The harvest time of a plant usually affects the yield of its metabolites. Polygonum tinctorium ( Polygonum tinctorium Ait., P. tinctorium ) is an important cultivated plant that produces the natural blue dye indigo and the medicinal metabolite indirubin. To explore the mechanism of indigo and indirubin accumulation, the leaves of P. tinctorium at different harvest times, namely July, August, and October, were investigated by metabolite and de novo transcriptome analyses. The results revealed that the indigo and indirubin content varied at different harvest times. Moreover, their contents were significantly higher (p < 0.05) in August than in either July or October. Transcriptome analysis showed different gene expression patterns of P. tinctorium leaves in August. Almost all the identified differentially expressed genes (DEGs) that were responsible for the biosynthesis of indigo and indirubin, including four anthranilate synthase (AS) genes, two indole-3-glycerolphosphate synthase (IGPS) genes, three tryptophan synthase (TS) genes and seven cytochrome P450 (CYP450) genes were expressed at significantly higher levels in August ( p < 0.05), which correlated with the increase of the two compounds. Overall, the results of this study provide useful information for understanding the accumulation mechanism of indigo and indirubin in P. tinctorium , which may promote the development of agricultural breeding and industrial production of P. tinctorium. Moreover, sequence data obtained by de novo transcriptome sequencing of P. tinctorium provides a platform for future molecular biological studies.

Organophosphate esters (OPEs) have been frequently detected in crops. However, few studies have focused on the uptake and translocation of OPEs in plants following foliar exposure. Herein, to investigate the foliar uptake, accumulation and translocation mechanisms of OPEs in plant, the cucumber ( Cucumis sativus ) was selected as a model plant for OPEs exposure via foliar application under control conditions. The results showed that the content of OPEs in the leaf cuticle was higher than that in the mesophyll on exposed leaf. Significant positive correlations were observed between the content of OPEs in the leaf cuticle and their log K ow and log K cw values ( P < 0.01), suggesting that OPEs with high hydrophobicity could not easily move from the cuticle to the mesophyll. The moderately hydrophobic OPEs, such as tris (2-chloroisopropyl) phosphate (TCPP, log K ow = 2.59), were more likely to move not only from the cuticle to the mesophyll but also from the mesophyll to the phloem . The majority of the transported OPEs accumulated in younger leaves (32–45 %), indicating that younger tissue was the primary target organ for OPEs accumulation after foliar exposure. Compared to chlorinated OPEs (except TCPP) and aryl OPEs, alkyl OPEs exhibited the strongest transport capacity in cucumber seedling due to their high hydrophilicity. Interestingly, tri-p-cresyl phosphate was found to be more prone to translocation compared to tri-m-cresyl phosphate and tri-o-cresyl phosphate, despite having same molecular weight and similar log K ow value. These results can contribute to our understanding of foliar uptake and translocation mechanism of OPEs by plant.

To explore variation patterns of uptake, translocation, and accumulation processes responding to organophosphate esters (OPEs) among Poaceae plants, hydroponic and computer simulation experiments were executed. Plant growth, OPEs’ concentration, and bioinformation and transcript of lipid transporters in the three terrestrial barley, wheat, and maize and aquatic rice seedlings were studied after exposure to seven OPE congeners. Four types of plants could accumulate seven OPE congeners. OPEs could promote rice growth by upregulating IAA27 hormone gene. However, maize growth was inhibited due to upregulating IAA17 hormone gene. In general, OPEs with log K ow > 4 tended to accumulate in roots of the four types of plants. Furthermore, the uptake, translocation, and accumulation mechanism of OPEs in different plants showed species-specific, depending on chemical properties of OPEs and biological factors specifically referring to the binding ability and gene expression of lipid transporters. The uptake and accumulation of OPE in aquatic rice roots were mainly influenced by biological factors. On the contrary, terrestrial plants relied on log K ow more than biological factors. Meanwhile, TIL of Poaceae plants could be a common and key protein that contributed to OPEs accumulation.

Hexafluoropropylene oxide (HFPO) homologues, which are important alternatives to perfluorooctanoic acid, have been frequently identified in crops. Although exposure to HFPO homologues via crops may pose non-negligible threats to humans, their impact on crops is still unknown. In this study, the accumulation, transport, and distribution mechanisms of three HFPO homologues in lettuce were investigated at the plant, tissue, and cell levels. More specifically, HFPO trimer acid and HFPO tetramer acid were primarily fixed in roots and hardly transported to shoots (TF, 0.06–0.63). Conversely, HFPO dimer acid (HFPO-DA) tended to accumulate in lettuce shoots 2–264 times more than the other two homologues, thus resulting in higher estimated daily intake values. Furthermore, the dissolved organic matter derived from root exudate enhanced HFPO-DA uptake by increasing its desorption fractions in the rhizosphere. The transmembrane uptake of HFPO homologues was controlled by means of a transporter-mediated active process involving anion channels, with the uptake of HFPO-DA being additionally facilitated by aquaporins. The higher accumulation of HFPO-DA in shoots was attributed to the larger proportions of HFPO-DA in the soluble fraction (55–74%) and its higher abundance in both vascular tissues and xylem sap. Our findings expand the understanding of the fate of HFPO homologues in soil–crop systems and reveal the underlying mechanisms of the potential exposure risk to HFPO-DA.
