Biomimetic inorganic nanoenzyme is a kind of nanomaterial with long-term stability, easy preparation and low cost, which could instead of natural biological enzyme. Metal-organic framework (MOFs) as effectively nanoenzyme was attracted more attention for the adjustability and large specific surface area. This design is based on the catalase-like catalytic activity of 2D metal–organic frameworks (MOFs) and the high sensitivity of surface enhanced Raman spectroscopy (SERS) biosensors to construct a novel SERS biosensor capable of efficiently detecting mercury (Hg 2+ ). In this study, 2D MOFs nanozyme was instead of 3D structure with more effecient catalytic site, which can catalyze o-Phenylenediamine (OPD) to OPDox with the assistance of H 2 O 2 . Besides, a magnetic composite nanomaterial Fe 3 O 4 @Ag@OPD was prepared as a signal carrier. In the presence of Hg 2+ , T-Hg 2+ -T base pairs were used to connect the two materials to realize Raman signal change. Based on this principle, the SERS sensor can realize the sensitive detection of Hg 2+ , the detection range is 1.0 × 10 -12 ∼ 1.0 × 10 -2 mol‧L -1 , and the detection limit is 1.36 × 10 -13 mol‧L -1 . This method greatly improves the reliability of SERS sensor for detecting the target, and provides a new idea for detecting metal ions in the environment.

Single-cell metabolomics could be used to discover the chemical strategies of cells for coping with chemical or environmental stress because metabolomics provides a more immediate and dynamic picture of cell functionality. However, these small-molecule metabolites are the most difficult to measure because they characterize rapid metabolic dynamics, structural diversity, and incapacity to achieve signal amplification or labeling. In order to solve above problems, we presented a surface-enhanced Raman scattering (SERS)-microfluidic droplet platform to realize the label-free simultaneous analysis of multiplexed metabolites at the single-cell level via a versatile magnetic SERS substrate composed by silver nanoparticles (AgNPs, 30 nm)-decorated 400 nm Fe3O4 magnetic microspheres. This metal-magnetic composite substrate is beneficial to efficient adsorption of single cell metabolites, fast separation from complex matrixes, and high SERS sensitivity. Also, the endocytosis effect can be fully prohibited due to its relatively large size. This work achieves label-free, nondestructive, simultaneous determination of three single-cell metabolites, pyruvate, adenosine triphosphate (ATP), and lactate, owing to their intrinsic SERS fingerprints. The “hotspots” effect induced by the magnetic aggregation of Fe3O4@AgNPs allows highly sensitive SERS detection. Encapsulating metabolites in such a limited and isolated droplet accelerates the process of diffusion and adsorption equilibriums. Activation with saponin for metabolites was assessed on different cell lines. The SERS-microdroplet platform is a powerful tool for exploring single-cell heterogeneity at the metabolic level.

To date, great achievements with GC-MS, HPLC-MS, and fluorescence biosensing techniques have been made to detect illegal additives of salbutamol (SAL) in swine meat. However, these methods are not suitable for rapid on-site screening due to either costly instruments or rather complicated and/or time consuming sample pretreatments. Herein, a simple, rapid and ultrasensitive approach based on an azo-coupling reaction and surface-enhanced resonance Raman scattering (SERRS) is presented. By combining with a magnetic SERS substrate, an indirect detection for SAL, with a LOD of 1.0 × 10−11 M (2.39 pg mL−1), was realized. Moreover, a colorimetric method for naked eye detection was successfully carried out for rapid screening of SAL in concentrations higher than 2.09 × 10−5 M (5 μg mL−1). In addition, the proposed method was successfully applied for the rapid determination of SAL in real swine meat. The entire process, including pretreatment, coupling reaction and SERRS detection, was performed within 7 min. Moreover, the SERRS fingerprint band being specific to corresponding functional group guarantees the selectivity for the target molecule. Therefore, the proposed strategy in the present study offers a new way to identify trace amounts of analytes, such as SAL as well as other illegal additives in health-related products and food.
