Cell perforation is a critical step for intracellular drug delivery and real-time biosensing of intracellular signals. In recent years, the nanostraws system has been developed to achieve intracellular drug delivery with minimal invasiveness to the cells. Repeated cell perforation via nano-system could allow delivery of multiple drugs into cells for cell editing, but the biosafety is rarely explored. In this work, a nanostraw-mediated nano-electroporation system was developed, which allowed repeated perforation of the same set of cells in a minimally invasive manner, while the biosafety aspect of this system was investigated. Highly controllable fabrication of Al2O3nanostraw arrays based on a porous polyethylene terephthalate (PET) membrane was integrated with a microfluidic device to construct the nanostraw-electroporation system. The pulse conditions and intervals of nano-electroporation were systematically optimized to achieve efficient cells perforation and maintain the viability of the cells. The cells proliferation, the early apoptosis activities after nanostraw-electroporation and the changes of gene functions and gene pathways of cells after repeated nano-electroporation were comprehensively analyzed. These results revealed that the repeated nanostraw-electroporation did not induce obvious negative effects on the cells. This work demonstrates the feasibility of repeated nano-electroporation on cells and provides a promising strategy for future biomedical applications. Keywords:nanostraw device;nano-electroporation;repeated electroporation;cellular safety;microfluidic device

Single-cell metabolite analysis plays an important role in biological study. While mass spectrometry is a powerful tool for identification and quantitation of metabolites, the low absolute analyte amounts in single cell and difficulty in sampling represent significant challenges in single cell analysis. In this study, we developed an effective method with a simple sampling procedure for analyzing single cells. A single cell was driven to a capillary tip through electro-migration, followed by releasing the cell contents through electroporation, into a sealed small volume (∼1.5 pL) to prevent dilution. Subsequent mass spectrometry analysis was performed directly with nanoelectrospray ionization. This method was applied for analyzing a variety of cells and monitoring the metabolic changes in response to perturbed cell culturing conditions. This method opens a new avenue for easy and rapid analysis of single cells with high sensitivity.

Intestinal perforation is a serious medical emergency, and traditional surgery often causes adhesion and other complications. Innovative hydrogels improve postoperative care and rehabilitation with their anti-adhesion, antibacterial, and hemostatic properties. We have developed an advanced anti-adhesion hydrogel, AA-A30, composed of polypyrrole-modified gelatin (PPy-GelMA), carboxymethyl chitosan (CMCS), and NHS-functionalized polyethylene glycol (PEG-NHS). This hydrogel is specifically tailored for intestinal perforations. Upon hydrolysis, PEG-NHS forms a protective barrier that effectively prevents adhesion to surrounding normal tissues. Furthermore, the integration of PPy-GelMA significantly extends the degradation duration of the hydrogel, from 24 to 48 h. In a mouse model of intestinal perforation, the AA-A30 hydrogel demonstrated remarkable efficacy in inhibiting inflammation and preventing tissue adhesion by modulating the expression of both inflammatory and tissue adhesion-related factors, such as IL-1β, TNF-α, and the ratio of tPA to PAI-1. These findings underscore the considerable potential of AA-A30 for the therapeutic management of intestinal perforations.

Determining the mechanism that effects embolism repair in the xylem vessels of plants is of great significance in predicting plant distribution and the screening of drought-resistant plants. However, the mechanism of perforation plates of xylem vessels in the acceleration of embolism repair is still not clear by using conventional methods of anatomy and visualization technology. Microfluidic devices have shown their ability to simulate physiological environments and conduct quantitative experiments. This work proposes a biomimetic microfluidic device to study the mechanism of perforation plates of xylem vessels in the acceleration of embolism repair. The results proffered that the perforation plates increase the rate of embolism removal by increasing the pressure differential through the vessel, and the rate of embolism removal is related to the structural parameters of the perforation plate. A combination of the perforation size, the vessel diameter and the perforation plate angle can be optimised to generate higher pressure differentials, which can accelerate the process of embolism repair. This work provides a new method for studying the mechanism of microstructures of natural plants. Furthermore, the mechanism that perforation plates accelerate embolism repair was applied to an electrochemical flow sensor for online determination of heavy metal ions. Test results of this application indicate that the mechanism can be applied in the engineering field to solve the problems of reduced sensitivity of devices, inaccuracy of analysis results and poor reaction performance caused by bubbles that are generated or introduced easily in microdevices, which paves the way for applying the theory to engineering.

Covalent organic frameworks (COFs) with ordered arrays of sub-2 nm regular pores are drawing increasing attention in membrane separation, and it remains highly desirable for effective and controllable strategies to fabricate COF-based membranes. Herein, we demonstrate a perforated polymer-assisted transfer strategy enabling COF nanofilms for molecular separation. Solvothermal synthesis is used for the confined growth of TpPa, a stable, imine-linked COF, on the smooth surfaces of silicon substrates. Continuous, crystalline COF nanofilms are obtained, and their thicknesses are tunable in the range from a few tens to several hundred nanometers depending on monomer concentrations and reaction time. A block copolymer layer is coated on the COF nanofilms, which is then perforated to produce interconnected mesopores by the mechanism of selective swelling-induced pore generation. The perforated polymer coating functions as a protective but permeable layer enabling the easy transfer of the COF nanofilm onto porous substrates. Thus, we obtain a new type of composite membranes with the microporous COF nanofilm as the selective layer, sandwiched between the macroporous substrate and the mesoporous protective layer. The composite membranes exhibit good separation performances with water permeance up to ∼51 L m–2 h–1 bar–1 and high rejection rates to various dyes. This work demonstrates a new method to prepare COF-based membranes for molecular separation, and the invented perforated polymer-assisted transfer technology is expected to find applications in transferring other ultrathin materials to demanded substrates.

Irreversible electroporation (IRE) has emerged as a promising therapeutic modality for pancreatic cancer. However, traditional IRE techniques rely on high-voltage electric fields and require precise alignment of multiple electrodes, which complicates the procedure and increases associated risks. To address these challenges, we developed a novel “bipolar” IRE electrode that combines both the cathode and anode into a single device, simplifying the procedure and potentially reducing operational risks. Additionally, we incorporated a conductive metal–organic framework (MOF) to enhance the electric field distribution of the electric field, thereby improving the efficacy of tumor ablation. Mechanistic studies revealed that this combined approach induces tumor cell apoptosis and improves the consistency of ablation outcomes. Both in vitro and in vivo experiments demonstrated that the bipolar electrode, in combination with the conductive MOF, achieved a significant apoptosis rate of 59.95% ± 2.41 in vitro and resulted in an 85.77% ± 0.21 reduction in tumor volume in in vivo models, without any adverse effects. This approach provides a more optimized and potentially more effective solution for treating pancreatic cancer.

Achieving strong adhesion of bioadhesives on wet tissues remains a challenge and an acute clinical demand because of the interfering interfacial water and limited adhesive-tissue interactions. Here we report a self-gelling and adhesive polyethyleneimine and polyacrylic acid (PEI/PAA) powder, which can absorb interfacial water to form a physically cross-linked hydrogel in situ within 2 seconds due to strong physical interactions between the polymers. Furthermore, the physically cross-linked polymers can diffuse into the substrate polymeric network to enhance wet adhesion. Superficial deposition of PEI/PAA powder can effectively seal damaged porcine stomach and intestine despite excessive mechanical challenges and tissue surface irregularities. We further demonstrate PEI/PAA powder as an effective sealant to enhance the treatment outcomes of gastric perforation in a rat model. The strong wet adhesion, excellent cytocompatibility, adaptability to fit complex sites, and easy synthesis of PEI/PAA powder make it a promising bioadhesive for numerous biomedical applications.

It is quite necessary to develop a safe and efficient technique for disinfection of drinking water to avoid waterborne pathogens of infectious diseases. Herein, ZnO nanoarray electrodes with different sizes were investigated for low-voltage and high-efficiency electroporation disinfection. The results indicated that the ZnO nano-pyramid with small tip width and proper length exhibited over 99.9% disinfection efficiency against Escherichia coli under 1 V and a flow rate of 10 mL/min (contact time of 1.2 s). The suitable size of the nanoarray for electroporation disinfection was optimized by establishing the correlation between four kinds of ZnO nanoarrays and their efficiency of electroporation disinfection, which can guide the preparation of next-generation electroporation-disinfecting electrodes.

Pore formation can facilitate the release of various intracellular substances upon cell death, which is of critical benefit for tumor immunotherapy by immunogenic cell death (ICD). Given the effective endogenous antigen release is of primary importance for ICD-eliciting immunogenicity, other than by enhancing the cytotoxicity of ICD, we designed an immunogenic induction strategy via pore formation based on saponin, a composite medium of the membrane-disrupting agent. In this study, saponin/polyphenol (ZS-TA) at proper concentrations can directly cause membrane perforation by removal of cholesterol from the membrane, thereby lead to the release of intracellular substances. Simultaneously, in situ nano-antigens (nano-Ag) would be formed through the mechanism of protein-polyphenol interaction and the nano-Ag would be served as reservoirs of antigen to trigger long-term immune effects. Meanwhile, this membrane perforation could also enhance the uptake of chemotherapeutic drugs, which could be served as a general approach for drug delivery. Therefore, this work insights for the design of enhanced drug delivery system and in situ vaccines to sensitize tumor chemo-immunotherapy.

Conventional oxidative disinfection methods are usually inefficient to eliminate intracellular antibiotic resistance genes (i-ARGs) due to competitive oxidation of cellular components of antibiotic-resistant bacteria (ARB), resulting in the ubiquitous occurrence of ARGs in drinking water systems. Herein, we developed the strategy of coupling electroporation and electrochemical oxidation on a Co3O4-nanowires-modified electrode to destroy the multiresistant Escherichia coli cells and promote subsequent i-ARG (blaTEM-1 and aac(3)-II) degradation. The lightning-rod effect over nanowire tips can form finite regions with a locally enhanced electric field and highly concentrated charge density, in turn facilitating the electroporation for ARB cell damage and electrochemical reactivity for reactive chlorine/oxygen species generation. Characterization of the ARB membrane integrity and morphology revealed that electroporation-induced cell pores were further enlarged by the oxidation of reactive species, resulting in i-ARG removal at lower applied voltages and with 6–9 times lower energy consumption than the conventional electrochemical oxidation approach with a Co3O4-film-modified electrode. The satisfactory application and effective inhibition of horizontal gene transfer in tap water further demonstrated the great potential of our strategy in the control of the ARG dissemination risk in drinking water systems.

Acute hemorrhage that occurs after trauma is a life-threatening condition. Hence, to halt massive bleeding, there is a critical need to develop a suitable therapy. In this study, we developed self-propelling chestnut-like particles (Pro-MAS) comprising a macro-acanthosphere (MAS) coated with calcium carbonate and protonated tranexamic acid to puncture red blood cells (RBCs) and thus activate hemostasis. In vitro assessments revealed that Pro-MAS was biocompatible, biodegradable, and nontoxic; furthermore, it was capable of puncturing RBCs to release procoagulants and activate platelet aggregation for hemostasis. Animal tests showed that self-propelling Pro-MAS effectively traveled through blood flow to the deep ends of wounds; hemorrhage was controlled within 90 s and 4 min in the injured liver and bleeding femoral artery, respectively. Compared with a commercial hemostat, superior hemostasis was achieved with Pro-MAS, which could be ascribed to its functional and structural features. Overall, traveling Pro-MAS possessed sufficient impact force to puncture RBCs and sufficient momentum to reach the targeted bleeding sites. The present study demonstrated the ability of a novel platform, self-propelling MAS particles, to trigger hemostasis by puncturing RBCs. To the best of our knowledge, this is the first trial in which the release of endogenous procoagulants is promoted without the addition of exogenous procoagulants for severe hemorrhage control.

Pancreatic cancer is a deadly disease with little response to standard therapies. Irreversible electroporation (IRE) has emerged as a novel ablative technique for the clinical treatment of pancreatic cancer. Combinations of IRE and immunotherapies, including anti-programmed death 1 ( α PD1) immune checkpoint blockade, have shown promising efficacy in both preclinical and clinical studies. However, tumor recurrence remains an obstacle that needs to be overcome. It herein is shown that IRE induces a substantial infiltration of neutrophils into pancreatic tumors. These neutrophils are then polarized into a protumor phenotype by immunosuppressive cues, in particular transforming growth factor β (TGF- β ). Using glutathione-responsive degradable mesoporous silica nanoparticles loaded with SB525334, an inhibitor of TGF- β 1 receptor, it is demonstrated that local inhibition of TGF- β within the tumor microenvironment promotes neutrophil polarization into an antitumor phenotype, enhances pancreatic cancer response to combined IRE and α PD1 therapy, and induces long-term antitumor memory. The therapeutic efficacy is also attributed to tumor infiltration by CD8 + cytotoxic T cells, depletion of regulatory T cells, and maturation of antigen-presenting dendritic cells. Thus, modulating neutrophil polarization with nanomedicine is a promising strategy for treating pancreatic cancer.
