The role of polysaccharides in quality control of ginseng is underestimated. Large-scale comparison on the polysaccharides of Panax ginseng (PG), P. quinquefolius (PQ), P. notoginseng (PN), Red ginseng (RG), P. japonicus (ZJS), and P. japonicus var. major (ZZS), was performed by both chemical and biological approaches. Holistic fingerprinting at polysaccharide and the hydrolyzed oligosaccharide and monosaccharide levels utilized various chromatography methods, while OGD and OGD/R models on H9c2 cells were introduced to evaluate the protective effects on cell viability and mitochondrial function. Polysaccharides from six ginseng species exhibited remarkable content difference (RG > PG/ZZS/ZJS/PQ > PN), but weak differentiations in molecular weight distribution and oligosaccharide profiles, while Glc and GalA were richer for monosaccharide compositions of PG and RG polysaccharides, respectively. RG polysaccharides (25/50/100 μg/mL) showed significant cardiomyocyte protection by regulating mitochondrial functions. These new evidences may provide support for the supplementary role of polysaccharides in quality control of ginseng.

Background Heightened glycolysis-driven activation of pro-inflammatory M1 macrophages is a characteristic of rheumatoid arthritis (RA). Dysbiosis of the intestinal microbiota is considered a trigger for the immune response in RA. Panax japonicus is used in ethnomedicine to treat RA, but its molecular mechanisms regarding macrophage polarization and the intestinal microbiota remain to be explored. Methods In vitro and in vivo models were established using lipopolysaccharide (LPS)-induced RAW 264.7 macrophage cells and adjuvant-induced arthritis (AIA) rats, and treated with total saponins from Panax japonicus (TSPJ). Inflammatory cytokines were quantified by enzyme-linked immunosorbent assay (ELISA). Glucose, lactate, and LPS were measured by biochemical kits. The M1/M2a ratio in RAW 264.7 cells was assessed by flow cytometry. Joint bone erosion was evaluated by micro-CT. Immunohistochemistry and western blotting were used to evaluate glycolytic regulators in rat joints. Colonic contents and mucosa were analyzed by 16S rRNA sequencing. Results TSPJ inhibited inflammatory response, lactate production, and glucose consumption in M1 macrophages, and restored the M2a/M1 balance. In vivo , TSPJ alleviated joint swelling in AIA rats, reduced serum inflammatory cytokines, lactate, and LPS levels, while increasing anti-inflammatory cytokine levels. The protein expressions of hypoxia-inducible factor-1α (HIF-1α), glucose transporter 1 (GLUT1), hexokina se 2 (HK2), and lactate dehydrogenase A (LDHA) in the ankle joints were down-regulated after TSPJ intervention. Furthermore, TSPJ restored microbiota diversity and community structure, and enriched probiotic abundances like Eubacterium_coprostanoligenes_group and Christensenella . Conclusion TSPJ alleviates arthritis by inhibiting glycolysis pathway in M1 macrophages, modulating colon microbiota, and promoting the abundance of mucosa-associated probiotics.

Panacis Japonici Rhizoma (Zhu-Jie-Shen in Chinese), the root ofP. japonicusC.A. Mey., is commonly used in traditional Chinese Medicine. Saponins are the major bioactive compounds in this herb. The similarity of polarity and structure of the natural products in herb caused the difficulty of purification and resulted in the shortage and high cost of the reference compounds, which has greatly hindered efforts toward quantification in quality control. A novel strategy using a standardized reference fraction for qualification of the major saponins in Panacis Japonici Rhizoma was proposed to easily and effectively control the quality of PJR. The strategy is feasible and reliable, and the methodology of the developed approach is also validated. The standardized reference fraction was used for quantification, which might solve the shortage of the pure reference compounds in the quality control of herbal medicines. Keywords:Panax japonicusC. Ethnopharmacological relevance Panax japonicus C. A. Meye (PJ) has unique effects on diseases by “qi” stagnation and blood stasis in ancient. Modern studies have shown that PJ can treat diabetic kidney disease (DKD) caused by deficiency and blood stasis. Aim of the study This study evaluated the potential effects of PJ on DKD, a microvascular complication, and investigated its possible mechanisms. Materials and methods In this study, the chemical constituents of PJ were analyzed by HPLC. In vivo studies, we constructed a diabetic mice model by HDF combined with STZ , then administered PJ to diabetic mice for 6 weeks. Blood lipid, BUN, 24h urine protein, and renal tissue HE staining were detected to comprehensively evaluate the protective effect of PJ on DKD. Metabolomics investigated the metabolic pathways influenced by PJ in the treatment of DKD. Moreover, the potential targets and signal pathways were investigated using network pharmacology . Finally, molecular docking predicts affinity of active compounds and core targets, and western blotting was used to detect core target expression levels. Results In vivo study, PJ can reduce hyperlipidemia , serum BUN, and 24-h urinary protein in diabetic mice, and protect the pathological changes in renal tissue. Metabolomics results showed that PJ had significant regulatory effect on unsaturated fatty acids , glycerophospholipid metabolism, and purine metabolism. Network pharmacology showed that MAPK1, MAPK8, Bcl-2, and Caspase 3 were the core targets in PJ against DKD. Molecular docking revealed that Bcl-2 and Caspase 3 have a strong affinity for Chikusetsusaponin Iva , Ginsenoside Rb1 , and Ginsenoside Rg1 . Moreover, when compared to the model group, the PJ group had higher levels of anti-apoptosis protein Bcl-2 and lower levels of pro-apoptosis protein Caspase 3. Conclusion PJ can reduce blood lipids, regulate the biosynthesis of unsaturated fatty acids and purine metabolism, thereby alleviating the renal injury of diabetic mice. Moreover, it can regulate the Bcl-2/caspase 3 apoptosis signaling pathway to prevent the apoptosis of renal cells and protect the renal function of diabetic mice.
