Insufficient early osteogenesis seriously affects the later stage osteogenic quality and osseointegration of dental implants. To promote early osteogenesis, we first designed a Ti dental implant with a built-in magnet (mTi) to produce a local static magnetic field (SMF). Then, a dental implantation system comprising the mTi implant and the superparamagnetic hydroxyapatite (HA:Yb/Ho–Fe, named HYH-Fe) particles was implanted into the alveolar bone of beagles. The results showed that the mTi + HYH-Fe group displayed better early osteogenesis and later stage osseointegration than the Ti + HA and mTi + HA groups. A combination of the local SMF (mTi) and superparamagnetic HYH-Fe particles had a positive effect on the pro-osteogenesis of Ti implants. The results also indicated that week 10 could be adopted as the key time point to evaluate the early osteogenic effect of the mTi + HYH-Fe implantation system, which would be a promising prospect for promotion of osteogenesis, in vivo tracking investigation of material-bone relationships, and clinical applications.

In conventional dental implant procedures, glass-ceramic dental crowns were traditionally bonded to titanium (Ti) abutment using organic glue cementation. However, degradation of the adhesive could result in crown loss. Therefore, it is crucial to explore alternative strategies for bonding the crown and abutment. In this study, we propose employing dissimilar brazing as a new approach for joining these two components. To assess its feasibility, Ti/ZrO 2 –SiO 2 nanocrystalline glass-ceramic (ZS-NCGC) joints were fabricated by brazing using Ni/Zr composite foils. The corrosion behavior and mechanism of the Ti/ZS-NCGC joint in artificial saliva were investigated. The complex interface of the Ti/ZS-NCGC joint increased the corrosion current and decreased the impedance, resulting in slightly lower corrosion resistance than the Ti substrate. However, after soaking the Ti/ZS-NCGC joint in acidic artificial saliva for 7 days, no pitting corrosion appeared, demonstrating excellent corrosion resistance. In addition, after adding highly destructive F ions, corrosion mainly occurred on layered α-(Ti, Zr), blocky (Ti, Zr) 2 Ni, and (α-(Ti, Zr) + (Ti, Zr) 2 Ni) eutectoid structure. The findings presented herein demonstrate the significant potential of dissimilar brazing in facilitating the robust bonding between glass-ceramic dental crowns and Ti abutment.

Following dental implantation, the characteristic bacterial milieu of the oral cavity may lead to peri-implant inflammation, which can negatively impact osseointegration and cause implant failure. To improve soft tissue sealing around the implant, enhance osseointegration, and improve implant success rates, this paper proposes a composite multifunctional coating (PHG) prepared using gelatin and polydopamine/hydroxyapatite nanoparticles, investigates the effects of this novel coating on cell adhesion, proliferation, antibacterial activity, osteogenic differentiation, and evaluates its immune-related properties. The PHG coating was proved to have satisfactory hydrophilicity and wettability for cell attachment. Furthermore, it improved the expression of adhesion-related genes and proteins in human gingival fibroblasts, indicating its adhesion-promoting effect. Additionally, bone marrow mesenchymal stem cells exhibited strong osteogenic differentiation potential and mineralization on PHG-coated surfaces. Notably, the PHG coating exhibited antibacterial activity against Streptococcus mutans , as well as anti-inflammatory effects, potentially via the regulation of macrophages. Therefore, the proposed PHG coating may promote soft tissue sealing and bone bonding, providing a potential strategy for the surface modification of dental implants.

Objective This study aims to investigate the influence of fluoride exposure on implant osseointegration. Methods A total of 24 male New Zealand white rabbits were randomly divided into the control group and the fluoride exposure group. Rabbits in the control group were fed with tap water, while those in the fluoride exposure group were given 200 mg/L sodium fluoride solution. After 2-month feeding, implants were inserted into the extraction socket immediately after extraction of rabbit mandibular anterior teeth. Four rabbits in each group were sacrificed to collect the implants samples at 1, 2, and 3 months post-implantation, respectively. Radiographic and histomorphometry examinations were performed to evaluate the condition of implant osseointegration. Results Bone volume around the implants increased in a time-dependent manner in both groups. Micro-CT images illustrated that the bone mineral density (BMD) in the fluoride exposure group was significantly lower than that in the control group after implantation for 2 and 3 months. The bone–implant contact ratio (BIC) in the fluoride exposure group was much lower than that of the control group at 3 months post-implantation according to histomorphometry examination. Conclusions In rabbit animal model, high fluoride exposure affected the quality of bone surrounding the implant and significantly reduced bone integration of the implant, especially in the late stage of osseointegration.

Peri-implantitis is an inflammatory disease characterized by progressive bone loss, challenging implant longevity and incurring significant treatment burden. Current reconstruction therapy involves mechanical scaling followed by artificial bone grafting. However, residual bacteria and inflammation memory often lead to recurrence, highlighting the critical need for the artificial bone substitute with sustained anti-inflammatory and antibacterial properties. By utilizing the phosphate groups of mesoporous hydroxyapatites for in situ loading of CeO 2 , followed by a reduction method to fabricate a CeO 2 -Cu heterojunction interface, this study develops Bonyzymes with anti-inflammatory, antibacterial, and osteogenic properties. The incorporation of Cu modulates the electronic environment of the CeO 2 surface, increasing superoxide dismutase and catalase activities of Bonyzymes. In vitro, the Bonyzymes effectively regulated oxidative stress by scavenging excess reactive oxygen species, exerting anti-inflammatory effects. Additionally, it exhibits antibacterial activity against Staphylococcus aureus , Escherichia coli and Porphyromonas gingivalis , and facilitates osteogenic differentiation in bone marrow stem cells. In vivo experiments in rat peri-implantitis models confirm its ability to promote bone regeneration in the infected peri-implant area. Overall, this work develops novel artificial bone substitutes, Bonyzymes, which not only promotes bone regeneration but also exhibits anti-inflammatory and antibacterial properties, offering a promising strategy for peri-implantitis treatment.

Titanium dental implants are prone to infections by pathogenic bacteria, leading to the formation of biofilm and development of peri-implantitis. Due to their excellent biocompatibility, peroxidase-like activity, and photothermal capabilities, carbon-based nanozymes emerge as a low-risk alternative to traditional antibiotics to treat drug-resistant bacterial infections. However, a crucial question regarding carbon-based nanomaterials with antibacterial properties lies in how to optimize the process parameters to maximize their bactericidal efficacy while minimizing adverse effects such as energy consumption, cytotoxicity, and damage to titanium implants. We have synthesized five groups of carbon-based nanozymes with distinct microstructures through pyrolysis and comprehensively evaluated their performance in five key aspects: energy consumption, photothermal performance, peroxidase-like activity, environmental sensitivity, and cytotoxicity. The results of our experiments provide valuable references for the rational design of carbon-based nanozymes for the treatment of peri-implantitis.

Background Titanium (Ti) is now widely used as implant material due to its excellent mechanical properties and superior biocompatibilities, while its inert bioactivities might lead to insufficient osseointegration, and limit its performance in dental applications. Methods We introduced a robust and simple approach of modifying titanium surfaces with polysaccharide complexes. Titanium samples were subjected to hydrothermal treatment to create a uniform porous structure on the surface, followed by coating with a bioinspired and self-assembly polydopamine layer. Strontium Eucommia Ulmoides Polysaccharide (EUP-Sr) complexes are then introduced to the polydopamine-coated porous titanium. Multiple morphological and physiochemical characterizations are employed for material evaluation, while cell proliferation and gene expression tests using macrophages, primary alveolar bone osteoblasts, and vascular endothelial cells are used to provide an overall insight into the functions of the product. The significances of statistical differences were analyzed using student’s t-test. Results Microscopic and spectrometric characterizations confirmed that the Ti surface formed a porous structure with an adequate amount of EUP-Sr loading. The attachment was attributed to hydrogen bonding between the ubiquitous glycosidic linkage of the polysaccharide complex and the ring structure of polydopamine, yet the loaded EUP-Sr complex can be gradually released, consequently benefiting the neighboring microenvironment. Cell experiments showed no cytotoxicity of the material, and the product showed promising anti-inflammation, osseointegration, and angiogenesis properties, which were further confirmed by in vivo evaluations. Conclusion We believe the EUP-Sr modified titanium implant is a promising candidate to be used in dental applications with notable osteoimmunomodulation and angiogenesis functions. And the novel technique proposed in this study would benefit the modification of metal/inorganic surfaces with polysaccharides for future research.

Introduction The nanostructural modification of the oral implant surface can effectively mimic the morphology of natural bone tissue, allowing osteoblasts to achieve both proliferation and differentiation capabilities at the bone interface of the dental implant. To improve the osteoinductive activity on the surface of titanium implants for rapid osseointegration, we prepared a novel composite coating (MAO-PDA-NC) by micro-arc oxidation technique and immersion method and evaluated the proliferation, adhesion, and osteogenic differentiation of osteoblasts on this coating. Methods The coatings were prepared by micro-arc oxidation (MAO) technique and immersion method, and characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM) for different coatings; the loading of PDA was examined using Fourier transform infrared spectroscopy (FTIR); the ion release capacity of the coatings was determined by inductively coupled plasma emission spectrometry (ICP-OES); the interfacial bonding of the coatings was examined using nanoscratch experiment strength. The cytotoxicity of the coating was examined by live/dead staining kit; cell proliferation viability was examined by CCK-8 kit; adhesion and osteogenic effect of the coating were examined by immunofluorescence staining and RT-PCR; osteogenic differentiation was examined by alkaline phosphatase staining. Results The surface morphology of titanium implants was modified by micro-arc oxidation technology, and a new MAO-PDA-NC composite coating was successfully prepared. The results showed that the MAO-PDA-NC coating not only optimized the physical and chemical properties of the titanium implant surface but also significantly stimulated the biological properties of osteoblast adhesion, proliferation, and osteogenic differentiation on the coating surface. Conclusion These results show that MAO-PDA-NC composite coating can significantly improve the surface properties of titanium implants and achieve a stable bond between implant and bone tissue, thus accelerating early osseointegration.

Objectives This study aimed to assess the effect of the low-density lipoprotein receptor-related protein 5 (Lrp5) A241T gain-of-function mutation, which induces a high-bone-mass phenotype, on implant osseointegration in a murine model. Methods Heterozygous Lrp5 A241T mutant ( Lrp5 A/+ ) and wild-type (WT) littermate male mice on a C57BL/6J background were used. Micro-computed tomography (micro-CT) was performed to assess tibial trabecular bone architecture. Sandblasted and acid-etched (SLA) titanium implants were surgically inserted into the proximal tibiae, and samples were harvested after 4 weeks for micro-CT and histological analysis. Primary osteoblasts were seeded onto SLA titanium sheets and induced for osteogenesis for 7 days. Osteogenic marker expression was assessed by quantitative real-time polymerase chain reaction and western blotting. Results The Lrp5 A/+ group exhibited significantly improved trabecular bone architecture compared to WT controls. At 4 weeks post-implantation, micro-CT revealed greater peri-implant bone formation in the Lrp5 A/+ group, while hard tissue sections showed increased bone area and bone-implant contact. HE and Masson staining further confirmed greater new bone formation. After 7 days of in vitro osteogenic induction, Lrp5, Runx2, and Sp7 were significantly upregulated in the Lrp5 A/+ group. Conclusions The Lrp5 A241T mutation significantly enhanced implant osseointegration in a transgenic murine model.

A novel magnetic drug-loaded osteoinductive Fe3O4/CaCO3 hybrid microspheres system (MDHMs) with excellent drug delivery property has been fabricated by a modified coprecipitation method. The MDHMs are uniform vaterite microspheres with well-defined mesoporous nanostructures. The formation of vaterite is based on the templating effect of casein micelles, which is promising in bone tissue engineering. Cyclodextrins (CDs) increase the porosity of the microspheres with a pore size of about 7.3 nm and a surface area of about 50.73 m2 g−1. The mesoporous structure gives the MDHMs great drug loading efficiency of about 80.75–88.74% and the minocycline-loaded MDHMs display a sustained drug release property (about 95% in 5 d). Moreover, the MDHMs also possess stable magnetic property with a saturation magnetization of about 4.41 emu g−1 and a remarkable magnetism-responsive behavior. These results demonstrate that with superior drug delivery property, osteoinductive potential and magnetic property, the MDHMs have a great potential in the treatment of peri-implantitis.

Peri-implantitis, a leading cause of implant failure, currently lacks effective therapeutic strategies. Given that bacterial infection and reactive oxygen species overabundance serve as primary pathogenic and triggering factors, respectively, an adhesive hydrogel has been created for in-situ injection. The hydrogel is a gallic acid-grafted chitosan (CS-GA) hydrogel containing tannic acid miniaturized particles (TAMP). This provides antibacterial and antioxidant properties. Therefore, this study aims to evaluate the potential role of this hydrogel in preventing and treating peri-implantitis via several experiments. It undergoes rapid formation within a span of over 20 s via an oxidative crosslinking reaction catalyzed by horseradish peroxidase and hydrogen peroxide, demonstrating robust adhesion, superior cell compatibility, and a sealing effect. Furthermore, the incorporation of TAMP offer photothermal properties to the hydrogel, enabling it to enhance the viability, migration, and antioxidant activity of co-cultured human gingival fibroblasts when subjected 0.5 W/cm 2 808 nm near-infrared (NIR) irradiation. At higher irradiation power, the hydrogel exhibits progressive improvements in its antibacterial efficacy against Porphyromonas gingivalis and Fusobacterium nucleatum . It attains rates of 83.11 ± 5.42 % and 83.48 ± 6.855 %, respectively, under 1 W/cm 2 NIR irradiation. In summary, the NIR-controlled CS-GA/TAMP hydrogel, exhibiting antibacterial and antioxidant properties, represents a promising approach for the prophylaxis and management of peri-implantitis.

Achieving optimal implant restoration hinges on both robust osseointegration as the structural foundation and favorable osseoperception for advanced masticatory function. The significance of nerve innervation in the peri-implant environment as the cornerstone of osseointegration and osseoperception is often underestimated. Despite the integral role of endogenous electric fields (EFs) in the human body, particularly in electrosensitive tissues like nerve tissue, the current approach involving external electrical stimulation is invasive and not clinically applicable. The present study introduces a conductive hydrogel designed to respond to endogenous EFs, aiming to foster nerve regeneration around dental implants to coordinate both osseointegration and osseoperception. The hydrogel promotes neurite outgrowth by upregulating intracellular Ca 2+ concentration and activating subsequent pathways. Furthermore, the enhanced release of neuropeptides from neurocells improves the osteogenesis of osteoblasts. The impact of the conductive hydrogel on osseointegration and osseoperception is also thoroughly investigated in vivo. This conductive hydrogel represents a unique strategy for enhancing osseointegration and osseoperception within the endogenous EFs of the peri-implant environment. This advancement opens the door to achieving the physiological and psychological integration of dental implants.
