Minimalistic Implant for Percutaneous Magnetic Hyperthermia-Based Combination Therapy of Hepatocellular Carcinoma

Guangchao Xie,Bingjie Li,Shuyue Guo,Wenjing Hou,Yao Wang,Jiamei Liu,Jialin Zhu,Luchen Chang,Xiaoqing Wang,Xiaoyi Wang,Jinbin Pan,Xi Wei

Percutaneous local thermal therapy, containing radio frequency and microwave ablation, is widely utilized in the clinical management of hepatocellular carcinoma (HCC) due to its minimal invasiveness and favorable therapeutic outcomes. However, its further development is limited by a prolonged ablation duration and the risk of excessive heating. Magnetic hyperthermia therapy (MHT) provides a new perspective for percutaneous tumor thermal ablation due to its superior tissue penetration capability and safety. Herein, an iron foam–agarose gel–drug (IF-Aga-drug) implant is prepared using a minimalistic method for percutaneous combination therapy of HCC. The excellent conductivity of IF endows it with strong heating capability owing to eddy current loss in an alternating magnetic field (AMF), while the abundant pores provide ample space for drug loading. Agarose gel imparts the IF platform with universal and efficient drug-loading capacity and controlled drug-release capability that is responsive to magnetic hyperthermia. Doxorubicin (DOX) is utilized as a representative drug to construct the IF-Aga-DOX implant, which is successfully employed in ultrasound-guided, magnetic hyperthermia-based combination chemotherapy for orthotopic HCC in rabbits under ultralow-power magnetic field intensities (Happl·fappl = 2.25 × 108 A m–1 s–1). The developed IF-Aga-drug implant platform offers a convenient and versatile strategy for percutaneous tumor therapy.

Multifunctional Fe2O3@PPy-PEG nanocomposite for combination cancer therapy with MR imaging

Jun Zhou,Jinghua Li,Xingwei Ding,Junjie Liu,Zhong Luo,Yun Liu,Qichun Ran,Kaiyong Cai

In recent years, magnetic hyperthermia nanoparticles have drawn great attention for cancer therapy because they have no limitation of tissue penetration during the therapy process. In this study, cubic nanoporous Fe2O3 nanoparticles derived from cubic Prussian blue nanoparticles were used as magnetic cores to generate heat by alternating the current magnetic field (AMF) for killing cancer cells. In addition, polypyrrole (PPy) was coated on the surfaces of the cubic Fe2O3 nanoparticles to load doxorubicin hydrochloride (DOX). The PEG component was then physically adsorbed onto the surfaces of the nanoparticles, resulting in a Fe2O3@PPy-DOX-PEG nanocomposite. The nanocomposite was triggered by acid stimulus and AMF to release DOX, resulting in a remarkable combination therapeutic effect via chemotherapy and magnetic hyperthermia. Furthermore, the nanocomposite could realize magnetic resonance imaging (MRI) due to the magnetic core structure. The study provides an alternative for the development of new nanocomposites for combination cancer therapy with MR imaging in vivo.

Polyetherimide- and folic acid-modified Fe3O4 nanospheres for enhanced magnetic hyperthermia performance

Qinganzi Wang,Yuemei Cheng,Wenhua Wang,Xiaolin Tang,Yongxiu Yang

Recent studies have highlighted the development prospects of magnetic hyperthermia in cancer therapy. A few studies on the application of Fe 3 O 4 nanospheres for the magnetic hyperthermia of gynecological malignancies have achieved certain efficacy, but there was no visible progress currently. In this work, Fe 3 O 4 nanospheres modified with polyetherimide (PEI) and folic acid (FA) were synthesized using a hydrothermal method for possible utility in biocompatible and active tumor-targeting magnetic induction hyperthermia. The PEI- and FA-coated Fe 3 O 4 nanospheres showed high crystallinity, well-dispersed spherical structures and ideal M s value. As a result, the designed Fe 3 O 4 @ PEI@FA nanospheres achieved higher specific absorption rate (SAR) values at 360 kHz and 308 Oe, as well as excellent biocompatibility in Hela, SKOV3, HEC-1-A and NIH3T3 cells. These nanospheres can be used as an optimal heating agent for the magnetic hyperthermia treatment of gynecological cancers.

Facile preparation of novel type polymethyl methacrylate/CoFe2O4/BNNS composite cements and their caloric performance in alternating magnetic fields

Jingxiang Wang,Min Li,Xunwei Liu,Jiangmin Yu,Dicheng Yang,Chunping Yuan,Xiaoying Yin,Yinan Yan

Magnetic hyperthermia generated by functionalized bone cement is a promising approach to treat postosteocarcinoma cancer regeneration. However, it is crucial to minimize the magnetic field intensity using localized high-caloric-generating nanoparticles to avoid the potential hazards of electromagnetic field exposure to living organisms. Herein, in situ hydrothermally synthesized CoFe 2 O 4 (CoFe) nanoparticles and boron nitride nanosheet (BNNS)–supported CoFe complexes (BNCoFe) are incorporated into polymethyl methacrylate (PMMA) cement to prepare a composite implant with high-intensity magnetic–thermal performance and safety. The results show that the CoFe nanoparticles (size = ~70 nm) anchored on BNNS surfaces exhibit a maximum magnetic–thermal ablation temperature of ~42°C within 50 s under an alternating magnetic field of 400 KHz (field frequency) and 30 Oe (field intensity) in vitro. Furthermore, the incorporation of BNNSs into the PMMA matrix notably increases the thermal conductivity of PMMA from 0.07 to 0.25 W mK −1 and improves its mechanical properties (compressive strength increases from 74.0 ± 1.6 to 81.0 ± 1.0 MPa), which is attributed to the crystalline structure of BNNSs in the PMMA matrix. Inductively coupled plasma analysis shows a considerably reduced Co 2+ ion release from PMMA/BNCoFe, indicating its potential as a safe implantation material for hyperthermia treatment.

Effective elimination of liver cancer stem-like cells by CD90 antibody targeted thermosensitive magnetoliposomes

Rui Yang,Li Y. An,Qin F. Miao,Feng M. Li,Yong Han,Hui X. Wang,Dang P. Liu,Rong Chen,Sha Q. Tang

Aim To investigate the use of thermosensitive magnetoliposomes (TMs) loaded with magnetic iron oxide (Fe 3 O 4 ) and the anti-cancer stem cell marker CD90 (CD90@TMs) to target and kill CD90 + liver cancer stem cells (LCSCs). Methods The hepatocellular carcinoma cell line Huh7 was used to separate CD90 + LCSCs by magnetic-activated cell sorting. CD90@TMs was characterized and their ability to target CD90 + LCSCs was determined. Experiments were used to investigate whether CD90@TMs combined with magnetic hyperthermia could effectively eliminate CD90 + LCSCs. Results The present study demonstrated that CD90 + LCSCs with stem cells properties were successfully isolated. We also successfully prepared CD90@TMs that was almost spherical and uniform with an average diameter of 130±4.6 nm and determined that magnetic iron oxide could be incorporated and retained a superparamagnetic response. CD90@TMs showed good targeting and increased inhibition of CD90 + LCSCs in vitro and in vivo compared to TMs. Conclusion CD90@TMs can be used for controlled and targeted delivery of anticancer drugs, which may offer a promising alternative for HCC therapy.

Nonmetallic magnetic hyperthermia and chemo-immunotherapy of tumors

Guangchao Xie,Bingjie Li,Shuyue Guo,Wenjing Hou,Molin Li,Yao Wang,Jun Liu,Jihui Hao,Xi Wei

Combined therapy based on magnetic hyperthermia holds significant promise for tumor treatment. Nevertheless, the existing magnetic hyperthermia platforms are often constrained by suboptimal heating efficiency and concerns over the potential metal ion toxicity. Herein, a minimalistic and nonmetallic graphite-agarose gel-drug (Gra-Aga-Drug) implant is prepared by mold-assisted casting method for magnetic hyperthermia-based combination therapy of tumors. This implant can achieve magnetic hyperthermia through the strong eddy current thermal effect of Gra, while the agarose gel provides the implant with universal and efficient drug-loading capacity and magnetic hyperthermia-responsive release ability. The chemotherapeutic drug doxorubicin (DOX) and the immune adjuvant imiquimod (R837) are chosen to prepare Gra-Aga-DOX-R837 implant for combination therapy of tumors in vivo . The magnetic hyperthermia and DOX induce immunogenic cell death (ICD) of tumors, combined with the immune adjuvant R837, eliciting a robust anti-tumor immune response to efficiently ablate primary tumors and suppress distant tumors growth. Importantly, the implant platform effectively addresses the challenges of limited heating efficiency and metal ion toxicity associated with traditional magnetic hyperthermia materials. This study marks the pioneering construction of nonmetallic combination therapy platform with excellent magnetothermal performance and favorable biocompatibility.

Three-Dimensional Printable Magnetic Hydrogels with Adjustable Stiffness and Adhesion for Magnetic Actuation and Magnetic Hyperthermia Applications

Xueting Xuan,Yi Li,Xing Xu,Zhouyi Pan,Yu Li,Yonghao Luo,Li Sun

Stimuli-responsive hydrogels hold immense promise for biomedical applications, but conventional gelation processes often struggle to achieve the precision and complexity required for advanced functionalities such as soft robotics, targeted drug delivery, and tissue engineering. This study introduces a class of 3D-printable magnetic hydrogels with tunable stiffness, adhesion, and magnetic responsiveness, prepared through a simple and efficient “one-pot” method. This approach enables precise control over the hydrogel’s mechanical properties, with an elastic modulus ranging from 43 kPa to 277 kPa, tensile strength from 93 kPa to 421 kPa, and toughness from 243 kJ/m3to 1400 kJ/m3, achieved by modulating the concentrations of acrylamide (AM) and Fe3O4nanoparticles. These hydrogels exhibit rapid heating under an alternating magnetic field, reaching 44.4 °C within 600 s at 15 wt%, demonstrating the potential for use in mild magnetic hyperthermia. Furthermore, the integration of Fe3O4nanoparticles and nanoclay into the AM precursor optimizes the rheological properties and ensures high printability, enabling the fabrication of complex, high-fidelity structures through extrusion-based 3D printing. Compared to existing magnetic hydrogels, our 3D-printable platform uniquely combines adjustable mechanical properties, strong adhesion, and multifunctionality, offering enhanced capabilities for use in magnetic actuation and hyperthermia in biomedical applications. This advancement marks a significant step toward the scalable production of next-generation intelligent hydrogels for precision medicine and bioengineering.

Micelle-like Nanoparticles for Drug Delivery and Magnetically Enhanced Tumor Chemotherapy

Liqin Xie,Xirui Zuo,Beilei Wang,Dan Li,Wenke Chang,Shenglu Ji,Dan Ding

Using the coordination bonds between transition metal atoms and electron-rich functional groups, we synthesized two kinds of micelle-like nanoparticles. Using magnetic Fe3O4 as the core, poly(methyl methacrylate) (PMMA) and poly(acrylic acid) (PAA) brushes were grafted via activators regenerated by electron transfer for atom transfer radical polymerization (ARGET-ATRP), which formed micelle-like magnetic nanoparticles Fe3O4/PAA–PMMA with a hydrophobic outer layer and Fe3O4/PMMA–PAA with a hydrophilic outer layer. Both the micelle-like nanoparticles had amphiphilic properties and can be used to load hydrophilic or hydrophobic drugs. Even loaded with hydrophobic drugs, the micelle-like nanoparticles can still be dispersed in aqueous solution, and Fe3O4/PAA–PMMA had a higher loading content. As the drug carrier, these two micelle-like nanoparticles can be used for magnetically targeted drug delivery and magnetic resonance imaging due to superparamagnetic Fe3O4. In addition, due to the magnetic retention effect, the drug-loaded micelle-like nanoparticles remained at the tumor site, increasing the local drug concentration. At the same time, the drug-loaded micelle-like nanoparticles generated a magnetocaloric effect under the alternating magnetic field, which not only killed tumor cells by magnetic hyperthermia but also promoted the rapid release of drugs at the tumor site. In general, magnetically enhanced chemotherapy showed the best therapeutic effect on tumors.

One-Minute Preparation of Iron Foam-Drug Implant for Ultralow-Power Magnetic Hyperthermia-Based Combination Therapy of Tumors in Vivo

Guangchao Xie,Bingjie Li,Xuejun Zhang,Jiaojiao Yu,Shao-Kai Sun

The magnetic hyperthermia-based combination therapy (MHCT) is a powerful tumor treatment approach due to its unlimited tissue penetration depth and synergistic therapeutic effect. However, strong magnetic hyperthermia and facile drug loading are incompatible with current MHCT platforms. Herein, an iron foam (IF)-drug implant is established in an ultra-facile and universal way for ultralow-power MHCT of tumors in vivo for the first time. The IF-drug implant is fabricated by simply immersing IF in a drug solution at an adjustable concentration for 1 min. Continuous metal structure of IF enables ultra-high efficient magnetic hyperthermia based on eddy current thermal effect, and its porous feature provides great space for loading various hydrophilic and hydrophobic drugs via “capillary action”. In addition, the IF has the merits of low cost, customizable size and shape, and good biocompatibility and biodegradability, benefiting reproducible and large-scale preparation of IF-drug implants for biological application. As a proof of concept, IF-doxorubicin (IF-DOX) is used for combined tumor treatment in vivo and achieves excellent therapeutic efficacy at a magnetic field intensity an order of magnitude lower than the threshold for biosafety application. The proposed IF-drug implant provides a handy and universal method for the fabrication of MHCT platforms for ultralow-power combination therapy.

Enhanced tumor penetration for efficient chemotherapy by a magnetothermally sensitive micelle combined with magnetic targeting and magnetic hyperthermia

Wang Yu,Wang Rui,Chen Lixin,Chen Lili,Zheng Yi,Xin Yuanrong,Zhou Xiqiu,Song Xiaoyun,Zheng Jinzhou

The high accumulation and poor penetration of nanocarriers in tumor is a contradiction of nanomedicine, which reduces the efficacy of chemotherapy. Due to the positive effect of hyperthermia on in vivo drug diffusion, we designed a magnetothermally sensitive micelle (MTM) by integrating magnetic targeting (MT), magnetic hyperthermia (MH), and magnetothermally responsive drug release to facilitate simultaneous drug accumulation and penetration in tumor. Accordingly, we synthesized a cyanine7-modified thermosensitive polymer with phase transition at 42.3°C, and utilized it to prepare drug-loaded MTMs by encapsulating superparamagnetic MnFe2O4 nanoparticles and doxorubicin (DOX). The obtained DOX–MTM had not only high contents of DOX (9.1%) and MnFe2O4 (38.7%), but also some advantages such as superparamagnetism, high saturation magnetization, excellent magnetocaloric effect, and magnetothermal-dependent drug release. Therefore, DOX–MTM improved in vitro DOX cytotoxicity by enhancing DOX endocytosis under the assistance of MH. Furthermore, MT and MH enhanced in vivo DOX–MTM accumulation and DOX penetration in tumor, respectively, substantially inhibiting tumor growth (84%) with excellent biosafety. These results indicate the development of an optimized drug delivery system with MH and MH-dependent drug release, introducing a feasible strategy to enhance the application of nanomedicines in tumor chemotherapy.

Xinqiang Liang,Wenting Xu,Siyi Li,Mekhrdod S. Kurboniyon,Kunying Huang,Guilan Xu,Wene Wei,Shufang Ning,Litu Zhang,Chen Wang

Hyperthermia therapy is a hotspot because of its minimally invasive treatment process and strong targeting effect. Herein, a synergistic magnetic and photothermal therapeutic nanoplatform is rationally constructed. The well-dispersive mSiO 2 -SmCo x nanoparticles (NPs) were synthesized through a one-step procedure with the regulated theoretical molar ratio of Sm/Co among 1:1, 1:2, and 1:4 for controlling the dispersion and magnetism properties of SmCo x NPs in situ growth in the pore structure of mesoporous SiO 2 (mSiO 2 ), where mSiO 2 with diverse porous structures and high specific surface areas serving for locating the permanent magnetic SmCo x NPs. The mSiO 2 -SmCo x (Sm/Co = 1:2) NPs with highly dispersed and uniform morphology has an average diameter of ∼73.08 nm. The photothermal conversion efficiency of mSiO 2 -SmCo x (Sm/Co = 1:2) NPs was determined to be nearly 41%. The further in vitro and in vivo anti-tumor evaluation of mSiO 2 -SmCo x (Sm/Co = 1:2) NPs present promising potentials for hyperthermia-induced tumor therapy due to magnetic and photothermal effects.

Nonmetallic graphite for tumor magnetic hyperthermia therapy

Guangchao Xie,Shuyue Guo,Bingjie Li,Wenjing Hou,Yanqi Zhang,Jinbin Pan,Xi Wei,Shao-Kai Sun

Magnetic hyperthermia therapy (MHT) has garnered immense interest due to its exceptional spatiotemporal specificity, minimal invasiveness and remarkable tissue penetration depth. Nevertheless, the limited magnetothermal heating capability and the potential toxicity of metal ions in magnetic materials based on metallic elements significantly impede the advancement of MHT. Herein, we introduce the concept of nonmetallic materials , with graphite (Gra) as a proof of concept , as a highly efficient and biocompatible option for MHT of tumors in vivo for the first time. The Gra exhibits outstanding magnetothermal heating efficacy owing to the robust eddy thermal effect driven by its excellent electrical conductivity . Furthermore, being composed of carbon, Gra offers superior biocompatibility as carbon is an essential element for all living organisms. Additionally, the Gra boasts customizable shapes and sizes, low cost, and large-scale production capability, facilitating reproducible and straightforward manufacturing of various Gra implants. In a mouse tumor model, Gra-based MHT successfully eliminates the tumors at an extremely low magnetic field intensity , which is less than one-third of the established biosafety threshold. This study paves the way for the development of high-performance magnetocaloric materials by utilizing nonmetallic materials in place of metallic ones burdened with inherent limitations.
