Imazethapyr disrupts plant phosphorus homeostasis and acquisition strategies

Yihao Li,Nan Zhang,Jiarui Xu,Lijuan Liu,Xiaochuang Cao,Xianyong Lin,Chengliang Sun

The deficiency of essential mineral nutrients caused by xenobiotics often results in plant mortality or an inability to complete its life cycle. Imazethapyr , a widely utilized imidazolinone herbicide, has a long-lasting presence in the soil-plant system and can induce toxicity in non-target plants. However, the effects of imazethapyr on mineral nutrient homeostasis remain poorly comprehended. In this study, Arabidopsis seedlings exposed to concentrations of 4 and 10 μg/L imazethapyr showed noticeable reductions in shoot development and displayed a distinct dark purple color, which is commonly associated with phosphorus (P) deficiency in crops. Additionally, the total P content in both the shoots and roots of Arabidopsis significantly decreased following imazethapyr treatment when compared to the control groups. Through the complementary use of physiological and molecular analyses, we discovered that imazethapyr hinders the abundance and functionality of inorganic phosphorus (Pi) transporters and acid phosphatase . Furthermore, imazethapyr impairs the plant's Pi-deficiency adaptation strategies, such as inhibiting Pi transporter activities and impeding root hair development, which ultimately exacerbate P starvation. These results provide compelling evidence that residues of imazethapyr have the potential to disrupt plant P homeostasis and acquisition strategies. These findings offer valuable insights for risk assessment and highlight the need to reconsider the indiscriminate use of imazethapyr, particularly under specific scenarios such as nutrient deficiency.

Beneficial Bacterium Azospirillum brasilense Induces Morphological, Physiological and Molecular Adaptation to Phosphorus Deficiency in Arabidopsis

Sun Nan,Huang Lin,Zhao Hongcheng,Zhang Nan,Lin Xianyong,Sun Chengliang

Although most cultivated soils have high levels of total phosphorus (P), the levels of bioavailable inorganic P (Pi) are insufficient. The application of plant-growth-promoting rhizobacteria (PGPR) is an eco-friendly strategy for P utilization; however, PGPR-mediated plant responses that enhance Pi acquisition remain unexplored. Here, we investigated the effect of Azospirillum brasilense on Arabidopsis adaptation to Pi deficiency. Results showed that A. brasilense inoculation alleviated Pi-deficiency-induced growth inhibition and anthocyanin accumulation and increased the total P content in Arabidopsis plants. A comprehensive analysis of root morphology revealed that A. brasilense increased root hair density and length under Pi-limited conditions. We further demonstrated that A. brasilense enhanced the acid phosphatase activity and upregulated the expression of several Pi transporter genes, such as PHOSPHATE1 (PHO1), PHOSPHATE TRANSPORTER 1:(PHT1:1) and PHT1;4. However, A. brasilense did not enhance the growth o total P content in pht1;1, pht1;4 and pht1;1pht1;4 mutants. Moreover, A. brasilense could not increase the P content and PHT1;1 expression in the root hairless mutant rsl4rsl2, because of the occurrence of low-Pi-induced PHT1;1 and PHT1;4 in root hairs. These results indicate that A. brasilense can promote root hair development and enhance acid phosphatase activity and Pi transporter expression levels, consequently improving the Pi absorption capacity and conferring plant tolerance to Pi deficiency.

Characterization of invisible symptoms caused by early phosphorus deficiency in cucumber plants using near-infrared hyperspectral imaging technology

Jiyong Shi,Yueying Wang,Zhihua Li,Xiaowei Huang,Tingting Shen,Xiaobo Zou

In the early stage of P deficiency in cucumbers, the P deficiency symptoms in leaves are similar to the symptoms in control leaves at the early stage of aging and are difficult to identify with naked eyes or computer image processing techniques. In order to realize the quick diagnosis of P deficiency in plants at the early stage, the NIR hyperspectral images of control leaves and P-deficient leaves were collected, and the feature information of the NIR hyperspectral images was extracted by PCA and ICA respectively. Through PCA and HCA verification, the IC1 component diagram of P-deficient leaves NIR hyperspectral image could effectively characterize the features of invisible water-stained plaques caused by early P-deficient leaves. Region of interest from IC1 was selected to extract spectral information for classification, and the diagnostic rate was remarkably improved. Finally, 240 leaves were diagnosed by using the BP-ANN model with a diagnostic rate of 97.5%. In addition, the experiment verified that it was possible to diagnose whether the plant was in the state of P deficiency 21 days in advance, and timely guidance of top dressing was of great significance to increase yield.

Root metabolites regulated by FERONIA promote phosphorus-solubilizing rhizobacteria enrichment induced by Arabidopsis thaliana coping with phosphorus deficiency

Lingyun Zhang,Xuelei Deng,Jia Xiao,Wei Zhao,Pan Zou,Ruizhe Liao,Keying Xie,Hongdong Liao

The recruitment of the phosphorus-solubilizing rhizobacteria plays an important role in response to phosphorus deficiency. Through the treatments of Arabidopsis thaliana (Col-0) and the FERONIA (FER) functional deficient mutants ( fer-4 and fer-5 ) with the soil suspension in various phosphorus conditions, we discovered that FER could promote phosphorus-solubilizing rhizobacteria enrichment to rescue the defective plant during phosphorus deficiency. The amplicon sequencing data reflected that the phosphorus-solubilizing rhizobacterial genus Alcaligenes was significantly enriched of Col-0 than fer-4 in low phosphorus conditions. Metabolomics analysis revealed that there were more α-D-Glucose (α-D-Glc) and L-Leucine (L-Leu) in Col-0 roots than those in fer-4 roots. The alterations of α-D-Glc and L-Leu mediated by FER had high-positive correlations to the enrichment of Alcaligenes . We successfully isolated a phosphorus-solubilizing rhizobacteria strain identified as Alcaligenes faecalis PSB15. The α-D-Glc and L-Leu could promote the strain PSB15 growth on LB agar plates and assist fer-4 in recovering from phosphorus starvation in the low phosphorus (LP) liquid medium vermiculite with tricalcium phosphate (TCP). The α-D-Glc and L-Leu could be considered as promising compounds to enrich beneficial phosphorus-solubilizing rhizobacteria, such as Alcaligenes , and provide a reference for overcoming the plight of phosphorus deficiency in crops in the field of agricultural production in the future.

NaCl Facilitates Cell Wall Phosphorus Reutilization in Abscisic Acid Dependent Manner in Phosphorus Deficient Rice Root

Yang Xiaozheng,Liu Yusong,Huang Jing,Tao Ye,Wang Yifeng,Shen Renfang,Zhu Xiaofang

Phosphorus (P) starvation in rice facilitates the reutilization of root cell wall P by enhancing the pectin content. NaCl modulates pectin content, however, it is still unknown whether NaCl is also involved in the process of pectin regulated cell wall P remobilization in rice under P starved conditions. In this study, we found that 10 mmol/L NaCl increased the shoot and root biomasses under P deficiency to a remarkable extent, in company with the elevated shoot and root soluble P contents in rice. Further analysis indicated that exogenous NaCl enhanced the root cell wall P mobilization by increasing the pectin methylesterase activity and uronic acid content in pectin suggesting the involvement of NaCl in the process of cell wall P reutilization in P starved rice roots. Additionally, exogenous NaCl up-regulated the expression of P transporter OsPT6, which was induced by P deficiency, suggesting that NaCl also facilitated the P translocation prominently from root to shoot in P starved rice. Moreover, exogenous abscisic acid (ABA) can reverse the NaCl-mediated mitigation under P deficiency, indicating the involvement of ABA in the NaCl regulated root cell wall P reutilization. Taken together, our results demonstrated that NaCl can activate the reutilization of root cell wall P in P starved rice, which is dependent on the ABA accumulation pathway.
