A Multi-Region Magnetoimpedance-Based Bio-Analytical System for Ultrasensitive Simultaneous Determination of Cardiac Biomarkers Myoglobin and C-Reactive Protein

Zhen Yang,Huanhuan Wang,Pengfei Guo,Yuanyuan Ding,Chong Lei,Yongsong Luo

Cardiac biomarkers (CBs) are substances that appear in the blood when the heart is damaged or stressed. Measurements of the level of CBs can be used in course of diagnostics or monitoring the state of the health of group risk persons. A multi-region bio-analytical system (MRBAS) based on magnetoimpedance (MI) changes was proposed for ultrasensitive simultaneous detection of CBs myoglobin (Mb) and C-reactive protein (CRP). The microfluidic device was designed and developed using standard microfabrication techniques for their usage in different regions, which were pre-modified with specific antibody for specified detection. Mb and CRP antigens labels attached to commercial Dynabeads with selected concentrations were trapped in different detection regions. The MI response of the triple sensitive element was carefully evaluated in initial state and in the presence of biomarkers. The results showed that the MI-based bio-sensing system had high selectivity and sensitivity for detection of CBs. Compared with the control region, ultrasensitive detections of CRP and Mb were accomplished with the detection limits of 1.0 pg/mL and 0.1 pg/mL, respectively. The linear detection range contained low concentration detection area and high concentration detection area, which were 1 pg/mL–10 ng/mL, 10–100 ng/mL for CRP, and 0.1 pg/mL–1 ng/mL, 1 n/mL–80 ng/mL for Mb. The measurement technique presented here provides a new methodology for multi-target biomolecules rapid testing.Keywords:myoglobin;C-reactive protein;Dynabeads;magnetoimpedance;microfluidic device

An integrated giant magnetoimpedance biosensor for detection of biomarker

Tao Wang,Zhen Yang,Chong Lei,Jian Lei,Yong Zhou

A Dynabeads-labeled magnetic immunoassay (MIA) has been developed by using an integrated giant magnetoimpedance (GMI) biosensor for the detection of alpha-fetoprotein (AFP). The GMI biosensor (Cr/Cu/NiFe/Cu/NiFe/Al 2 O 3 /Cr/Au films) integrated magnetic sensing elements and a biomolecular immunoplatform. Au film was modified with 11-Mercaptoundecanoic acid (11-MUA) for the immobilization of AFP monoclonal antibody . Double antibody sandwich immunoassay was used to specifically capture and label AFP antigen. Functionalized Dynabeads were conjugated to AFP antigen by streptavidin–biotin binding assay. GMI responses were measured for sensitive detection of AFP from 1 to 10   ng/ml. Our results revealed that the presence of AFP on the biosensor improved the GMI effect owing to the induced magnetic dipole of superparamagnetic Dynabeads, and the GMI ratio was greatly increased at high frequency. Specificity of MIA was tested through the use of 1% bovine serum albumin (BSA). The underlying biophysical mechanisms responsible for the enhanced GMI effect in the detection of AFP were discussed. This work provides a complex lab-on-chip MIA for the detection of biomarker, which may open up a new way for the development of GMI-based MIA in clinical trials .

An integrated magnetoimpedance biosensor microfluidic magnetic platform for the evaluation of cardiac markers cTnI

Yang Zhen,Jingyuan Chen,mengyu Liu,Jiabao Huang,Jieping Liang,Mengjiao Zhu,Yuanwei Shen,Danqing Li,Chong Lei,Xuecheng Sun

An integrated magnetoimpedance (MI) biosensor microfluidic magnetic platform was proposed for the evaluation of of cardiac markers cardiac troponin-I (cTnI). The bioanalytes evaluation platform mainly contained external permanent magnets (PMs), MI elements, peelable SiO2film units and microfluidic chip (MFC). The MI elements were made of micro-electro-mechanical system (MEMS)-based multilayered [Ti(6 nm)/FeNi(100 nm)]5/Cu(400 nm)/[Ti(6 nm)/FeNi(100 nm)]5 thin films and designed as meander structures with closed magnetic flux. The MFC was fabricated by using 3D printing and inverted molding techniques, and designed with a solution mixing reaction region, a magnetic separation region and detection regions. Peelable SiO2 film units with the same size as the MI sensing element are used for bioanalytes immunoreactivity interface. Two large PMs were placed directly below the MI sensing unit to provide a bias magnetic field, and the smaller PM was embedded in MCF for magnetic separation function. The different concentration biological target (cTnI antigen), PBS buffer and Dynabeads labeled polyclonal cTnI antibody solution are injected sequentially into the MCF. After immunoreactivity and magnetic separation, a classical sandwich immunoreaction process occured on the surface of the monoclonal antibody-modified SiO2 film via self-assembling technology in the reaction region of MFC. The fundamental principle for evaluation of cTnI was based on variation of the MI signal under different concentration biological target coupled with different number of Dynabeads. It was demonstrated that the mentioned MI-based magnetic platform can perform detection analyses over a range of cTnI concentrations (lowest concentration 0.1 ng/ml and highest concentration 100 ng/ml). The proposed MI-based magnetic platform provides a sensitive, reliable, stable and reusable bioanalytical platform and has potential for future biomedical applications.

Giant magnetoimpedance based immunoassay for cardiac biomarker myoglobin

Zhen Yang,Huanhuan Wang,Xinwei Dong,Hailong Yan,Chong Lei,Yongsong Luo

Herein, a novel method for the detection of the cardiac biomarker myoglobin (Mb) is proposed by using an integrated giant magnetoimpedance (GMI) based immunosensor. The GMI sensor integrates magnetic sensing elements and a gold film. Micro-patterned GMI sensing elements were prepared from a cobalt-based commercial amorphous ribbon (Metglas® 2714A) using micro-electro-mechanical system (MEMS) technology. The gold film was deposited on the GMI sensing element to act as a support for the immunoplatform. The classical sandwich assay was used for the detection of Mb targeted with Dynabeads by using the antibody–antigen pair combination of biotin–streptavidin. The GMI ratios of the sensors with different concentrations of the antigen against Mb were investigated. As a result, the GMI ratios are significantly improved at high frequency if Mb is present. The sensor has a linear response in the 1 to 10 ng ml−1 concentration range, and the detection limit is 0.5 ng ml−1 at a working frequency of 10 MHz. The method is successfully applied to the determination of Mb, and can be used to detect other cardiac markers like Troponin and CK-MB.

An integrated magnetic microfluidic chip for rapid immunodetection of the prostate specific antigen using immunomagnetic beads

Feng Zhu,Zhi Shaotao,Guo Lei,Zhou Yong,Lei Chong

The authors describe an integrated microfluidic chip for immunodetection of the prostate specific antigen (PSA) by using giant magnetoimpedance (GMI) sensor. This chip contains an immunoreaction platform and a biomarker detection system. The immunoreaction platform contains an incubation chamber and a reactive chamber to implement immunological reaction in microfluidics. The system can detect PSA rapidly with ultra-high sensitivity. Both are fabricated by MEMS technology. Immunomagnetic beads (If PSA binds to its antibody (that is labeled with immunomagnetic beads; IMBs) it will be trapped on the surface of self-assembled film. Trapped IMBs generate a stray magnetic field under the magnetization of the external applied magnetic field and can be detected by the GMI sensor. The chip can detect PSA with a detection limit as low as 0.1 ng ∙ mL −1 and works in the 0.1 ng ∙ mL −1 to 20 ng ∙ mL −1  concentration range. Compared to established GMI biosensors, the magnetic microfluidic chip reduces assay time, and lends itself to fast detection. It also avoids complex handling steps, enhances reaction efficiency and decreases experimental errors. An integrated magnetic microfluidic chip which contains immunoreaction platform and biomarker detection system was designed and microfabricated by micro-electromechanical systems (MEMS) technology to detect prostate specific antigen (PSA) rapidly, and has promise in Point-of-care (PoC) diagnostic applications.

Ultrasensitive determination of carcinoembryonic antigens using a magnetoimpedance immunosensor

Tao Wang,Lei Guo,Chong Lei,Yong Zhou

Herein, a giant magnetoimpedance (GMI) sensor combined with the sandwich immunoassay was used for ultrasensitive detection of carcinoembryonic antigens (CEA). Oblong-shaped Au films were enclosed with SU-8 photoresists to form several microcavities that were treated with different concentrations of CEA. The quantification of low-concentration CEA (1 pg mL−1–10 ng mL−1) was achieved by using one single GMI sensor, demonstrating a good linear relationship between the GMI response and the CEA concentration. The lower detection limit for CEA was found to be as low as 1 pg mL−1, which provides extreme sensitivity. We observed that high-concentration CEA (>10 ng mL−1) can result in a cluster of Dynabeads, thereby reducing the target signals since the adjacent magnetic dipole fields canceled each other out. Target specificity of the GMI-based magnetic immunoassay has also been verified by testing nonspecific targets. The results obtained here suggest that the GMI sensor could serve as an ultrasensitive immunosensor for quantitative determination of biomarkers.

A flexible giant magnetoimpedance-based biosensor for the determination of the biomarker C-reactive protein

Yang Zhen,Liu Yan,Lei Chong,Sun Xue-cheng,Zhou Yong

We report on a method for the determination of magnetic bead-labeled C-reactive protein (CRP), a biomarker of cardiovascular diseases and inflammations. It is using a flexible giant magnetoimpedance (GMI)-based platform. Micro-patterned GMI sensing elements were prepared from a cobalt-based commercial amorphous ribbon (Metglas® 2714A) using micro electro-mechanical system (MEMS) technology. A gold film was then deposited on the GMI sensing element to act as a support for the immuno platform. Sandwich assays are performed using antibody-antigen combinations and biotin-streptavidin interactions on the gold film substrate surface via self-assembled layers. The GMI ratios of the sensors with different concentrations of antigen against CRP were investigated. The results show that the presence of CRP antigens on the biosensor improves the GMI effect owing to the induced magnetic dipole of superparamagnetic beads, and that the GMI ratios show distinct changes at high frequency. This bioassay for CRP has a linear detection range between 1 to 10 ng·mL −1 . This new method in our perception provides a widely applicable basis for rapid diagnostic testing and will pave the way for future development of electrochemical point-of-care diagnostic devices for cardiac diseases. Study on the Mechanism, Properties, and Application as EMW-Absorbing Matrix Materials of Protonic Acid-Doped Heat-Resistant Conductive Polyurethanes

Shuai Yuan,Jin Yang

Protonic acid doping of heat-resistant and conductive polyurethane elastomers (PCPU) can significantly enhance their electrical conductivity. By doping with different types of protonic acids and varying molar amounts of camphorsulfonic acid (CSA), the doping mechanism and molecular structure changes of PCPU were systematically investigated using multiple characterization techniques such as Fourier-transform infrared (FTIR), X-ray diffraction (XRD), UV–vis and so on. Meanwhile, the effects of protonic acid doping on the thermal stability and mechanical properties of PCPU were further evaluated using thermo-gravimetric analysis (TGA), Differential scanning calorimetry (DSC), and stress–strain testing. Although protonic acid doping reduces the thermal stability and fracture strength of PCPU, the acid-doped PCPU can form an ionic gel network, which facilitates the uniform dispersion of electromagnetic wave (EMW) absorbers within the matrix. Additionally, protonic acid doping endows the electromagnetic wave absorbing materials (EMWAMs) prepared from PCPU and nanographite (Gr) with tunable electromagnetic matching characteristics and enhanced wave attenuation capabilities, exhibiting excellent EMW absorption performance across a broad frequency range. For instance, the 10% Gr-PCPU-2CSA composite achieves a minimum reflection loss of −27.98 dB and an effective absorption bandwidth of 5.3 GHz at a thickness of 2.5 mm. Therefore, this study provides novel insights for the development of high-performance EMW-absorbing matrix materials.

Ultrasensitive detection and quantification of E. coli O157:H7 using a giant magnetoimpedance sensor in an open-surface microfluidic cavity covered with an antibody-modified gold surface

Yang Zhen,Liu Yan,Lei Chong,Sun Xue-cheng,Zhou Yong

We report on a method for ultrasensitive detection and quantification of the pathogen Escherichia coli ( E. coli ), type O157:H7. It is using a tortuous-shaped giant magnetoimpedance (GMI) sensor in combination with an open-surface microfluidic system coated with a gold film for performing the sandwich immunobinding on its surface. Streptavidin-coated super magnetic Dynabeads were loaded with biotinylated polyclonal antibody to capture E. coli O157:H7. The E. coli -loaded Dynabeads are then injected into the microfluidics system where it comes into contact with the surface of gold nanofilm carrying the monoclonal antibody to form the immunocomplex. As a result, the GMI ratio is strongly reduced at high frequencies if E. coli O157:H7 is present. The sensor has a linear response in the 50 to 500 cfu·mL −1 concentration range, and the detection limit is 50 cfu·mL −1 at a working frequency of 2.2 MHz. In our perception, this method provides a valuable tool for developing GMI-based microfluidic sensors systems for ultrasensitive and quantitative analysis of pathogenic bacteria. The method may also be extended to other sensing applications by employing respective immunoreagents. Fig. (a) Graphical illustration of the test setup. (b) Relationship of GMI signal vs. E. coli O157:H7 concentration.
