In this study, a polymeric ionic liquid (PIL) called Poly[ECH-BIM]Cl was polymerized by ring-opening and quaternization reaction of benzimidazole and epichlorohydrin without additional ion-exchange reaction. Both 1 H NMR and fourier transform infrared (FTIR) confirmed the chemical structure of Poly[ECH-BIM]Cl. Poly[ECH-BIM]Cl in water was thermoresponsive and its UCST transition was studied by UV–vis spectrometry, dynamic light scattering (DLS) and scanning electron microscopy (SEM) at variable temperature. Moreover, the first introduced Temp-resistance behavior during ions pairing found Poly[ECH-BIM]Cl had properties similar to those of the precipitation of inorganic salts from their saturated solutions. The variable-temperature 1 H NMR analysis demonstrated that the tailored interchain π-π interactions promoted the aggregation of macromolecular chains and particles formed from Poly[ECH-BIM]Cl aqueous solution. The results showed that increasing the interaction between cations of PIL can endow its thermosensibility and the possibility of being a programmable shuttle.

In this work, we have investigated a pH-modulated complexation between two oppositely charged strong polyelectrolytes to demonstrate the effect of counterion-mediated hydrogen bonding (CMHB) on polyelectrolyte complexation. We have found that such a pH-modulated complexation cannot be understood without considering the CMHB. Thermodynamically, the effect of CMHB on the polyelectrolyte complexation is manifested by the alteration of both enthalpic and entropic contributions to the free energy change. The pH-dependent intrinsic ion-pairing and complex coacervation processes of the polyelectrolyte complexation can be understood when considering the CMHB. Our study demonstrates that both the extent of polyelectrolyte complex formation in bulk solutions and the formation of polyelectrolyte multilayers on surfaces are controlled by the pH-dependent intrinsic ion-pairing process. Furthermore, on the basis of the pH-dependent intrinsic ion pairing, the properties of the multilayers can be tuned by pH. This work provides a new strategy to control the polyelectrolyte complexation with counterions and will inspire new ideas for building advanced polyelectrolyte materials.

Local analgesia is one of the most desirable methods for postoperative pain control, while the existing local anesthetics have a short duration of analgesic effect. Nano-drug carriers have been widely used in various fields and provide an excellent strategy for traditional drugs. Although the existing liposomes for local anesthetics have certain advantages, their instability and complexity of the preparation process still cannot be ignored. Here, we developed novel ropivacaine hydrochloride liposomes with improved stability and sustained release performance by combining ropivacaine hydrochloride with sodium oleate in liposomes via hydrophobic ion-pairing (HIP). The liposomes are easy to prepare, inexpensive, and suitable for mass production. The infrared (IR), particle size, and Zeta potential measurements adequately characterized the complex, which showed a diameter of 81.09 nm and a zeta potential of −83.3 mV. Animal behavioral experiments, including the hot plate test and von Frey fiber test, demonstrated that the liposome system had a prolonged analgesic effect of 2 h versus conventional liposome preparations, consistent with the results of in vitro release experiments. In addition, in vitro cytotoxicity evaluations in RAW264.7 cells and in vivo evaluations revealed the biocompatibility and safety of the ropivacaine-sodium oleate ion-paired liposome (Rop-Ole-Lipo) system as a suitable local anesthetic for local pain management. Our findings provide a new idea for the preparation of local anesthetics.

Polyelectrolyte-doped microcapsules (PDM) was fabricated by coaxial electrospray of a mixture of glycerol and water containing 10 mg/mL cationic polyelectrolyte poly(allylamine hydrochloride) (PAH) fed as the core phase solution, and a N,N-dimethylacetylamide solution of 10 wt% polyurethane fed as the shell phase solution. Multi-enzyme system involving Candida Antarctica lipase B (CALB), glucose oxidase (GOD), and horseradish peroxidase (HRP) for cascade reaction was assembled in the PDM at three different places, namely, surface, shell, and lumen. Placing of enzyme inside aqueous lumen of the PDM was realized by in situ encapsulation through adding the enzyme in the core-phase solution for coaxial electrospray. By ion-pairing of enzyme with cationic surfactant CTAB, an organic soluble enzyme-CTAB complex was prepared, so that in situ embedding of enzyme in the shell of the PDM was realized by adding it into the shell phase solution. Surface attachment of enzymes was achieved by layer-by-layer (LbL) technology, which is based on the ion-exchange interactions between oppositely charged enzymes and PAH that was doped in PDM. The enzyme-decorated microcapsule was then studied as a micro-bioreactor, in which 1-Oxododecyla-α-glucopyranoside was converted by CALB to glucose, which was oxidised by GOD to gluconolactone in a second step. The hydrogen peroxide produced was then used by HRP to oxidize ABTS to form coloured radical cation ABTS • + for activity analysis. The successful fabrication of the PDM and precise localization of enzymes in the PDM by different strategies were fully characterized. By varying the immobilization strategy, totally six PDM bioreactors with three enzymes precisely positional assembled in different strategies were constructed and their activities for the cascade reaction were investigated and compared. The PDM micro-bioreactor prepared by novel electrospray technologies provide a smart platform for positional assembly of multi-enzyme cascade reaction in a precise and well-controlled manner.
