

Tao Guo,Jiayuan Chen,Lan Luo,Qiangwang Geng,Linlin Wang,Fenghua Zhang,Narisu Hu,Yanju Liu,Jinsong Leng

Shape memory polymer (SMP) microspheres possess controllable physicochemical properties, internal cavities capable of loading drugs or nanoparticles, and inherent intelligence, making them suitable for various biomedical applications. In this research, we utilized tri-n-butyl citrate (TBC) to lower the glass transition temperature of shape memory polylactic acid (PLA) to approximate oral temperature. Following this, we introduced oleic acid-modified Fe 3 O 4 nanoparticles to confer magnetism to the material. Subsequently, SMP microspheres with a core–shell structure encapsulating Fe 3 O 4 particles were prepared using the emulsion-solvent evaporation method, and we carefully controlled their particle size and morphology to match the diameter of dentinal tubules. Our investigation focused on applying these magnetically guided microspheres, which exhibited excellent shape memory performance, for sealing dentinal tubules. These microspheres displayed the ability to move directionally under the influence of a magnetic field. As a result, they could be attracted into the interior of dentinal tubules in their temporary shape and then revert to their initial shape upon heating at 40 °C, thereby achieving the closure of dentinal tubules effectively. The microsphere entry rate during the tubule closure process was found to be 93.33 ± 2.82 %, with a corresponding sealing rate of 85.41 ± 7.11 %. The successful demonstration of magnetic-guided movement and the utilization of the shape memory function for dentinal tubule sealing significantly expand the potential applications of SMP microspheres in the field of biomedical science.

Directed Neural Stem Cells Differentiation via Signal Communication with Ni–Zn Micromotors

Ye Feng,Chao Gao,Dazhi Xie,Lu Liu,Bin Chen,Suyi Liu,Haihong Yang,Zhan Gao,Daniela A. Wilson,Yingfeng Tu,Fei Peng

Neural stem cells (NSCs), with the capability of self-renewal, differentiation, and environment modulation, are considered promising for stroke, brain injury therapy, and neuron regeneration. Activation of endogenous NSCs, is attracting increasing research enthusiasm, which avoids immune rejection and ethical issues of exogenous cell transplantation. Yet, how to induce directed growth and differentiation in situ remain a major challenge. In this study, a pure water-driven Ni–Zn micromotor via a self-established electric–chemical field is proposed. The micromotors can be magnetically guided and precisely approach target NSCs. Through the electric–chemical field, bioelectrical signal exchange and communication with endogenous NSCs are allowed, thus allowing for regulated proliferation and directed neuron differentiation in vivo. Therefore, the Ni–Zn micromotor provides a platform for controlling cell fate via a self-established electrochemical field and targeted activation of endogenous NSCs.

Composite Multifunctional Micromotors from Droplet Microfluidics

Minhan Zou,Jie Wang,Yunru Yu,Lingyu Sun,Huan Wang,Hua Xu,Yuanjin Zhao

Inspired by natural biological machines, lots of effort has been invested in developing artificially functional micromotors which can convert energy into movement for carrying out tasks in diverse areas. Here, we present a capillary microfluidic system with dual inner injections for one-step generation of composite structured polymer micromotors with two distinct cores of platinum (Pt) nanoparticle-integrated and iron oxide (Fe3O4) nanoparticle-dispersed hydrogels. Because the flow rates of the prepolymerized fluids can be precisely tuned in the microfluidics, the diameters of the micromotors as well as the sizes and numbers of the inner cores can be well tailored to optimize the parameters of the resultant micromotors. When exposed to a hydrogen peroxide (H2O2) medium, the Pt-integrated cores of the micromotors could provide propulsion by expelling bubbles produced from the catalytic decomposition of H2O2, while the Fe3O4-dispersed cores could impart magnetic guidance for the micromotors. Benefiting from the close cooperation of these two types of cores, the micromotors were imparted with a strong propulsion and prominent recyclability for the delivery of both microscale and macroscale objects. These results manifest that this kind of composite micromotor has great diversity in various applications.
