A novel asymmetrical arc-shaped electrode ion trap for improving the performance of a miniature mass spectrometer

Zhong-yao Zhang,Cuiping Li,Chuan-fan Ding,Fuxing Xu,Baoqiang Li,Qibin Huang,Hengxin Xia

RATIONALE The pivotal challenge associated with miniature mass analyzers is their proper design and construction without sacrificing performance. In order to analyze and improve the performance of a miniature linear ion trap with odd and even multipole fields, we designed a novel asymmetrical arc-shaped electrode ion trap (AAEIT), and tested the properties of AAEITs with different dimensions. METHODS A series of asymmetrical ion traps using arc-shaped electrodes were designed to optimize the properties (resolutions and intensity) of the coupling effects between odd and even multipole fields. Using arginine and reserpine, we evaluated the performance of mass resolution, ion intensity ratio and deduced the collision-induced dissociation (CID) efficiency using a self-constructed electrospray ionization mass spectrometry (ESI-MS) platform. RESULTS An AAEIT with field radius dimensions of 5 mm × 5.75 mm exhibits a good performance: its maximum resolution of 833 (FWHM) at m/z 175 was achieved for the side of small electrode. With this AAEIT, a tandem mass (MS/MS) capability with 91.0% CID efficiency was obtained with reserpine ( m/z 609). CONCLUSIONS The results indicated that the AAEIT, comprising both odd and even multipole fields, could act as a qualified linear ion trap mass analyzer with compact structure, high resolution, and high tandem mass analysis efficiency. It has a great potential in miniature mass spectrometry. Copyright © 2014 John Wiley & Sons, Ltd.

SAM–SFM: High-Efficiency and High-Resolution Tandem Mass Spectrometry Enabled by Sinusoidal Amplitude Modulation of Multiple Sinusoidal Frequency-Modulated Waveforms

Ang Li,Jian Sun,Haoqiang Yan,Dayu Li,Wei Xu

In miniature ion trap mass spectrometry, achieving a balance between isolation resolution and efficiency is a formidable challenge. The presence of absorption curves causes target ions to inadvertently absorb energy from AC signal components near their resonant frequencies. To mitigate this issue, SAM–SFM waveforms introduce a parameter known as the decreasing factor. Unlike SWIFT waveforms, SAM–SFM’s spectral profile intentionally departs from a rectangular window, adopting an arch-shaped excitation window to minimize the impact on target ions and improve ion isolation efficiency. SAM–SFM waveforms have the advantage of low computational complexity, enabling real-time computation using an embedded FPGA technology. Regardless of any parameter changes, the FPGA can consistently guarantee waveform output within 1 μs. This not only enhances throughput but also eliminates the need for a PC in miniature mass spectrometry devices. The performance of SAM–SFM has been validated on an improved “Brick” miniature ion trap mass spectrometer. Comparative experiments with SWIFT waveforms confirm the lossless unit-mass isolation capabilities of SAM–SFM. This waveform has the capability to simultaneously isolate multiple target ions, even allowing for the lossless isolation of ions with lower abundance within isotopic clusters, albeit at the cost of requiring extended isolation durations.

Novel control modes to improve the performance of rectilinear ion trap mass spectrometer with dual pressure chambers

Huo Xinming,Tang Fei,Zhang Xiaohua,Chen Jin,Zhang Yan,Guo Cheng’an,Wang Xiaohao

The rectilinear ion trap (RIT) has gradually become one of the preferred mass analyzers for portable mass spectrometers because of its simple configuration. In order to enhance the performance, including sensitivity, quantitation capability, throughput, and resolution, a novel RIT mass spectrometer with dual pressure chambers was designed and characterized. The studied system constituted a quadrupole linear ion trap (QLIT) in the first chamber and a RIT in the second chamber. Two control modes are hereby proposed: Storage Quadrupole Linear Ion Trap-Rectilinear Ion Trap (SQLIT-RIT) mode, in which the QLIT was used at high pressure for ion storage and isolation, and the RIT was used for analysis; and Analysis Quadrupole Linear Ion Trap-Rectilinear Ion Trap (AQLIT-RIT) mode, in which the QLIT was used for ion storage and cooling. Subsequently, synchronous scanning and analysis were carried out by QLIT and RIT. In SQLIT-RIT mode, signal intensity was improved by a factor of 30; the limit of quantitation was reduced more than tenfold to 50 ng mL −1 , and an optimal duty cycle of 96.4% was achieved. In AQLIT-RIT mode, the number of ions coexisting in the RIT was reduced, which weakened the space-charge effect and reduced the mass shift. Furthermore, the mass resolution was enhanced by a factor of 3. The results indicate that the novel control modes achieve satisfactory performance without adding any system complexity, which provides a viable pathway to guarantee good analytical performance in miniaturization of the mass spectrometer.

Development and Investigation of a Mesh-Electrode Linear Ion Trap (ME-LIT) Mass Analyzer

Liang Wang,Fuxing Xu,Xinhua Dai,Xiang Fang,Chuan-Fan Ding

A mesh-electrode linear ion trap (ME-LIT) mass analyzer was developed and its performance was primarily characterized. In conventional linear ion trap mass analyzers, the trapped ions are mass-selected and then ejected in a radial direction by a slot on a trap electrode. The presence of slots can strongly affect the electric field distribution in the ion trapping region and distort the mass analysis performance. To compensate for detrimental electric field effects, the slot is usually designed and fabricated to be as small as possible, and also has very high mechanical accuracy and symmetry. A ME-LIT with several mesh electrodes was built to compensate for the effects caused by slots. Each mesh electrode was fabricated from a plate electrode with a relatively large slot and the slot was covered with a conductive mesh. Our preliminary experimental results show that the ME-LIT could considerably diminish the detrimental electric field effects caused by slots, and increase the mass resolving power and ion detection efficiency. Even with 4-mm-wide slots, a mass resolution in excess of 600 was obtained using the ME-LIT. Mass resolution could be remarkably improved using mesh electrodes in ion traps with asymmetric electrodes. The stability diagram of the ME-LIT was mapped, and highly efficient tandem mass spectrometry was demonstrated. The ME-LIT was qualified as a LIT mass analyzer. The ME-LIT can improve the mass resolution and decrease the requirements of mechanical accuracy and symmetry of slots, so it shows potential for a wide range of practical uses.

An Ultra-High-Resolution Mass Spectrometer with an Intermediate-Pressure Matrix-Assisted Laser Desorption/Ionization Source

Zhengge Chen,Yi Hong,Futao Lin,Xiongfei Zha,Fengcheng Lin,Yang Li,Bin Hu,Qi Huang,Lei Li,Zhengxu Huang,Li Ding,Zhen Zhou

The demand for high-resolution, high-throughput and spatially resolved mass analysis has been a key focus in developing mass spectrometers. We have developed a novel high-performance mass spectrometer by integrating matrix-assisted laser desorption/ionization (MALDI) source with a new planar electrostatic ion trap mass analyzer for ultrahigh-resolution MALDI source mass analysis. Performance characterization experiments show that with a transient time of 800 ms, the resolution for 392 Th can exceed 530 K by utilizing high-order harmonics, and the spectrometer can distinguish the isotopic peaks in the Met-Arg-Phe-Ala acetate salt (MRFA) M+2 peak family. Preliminary results also show an overall mass range from 133 to 9500 Th by stitching together multiple single scans with at least 6-fold mass range, and a low detection limit of 32 fg. Initial tests on analysis of edible oils demonstrate its capability to distinguish plant and animal oils based on triacylglycerols.

Discontinuous Atmospheric Pressure Interface in-Trap Single Photon Ionization Ion Trap Mass Spectrometry (DAPI in-Trap SPI ITMS)

Tianhan Zhang,Chengyi Xie,Yong Wu,Wenqing Gao,Chenlu Wang,Keqi Tang,Jiancheng Yu

A compact in-trap single photon ionization (SPI) system, termed discontinuous atmospheric pressure interface (DAPI) ion trap mass spectrometry (ITMS), for the rapid determination of trace volatile organic compounds (VOCs). DAPI enables the maintenance of high analyte pressure in the SPI source while preserving the vacuum. Utilizing vacuum ultraviolet (VUV) lamp as the ion source, we ionize the analyte within an enclosed miniature 3D ion trap for efficient analysis. Experimental findings demonstrate that the system achieves a mass resolution of less than 0.5 atomic mass units for full width at half maxima (FWHM), providing unit resolution. For the response for benzene, the system achieves almost instantaneous response, with rise and fall times of only 2 s, which is a significant advantage compared to the membrane inlet (MI) approach necessary for on-site detection. Additionally, tandem mass spectrometry (MS/MS) with unit m/z isolation window was achieved and resolved dimethyl disulfide and phenol with similar molecular weights. Our results demonstrate that the system provides excellent performance and we anticipate its application in rapidly determining VOCs.

Rapid Mass Analysis of Reserpine and Arginine by Ion Trap Mass Spectrometer

Fu-Xing XU,Qian-Kun DANG,Yin-Juan CHEN,Kai YANG,Qiang WANG,Bin CHEN,Yuan-Yuan WANG,Chuan-Fan DING

A fast mass analysis method using digital ion trap technology was reported. The mass analysis was realized by scanning the frequency of digital RF voltage and dipole ion resonance voltage during ion injection period without the conventional ion cooling, ion resonance ejection or ion cleaning process. It can decrease about three fourth of traditional analysis time using ion trap mass spectrometry. The ion signal was optimized through the modification of frequency scanning rate, ion gate voltage and other experimental parameters. For example, an optimal mass spectroscopy of reserpine ( m / z 609) and arginine ( m / z 174) were obtained when the ion gate voltage was 9 V and the digital RF frequency was scanned from 1 MHz to 400 kHz at rate of 2385 Th s −1 . The results indicated that the obtained mass spectra were in very good agreement with conventional analytical method using ion trap mass analyzer.

Ion Mobility Separation Using a Dual-LIT Miniature Mass Spectrometer

Jingjin Fan,Penglong Lian,Ming Li,Xinwei Liu,Xiaoyu Zhou,Zheng Ouyang

Ion mobility (IM) has been increasingly used in combination with mass spectrometry (MS) for chemical and biological analysis. While implementation of IM with MS usually requires complex instrumentation with delicate controls, in this study we explored the potential of performing IM separation using dual-linear ion traps (LITs) in a miniature mass spectrometer, which was originally developed for performing comprehensive MS/MS scan functions with a simple instrumentation configuration. The IM separation was achieved by ion transfer between the LITs with dynamic gas flow. Its performance was characterized for analysis of a broad range of chemical and biological compounds including small organic compounds such as trisaccharides, raffinose, cellotriose, and melezitose, as well as protein conformers. The demonstrated technique serves as another example of developing powerful hybrid instrument functions with simple configurations and miniaturized sizes.

Preparation and characterization of partial de-O-sulfation of heparin oligosaccharide library

Jianghui Lin,Liyang Zheng,Quntao Liang,Lilong Jiang,Zheng Wei

The O -sulfation, including 2- O - and 6- O -sulfation, in heparan sulfate (HS) have important biological and pathophysiological roles. Therefore, the ability to chemically generate a series of oligosaccharides , which have a similar structure to the naturally-occurring, 2- O - and 6- O -sulfating oligosaccharides from HS, would greatly contribute to investigating their natural role in HS. In this study, a heparin oligosaccharide library, including dp2, dp4 and dp6, were prepared from the chemical modification of the fully sulfated dp2, dp4 and dp6. Chemical reaction conditions were optimized to generate different patterns of 2- O - and 6- O -sulfated oligosaccharides, then the disaccharide composition and structure of the library was detected by high-performance liquid chromatography-ion trap/time-of-flight mass spectrometry (LC-IT-TOF/MS) analysis. This provides a foundation for further structural and functional studies of O -sulfated groups in HS.

Characteristics of Ion Activation and Collision Induced Dissociation Using Digital Ion Trap Technology

Fuxing Xu,Qiankun Dang,Xinhua Dai,Xiang Fang,Yuanyuan Wang,Li Ding,Chuan-Fan Ding

Collision induced dissociation (CID) is one of the most established techniques for tandem mass spectrometry analysis. The CID of mass selected ion could be realized by ion resonance excitation with a digital rectangular waveform. The method is simple, and highly efficient CID result could be obtained by optimizing the experimental parameters, such as digital waveform voltage, frequency, and q value. In this work, the relationship between ion trapping waveform voltage and frequency at preselected q value, the relationship between waveform frequency and the q value at certain ion trapping voltage for optimum CID efficiency were investigated. Experiment results showed that the max CID efficiency of precursor reserpine ions can be obtained at different trapping waveform voltage and frequency when q and β are different. Based on systematic experimental analysis, the optimum experimental conditions for high CID efficiency can be calculated at any selected β or q. By using digital ion trap technology, the CID process and efficient fragmentation of parent ions can be realized by simply changing the trapping waveform amplitude, frequency, and the β values in the digital ion trap mass spectrometry. The technology and method are simple. It has potential use in ion trap mass spectrometry.

Printed Circuit Board Ion Trap Mass Analyzer: Its Structure and Performance

Dan Jiang,Gong-Yu Jiang,Xiao-Xu Li,Fu-xing Xu,Liang Wang,Li Ding,Chuan-Fan Ding

An ion trap (IT) mass analyzer can be simply built with low cost material—the printed circuit board (PCB). A printed circuit board ion trap (PCBIT) can perform ion trapping, mass analysis, and tandem mass spectrometry as a conventional ion trap mass analyzer. In a PCBIT, each PCB electrode was fabricated to specially designed patterns with several separate electric strips. The strips’ electrodes were insulated from each other and applied with different voltages during the experiment. Therefore, the electric field distribution inside the ion trap region may be adjusted and optimized by simply adjusting the voltage on each strip. The performance of the PCBIT can also be optimized since the property of an ion trap is strongly dependent on the field distribution. The fabrication, operation, and performance of the PCBIT are described and characterized in this paper. A prototype PCBIT was built with two pairs of 64 mm × 12 mm PCB rectangular plates and one pair of 10 mm × 10 mm stainless steel square plates. A mass analysis with a resolving power of over 1500 and a mass range of around 3000 Th was observed. The mass-selected isolation and collision-induced dissociation (CID) of ions were also tested using the homemade PCBIT system. The adjustable electric field distribution, simple structure, and low cost of PCBIT make it certainly suitable for the further miniaturization of the portable mass spectrometer.

Characterization of the impact of the ejection slit on miniature rectilinear ion trap analysis

Xinming Huo,Fei Tang,Jin Chen,Xiaohua Zhang,Xiaohao Wang

The rectilinear ion trap (RIT) has always been one of the ideal candidates for mass analyzer miniaturization because of its simple configuration. In this work, the effect of the ejection-slit dimensions on the ion ejection efficiency of a miniature rectilinear ion trap (mRIT) with full-scale dimensions of 5   mm   ×   4   mm   ×   25   mm was investigated by using SIMION8.1 simulation. Six mRITs with the same dimensions as those simulated and with different slit widths, whose X electrodes were fabricated by the stainless steel chemical etching process, were tested and characterized on a homemade mass spectrometer platform. It is shown that the ejection slit dimensions have a significant impact on both the signal intensity and mass resolution. For an mRIT with these full-scale dimensions, the optimized slit dimensions are 0.6   mm   ×   18   mm. The optimized mRIT demonstrated capabilities including a mass/charge range of over 1000   Th, unit mass resolution at m / z 332, and a passable tandem mass spectrometry capability, which are better than the non-optimized RIT with the same dimensions.
