Electromagnetic Field-Programmed Magnetic Vortex Nanodelivery System for Efficacious Cancer Therapy

Xiaoli Liu,Yifan Zhang,Yu Guo,Wangbo Jiao,Xiao Gao,Wee Siang Vincent Lee,Yanyun Wang,Xia Deng,Yuan He,Ju Jiao,Ce Zhang,Guoqing Hu,Xing-Jie Liang,Haiming Fan

Effective delivery of anticancer drugs into the nucleus for pharmacological action is impeded by a series of intratumoral transport barriers. Despite the significant potential of magnetic nanovehicles in electromagnetic field (EF)-activated drug delivery, modularizing a tandem magnetoresponsive activity in a one-nanoparticle system to meet different requirements at both tissue and cellular levels remain highly challenging. Herein, a strategy is described by employing sequential EF frequencies in inducing a succession of magnetoresponses in the magnetic nanovehicles that aims to realize cascaded tissue penetration and nuclear accumulation. This nanovehicle features ferrimagnetic vortex-domain iron oxide nanorings coated with a thermo-responsive polyethylenimine copolymer (PI/FVIOs). It is shown that the programmed cascading of low frequency (L f )-EF-induced magnetophoresis and medium frequency (M f )-EF-stimulated magneto-thermia can steer the Doxorubicin (DOX)-PI/FVIOs to the deep tissue and subsequently trigger intracellular burst release of DOX for successful nuclear entry. By programming the order of different EF frequencies, it is demonstrated that first-stage L f -EF and subsequent M f -EF operation enables DOX-PI/FVIOs to effectively deliver 86.2% drug into the nucleus in vivo . This nanodelivery system empowers potent antitumoral activity in various models of intractable tumors, including DOX-resistant MCF-7 breast cancer cells, triple-negative MDA-MB-231 breast cancer cells, and BxPC-3 pancreatic cancer cells with poor permeability.

Magnetic vortex nanoring coated with gadolinium oxide for highly enhanced T1-T2 dual-modality magnetic resonance imaging-guided magnetic hyperthermia cancer ablation

Jianfeng Bao,Shuangshuang Guo,Xiangyang Zu,Yuchuan Zhuang,Dandan Fan,Yong Zhang,Yupeng Shi,Xin Pang,Zhenyu Ji,Jingliang Cheng

Nowadays, about 30% of magnetic resonance imaging (MRI) exams need contrast agents (CAs) to improve the sensitivity and quality of the images for accurate diagnosis. Here, a multifunctional nano-agent with ring-like vortex-domain iron oxide as core and gadolinium oxide as shell (vortex nanoring Fe 3 O 4 @Gd 2 O 3 , abbreviated as VNFG) was firstly designed and prepared for highly enhanced T 1 -T 2 dual-modality magnetic resonance imaging (MRI)-guided magnetic thermal cancer therapy. After thorough characterization, the core-shell structure of VNFG was confirmed. Moreover, the excellent heat generation property (SAR=984.26 W/g) of the proposed VNFG under alternating magnetic fields was firmly demonstrated. Furthermore, both in vitro and in vivo studies have revealed a good preliminary indication of VNFG’s biological compatibility, dual-modality enhancing feature and antitumor efficacy. This work demonstrates that the proposed VNFG can be a high-performance tumor diagnosis and theranostic treatment agent and may have great potential for clinical application in the future.

Copper doped magnetic vortex nanoring based nanotherapeutics for bacterial infection tri-therapy: interplay of magnetic hyperthermia, chemodynamic therapy and photothermal therapy

Jing Wang,Wenqian Zhao,Hui Tu,Xiangyang Zu,Jinghua Li,Kun Lei,Jing Li,Yuchuan Zhuang,Yanbo Dong,Andrey Tulupov,Fengshou Zhang,Jianfeng Bao

Infectious bacteria pose an increasing threat to public health, and hospital-acquired bacterial infections remain a significant challenge for wound healing. In this study, we developed an advanced nanoplatform utilizing copper doped magnetic vortex nanoring coated with polydopamine (Cu–MVNp) based nanotherapeutics for bacterial infection tri-therapy. This multifunctional nanoplatform exhibits remarkable dual-stimulus thermogenic capabilities and Fenton-like peroxidase activity. Exposure to an alternating magnetic field (AMF) and near-infrared (NIR) light allows the nanoring to elevate environmental temperatures through hysteresis losses and the non-radiative decay effects of the PDA coating. At a concentration of 150 μg mL−1, Cu–MVNp increases the temperature by 18.2 °C under an AMF, achieving a specific absorption rate (SAR) of 640.9 W g−1. On the other hand, under 808 nm NIR irradiation, the temperature rises by 42.6 °C, with a photothermal conversion efficiency of 46.45%. Furthermore, by incorporating copper ions (Cu), which can damage cell membranes themselves, Cu–MVNp was endowed with Fenton-like functions and can catalyze the formation of hydroxyl radicals (˙OH) from low concentrations (1 mM) of hydrogen peroxide (H2O2), thus enhancing the effectiveness of chemodynamic therapy (CDT). Cu–MVNp exhibits significant antibacterial efficacy, achieving notable kill rates against E. coli and S. aureus, with enhanced effects under NIR and nearly complete eradication with an AMF. In vivo tests using a mouse wound model confirm its potent bactericidal properties and good biocompatibility. The Cu–MVNp nanoring shows promise as an antibacterial treatment, potentially effective at inhibiting bacterial growth.

Ferrimagnetic Vortex Nanorings Facilitate Efficient and Safe Deep-Brain Magnetothermal Stimulation in Freely Moving Mice

Galong Li,Xin Qiao,Yu Zhao,Dongyan Li,Guigen Zhang,Xiaoli Liu,Fulin Chen,Huaning Wang,Hongbing Lu,Jin Zhou,Changyong Wang,Haiming Fan

Magnetothermal neuromodulation is a minimally invasive, deep-brain accessible, and tether-free technique. The precisely timed activation of thermosensitive ion channels, such as TRPV1, with local heat generated using magnetic nanoparticles is crucial for efficient neuromodulation. Nevertheless, this technique is greatly hindered by its long stimulus-response time and high safety risks due to the poor heat-generating performance of the nanomediators. Herein, we report the establishment of a ferrimagnetic vortex iron oxide nanoring (FVIO)-mediated magnetothermal neurostimulation technique that is efficient and safe. Compared with widely used superparamagnetic iron oxide nanomediators (SPIOs), the FVIOs triggered Ca 2+ influx into HEK293T cells and cortical neurons at an Fe concentration of 51 µg mL −1 , which is 20.27-fold lower than that needed for SPIOs. In vivo magnetothermal stimulation in the central nucleus of the amygdala of mice further demonstrated that FVIOs with the optimal dose of 0.05 µg evoked fear behaviors with an average latency of 2.51 s, which was 2.3-fold faster than that in the SPIO (0.80 µg)-treated group. More importantly, FVIOs-mediated stimulation not only exhibited negligible histopathological alterations and proinflammatory cytokine expression but also successfully elicited fear behaviors in transgene-free mice. The FVIO-mediated efficient and safe neuromodulation has the potential for future neuroscience exploitation and neurological disease treatment.

Magnetic-vortex nanodonuts enhance ferroptosis effect of tumor ablation through an imaging-guided hyperthermia/radiosensitization strategy

S.S. Guo,M.M. Chen,Y.H. Yang,Y.Y. Zhang,X. Pang,Y.P. Shi,Y.C. Zhuang,D.D. Fan,J.F. Bao,Z.Y. Ji

Pursuing more efficient multifunctional treatment is the main challenge of preclinical nanoparticle-mediated theranostics research. Here, nanoscale magnetite vortex donut shape was synthesized as a platform, and then ultrasmall gold nanoparticles were successfully embedded into the nanoring surface, thereby obtaining gold-modified magnetic vortex donut (GMVD). GMVD has a high photothermal conversion efficiency (η = 42.2%), which makes it have excellent photothermal ablation effect on tumors both in vivo and in vitro . Simultaneously, GMVD forms reactive oxygen species (ROS) under the 808 nm laser triggering, inducing ferroptosis. The addition of gold element also makes GMVD have the effect of radiotherapy (RT) sensitization. In conclusion, the synergistic treatment of RT and PTT greatly enhanced tumor ablation, indicating that GMVD has good biocompatibility and antitumor efficacy. This work demonstrates that the proposed GMVD can be a high-performance tumor diagnosis and theranostic treatment agent and may have great potential for clinical application in the future.

Interfacial engineered iron oxide nanoring for T2-weighted magnetic resonance imaging-guided magnetothermal-chemotherapy

Bao Jianfeng,Tu Hui,Li Jing,Dong Yanbo,Dang Le,Yurievna Korjova Elena,Zhang Fengshou,Xu Lei

Due to no penetration depth limitation, low cost, and easy control, magnetic nanoparticles mediated magnetic hyperthermia therapy (MHT) has shown great potential in experimental and clinal treatments of various diseases. However, the low heating conversion efficiencies and short circulation times are major drawback for most existing magnetic-thermal materials. Additionally, single MHT treatment always leads to resistance and recurrence. Herein, a highly efficient magnetic-thermal conversion, ferrimagnetic vortex nanoring Fe3O4 coated with hyaluronic acid (HA) nanoparticles (Fe3O4@HA, FVNH NPs) was firstly constructed. Additionally, the doxorubicin (DOX) was successfully enclosed inside the FVNH and released remotely for synergetic magnetic–thermal/chemo cancer therapy. Due to the ferrimagnetic vortex-domain state, the ring shape Fe3O4 displays a high specific absorption rate (SAR) under an external alternating magnetic field (AMF). Additionally, antitumor drug (DOX) can be encapsulated inside the single large hole of FVNH by the hyaluronic acid (HA) shell and quickly released in response the tumor acidic microenvironments and AMF. What’s more, the non-loaded FVNH NPs show good biocompatibility but high cytotoxicity after loading DOX under AMF. Furthermore, the synthesized FVNH can efficiently reduce the transverse relaxation time and enhance negative magnetic resonance imaging (MRI). The impressive in vivo systemic therapeutic efficacy of FVNH was also proved in this work. Taken together, the results of this study demonstrate that the synthesized FVNH NPs offer the promise of serving as multifunctional theranostic nanoplatforms for medical imaging-guided tumor therapies.
