Structural characterization and antioxidant activities of polysaccharides from okra (Abelmoschus esculentus (L.) Moench) pericarp

Lu-Yao Ma,Rou Xu,Hai-Feng Lin,Ming-Yong Xie,Shao-Ping Nie,Jun-Yi Yin

In this study, okra ( Abelmoschus esculentus  (Linn.) Moench) pods (pericarp) obtained after removing seeds and flesh were applied for polysaccharide extraction. The 60 °C water extract polysaccharide (OPW-60) was obtained from okra peel at 60 °C, and then 100 °C water extract polysaccharide (OPW-100) was obtained from the residue at 100 °C. Acid extraction at 60 °C was finally used to obtain the polysaccharide by acid leaching at 60 °C (OPA-60). Then, structural characteristics and antioxidant activities investigations were performed on the extracted polysaccharides. The results showed that monosaccharide composition of three polysaccharides was different. OPW-60 was mainly type I rhamnogalacturonan (RG-I) pectin, while OPW-100 and OPA-60 consist of RG-I and homogalacturoan (HG) pectins. 1 H NMR analysis confirmed the existence of rhamnose and acetyl group among them. The structure diversity resulted in the difference of viscosity, molecular weight, surface morphology and antioxidant activities. OPW-60 exhibited the highest apparent viscosity, which might be attributed to the differences in molecular weight distribution and monosaccharide composition. OPW-60 was most effective in scavenging 2,2-Diphenyl-1-picrylhydrazyl radicals and chelating ferrous ion ability, while OPW-100 exhibited the highest hydroxyl radical scavenging activity. The results showed that polysaccharides with different structure and antioxidant ability could be obtained from okra under different extraction conditions.

Preparation, characterization and improvement in intestinal function of polysaccharide fractions from okra

Han Gao,Wencheng Zhang,Zeyu Wu,Haiyan Wang,Ailing Hui,Ling Meng,Pengpeng Chen,Zhaojun Xian,Yiwen He,Honghong Li,Biao Du,Huawei Zhang

The purpose of this study was to investigate the effect of okra polysaccharides with similar molecular weight on intestinal ecology. After extraction, ethanol precipitation, deproteinization and purification, two polysaccharide fractions, named as water-extracted polysaccharides (WEP) and alkaline-extracted polysaccharides (AEP), were obtained. According to the results of high performance liquid chromatography, AEP contained mannose, rhamnose, galacturonic acid and galactose, while WEP was composed of galactose, rhamnose, fructose, arabinose, glucose, galacturonic acid and mannose. The compositions of intestinal microbiota were affected by the change of daily substrates. Meanwhile, the concentration of ammonia and gastrointestinal transit time decreased, the total short chain fatty acid (SCFA) levels, moisture of caecum content increased. These findings suggested that the effect of okra polysaccharides on intestinal microbiota was achieved by the output of detrimental compounds and the supply of nutrients. Besides, AEP supported a more pivotal role in manipulating gut microbiota than WEP.

Nano-fumed silica coated novel cellulosic okra fabrics with enhanced hydrophobic, mechanical and thermal properties for high performance bio-composite applications

Rahman A. N. M. Masudur,Zhang Xueping,Qin Xiaohong

The hydrophilic property of lignocellulosic fibers is the main obstacle to improve the performance of structural bio-composites. To effectively utilize such fibers in high-performance composites, their hydrophobicity must be significantly increased. This study presents the hydrophobicity enhancement of okra woven fabric (OWF) by the application of hydrophobic nano-fumed silica (NFS) coating. Different concentrations of NFS were investigated, and SEM analysis confirmed that 1% NFS coated OWF presented the greatest quantity of nanoparticle deposition. The 1% NFS treated OWF exhibited the highest water contact angle of 137.2 ± 1° (~ 148% increased than uncoated fabric) and the greatest tensile strength of 21.85 ± 1.64 MPa in the warp direction and 15.66 ± 1.73 MPa in the weft direction (~ 46.35% and ~ 42.75% respectively superior to uncoated one). The TGA analysis demonstrated that the thermal characteristics have also been enhanced by NFS coating. The interaction between NFS and OWF has been investigated using FTIR analysis, while their elemental composition has been assessed using EDX and XPS techniques. Graphical abstract

Okra polysaccharides/gelatin complex coacervate as pH-responsive and intestine-targeting delivery protects isoquercitin bioactivity

Jingwen Li,Xiaoran Yang,Xiao Li,Zihan Zhang,Zeliang Wei,Zhihua Xing,Sha Deng,Feixia Duan

Okra polysaccharides (OPs) belong to RG I pectin branched with neutral saccharide side chains, which possesses distinctive structure and physicochemical properties from the commonly used HG pectin. Until now, the application of RG I pectin as wall material of microcapsule remains unclear. Here, we obtained OPs/gelatin complex coacervate at the maximum yield of 86.8% (pH 3.5, gelatin/OPs ratio 9:1 and 2% (w/v) total polymer concentration) by response surface methodology. Isoquercitin (IQ)-loaded OPs/gelatin complex coacervate (OGIQ) showed porous spongy-like surface structure with average particle size, encapsulation efficiency and surface porosity at 334 nm, 81.6% and 31.9%, respectively. OGIQ was found to be pH-responsive and intestine-targeting. The IQ-release rate of OGIQ was assayed to be 89.4% in intestine fluid and below 2% in acidic and simulated gastric digestion, respectively. Accordingly, embedding in OGIQ protected IQ in digestion and improved its postdigestive α-glucosidase inhibitory rate by 88.7%. The differential scanning calorimetry curves showed that OGIQ effectively prevented IQ from thermal decomposition. The XRD, FT-IR and CD spectra indicated that IQ was embedded in OGIQ in amorphous state by hydrogen bonds and electrostatic interaction. Compared with HG, the neutral saccharide side chains of OPs could induce different secondary conformation change of gelatin during complex coacervation.

Colorimetric film based on polyvinyl alcohol/okra mucilage polysaccharide incorporated with rose anthocyanins for shrimp freshness monitoring

Shaolei Kang,Hualin Wang,Li Xia,Minmin Chen,Linlin Li,Junfeng Cheng,Xingjiang Li,Shaotong Jiang

In this work, a colorimetric film was designed for shrimp freshness monitoring by incorporating rose anthocyanins (RAs) in polyvinyl alcohol/okra mucilage polysaccharide (PVA/OMP) composite film. The presence of OMP changed the film-forming solution from Newtonian to non-Newtonian fluid. The addition of OMP and RAs decreased the crystalline of PVA due to the hydrogen bonds among RAs, OMP and PVA. An appropriate content of RAs and OMP could improve the film mechanical and barrier properties. The colorimetric film showed distinguishable color changes at pH 2–12 and was high sensitive to volatile ammonia. The target film of PVA/OMP-RAs could effectively monitor shrimp freshness in real time and the color changes were easily distinguished by naked eye, suggesting its potential in intelligent packaging for freshness monitoring of aquatic products and meat foods.

Effect of Lactiplantibacillus plantarum fermentation on the physicochemical, antioxidant activity and immunomodulatory ability of polysaccharides from Lvjian okra

Xingjie Wang,Kaidi Hu,Yuan Chen,Jinghui Lai,Mengmei Zhang,Jianlong Li,Qin Li,Ning Zhao,Shuliang Liu

Okra polysaccharides exhibits a range of biological activities. To date, its processing using microbial fermentation has not been explored. This study investigated the fermentation of okra juice with various lactic acid bacteria , followed by the extraction and characterization of crude polysaccharides (termed OPS-F), in contrast to their non-fermented counterpart (OPS). Changes in physicochemical properties, antioxidant activity and immunomodulatory ability were noted. The results demonstrated that OPS-F had a 7.42–12.53 % increase in total polysaccharides content compared to OPS. However, high-performance size-exclusion chromatography indicated a reduction in the molecular weight of OPS-F (7.9–9.5 × 10 5  Da) relative to OPS (1.66 × 10 6  Da). Compared to OPS, OPS-F had reduced levels of mannose , glucose, glucuronic acid and arabinose , but increased rhamnose , galacturonic acid and galactose , exhibiting enhanced solubility and lower apparent viscosity. Fourier transform infrared spectroscopy and nuclear magnetic resonance analysis showed minimal changes in polysaccharide structure post-fermentation. Moreover, despite a decrease in antioxidant activity post-fermentation, OPS-F exhibited superior immunomodulatory potential. In conclusion, fermenting okra juice with lactic acid bacteria alters the physicochemical properties of crude polysaccharides and enhances their immunomodulatory activity, offering a promising approach for developing new functional food resources.

Nitric Oxide Modulates Postharvest Physiology to Maintain Abelmoschus esculentus Quality Under Cold Storage

Xianjun Chen,Fenghuang Mo,Ying Long,Xiaofeng Liu,Yao Jiang,Jianwei Zhang,Cheng Zhong,Qin Yang,Huiying Liu

Cold storage is widely used for the postharvest preservation of fruits and vegetables; however, okra, as a tropical vegetable, is susceptible to chilling injury under low-temperature storage conditions, leading to quality deterioration, reduced nutritional value, and significant economic losses. Nitric oxide (NO), as an important signaling molecule, plays a crucial role in the postharvest preservation of fruits and vegetables. To investigate the effects of different concentrations of nitric oxide on the postharvest quality of okra under cold storage, fresh okra pods were treated with sodium nitroprusside (SNP), a commonly used NO donor, at concentrations of 0 (control), 0.5 (T1), 1.0 (T2), 1.5 (T3), and 2.0 mmol·L−1(T4). The results showed that low-concentration NO treatment (T1) significantly reduced weight loss, improved texture attributes including hardness, springiness, chewiness, resilience, and cohesiveness, and suppressed the increase in adhesiveness. T1 treatment also effectively inhibited excessive accumulation of cellulose and lignin, thereby maintaining tissue palatability and structural integrity. Additionally, T1 significantly delayed chlorophyll degradation, preserved higher levels of soluble sugars and proteins, and enhanced the activities of key antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), contributing to improved oxidative stress resistance and membrane stability. In contrast, high-concentration NO treatments (T3 and T4) led to pronounced quality deterioration, characterized by accelerated membrane lipid peroxidation as evidenced by increased malondialdehyde (MDA) content and relative conductivity, and impaired antioxidant defense, resulting in rapid texture degradation, chlorophyll loss, nutrient depletion, and oxidative damage. These findings provide theoretical insights and practical guidance for the precise application of NO in extending shelf life and maintaining the postharvest quality of okra fruits.

Preparation, amino acid composition, and in Vitro antioxidant activity of okra seed meal protein hydrolysates

Hongliang Yao,Jiani Yang,Jiajia Zhan,Qu Lu,Min Su,Yaojiong Jiang

To improve the utilization of okra seed, acidic and enzymatic hydrolyses of producing protein hydrolysates were respectively optimized by orthogonal experiment and response surface methodology using the degree of hydrolysis (DH) as evaluating index. Amino acid composition and antioxidant capacity in vitro of two kinds of hydrolysates were both analyzed. The degree of acidic hydrolysis was 58.53 ± 1.92% under the following optimized condition: hydrolyzing time 40 hr, temperature 95°C, ratio of acid solution to okra seed meal (OSM) powder was 5:1 (V:W/ml:g), and hydrochloric acid concentration was 18% (W/W). The degree of enzymatic hydrolysis was 16.26 ± 0.56% under the optimized condition: hydrolyzing time 8.20 hr, ratio of buffer to OSM powder was 10:1, and enzyme dosage was 3,100 International Units (IU) g −1 . Enzymatic hydrolysates had a fuller range of amino acids and antioxidant capacity than acidic hydrolysates. The results provide technical support for the expansion of okra seed utilization.

3D Printing of Naturally Derived Adhesive Hemostatic Sponge

Minyu Zhou,Tao Yuan,Luoran Shang

Hydrogel hemostatic sponges have been recognized for its effectiveness in wound treatment due to its excellent biocompatibility, degradability, as well as multi-facet functionalities. Current research focuses on optimizing the composition and structure of the sponge to enhance its therapeutic effectiveness. Here, we propose an adhesive hydrogel made from purely natural substances extracted from okra and Panax notoginseng. We utilize 3-dimensional (3D) printing technology to fabricate the hemostatic hydrogel scaffold, incorporating gelatin into the hydrogel and refining the mixing ratio. The interaction between gelatin and okra polyphenols contributes to successful injectability as well as stability of the printed scaffold. The okra in the scaffold exhibits favorable adhesion and hemostatic effects, and the total saponins of Panax notoginseng facilitate angiogenesis. Through in vitro experiments, we have substantiated the scaffold's excellent stability, adhesion, biocompatibility, and angiogenesis-promoting ability. Furthermore, in vivo experiments have demonstrated its dual functionality in rapid hemostasis and wound repair. These features suggest that the 3D-printed, natural substance-derived hydrogel scaffolds have valuable potential in wound healing and related applications.

Effect of drying methods on physicochemical properties and antioxidant activities of okra pods

Xiao Gong,Xiaobing Huang,Taohua Yang,Jing Wen,Wei Zhou,Jihua Li

The effects of hot air drying (HD), freeze-drying (FD), microwave drying (MWD), natural convective drying (NCD), and vacuum drying (VD) on the physicochemical, nutritional, and antioxidant properties of okra pods were investigated. Changes in the surface morphology, color, rehydration ratio, protein, pectin, ascorbic acid, and phenolics, as well as antioxidant activities of okra pods measured by 2,2′-azinobis (3-ethylbenzo thiazoline-6-sulphonic acid) diammonium salt (ABTS), 1,1-diphenyl-2-picrylhydrazyl (DPPH), and Ferric Ion Reducing Antioxidant Power (FRAP) assays were evaluated. In general, FD products exhibited a better retention of the texture characteristics, nutritional, and antioxidant activities, and was suitable for okra pods drying, although FD was the longer time-consuming method among the five drying methods. Furthermore, NCD products were poorer in color, texture, and nutrition. Future studies should continuously focus on various combinations of different types of drying methods to supply okra pods products with the most affordable cost and the highest quality. Practical applications Physicochemical properties and antioxidant capacities of okra pods are affected by the drying process. The drying process with nonthermal, low temperature, and low content of oxygen is beneficial to the retention of nutrients and functional components. In general, freeze-drying exhibited a better texture characteristics and good antioxidant activities, although higher energy and longer time consumption was the current challenge. How to combine the different types of drying methods has become an inevitable trend and research focus.
