Currently, magnetic applications have great potential for development in the field of drug carriers. In this paper, Fe3O4-PVA@SH, an amphiphilic magnetically targeting drug carrier, was prepared by using Fe3O4 and PVA with thiohydrazide-iminopropyltriethoxysilane(TIPTS). The loading capacity of Fe3O4-PVA@SH on Aspirin and the drug release kinetics of loaded drugs were studied. The obtained Fe3O4-PVA@SH exhibits excellent drug release properties in simulating the human body fluid environment (pH 7.2). Since magnetically targeting drug carriers are readily available and have excellent biocompatibility and the characteristics of drug release. This work’s development, preparing amphiphilic magnetically targeting drug carriers in drug delivery and other fields, has great significance.

Bacterial biofilms are difficult to treat with traditional antibiotics. Ultrasound (US)-responsive antibacterial agents have emerged as promising alternatives for the treatment of bacterial biofilm infections. However, they often suffer from antibiotic dependence, lack of targeting, and low antibacterial efficiency. Herein, we develop Fe3O4/MnCO3 microbubbles (FMMB) by self-assembly of Fe3O4 nanoparticles (Fe3O4 NPs) and MnCO3 nanoparticles (MnCO3 NPs). By the direction of magnetic field (MF), FMMB can be targeted to the Staphylococcus aureus (MRSA) biofilm. Under US irradiation, FMMB can destroy the structure of MRSA biofilms by cavitation-induced mechanical effects and kill bacteria by MnCO3 NPs generated reactive oxygen species (ROS) through the sonodynamic effect. In the mice model with catheter MRSA biofilm infection, FMMB removed 70.4% of the biofilm with MF and US, and the bacteria inactivation efficiency reached as high as 99.999% (4.9 Log). This work develops multifunctional microbubbles with both US-responsive mechanical and sonodynamic effects for biofilm destruction and MF-responsive properties for biofilm targeting, which provides a promising strategy for the design of antibiofilm agents to effectively treat bacterial biofilm infections.

The booming development of nanomedicine offers great opportunities for cancer diagnostics and therapeutics. Herein, a magnetic targeting-enhanced cancer theranostic strategy using a multifunctional magnetic-plasmonic nano-agent is developed, and a highly effective in vivo tumor photothermal therapy, which is carefully planed based on magnetic resonance (MR)/photoacoustic (PA) multimodal imaging, is realized. By applying an external magnetic field (MF) focused on the targeted tumor, a magnetic targeting mediated enhanced permeability and retention (MT-EPR) effect is observed. While MR scanning provides tumor localization and reveals time-dependent tumor homing of nanoparticles for therapeutic planning, photoacoustic imaging with higher spatial resolution allows noninvasive fine tumor margin delineation and vivid visualization of three dimensional distributions of theranostic nanoparticles inside the tumor. Utilizing the near-infrared (NIR) plasmonic absorbance of those nanoparticles, selective photothermal tumor ablation, whose efficacy is predicted by real-time infrared thermal imaging intra-therapeutically, is carried out and then monitored by MR imaging for post-treatment prognosis. Overall, this study illustrates the concept of imaging-guided MF-targeted photothermal therapy based on a multifunctional nano-agent, aiming at optimizing therapeutic planning to achieve the most efficient cancer therapy.

Graphical Dual response : Monodisperse poly(acrylic acid)-modified Fe 3 O 4 (PAA@Fe 3 O 4 ) hybrid microspheres with dual responses (magnetic field and pH) were successfully fabricated (see figure). The drug release behavior was strongly dependent on pH value due to the unique properties of PAA. Furthermore, doxorubicin (DOX)-loaded PAA@Fe 3 O 4 composites show an enhanced magnetically guided anticancer effect. Monodisperse poly(acrylic acid)-modified Fe 3 O 4 (PAA@Fe 3 O 4 ) hybrid microspheres with dual responses (magnetic field and pH) were successfully fabricated. The PAA polymer was encapsulated into the inner cavity of Fe 3 O 4 hollow spheres by a vacuum-casting route and photo-initiated polymerization. TEM images show that the samples consist of monodisperse porous spheres with a diameter around 200 nm. The Fe 3 O 4 spheres, after modification with the PAA polymer, still possess enough space to hold guest molecules. We selected doxorubicin (DOX) as a model drug to investigate the drug loading and release behavior of as-prepared composites. The release of DOX molecules was strongly dependent on the pH value due to the unique property of PAA. The HeLa cell-uptake process of DOX-loaded PAA@Fe 3 O 4 was observed by confocal laser scanning microscopy (CLSM). After being incubated with HeLa cells under magnet magnetically guided conditions, the cytotoxtic effects of DOX-loaded PAA@Fe 3 O 4 increased. These results indicate that pH-responsive magnetic PAA@Fe 3 O 4 spheres have the potential to be used as anticancer drug carriers.

The high accumulation and poor penetration of nanocarriers in tumor is a contradiction of nanomedicine, which reduces the efficacy of chemotherapy. Due to the positive effect of hyperthermia on in vivo drug diffusion, we designed a magnetothermally sensitive micelle (MTM) by integrating magnetic targeting (MT), magnetic hyperthermia (MH), and magnetothermally responsive drug release to facilitate simultaneous drug accumulation and penetration in tumor. Accordingly, we synthesized a cyanine7-modified thermosensitive polymer with phase transition at 42.3°C, and utilized it to prepare drug-loaded MTMs by encapsulating superparamagnetic MnFe2O4 nanoparticles and doxorubicin (DOX). The obtained DOX–MTM had not only high contents of DOX (9.1%) and MnFe2O4 (38.7%), but also some advantages such as superparamagnetism, high saturation magnetization, excellent magnetocaloric effect, and magnetothermal-dependent drug release. Therefore, DOX–MTM improved in vitro DOX cytotoxicity by enhancing DOX endocytosis under the assistance of MH. Furthermore, MT and MH enhanced in vivo DOX–MTM accumulation and DOX penetration in tumor, respectively, substantially inhibiting tumor growth (84%) with excellent biosafety. These results indicate the development of an optimized drug delivery system with MH and MH-dependent drug release, introducing a feasible strategy to enhance the application of nanomedicines in tumor chemotherapy.

Recent advances in microencapsulation technology have emerged as a promising approach for achieving self-healing of electrical and mechanical damage in epoxy resin. Nevertheless, current implementations face dual challenges: (1) Dependency on external stimuli (elevated temperature, pH variations, or humidity) for microcapsule activation, precluding non-contact self-healing mechanisms; (2) Compromised matrix performance through reduced mechanical strength and dielectric integrity. To address these issues, this work innovatively presents a method to fabricate magnetic-targeted natural photosensitive microcapsules. By incorporating Fe 3 O 4 @SiO 2 magnetic nanoparticles into the shells of photosensitive microcapsules via interfacial polymerization with a core-shell structure, the microcapsules gain targeting migration ability and UV shielding function. The resulting magnetic-targeted UV light-sensitive microcapsules/epoxy composite insulating materials can achieve non-contact damage repair using natural light. Moreover, the magnetic targeting enables directional migration of the microcapsules to the damage site in the matrix, significantly enhancing their utilization rate. Results show that, at a microcapsule doping concentration of 2 wt%, the tensile properties of the epoxy resin increased by 16.3% compared to pure epoxy resin, with minimal change in insulation strength and dielectric properties. Furthermore, the magnetically targeted natural photosensitive microcapsules can effectively repair surface cracks and electrical tree damage in epoxy resin, restoring the composite material's insulating property to 92.1% of its pre-damage state after self-healing.

Magnetic polymer nanofibres intended for drug delivery have been designed and fabricated by electrospinning. Magnetite (Fe3O4) nanoparticles were successfully incorporated into electrospun nanofibre composites of two cellulose derivatives, dehydroxypropyl methyl cellulose phthalate (HPMCP) and cellulose acetate (CA), while indomethacin (IDN) and aspirin have been used as model drugs. The morphology of the neat and magnetic drug-loaded electrospun fibres and the release characteristics of the drugs in artificial intestinal juice were investigated. It was found that both types of electrospun composite nanofibres containing magnetite nanoparticles showed superparamagnetism at room temperature, and their saturation magnetisation and morphology depend on the Fe3O4 nanoparticle content. Furthermore, the presence of the magnetite nanoparticles did not affect the drug release profiles of the nanofibrous devices. The feasibility of controlled drug release to a target area of treatment under the guidance of an external magnetic field has also been demonstrated, showing the viability of the concept of magnetic drug-loaded polymeric composite nanofibres for magneto-chemotherapy.

The ability to selectively destroy cancer cells while sparing normal tissue is highly desirable during the cancer therapy. Here, magnetic targeted photothermal therapy was demonstrated by the integration of MoS 2 (MS) flakes and Fe 3 O 4 (IO) nanoparticles (NPs), where MoS 2 converted near-infrared (NIR) light into heat and Fe 3 O 4 NPs served as target moiety directed by external magnetic field to tumor site. The MoS 2 /Fe 3 O 4 composite (MSIOs) functionalized by biocompatible polyethylene glycol (PEG) were prepared by a simple two-step hydrothermal method. And the as-obtained MSIOs exhibit high stability in bio-fluids and low toxicity in vitro and in vivo . Specifically, the MSIOs can be applied as a dual-modal probe for T 2 -weighted magnetic resonance (MR) and photoacoustic tomography (PAT) imaging due to their superparamagnetic property and strong NIR absorption. Furthermore, we demonstrate an effective result for magnetically targeted photothermal ablation of cancer. All these results show a great potential for localized photothermal ablation of cancer spatially/timely guided by the magnetic field and indicated the promise of the multifunctional MSIOs for applications in cancer theranostics.

Although there has been significant progress in the development of tumor immunotherapies, many challenges still exist for the treatment of solid tumors. Natural killer (NK) cells possess broad-spectrum cytotoxicity against tumors, but their limited migration and infiltration abilities restrict their application in solid tumor therapies. Here, we combined a facile and efficient magnetic-targeting strategy with NK cell-based therapy to develop a novel immunotherapy approach for treating solid tumors. Anti-CD56 antibodies were conjugated with Fe3O4 nanoparticles, which could specifically bind with NK-92 cells endowing them with a magnetic field driven targeting ability. These NK–Fe3O4 biohybrid nanoparticles were able to facilitate directional migration to the tumor site in vivo under external magnetic field guidance and efficiently inhibit tumor growth. These functionalized NK cells represent a novel approach for solid tumor therapy and may provide a promising modality for cancer interventions in the future.

Near-infrared (NIR) light induced phototherapy has attracted considerable attention due to its deep therapeutic depth. To improve the therapeutic outcome and address non-selective side effects, the combination of complementary phototherapeutic strategies in a single nanoagent with precise targeting ability may provide an effective approach for cancer therapy. Thus we have developed an 808 nm NIR light triggered nanosystem based on IR806 dye functionalized MnFe2O4 (MFO-IR) for synchronous magnetic targeted and magnetic resonance (MR) imaging guided in vivo photodynamic/photothermal synergistic therapy. In this construction strategy, carboxylic acid functionalized NIR dye IR806 is explored as an 808 nm NIR-excited photosensitizer (PS) for the first time, which can also provide a conjugation site for MnFe2O4 nanoparticles (MFO NPs). Here, monodisperse MFO NPs have multiple capacities as dye carriers, targeting ligands, MRI contrast agents and photothermal agents. MFO-IR nanocomposites (NCs) with negligible toxicity present efficient NIR-mediated photothermal damage and ROS cytotoxicity via the relevant in vitro experimental investigations. With ideal magnetic targeting effects and remarkable NIR light-responsive properties, these MFO-IR NCs exhibit high in vivo tumor localization and could destroy subcutaneous solid tumors completely under an external magnetic field and 808 nm laser irradiation. Consequently, this magnetic nanosystem has great potential for simultaneous diagnosis and precise cancer phototherapy.

Near-infrared (NIR) light has great potential in biomedical applications due to its advantages of deep penetration depth and low photodamage to biological tissues. In this paper, we constructed a novel core-shell structured drug nanocarrier, Fe 3 O 4 @Au@SiO 2 , for the controlled delivery of etoposide (VP16), a chemotherapeutic drug for cancer patients. The novel core-shell structured drug delivery platform is composed of a mesoporous silica shell and a magnetic Fe 3 O 4 core using Au nanoparticles (AuNPs) as the interlayer, which is characterized by atomic force microscopy (AFM), X-ray diffraction (XRD), Fourier-transform infrared (FTIR) spectroscopy, N 2 adsorption/desorption isotherms and the magnetic measurements with vibrating-sample magnetometer (VSM). The synergistic effects of AuNPs, mesoporous silica and Fe 3 O 4 make the core-shell structured nanocomposites an excellent candidate for targeted and NIR light irradiation-controlled drug delivery. For the proposed nanocarrier of VP16, the mesopores in silica can enhance the encapsulation capacity of the nanocarrier and the AuNPs can effectively convert the NIR light into heat to speed up the drug deliver; meanwhile, the incorporation of Fe 3 O 4 with high magnetization to the drug delivery platform realize drug targeting under an applied external magnetic field.

Low back pain (LBP) resulting from sciatic nerve compression presents major challenges in pain management, as traditional therapies provide only short-term relief and pose risks of systemic toxicity. In this study, an innovative Fe 3 O 4 @ZIF-8-RVC (FZR) dual-responsive nanoplatform is introduced that integrates magnetic targeting with pH-sensitive, sustained drug release to overcome these limitations. The FZR nanoplatform encapsulates ropivacaine (RVC) within the ZIF-8-coated Fe 3 O 4 core, enabling precise and prolonged analgesia at the injury site through magnetic guidance and acid-triggered release. In vitro and in vivo assessments indicate that FZR achieves high drug loading, sustained release in acidic environments, and excellent biocompatibility, significantly extending analgesic effects in chronic nerve injury models while minimizing systemic exposure. Behavioral tests and molecular analyses in LBP rat models confirm that FZR effectively suppresses pain-related neuronal activity and central sensitization markers. This dual-responsive nanoplatform FZR offers a safe, long-lasting, and targeted therapeutic approach, holding strong potential for advancing pain relief in LBP and related neuropathic pain conditions.
