Multichannel Sensor Arrays Based on Surface-Engineered Fluorescent Metal Clusterzymes for Differential Sensing of Cancer Cells

Liang Yunyan,Zhou Xiaomeng,Liu Wenfeng,Shang Li

Rapid and effective differentiation of cancerous cells is essential for early diagnosis and treatment of tumors. Here, a multichannel sensor array is developed based on four surface-engineered gold-silver nanoclusters (AuAgNCs) to discriminate multiple cancerous and normal cells. Different binding affinities between various cells and AuAgNCs lead to changes in the intrinsic fluorescence of AuAgNCs as well as their peroxide-like activity. Based on the distinct response of dual fluorescence signals of four AuAgNCs, a multichannel sensor array is established, which is capable of accurately identifying both normal and cancerous cells, even cells of different subtypes and homologous cells from various individuals with high sensitivity. Moreover, this fluorescent clusterzyme-based sensor array enables quantitative detection of cancerous cells (e.g., HeLa cells) with good performance, which holds great potential as a promising platform for further cancer diagnosis.

An enzymatic activity regulation-based clusterzyme sensor array for high-throughput identification of heavy metal ions

Yueyuan Li,Zhaode Mu,Yonghua Yuan,Jing Zhou,Lijuan Bai,Min Qing

The accurate identification and sensitive quantification of heavy metal ions are of great significance, considering that pose a serious threat to environment and human health. Most array-based sensing platforms, to date, utilize nanozymes as sensing elements, but few studies have explored the application of the peroxidase-like activity of clusterzymes in identification of multiple analytes. Herein, for the first time, we developed a clusterzyme sensor array utilizing gold nanoclusters (AuNCs) as sensing elements for five heavy metal ions identification including Hg 2+ , Pb 2+ , Cu 2+ , Cd 2+ and Co 2+ . The heavy metal ions can differentially regulate the peroxidase-like activity of AuNCs, and that can be converted into colorimetric signals with 3,3′,5,5′-tetramethylbenzidine (TMB) as the chromogenic substrate. Subsequently, the generated composite responses can be interpreted by combining pattern recognition algorithms. The developed clusterzyme sensor array can identify five heavy metal ions at concentrations as low as 0.5 μM and their multi-component mixtures. Especially, we demonstrated the successful identification of multiple heavy metal ions in tap water and traditional Chinese medicine, with an accuracy of 100% in blind test. This study provided a simple and effective method for identification and quantification of heavy metal ions, rendering a promising technique for environmental monitoring and drug safety assurance.

Atomic-Level Engineering of Protein-Templated Gold Clusterzymes with Tunable Fluorescence and Multi-Enzyme Activities for Antiaging Treatment

Ziyu Kuai,Yibing Huang,Fengjie Hao,Wantong Jiang,Mengmeng Niu,Yu qin,Ao Song,Yuran Deng,Youzhong Wan,Quan Luo

Atomic precision synthesis and modulation of nanozymes are critical for evaluating their performance and developing functional applications. Herein, we developed an in situ strategy for the controlled synthesis of gold clusterzymes (AuCEs) within the confined cavities of the protein scaffolds. Using a stable dodecameric protein (SP1) as a ligand, we fused a short peptide containing the CCY sequence (SP1-CCY) to the N-terminus of SP1, which is located within its cyclic structure. In this design, cysteine and tyrosine synergistically chelate and reduce Au ions, while the confined protein structure restricts cluster growth, leading to the formation of a 25-gold atom (Au25) nanocluster with strong fluorescence and peroxidase-like activity. Unlike chemical ligands, protein scaffolds offer atomic-level precision and allow for site-specific modulation of the local environment through rational mutations, such as L9D, L9I, and E73I/L9I. This allows precise control of both fluorescence and enzymatic activity via protein engineering. Additionally, heteroatom doping of the Au core with copper endows the AuCEs with significant superoxide dismutase-like activity. Antioxidant assays demonstrate that the optimized AuCEs exhibit potent reactive oxygen species scavenging activity, providing significant protection to Caenorhabditis elegans from oxidative stress and highlighting their potential as therapeutic candidates for aging-related diseases.

Catalytically potent and selective clusterzymes for modulation of neuroinflammation through single-atom substitutions

Liu Haile,Li Yonghui,Sun Si,Xin Qi,Liu Shuhu,Mu Xiaoyu,Yuan Xun,Chen Ke,Wang Hao,Varga Kalman,Mi Wenbo,Yang Jiang,Zhang Xiao-Dong

Emerging artificial enzymes with reprogrammed and augmented catalytic activity and substrate selectivity have long been pursued with sustained efforts. The majority of current candidates have rather poor catalytic activity compared with natural molecules. To tackle this limitation, we design artificial enzymes based on a structurally well-defined Au 25 cluster, namely clusterzymes, which are endowed with intrinsic high catalytic activity and selectivity driven by single-atom substitutions with modulated bond lengths. Au 24 Cu 1 and Au 24 Cd 1 clusterzymes exhibit 137 and 160 times higher antioxidant capacities than natural trolox, respectively. Meanwhile, the clusterzymes demonstrate preferential enzyme-mimicking catalytic activities, with Au 25 , Au 24 Cu 1 and Au 24 Cd 1 displaying compelling selectivity in glutathione peroxidase-like (GPx-like), catalase-like (CAT-like) and superoxide dismutase-like (SOD-like) activities, respectively. Au 24 Cu 1 decreases peroxide in injured brain via catalytic reactions, while Au 24 Cd 1 preferentially uses superoxide and nitrogenous signal molecules as substrates, and significantly decreases inflammation factors, indicative of an important role in mitigating neuroinflammation.

A Biomimetic Nanoplatform with Improved Inflammatory Targeting Behavior for ROS Scavenging-Based Treatment of Ulcerative Colitis

Xiaofen Wang,Menglu Jiao,Fangzhen Tian,Xuan Lu,Huihuang Xiong,Fan Liu,Yiqun Wan,Xiaodong Zhang,Hao Wan

Ulcerative colitis (UC), a refractory disease, has become a global problem. Herein, a biomimetic nanoplatform (AU-LIP-CM) comprising Au cluster enzymes (AU)-loaded liposomes (AU-LIP) camouflaged with the fusion membrane (CM) consisting of neutrophil (NC) and red blood cell (RBC) membrane was designed for the treatment of UC. Briefly, revealed by second near-infrared (NIR-II) imaging through collection of fluorescence emitting > 1200 nm from AU, the improved inflammatory targeting behavior contributed by CM cloaking, which inherited abilities of inflammatory targeting and immune escape from NC and RBC, respectively, promoted specific accumulation of AU within inflammatory intestines with up to ∼11.5 times higher than that of bare AU. Afterwards, AU possessing superoxide dismutase- and catalase-like activities realized high-efficiency scavenging of reactive oxygen species (ROS), leading to repair of intestinal barriers, regulation of the immune system, and modulation of gut microbiota, which surpassed first-line UC drug. In addition, study of underlying therapeutic mechanism demonstrated the treatment with AU-LIP-CM could alter the gene signature associated with response to ROS for UC mice to a profile similar to that of healthy mice, deciphering related signal pathways. The strategy developed here provided insights of learning from properties of natural bio-substances to empower biomimetic nanoplatform to confront diseases. This article is protected by copyright.
