In this work, an anti-icing coating based on superhydrophilic polyzwitterion brushes is reported. Poly(sulfobetaine methacrylate) (PSBMA) brushes were synthesized by surface-initiated atom-transferred radical polymerization to create superhydrophilic films on silicon wafers. The thickness of the PSBMA brushes film increased linearly with the polymerization time, and the film remained superhydrophilic in a nonassociated state when the thickness was less than ∼100 nm. DSC and FTIR analyses revealed that PSBMA contains more nonfreezable bound water than typical polyelectrolytes such as poly(sodium styrenesulfonate) and poly(2-(methacryloyloxy)ethyltrimethylammonium chloride), leading to lower ice adhesion strength than the latter two. At −20 °C, the PSBMA brushes coating demonstrated low ice adhesion strength of 60 kPa, showing a significant reduction in ice adhesion by up to ∼75% compared to uncoated silicon wafer. The optimum PSBMA layer thickness for low ice adhesion was ∼100 nm. These findings suggest that polyzwitterions are excellent candidates for anti-icing coating application.

Diseases and complications related to catheter materials are severe problems in biomedical material applications, increasing the infection risk and medical expenses. Therefore, there is an enormous demand for catheter materials with antibacterial and antifouling properties. Considering this, in this work, we developed an approach of constructing antibacterial surfaces on polyurethane (PU) via surface-initiated atom transfer radical polymerization (SI-ATRP). A variety of cationic polymers were grafted on PU. The biocompatibility and antifouling properties of all resulting materials were evaluated and compared. We also used a theoretical algorithm to investigate the anticoagulant mechanism of our PU-based grafts. The hemocompatibility and anti-biofouling performance improved at a 86–112 μg/cm2grafting density. The theoretical simulation demonstrated that the in vivo anti-fouling performance and optimal biocompatibility of our PU-based materials could be achieved at a 20% grafting degree. We also discuss the mechanism responsible for the hemocompatibility of the cationic brushes fabricated in this work. The results reported in this paper provide insights and novel ideas on material design for applications related to medical catheters. Keywords:polyurethane;cationic brushes;graft density;hemocompatibility;anti-biofouling;catheter

The addition of nanoparticles to water-based lubricants is a commonly used method to improve lubrication, but to the best of our knowledge few studies have been reported to investigate the lubrication property of surface-modified nanodiamonds (ND) with polyzwitterionic brushes. In this study, a bioinspired copolymer containing dopamine and 2-methacryloyloxyethyl phosphorylcholine (MPC) was synthesized (DMA-MPC) and then spontaneously grafted onto the ND surface (ND-MPC) through simple stirring in order to enhance lubrication. The characterization of transmission electron microscopy, Fourier transform infrared spectroscopy, and thermogravimetric analysis indicated that the DMA-MPC was successfully modified on the ND surface. Furthermore, a series of tribological experiment were performed on a universal materials tester using glycerol, glycerol + ND, and glycerol + ND-MPC as the lubricants. It was found that the addition of ND to the lubricant (i.e., glycerol + ND and glycerol + ND-MPC) significantly reduced wear with a smaller wear scar and wear track on the tribopairs, and the coefficient of friction further decreased by about 40% when using glycerol + ND-MPC as the lubricant, which could be attributed to the hydration lubrication of the polyzwitterionic brushes modified on the ND surface and the rolling effect of nanoparticles. In conclusion, in this study a universal and versatile surface modification method was proposed on the basis of the synthesis of bioinspired copolymer DMA-MPC, which remarkably enhanced the lubrication property of ND nanoparticles when added to water-based lubricants.

Smart gene delivery vectors are gaining increasing attention in gene therapy, owing to their low cytotoxicity and intrinsic responsiveness. Our previously fabricated novel cationic brush polymer, comprising C Se bonds and tertiary amine EGIn- g -PDMAEMA, shows potential for gene transfection. In this study, its high efficiency for siRNA/pDNA transfection and low cytotoxicity in reactive oxygen species (ROS)-rich microenvironments is substantiated in vitro. Its superior binding capacity with siRNA/pDNA is confirmed by agarose gel electrophoresis assay. The threshold weight ratios for siRNA/pDNA migration delay are 15 and 3 (polymer-to-nucleic acid, w/w), respectively. Fluorescence microscopy and ribonucleotide reductase regulatory subunit M2 gene silencing essay verify the biodegradability and responsive control release of nucleic acids under hydrogen peroxide stimulation in Huh-7 cells. Compared with the gold standard, polyethylenimine 25 kDa, the target polymer displays superior transfection efficiency in ROS-rich tumor cells under serum-free conditions. Furthermore, the vector–nucleic acid complexes exhibit over 90% cell viability at a high concentration of 12 μg mL −1 and good colloidal stability in phosphate-buffered saline (PBS) and 10% fetal bovine serum-PBS for 24 h. The efficient control release and expression of nucleic acids in ROS environments and reduced cytotoxicity highlight the superiority of EGIn- g -PDMAEMA as a gene delivery platform for tumor gene therapy.

The conformation of polyzwitterionic brushes plays a crucial role in the adsorption/desorption of proteins on solid surfaces. By use of quartz crystal microbalance with dissipation (QCM-D) and surface plasmon resonance (SPR), we have systematically investigated the conformational behavior of poly(sulfobetaine methacrylate) (PSBMA) brushes as a function of ionic strength in the presence of different ions. The frequency change demonstrates that the effectiveness of anions to weaken the inter/intrachain association and to enhance the hydration of the grafted chains increases from kosmotrope to chaotrope in the low ionic strength regime, but the ordering of anions is almost reversed at the high ionic strengths. The dissipation change indicates that some heterogeneous structures are formed inside the brushes in the presence of chaotropic anions with the increase of ionic strength. In SPR studies, the change of resonance unit (ΔRU) with ionic strength is determined by the balance between the increase of thickness and the decrease of refractive index of the brushes. No anion specificity is observed in the SPR measurements because ΔRU is insensitive to the coupled water molecules inside the brushes. For the control of protein adsorption/desorption, our studies show that the brushes can more effectively resist the protein adsorption in the presence of a more chaotropic anion and a more chaotropic anion can also more effectively induce the protein desorption from the surface of the brushes. In addition, no obvious cation specificity can be observed in the conformational change of the brushes in either QCM-D or SPR measurements.
