Hybrid plasmonic-magnetic nanoparticles (NPs) can mediate tumors through the magneto-photothermal therapy, gaining rising attention in the treatment of oncology. Herein, carboxylated carbon coated plasmonic satellite/magnetic core NPs were successfully fabricated by a pulsed laser ablation in solution. Plasmonic element (Au, Pd, Ag and Cu), shell thickness (i.e., 1-2 layers over Fe2O3 core and 1-2 layers over Au satellite) and plasmonic/magnetic composition were systematically tailored to achieve optimal plasmonic/magnetic properties. These designed NPs were applied as heat generators as functions of NP concentrations, laser power densities and magnetic field intensities. Carboxylated shells facilitated good colloidal stability and cellular uptake of NPs. Carbon coated plasmonic satellite/magnetic core NPs exhibited excellent capability (66.1-193.6 mM−1·s−1) of magnetic resonance imaging for tumor visually in vivo. Cancer cells were killed in 300 s in vitro by enhanced the temperature of tumor rapidly to 55°C within 100 s with combined exposure dual modalities including laser radiation of 0.75 W/cm2 (785 nm) and magnetic field of 400 Oe. Carbon coated plasmonic satellite/magnetic core NPs present good biocompatibility in multiple organs. This work provided a versatile strategy for fabricating core/satellite theranostic nanoprobes with rationally multimodal imaging and therapeutic functionalities.

Tumor recurrence and bone defects are two key challenges in the surgical treatment of osteosarcoma (OS). Therefore, it is highly necessary to develop a multifunctional scaffold that can simultaneously eradicate tumor cells and promote bone regeneration. Herein, a hierarchically porous shape memory scaffold consisting of hydroxyapatite, silica, poly(D,L-lactide-co-trimethylene carbonate) and Fe3O4 (HSP-Fe3O4) is constructed by Pickering emulsion and 4D printing technique. The HSP-Fe3O4 scaffold demonstrates the advantages of multimodal anti-tumor therapy, including chemotherapy through the Fenton reaction, effective photothermal conversion for photothermal therapy under near-infrared laser irradiation, and magnetothermal therapy provided by an alternating magnetic field. Furthermore, photothermal hyperthermia also serve as triggers for the shape memory effect of the HSP-Fe3O4 scaffold, enabling the scaffold to precise adaptation of complex bone defects after minimally invasive surgical implantation. Additionally, the HSP-Fe3O4 scaffold with interconnected multiscale pore exhibits good biocompatibility and excellent bone repair capabilities. This study proved that the HSP-Fe3O4 scaffold provides positive insights for preventing tumor recurrence and facilitating bone regeneration after OS surgery.

Bone implant-associated bacterial infection is a common cause of transplant failure. Chemodynamic therapy (CDT) has significant antibacterial efficacy by producing reactive oxygen species (ROS). However, the ROS permeability is restricted by the natural barrier of bacterial biofilms. Herein, a hard-soft magnetic biphasic nanoparticle (CF@MF) was synthesized by in-situ growth of the soft MnFe 2 O 4 shell on the hard CoFe 2 O 4 core. Then, the CF@MF was incorporated into poly- l -lactide powders to prepare porous scaffolds by selective laser sintering. The scaffold exhibited superior magnetothermal conversion efficiency, which was attributed to the interfacial exchange coupling of CF@MF. Importantly, the scaffold effectively dispersed biofilms by magnetothermal therapy (MTT), favoring numerous ROS pre-generated by CDT to enter the interior to kill bacteria. This study provided a synergistic MTT/CDT strategy to eradicate deep bacterial infections in biomedical application.

CoFe2O4@MnFe2O4/polypyrrole nanocomposites, in which CoFe2O4@MnFe2O4 core/shell nanoparticles (NPs) were well-distributed in/on polypyrrole (PPy) nanoparticles, have been prepared by the polymerization of pyrrole by Fe3+ in the presence of polyvinyl alcohol (PVA) and CoFe2O4@MnFe2O4 NPs. In CoFe2O4@MnFe2O4/polypyrrole nanocomposites, the PPy with strong near-infrared absorption can convert 808 nm laser irradiation to heat with a photothermal conversion efficiency of ∼38%, and the CoFe2O4@MnFe2O4 NPs can convert the electromagnetic energy to heat simultaneously in an alternating magnetic field (200 kHz, 60.6 kA m−1) with a specific loss power of 930 W g−1. In vitro experiments showed that the cell viability of 4T1 cells incubated with 200 μg mL−1 of nanocomposites was ∼58% when irradiated by an 808 nm laser (0.2 W cm−2) for 10 min, and ∼68% in an alternating magnetic field (200 kHz, 60.6 kA m−1) for 0.5 h. A combination of 808 nm laser irradiation and alternating magnetic field can ablate nearly all the cells, which suggests the synergistic effect of photothermal and magnetothermal combined therapy.
