Autophagy in human skin squamous cell carcinoma: Inhibition by 3-MA enhances the effect of 5-FU-induced chemotherapy sensitivity

Lili Zhang,Jie Zhang,Li Chen,Jianli Wang

Autophagy is an intracellular multi-step catabolic degradation process that involves the degradation and recycling of cellular proteins and cytoplasmic damaged organelles to maintain cellular homeostasis and reduction of metabolic stress. Numerous studies have indicated the importance of autophagy in cancer, but the role of autophagy in human skin squamous cell carcinoma (SSCC) development and response to therapy it is still unclear. In the present study, we investigated the role of autophagy in SSCC and the relationship with chemotherapy sensitivity. The present study demonstrated that autophagy related gene the microtubule-associated protein 1 light chain 3 (LC3) expression was low in SSCC. The negative correlation with Bcl2 and survivin, and the chemotherapy drug 5-FU increased the level of autophagy and the autophagy inhibitor 3-MA inhibited this effect in SSCC cells, time- and dose-dependently. When SSCC cells were treated first with 3-MA and then with 5-FU, the inhibition of proliferation, migration, invasion and apoptosis of SSCC cells was enhanced. Our results suggested the possibility of autophagy as a potential target in SSCC therapy and 3-MA and 5-FU combination treatment may be an effective SSCC therapy via autophagy modulating.

Stretchable, Washable, and Ultrathin Triboelectric Nanogenerators as Skin-Like Highly Sensitive Self-Powered Haptic Sensors

Yang Jiang,Kai Dong,Xin Li,Jie An,Dequan Wu,Xiao Peng,Jia Yi,Chuan Ning,Renwei Cheng,Pengtao Yu,Zhong Lin Wang

Accompanying the boom in multifunctional wearable electronics, flexible, sustainable, and wearable power sources are facing great challenges. Here, a stretchable, washable, and ultrathin skin-inspired triboelectric nanogenerator (SI-TENG) to harvest human motion energy and act as a highly sensitive self-powered haptic sensor is reported. With the optimized material selections and structure design, the SI-TENG is bestowed with some merits, such as stretchability ( ≈ 800%), ultrathin ( ≈ 89 µ m), and light-weight ( ≈ 0.23 g), which conformally attach on human skin without disturbing its contact. A stretchable composite electrode, which is formed by homogenously intertwining silver nanowires (AgNWs) with thermoplastic polyurethane (TPU) nanofiber networks, is fabricated through synchronous electrospinning of TPU and electrospraying of AgNWs. Based on the triboelectrification effect, the open-circuit voltage, short-circuit current, and power density of the SI-TENG with a contact area of 2 × 2 cm 2 and an applied force of 8 N can reach 95 V, 0.3 µ A, and 6 mW m −2 , respectively. By integrating the signal-processing circuits, the SI-TENG with excellent energy harvesting and self-powered sensing capability is demonstrated as a haptic sensor array to detect human actions. The SI-TENG exhibits extensive applications in the fields of human–machine interface and security systems.

One-step fabrication of cell sheet-laden hydrogel for accelerated wound healing

Huijuan Wang,Deshun Sun,Weiming Lin,Chao Fang,Kui Cheng,Zhengzhou Pan,Daping Wang,Zhangfa Song,Xiaojun Long

Full-thickness skin wounds are have continued to be reconstructive challenges in dermal and skin appendage regeneration, and skin substitutes are promising tools for addressing these reconstructive procedures. Herein, the one-step fabrication of a cell sheet integrated with a biomimetic hydrogel as a tissue engineered skin for skin wound healing generated in one step is introduced. Briefly, cell sheets with rich extracellular matrix, high cell density, and good cell connections were integrated with biomimetic hydrogel to fabricate gel + human skin fibroblasts (HSFs) sheets and gel + human umbilical vein endothelial cells (HUVECs) sheets in one step for assembly as a cell sheet-laden hydrogel (CSH). The designed biomimetic hydrogel formed with UV crosslinking and ionic crosslinking exhibited unique properties due to the photo-generated aldehyde groups, which were suitable for integrating into the cell sheet, and ionic crosslinking reduced the adhesive force toward the substrate. These properties allowed the gel + cell sheet film to be easily released from the substrate. The cells in the harvested cell sheet maintained excellent viability, proliferation, and definite migration abilities inside the hydrogel. Moreover, the CSH was implanted into a full-thickness skin defects to construct a required dermal matrix and cell microenvironment. The wound closure rate reached 60.00 ± 6.26% on the 2nd day, accelerating mature granulation and dermis formation with skin appendages after 14 days. This project can provide distinct guidance and strategies for the complete repair and regeneration of full-thickness skin defects, and provides a material with great potential for tissue regeneration in clinical applications.

An Integrated Monolithic Synaptic Device for C-Tactile Afferent Perception and Robot Emotional Interaction

Yue Li,Lu Yang,Qianbo Yu,Yi Du,Ning Wu,Wentao Xu

C-tactile afferents are low-threshold mechanoreceptors that innervate the hairy skin of mammals, essential for emotional interactions. Replication of such a mechanism could facilitate emotional interactions between humans and embodied intelligence robotic systems. Herein, we demonstrate a monolithic synaptic device that replicates and integrates tactile sensing and neuromorphic processing functions for in-sensor computing. The device is operable by both mechanical and electrical inputs, with the mechanoelectrical operation mechanism stemming from the synergistic effect of dynamic ionic migration and injection. As a proof of concept, the device effectively converts spatiotemporal tactile stimuli into distinct electrical signals, which are subsequently encoded to enable the microcomputer to classify multiple discrete emotional states, such as happiness, calmness, and excitement. This monolithic integrated device, which converges mild tactile perception with neuromorphic processing, with high tactile sensitivity and low-energy consumption, establishes an approach for emotional interaction between intelligent robots and human beings.

A rapid self-healing hydrogel based on PVA and sodium alginate with conductive and cold-resistant properties

Guangfeng Wu,Kaiyun Jin,Li Liu,Huixuan Zhang

Self-healing hydrogels as renewable materials have attracted significant attention recently. However, traditional self-healing hydrogels require a long time for self-healing and cannot be used at low temperatures. Besides, their poor biocompatibility limits the application of hydrogels. Herein, we have prepared a hydrogel composed of polyvinyl alcohol (PVA) and modified sodium alginate. Due to the dynamic recombination of borate bonds, these hydrogels show a strong self-healing ability, with the shortest self-healing time up to 15 seconds. Also, glycerin (GI) was added into the hydrogel to increase the cold resistance of the hydrogel. At −25 °C, the hydrogel still displayed good frost resistance and elasticity. NaCl and other inorganic salts were added to endow the hydrogel with good electrical conductivity. The hydrogel also had good skin-like properties and could activate the capacitive screen of an electronic device.

A skin-matchable, recyclable and biofriendly strain sensor based on a hydrolyzed keratin-containing hydrogel

Yang Gao,Song Gu,Fei Jia,Guanghui Gao

The current hydrogel sensors have mostly met diverse requirements including mechanical adaptability, rapid responsiveness and strain sensitivity. However, the wearing comfort, long-term safe integration and environmental friendliness have received negligible attention. Here, a novel type of skin-matchable, recyclable and biofriendly sensor based on a hydrolyzed keratin-containing polyvinyl alcohol hydrogel was reported. The hydrogel demonstrated skin-like mechanical properties of a low modulus, high toughness, superior strength and persistent anti-fatigue-fracture ability (no crack after 1000 cycles) to minimize wearing discomfort and guarantee endurance. A wearable sensor based on the hydrogel conductor possessed ion-transporting behavior close to that of organism, remarkable strain-sensitivity (GF = 4.92) with a low detectable strain of 0.25%, rapid responsiveness, negligible hysteresis and operational durability, which were favorable for providing whole-body motion signals continuously and discerning action status precisely. Notably, the hydrogel sensor had incomparable biocompatibility for achieving safe integration with skin. Furthermore, the hydrogel sensor was fully recyclable, and could be re-used in new electronic devices without sacrificing the original function to prevent the production of electronic waste. It is believed that the simple design concept of this work will provide a new horizon for the development of “green” wearable and implantable electronic devices in the future.

Intrinsically stretchable light-emitting drawing displays

Zhang Jiaxue,Lu Qianying,Wu Ming,Sun Yuping,Wang Shaolei,Wang Xiaoliang,Lu Ming-Hui,Kong Desheng

Stretchable displays that combine light-emitting capabilities with mechanical compliance are essential building blocks of next-generation wearable electronics. However, their widespread applications are currently limited by complex device architecture, limited pixel density, and immature fabrication processes. In this study, we present the device design and material developments of intrinsically stretchable light-emitting drawing displays that can show arbitrary hand-drawing features. The alternating-current electroluminescent display uses a simplified architecture comprising coplanar interdigitated liquid metal electrodes, an electroluminescent layer, and a dielectric encapsulation layer. Ink patterns on the device are coupled with the interdigitated electrodes under alternating voltage stimulations, generating localized electric fields for bright emissions. Various inks are prepared for painting, stamping, and stencil printing. Arbitrary luminous features on the devices can be either long-lasting or transient in characteristics. These skin-like devices are made entirely of compliant materials that can withstand bending, twisting, and stretching manipulations. Due to the excellent mechanical deformability, the drawing displays can be conformally laminated on the skin as body-integrated optoelectronic communication devices for graphic information.

Human Skin-Like, Robust Waterproof, and Highly Breathable Fibrous Membranes with Short Perfluorobutyl Chains for Eco-Friendly Protective Textiles

Jing Zhao,Xianfeng Wang,Lifang Liu,Jianyong Yu,Bin Ding

Flexible smart membranes with superior waterproofness and extreme breathability are highly desirable for wearable uses. However, present waterproof and breathable materials suffer from limited performance efficiency, alarming environmental risks, and complicated fabrication procedures. We report on eco-friendly fibrous membranes with human skin-like, robust waterproof, and highly breathable capabilities that can be prepared via a facile electrospinning strategy. A novel polyurethane elastomer (C4FPU) possessing double-terminal short perfluorobutyl (−C4F9) chain is synthesized for the first time and incorporated into the polyurethane (PU) fiber matrix, endowing the membrane with mighty and durable hydrophobicity. Additionally, the employment of AgNO3 greatly decreased the maximum pore size (dmax), contributing to the dramatically enhanced waterproofness. The resulting PU/C4FPU/AgNO3 fibrous membranes exhibit comprehensive properties of exceptional hydrostatic pressure (102.8 kPa), excellent water vapor transmission rate (12.9 kg m–2 d–1), high mechanical property (9.8 MPa), and significant antibacterial efficacy against Escherichia coli and Staphylococcus aureus. The successful synthesis of these intriguing membranes may provide a promising candidate for the new generation of key building blocks of the upscale protective garments.

Tribuloside acts on the PDE/cAMP/PKA pathway to enhance melanogenesis, melanocyte dendricity and melanosome transport

Yan Cao,Jinpeng Lv,Yan Tan,Ruolin Chen,Xiaoxue Jiang,Duo Meng,Kun Zou,Min Pan,Liming Tang

Ethnopharmacological relevance Tribuloside, a natural flavonoid extracted from Chinese medicine Tribulus terrestris L., has shown potent efficacy in treating various diseases. In China, the fruits of Tribulus terrestris L. have long been utilized for relieving headache, dizziness , itchiness, and vitiligo . Water-based extract derived from Tribulus terrestris L. can enhance melanogenesis in mouse hair follicle melanocytes by elevating the expression of α-melanocyte stimulating hormone (α-MSH) and melanocortin-1 recepter (MC-1R). Nevertheless, there is a lack of information regarding the impact of tribuloside on pigmentation in both laboratory settings and living organisms. Aim of the study The present research aimed to examine the impact of tribuloside on pigmentation, and delve into the underlying mechanism. Materials and methods Following the administration of tribuloside in human epidermal melanocytes (HEMCs), we utilized microplate reader, Masson-Fontana ammoniacal silver stain, transmission electron microscopy (TEM) and scanning electron microscopy (SEM) to measure melanin contents, dendrite lengths, melanosome counts; L-DOPA oxidation assay to indicate tyrosinase activity, Western blotting to evaluate the expression of melanogenic and associated phosphodiesterase (PDE)/cyclic adenosine monophosphate (cAMP)/cyclic-AMP dependent protein kinase A (PKA) pathway proteins. A PDE-Glo assay to verify the inhibitory effect of tribuloside on PDE was also conducted. Additionally, we examined the impact of tribuloside on the pigmentation in both zebrafish model and human skin samples. Results Tribuloside had a notable impact on the production of melanin in melanocytes, zebrafish, and human skin samples. These functions might be attributed to the inhibitory effect of tribuloside on PDE, which could increase the intracellular level of cAMP to stimulate the phosphorylation of cAMP-response element binding (CREB). Once activated, it induced microphthalmia-associated transcription factor (MITF) expression and increased the expression of tyrosinase, Rab27a and cell division cycle protein 42 (Cdc42), ultimately facilitating melanogenesis, melanocyte dendricity, and melanin transport. Conclusion Tribuloside acts on the PDE/cAMP/PKA pathway to enhance melanogenesis, melanocyte dendricity, and melanosome transport; meanwhile, tribuloside does not have any toxic effects on cells and may be introduced into clinical prescriptions to promote pigmentation.

A hybrid binder of carboxymethyl chitosan and l-methionine enables a slight amount of Ag NPs to be durably effective on antibacterial cotton fabrics

Zhou Jing,Hu XinYue,Zhu YiYuan,Lyu HongFen,Zhang Lei,Fu FeiYa,Liu XiangDong

This work proposes a facile approach to improve the durability of antibacterial cotton fabric by using carboxymethyl chitosan (CMC) and l -methionine (Met) as a combination binder to immobilize silver nanoparticles (Ag NPs). The durable antibacterial function is achieved by grafting CMC and Met molecules onto a cotton fabric by a simple pad-dry-cure process, followed by the preparation of Ag NPs on the modified surface. This surface modification process provides the cotton fabric with an excellent antibacterial effect and outstanding laundering durability. Structural analyses of the modified surface revealed that the covalent bonds between Met, CMC, and cotton fibers were formed, and the thioether groups caused the stable immobilization of the Ag NPs. Notably, the modification process does not significantly damage the original fiber structure. The desired cotton properties such as vapor permeability, water absorptivity, and flexibility of the modified fabric were very close to the original cotton. Moreover, the safety to human skin was verified by cytotoxicity tests using fibroblast cells.

Colorimetric Ionic Organohydrogels Mimicking Human Skin for Mechanical Stimuli Sensing and Injury Visualization

Wenlian Qiu,Changgeng Zhang,Guoqing Chen,He Zhu,Qi Zhang,Shiping Zhu

Artificial skins with sensing ability have great potential in applications of wearable devices and soft robotics. Inspired by the functions of human skins including sensing stimuli via electrical signal and bruising for injury indication, an ionic conductive and mechanochromic organohydrogel is synthesized and demonstrated as ionic skin (I-skin). The gel consisting of mechanochromophore cross-linked micelles is mechanically robust, stretchable, and deformation durable with minor hysteresis, and it also displays good solvent retention. The change of relative resistance during elongation and compression suggests a high sensitivity. An optical change from pale yellow to bruise-like blue-purple color is observed under a large deformation. The ionic conductive organohydrogel as I-skin is attached to different parts of the human body with movements mimicking various body-bruising scenarios, demonstrating successful perception and visualization of mechanical stimuli. The work vividly presents a strain sensor with the functions of injury visualization and damage warning for mechanical impacts. The I-skin can be potentially used in the applications including prosthetic devices, wearable electronics, and intelligent robots.

Dopamine-Triggered Hydrogels with High Transparency, Self-Adhesion, and Thermoresponse as Skinlike Sensors

Chao Zhang,Yongsen Zhou,Haijie Han,Huanxi Zheng,Wanghuai Xu,Zuankai Wang

Mussel-inspired conductive hydrogels are attractive for the development of next-generation self-adhesive, flexible skinlike sensors. However, despite extensive progress, there are still some daunting challenges that hinder their applications, such as inferior optical transparency, low catechol content (e.g., poor adhesion), as well as limited sensation performances. Here, we report a dopamine-triggered gelation (DTG) strategy for fabricating mussel-inspired, transparent, and conductive hydrogels. The DTG design leverages on the dual functions of dopamine, which serves as both polymerization initiator and dynamic mediator to elaborate and orchestrate the cross-linking networks of hydrogels, allowing for pronounced adhesion, robust elasticity, self-healing ability, excellent injectability and three-dimensional printability, reversible and tunable transparent-opaque transition, and thermoresponsive feature. These preferable performances enable DTG hydrogels as self-adhesive, flexible skinlike sensors for achieving multiple sensations toward pressure, strain, and temperature, even an extraordinary visual perception effect, making it a step closer in the exploration of future biomimetic skin.
