Background
Magnaporthe oryzae , the rice blast fungus, might secrete certain proteins related to plant-fungal pathogen interactions.
Methodology/Principal Findings
In this study, we report the purification, characterization, and gene cloning of a novel hypersensitive response-inducing protein elicitor (MoHrip1) secreted by M. oryzae . The protein fraction was purified and identified by de novo sequencing, and the sequence matched the genomic sequence of a putative protein from M. oryzae strain 70-15 (GenBank accession No. XP_366602.1). The elicitor-encoding gene mohrip1 was isolated; it consisted of a 429 bp cDNA, which encodes a polypeptide of 142 amino acids with a molecular weight of 14.322 kDa and a pI of 4.53. The deduced protein, MoHrip1, was expressed in E. coli . And the expression protein collected from bacterium also forms necrotic lesions in tobacco. MoHrip1 could induce the early events of the defense response, including hydrogen peroxide production, callose deposition, and alkalization of the extracellular medium, in tobacco. Moreover, MoHrip1-treated rice seedlings possessed significantly enhanced systemic resistance to M. oryzae compared to the control seedlings. The real-time PCR results indicated that the expression of some pathogenesis-related genes and genes involved in signal transduction could also be induced by MoHrip1.
Conclusion/Significance
The results demonstrate that MoHrip1 triggers defense responses in rice and could be used for controlling rice blast disease.

The cell surface receptor chitin elicitor receptor kinase 1 (CERK1) is a well-known component of plant immunity. In a previous study, we found that OsCERK1 is also involved in regulating copper (Cu) uptake in rice, though the underlying mechanisms remain unclear. In this study, we identified proteins that interact with OsCERK1 and discovered a novel heavy metal-associated (HMA) domain-containing protein, OsHPP08. Our findings show that OsCERK1 phosphorylates and stabilizes OsHPP08. Structural analysis using AlphaFold, a yeast sensitivity assay of the Cu uptake-deficient yeast mutant, and Cu level measurements in the oshpp08 mutants and overexpression plants ( OsHPP08OE ) revealed that OsHPP08 facilitates Cu uptake. Moreover, rice seedling infection assays demonstrated that OsHPP08 positively contributes to blast resistance, and both OsCERK1 and OsHPP08 are necessary for Cu-modulated blast resistance. Further analyses suggest that OsCERK1 and OsHPP08 likely enhance blast resistance by regulating the antioxidant system and increasing the accumulation of H 2 O 2 . In conclusion, OsCERK1 promotes Cu uptake by stabilizing OsHPP08, and together they contribute to Cu-modulated blast resistance likely by modulating ROS accumulation. These results deepen our understanding of the interplay between biotic and abiotic signals in rice.

Camellia oleifera is expected to provide alternative aglycone to synthesize some saponins similar to that from Schima superba with inhibitory activity against Magnaporthe oryzae. Eight theasapogenol galactosides were synthesized via protection of adjacent hydroxyl groups by a benzylidene for regioselective glycosylation in the multi-hydroxyl sapogenin. Water soluble galactose chain connected far from liposoluble end was a key group in inhibiting the growth of M. oryzea unless theasapogenol was modified by two galactosyl groups or by one galactosyl group and one benzylidene group. The amphoteric characteristics of saponin such as saccharide group number, distance between bipolar groups play an important role in inhibiting mycelium growth of M. oryzae.

Rice blast caused by Magnaporthe oryzae B. Couch is one of the most devastating diseases on rice. Camptothecin (CPT), which was primarily isolated from Camptotheca acuminata , is well-known for its anti-tumor activities, and is also developed as a potential biological pesticide . We previously investigated the anti-microbial activities of CPT against 11 fungi, 3 oomycetes , and 4 bacteria, and found that CPT was strongly effective against M. oryzae , indicating its potential as a lead for developing fungicide against rice blast. However, the anti-fungal effects of CPT on M. oryzae need further elucidation. In this study, the anti-fungal activities of CPT against M. oryzae were further investigated, which revealed that CPT was effective against M. oryzae both in vitro and in vivo . The transcriptome of M. oryzae was analyzed after CPT treatment, which showed that CPT had a strong inhibitory effect on ‘translation’ and ‘carbohydrate metabolism/energy metabolism’ of M. oryzae . Some physiology characteristics of M. oryzae were also assayed, which confirmed that CPT inhibited RNA synthesis , protein synthesis , and carbohydrate metabolism/energy metabolism of M. oryzae , and caused membrane damage. The molecular simulation result showed that CPT binds to the interface of DNA-topoisomerase I complex of M. oryzae . In conclusion, CPT is a promising lead for developing fungicide against rice blast. CPT may bind to DNA-topoisomerase I complex of M. oryzae , thus affecting ‘translation’ and ‘carbohydrate metabolism/energy metabolism’, leading to cell death.

BACKGROUND Rice blast, caused by Pyricularia oryzae , is a devastating fungal disease threatening global rice production. Overreliance on chemical fungicides has raised environmental concerns and led to resistant strains, necessitating the development of sustainable alternatives. This study integrated marine microbiology and natural antifungal compounds to create eco-friendly alternatives to chemical fungicides for disease management. RESULTS We identified Pseudomonas aeruginosa R64 with broad-spectrum antimicrobial activity from mangrove soil in the Mai Po Nature Reserve. The R64 fermentation extract (RFE) exhibited multifaceted inhibition of P. oryzae , suppressing mycelial growth, conidiation, conidial germination and appressorial formation, while disturbing cell wall and membrane function. It also attenuated virulence by impairing appressorial penetration and invasive growth. Further chemical analysis identified phenazines and quinolines as the primary compounds in RFE, corroborated by PCR detection of corresponding phenazine biosynthetic gene clusters. Comparative bioassays with two main bioactive components of RFE, phenazine-1-carboxamide (PCN) and phenazine-1-carboxylic acid (PCA), against P. oryzae implicated PCN as the principal antifungal effector. RFE and PCN had higher efficacy than tricyclazole in P. oryzae growth inhibition, but were less effective than isoprothiolane. Furthermore, RFE and PCN displayed lower acute ecotoxicity to an environmental indicator organism than isoprothiolane, suggesting their potential as sustainable biopesticides for rice blast management. CONCLUSION Natural products from mangrove soil bacterium P. aeruginosa R64 inhibited key developmental and infection processes of P. oryzae , effectively reducing rice blast development. The promising disease inhibition and low ecotoxicity of mangrove-associated bacteria highlight their untapped potential for innovative, eco-friendly fungicide mining for sustainable agriculture. © 2025 The Author(s). Rice blast disease (RBD) caused by Magnaporthe oryzae , threaten food security by cutting agricultural output. Nano agrochemicals are now perceived as sustainable, cost-effective alternatives to traditional pesticides. This study investigated bioformulation of moringa chitosan nanoparticles (M-CsNPs) and their mechanisms for suppressing RBD while minimizing toxic effects on the microenvironment. M-CsNPs, sized 46 nm with semi-spherical morphology, significantly suppressed pathogen growth, integrity, and colonization at 200 mg L −1 in vitro . Greenhouse tests with foliar exposure to the same concentration resulted in a substantial 77.7 % reduction in RBD, enhancing antioxidant enzyme activity and plant health. Furthermore, M-CsNPs improved photosynthesis, gas exchange, and the nutritional profile of diseased rice plants. RNA-seq analysis highlighted upregulated defense-related genes in treated rice plants. Metagenomic study showcased reshaping of the rice microbiome, reducing Magnaporthe abundance by 93.5 %. Both healthy and diseased rice plants showed increased microbial diversity, particularly favoring specific beneficial species Thiobacillus , Nitrospira , Nocardioides , and Sphingomicrobium in the rhizosphere and Azonexus , Agarivorans , and Bradyrhizobium in the phyllosphere. This comprehensive study unravels the diverse mechanisms by which M-CsNPs interact with plants and pathogens, curbing M. oryzae damage, promoting plant growth, and modulating the rice microbiome. It underscores the significant potential for effective plant disease management.

Rice blast, caused by the filamentous fungusPyricularia oryzae, has long been one of the major threats to almost all rice-growing areas worldwide. Metconazole, 5-(4-chlorobenzyl)-2, 2-dimethyl-1-(1H-1, 2, 4-triazol-1-ylmethyl) cyclopentanol, is a lipophilic, highly active triazole fungicide that has been applied in the control of various fungal pathogens of crops (cereals, barley, wheat), such as theFusariumandAlternariaspecies. However, the antifungal activity of metconazole againstP. oryzaeis unknown. In this study, metconazole exhibited broad spectrum antifungal activities against sevenP. oryzaestrains collected from rice paddy fields and the wild type strain P131. Scanning electron microscopic analysis and fluorescein diacetate staining assays revealed that metconazole treatment damaged the cell wall integrity, cell membrane permeability and even cell viability ofP. oryzae, resulting in deformed and shrunken hyphae. The supplementation of metconazole in vitro increased fungal sensitivity to different stresses, such as sodium dodecyl sulfate, congo red, sodium chloride, sorbitol and oxidative stress (H2O2). Metconazole could inhibit key virulence processes ofP. oryzae, including conidial germination, germ tube elongation and appressorium formation. Furthermore, this chemical preventedP. oryzaefrom infecting barley epidermal cells by disturbing appressorium penetration and subsequent invasive hyphae development. Pathogenicity assays indicated a reduction of over 75% in the length of blast lesions in both barley and rice leaves when 10 μg/mL of metconazole was applied. This study provides evidence to understand the antifungal effects of metconazole againstP. oryzaeand demonstrates its potential in rice blast management.

Improving rainfastness of agro-chemicals on leaves to reduce chemical leaching onto the ecosystem and aerosol drift is a huge challenge in modern agriculture and environmental protection. In this study, tea saponin (TS), a natural product and efficient bio-pesticide, was blended with environmentally-friendly vegetable oil-based waterborne polyurethanes . The chemical structures, mechanical properties, thermal stability, slow-release behavior of the composite films with different TS contents and the inhibitory effect of composites on mycelia growth of rice blast were studied. It is found that the release rate of TS from the composite films could be tailored by controlling the TS content of the composite emulsion. The composites prepared exhibit excellent inhibitory effect on the mycelia growth of rice blast, even after rainfall washing. This is the first time that bio-based and degradable polymer films with sustained-release of TS are proposed to control rice blast diseases with enhanced rainfastness.

The infection and resistance of pathogenic fungi pose a significant threat to food crop safety, highlighting the urgent need for new green antifungal agents. Fusarium acid (FA) with its simple and novel structure correlates with the principles of green pesticides and has demonstrated substantial broad-spectrum antifungal activity. In a previous study, several promising lead compounds were identified. This study focused on further optimizing lead compound A1. Its inhibitory effect was evaluated against six different pathogenic fungi and several new promising compounds were identified. Notably, the anti-Magnaporthe oryzae (M. oryzae) activity of compound B4 was significantly enhanced with an EC50 value of 910 ng/mL. Rice safety tests indicated that B4 not only exhibited no adverse effect on rice growth and chlorophyll synthesis but also induced plant immunity by increasing the levels of polyphenol oxidase (PPO), peroxidase (POD), and superoxide dismutase (SOD), and soluble sugars. Further investigations into the antifungal mechanism of B4 indicated that it enhanced the permeability and damage of the mycelial membrane, thereby leading to electrolyte leakage and the loss of essential cellular substances. Subsequently, B4 induced the production of reactive oxygen species (ROS) in M. oryzae cell, including •OH, •O2–, and 1O2, while promoting increased levels of malondialdehyde (MDA) and SOD enzyme activity. Rice infection tests indicated that B4 effectively inhibited M. oryzae conidia activity and inhibited melanin production. These findings indicated that FA derivatives hold significant potential as a new strategy for M. oryzae disease management and offer valuable insights for the control of plant fungal diseases.

Background: Natural products are often favored in the study of crop pests and diseases. Previous studies have shown that citronellal has a strong inhibition effect onMagnaporthe oryzae. The objective of this study was to clarify its mechanism of action againstM. oryzae. Results: Firstly, the biological activity of citronellal againstM. oryzaewas determined by direct and indirect methods, and the results show that citronellal had a strong inhibition effect onM. oryzaewith EC50values of 134.00 mg/L and 70.48 μL/L air, respectively. Additionally, a preliminary study on its mechanism of action was studied. After citronellal treatment, electron microscopy revealed that the mycelium became thin and broken; scanning electron microscopy revealed that the mycelium was wrinkled and distorted; and transmission electron microscopy revealed that the mycelium cell wall was invaginated, the mass wall of mycelium was separated, and the organelles were blurred. The mycelium was further stained with CFW, and the nodes were blurred, while the mycelium was almost non-fluorescent after PI staining, and there was no significant difference in the relative conductivity of mycelium. In addition, chitinase was significantly enhanced, and the expression of chitin synthesis-related genes was 17.47-fold upregulated. Finally, we found that the efficacy of citronellal against the rice blast was as high as 82.14% according to indoor efficacy tests. Conclusion: These results indicate that citronellal can affect the synthesis of chitin inM. oryzaeand damage its cell wall, thereby inhibiting the growth of mycelium and effectively protecting rice from rice blasts. Keywords:magnaporthe oryzae;citronellal;cell wall;physiology;cytotoxicity

Rice is a major global staple crop, but rising temperatures and freshwater shortages have made drought one of the most severe abiotic stresses affecting agriculture. Additionally, rice blast disease and brown planthopper infestations significantly impact yields. Therefore, developing water-saving, drought-resistant, high-yielding, and disease-resistant rice varieties is critical for sustainable rice production. The new water-saving and drought-resistant (WDR) rice ‘Huhan 1516’, bred using marker-assisted selection (MAS) and marker-assisted backcrossing (MABC) techniques, addresses these challenges. This variety is highly adaptable to drought-prone and water-scarce regions such as the Yangtze and Huai River basins. With its high yield, drought tolerance, and broad-spectrum resistance to rice blast (conferred by thePi2gene) and brown planthopper (BPH), ‘Huhan 1516’ is suitable for various farming systems and environments. Field trials show that this variety outperforms control varieties by 2.2% in yield and exhibits moderate resistance to both rice blast and brown planthopper. ‘Huhan 1516’ has been recognized as a new water-saving and drought-resistant rice variety by the state, and as a released cultivar, it has great potential for market promotion and application.

A pesticide carrier system can offer advantages to active components, such as improving shelf life, extending the lasting period, and reducing toxicity, all of which could reduce the adverse effects of pesticides on the environment. Graphene oxide (GO) is a promising medical drug carrier material, and its research in the field of pesticides is still in the exploration stage. Herein, in order to load water-insoluble pesticides, a series of polylactic acid–graphene oxides (PLA–GOs) were produced incorporating small amounts (0.1–1 wt %) of GO using acetone–chloroform and N,N-dimethylformamide–dichloromethane, two dual-solvent systems. The prepared PLA–GO carrier materials were characterized by FT-IR, Raman spectroscopy, and differential scanning calorimetry analysis, and their effects on rice growth within 20 days were also evaluated. When the amount of GO added was 0.5% and the solvent system was acetone–chloroform, the amount of pyraclostrobin (Pyr) loaded on PLA–GO (A-0.5%) was very high (39.89%). The PLA–GO pesticide microsphere showed a significantly sustained release of Pyr (up to 30 d). Furthermore, the indoor bioassay results showed that the PLA–GO pesticide microsphere (EC50 = 1.2682) had the best fungicidal activity against Rhizoctonia solani, followed by Pyr technical (EC50 = 2.3137) and commercial Pyr microcapsule suspension (EC50 = 3.4570). In conclusion, it was found that PLA–GOs prepared using the acetone–chloroform system were more suitable as a pesticide carrier, which provided a preliminary basis for GO to realize the coating of water-insoluble pesticides.
