Efficient Enzymatic Enrichment of High-purity Nervonic Acid from Malania oleifera Seed Oil

Ximei Yang,Sen Lin,Ying Chen,Wen Chen,Dongming Lan,Yonghua Wang

Nervonic acid (NA) is a monounsaturated fatty acid vital for brain health and is of emerging importance in various industrial applications, including therapeutics, food, and cosmetics. Given the growing demands of the food and pharmaceutical industries, there's a pressing need for high-purity NA. Previously, NA constituents in plant seed oils were chemically transformed into nervonic acid ethyl ester (NAEE) to facilitate extraction from seed oils. In this study, we present an enzymatic approach to convert NA constituents in Malania oleifera seed oil to NAEE. Combined with the utilization of the semi-preparative chromatography, we achieved a remarkable purity of 97.52% NAEE. Compared to conventional chemical preparations characterized by multiple steps, prolonged processing times, and low yields and purities, our enzymatic method stands out as a more efficient and advantageous alternative. On top of that, this innovative approach is environmentally friendly and circumvents health and safety issues associated with chemical processes.

Enrichment and purification of nervonic acid from Acer truncatum seed oil by combining vacuum distillation and low-temperature crystallization: Experiments and process modeling

Yingxi Gao,Tong Wei,Jie Wang,Yuming Tu,Zhiyong Zhou,Chencan Du,Zhongqi Ren

Nervonic acid (NA) is a long-chain monounsaturated fatty acid with significant potential for neural fiber repair. In this study, a mixed fatty acid methyl ester was synthesized as the raw material through saponification of Acer truncatum Bunge seed oil. Based on the differences in boiling points and relative volatilities of various components, a four-stage vacuum batch distillation process was employed to enrich the nervonic acid methyl ester (NAME). The effect of distillation process parameters on enrichment efficiency was investigated, including distillation temperature, operating pressure, and reflux ratio. The purity of NAME achieved as 91.20% under optimal conditions and the corresponding yield was 48.91%. To further increase the purity, a low-temperature crystallization process was adopted and a final purity of NAME was obtained as 97.56%. Simulation of the above four-stage batch distillation was conducted using Aspen Plus software, and a continuous distillation processes was further simulated to establish a theoretical basis for future industrial-scale production. The results of experiments and simulation demonstrate that the integrated process of vacuum distillation and low-temperature crystallization exhibits remarkable separation performances, providing robust guidance for the production of high-purity NA.

Lipidomic analysis of polar lipids in human breast milk using UPSFC-Q-TOF-MS: A study focused on ceramide and cholesterol

Chenyu Jiang,Xinghe Zhang,Jiahui Yu,Ling-zhi Cheong,Xingguo Wang,Wei Wei

Herein, polar lipids, including glycerophospholipid, lysophospholipid, cholesterol (CHOL), sphingomyelin (SM), and ceramide (Cer), in human breast milk were subjected to lipidomic analysis via UPSFC-Q-TOF-MS. A total of 11 lipid classes and 195 molecular species were identified and quantified in human breast milk samples ( n  = 126) from three lactational stages across three regions in China. CHOL and SM together account for more than 50 % of the total polar lipids. The total polar lipids, SM, and phosphatidylethanolamine (PE) were ∼ 54.5, ∼17.8, and ∼ 3.5 mg/100 mL, respectively, which both significantly ( P  < 0.05) increased from colostrum to later lactational stages. DHA is highly enriched (68 % of the total DHA) in PE molecules. SM and Cer contain high ratios of nervonic acid (24:1 n-9), ∼35 % and ∼ 23 %, respectively. General patterns exist in the variation of polar lipid molecular species in human breast milk across different regions throughout the lactational stages.

Enrichment of nervonic acid in Acer truncatum Bunge oil by combination of two-stage molecular distillation, one-stage urea complexation and five-stage solvent crystallization

Yingxi Gao,Jiayi Shi,Jie Wang,Fan Zhang,Shichao Tian,Zhiyong Zhou,Zhongqi Ren

Nervonic acid is the world's first and only potent substance that can repair damaged nerve fibers and promote nerve cell regeneration with high nutritional value. The wide variety of fatty acids in plant oils and fats with similar structures makes the large-scale separation and purification of high-purity nervonic acid very difficult. A new combined process of molecular distillation, urea inclusion and solvent crystallization was established to prepare high-purity nervonic acid with the mixed fatty acids obtained after saponification and acidification of Acer truncatum Bunge oil as raw materials. First, according to the difference in the mean free path of fatty acids, molecular distillation was used to separate and remove C16 saturated fatty acid of palmitic acid and four C18–C20 fatty acids of stearic, oleic, linoleic, and linolenic acids. The content of C16–C20 fatty acids decreased from 72.92% to 19.22% after two-stage molecular distillation processes, in which the contents of saturated fatty acid of palmitic acid decreased to about 0.5%. Then, according to the difference in carbon chain length and saturation of fatty acid, the contents of C22–C24 saturated fatty acids of tetracosanoic and docosanoic acids decreased to 0.21% and 0.07% by urea inclusion with urea/free fatty acid preparation by saponification (SPOMFs) ratio as 0.6. In addition, all saturated fatty acids were basically separated. Finally, according to the difference in the solubility of fatty acids in solvents, the C18–C20 unsaturated fatty acids of oleic, linoleic, and linolenic acids and C22 unsaturated fatty acid of erucic acid were removed by solvent crystallization. The content of C18–C20 unsaturated fatty acids decreased to less than 5% with pentanol as the solvent after the first stage solvent crystallization. The content of erucic acid decreased to 3.47% with anhydrous ethanol as the solvent after the second to fifth stage solvent crystallization. The combined process of molecular distillation, urea inclusion and low temperature crystallization innovatively adopted an efficient, simple and easy-to-industrial solvent crystallization method to separate erucic and nervonic acids, obtaining nervonic acid with purity of 96.53% and final yield of 47.99%.

Study on the Extraction of Nervonic Acid from the Oil of Xanthoceras sorbifolium Bunge Seeds

Hui Gao,Jie Sun,Xuan Guo,Ziyan Zhang,He Liu,Zhiran Zhang,Mengkai Liu,Sen Zhou,Shengxin Li,Tingting Zhang

Seven fatty acids were detected by GC-MS inXanthoceras sorbifoliumBunge seed oil extracted at different temperatures, including Palmitic acid C16:0, Stearic acid C18:0, Oleic acid C18:1, Eicosenoic acid C20:1, Docosenoic acid C22:1, Tetracosenoic acid C24:1, and Linoleic acid C18:2. The highest content of nervonic acid (NA) was found inXanthoceras sorbifoliumBunge seed oil extracted at 70 °C. Three methods were selected to analyze the extraction rate of nervonic acid inXanthoceras sorbifoliumBunge seed oil, including urea complexation, low-temperature solvent crystallization, and a combined treatment using these two methods. The final content of nervonic acid obtained was 14.07%, 19.66%, and 40.17%, respectively. The combined treatment method increased the purity of nervonic acid inXanthoceras sorbifoliumBunge seed oil by 12.62 times. Meanwhile, thermogravimetric behavior analysis of samples extracted using different methods was conducted by thermogravimetric analyzer, which suggested that the thermal stability of the samples extracted by the combined treatment was enhanced. These results can provide a new process parameter and scientific basis for the extraction of NA. At the same time, FTIR and NMR were also used to characterize the combined extraction sample, and the structure of the samples was proved.

Evaluation of the encapsulation capacity of nervous acid in nanoemulsions obtained with natural and ethoxylated surfactants

Yu Jin,Feng Li,Xinman Lou,Yaqing Xiao,Xiaojing Wang,Fuguo Liu,Jun Wang,Huaide Xu

Nervonic acid (NA) has considerable potential for promoting brain development, but its utilization is limited attributed to its poor water solubility. Novel NA-loaded nanoemulsions (NA-NEs) were fabricated with planted-based soybean protein isolates (SPI), compared with whey protein isolates and Tween 20. The impact of emulsifiers on NA-NEs properties and stabilities was evaluated. Emulsions formulated with 0.5 mg/mL nervonic acid and 1.5 wt% SPI under pressure (120 MPa, 5 cycles) contained nano-scale droplets and considerable rheologic properties. The oil droplets were spherical and encapsulated within the emulsions, evidenced by atomic force microscope and confocal laser scanning microscopy. SPI-emulsions were stable under thermal treatment, NaCl concentrations, storage at 4 °C (30 days), and multiple scattering experimental conditions. SPI behaved similarly to the other two emulsifiers under the studied conditions. These findings demonstrate that SPI is a promising plant-based emulsifier for applications in nervonic acid-fortified food.

Optimization of ultrasonic circulating extraction of samara oil from Acer saccharum using combination of Plackett–Burman design and Box–Behnken design

Fengli Chen,Qiang Zhang,Shimin Fei,Huiyan Gu,Lei Yang

In this study, ultrasonic circulating extraction (UCE) technique was firstly and successfully applied for extraction of samara oil from Acer saccharum . The extraction kinetics were fitted and described, and the extraction mechanism was discussed. Through comparison, n -hexane was selected as the extraction solvent, the influence of solvent type on the responses was detailedly interpreted based on the influence of their properties on the occurrence and intensity of cavitation. Seven parameters potentially influencing the extraction yield of samara oil and content of nervonic acid, including ultrasound irradiation time, ultrasound irradiation power, ultrasound temperature, liquid-solid ratio, soaking time, particle size and stirring rate, were screened through Plackett–Burman design to determine the significant variables. Then, three parameters performed statistically significant, including liquid-solid ratio, ultrasound irradiation time and ultrasound irradiation power, were further optimized using Box–Behnken design to predict optimum extraction conditions. Satisfactory yield of samara oil (11.72   ±   0.38%) and content of nervonic acid (5.28   ±   0.18%) were achieved using the optimal conditions. 1% proportion of ethanol in extraction solvent, 120   °C of drying temperature and 6.4% moisture were selected and applied for effective extraction. There were no distinct differences in the physicochemical properties of samara oil obtained by UCE and Soxhlet extraction, and the samara oil obtained by UCE exhibited better antioxidant activities. Therefore, UCE method has enormous potential for efficient extraction of edible oil with high quality from plant materials.

Su Ji-Ling,Wang Juan,Fu Xiao-Na,Hou Li,Zhu Ping,Zhang Kai,Li Huo-Wang,Hou Ying,Liu Xiang-Yi,Xu Juan

This study explored the physicochemical properties, fatty acid compositions, and thermal characteristics of Malania oleifera seed oil (MOSO). Our data showed that the refractive index and specific gravity of MOSO at ambient temperature were 1.461 and 0.913, respectively. The oil content, acid, peroxide, saponification, iodine value, average molecular weight, and an average number of carbon–carbon double bond per triglyceride were 47.8%, 3.62 mg KOH/g oil, 4.61 meq O 2 /kg oil, 163.16 mg KOH/g oil, 104.28 g I 2 /100 g oil, 1031.51 g/mol, and 4.24, respectively. The fatty acid profiles of the methyl esters showed the presence of 96.09% unsaturated fatty acids and 2.16% saturated fatty acids. Nervonic acid (NA) was the major component (45.04%). These results suggested that MOSO is an excellent source of essential fatty acids and NA and may be used as a potential NA resource of health supplements and medicines. Graphical abstract

Interesterified trans-free fats rich in sn-2 nervonic acid prepared using Acer truncatum oil, palm stearin and palm kernel oil, and their physicochemical properties

Peng Hu,Xuebing Xu,Liangli Lucy Yu

This study obtained trans -free fats containing nervonic acid, a fatty acid with beneficial effects for mental health. Four fats containing 1.28–2.41% (w/w) nervonic acid were prepared by chemical or enzymatic interesterification of palm stearin (PS), Acer truncatum oil (ATO) and palm kernel oil (PKO). The chemical procedure was catalyzed using MeONa, whereas the enzymatic procedure was catalyzed with a 1,3–specific lipase. As a result, the interesterified fats showed a similar fatty acid profile to their corresponding physical blends measured by gas chromatography , but contained more nervonic acids at the sn -2 position, which was more conducive to metabolism and absorption. The content of tripalmitoylglycerol (PPP) and 1,3-dipalmitoyl-2-oleoyl glycerol (POP) or 1,2-dipalmitoyl-3-oleoyl glycerol (PPO) in the interesterified fats decreased significantly, resulting in improved plasticities. All interesterified fats showed a satisfactory content of tocopherols and oxidative stability. A combination of differential scanning calorimetry , X-ray diffraction spectroscopy and polarized light microscopy results suggested that the interesterified fats formed spherulitic needle-like crystals in the β′ form, while the physical blends formed dendritic crystals in the β form. These data indicated that the trans -free interesterified fats rich in nervonic acid may have a great potential in margarine applications.

Acer truncatum Bunge seed oil ameliorated oxaliplatin-induced demyelination by improving mitochondrial dysfunction via Pink1/Parkin mitophagy pathway

Dekun Cheng,Zhangtie Wang,Xin Guo,Yiwen Guo,Yu Zhang,Yuanhui Zhao,Ruijie Liu,Ming Chang

Dietary nutritional support for special populations is an effective and feasible method to improve quality of life of patients and reduce medical pressure. The effects of ATSO on chemotherapy population were evaluated by a zebrafish model of demyelination. The results showed that 100 μg/mL ATSO reversed tail morphology damage, axon degeneration, touch response delay, ROS level upregulation and the expression of myelin basic protein decrease in chemotherapy-induced zebrafish. In addition, the expression of myelin markers (including sox10, krox20, pmp22) in oxaliplatin-induced cells was markedly reversed by ATSO and its active components (gondoic acid, erucic acid, nervonic acid). ATSO and its active components could reverse demyelination by ameliorating mitochondrial dysfunction. Conversely, linoleic acid and linolenic acid promoted demyelination by exacerbating mitochondrial dysfunction. Moreover, the Pink1/Parkin pathway was recognized as the main reason for ATSO and its active components to improve mitochondrial function by activating mitophagy and restoring autophagic flow. Taken together, this study demonstrated that ATSO and its active components could be further developed as novel functional food ingredients to antagonize demyelination.

Improvement of stability and in vitro bioaccessibility of nervonic acid by nonionic surfactant in protein-based nanoemulsions

Yu Jin,Bin Shu,Xinman Lou,Kunhua Wang,Yunhui Zhai,Yingjuan Qu,Ruijuan Song,Fuguo Liu,Xiaobo Dong,Huaide Xu

Nervonic acid (NA, ω-9) is beneficial for brain health, but its poor water solubility limits its bioaccessibility. The protein-based emulsions were unstable under environmental stress. In this study, the effects of a nonionic surfactant (Tween 20, T20) on the characteristics, stability, and in vitro digestion properties of NA-loaded nanoemulsions prepared with proteins (whey or soybean protein isolate) were evaluated. The emulsions stabilized by protein-T20 mixtures had smaller droplets with more even distributions, more negative charges, and higher encapsulation efficiencies for NA. The addition of T20 improved the interfacial properties of the systems, as confirmed by spectral and rheological analyses. Emulsion stability under various conditions was enhanced by the improved interfacial composition and emulsifying ability. In particular, the effective interfacial layers produced by the protein-T20 mixture decreased lipid digestion but increased NA bioaccessibility. Therefore, the co-adsorption of proteins with suitable nonionic surfactants at oil-water interfaces is a promising approach for designing functional food delivery systems with improved stability and NA bioaccessibility.

Dietary Supplementation with Nervonic Acid Ameliorates Cerebral Ischemia-Reperfusion Injury by Modulating of Gut Microbiota Composition-Fecal Metabolites Interaction

Hui Zhou,Ziyi Chen,Jingbin Li,Rongjin Wang,Hongshi Bu,Chengjiang Ruan

Scope Cerebral ischemia-reperfusion (IR) injury stands as a prominent global contributor to disability and mortality. Nervonic acid (NA), a bioactive elongated monounsaturated fatty acid, holds pivotal significance in human physiological well-being. This research aims to explore the prophylactic effects and fundamental mechanisms of NA in a rat model of cerebral IR injury. Methods and results Through the induction of middle cerebral artery occlusion, this study establishes a rat model of cerebral IR injury and comprehensively assesses the pharmacodynamic impacts of NA pretreatment. This evaluation involves behavioral analyses, histopathological examinations, and quantification of serum markers. Detailed mechanisms of nervonic acid's prophylactic effects are revealed through fecal metabolomics and 16S rRNA sequencing analyses. Our findings robustly support nervonic acid's capacity to ameliorate neurological impairments in rats afflicted with cerebral IR injury. Beyond its neurological benefits, NA demonstrates its potential by rectifying metabolic perturbations across diverse pathways, particularly those pertinent to unsaturated fatty acid metabolism. Additionally, NA emerges as a modulator of gut microbiota composition, notably by selectively enhancing vital genera like Lactobacillus . Conclusion These comprehensive findings highlight the potential of incorporating NA as a functional component in dietary interventions aimed at targeting cerebral IR injury.
