Tailored multilayer nanoparticles against resistant P. aeruginosa by disrupting the thickened mucus, dense biofilm and hyperinflammation

Mao Xu,Shihui Yu,Pengyu Li,Yili Chen,Yujun Chen,Jieyi Pan,Xin Deng,Haiyan Hu

Therapeutic challenges of chronic pulmonary infections caused by multidrug-resistant Pseudomonas aeruginosa (MDR P. aeruginosa ) biofilms due to significantly enhanced antibiotic resistance. This resistance is driven by reduced outer membrane permeability, biofilm barriers, and excessive secretion of virulence factors. Thickened mucus in the airways exacerbates the problem by impeding antibiotic penetration, providing a breeding ground for biofilms, consequently aggravating infection. Moreover, biofilms recruit numerous immune cells, resulting in chronic inflammation and lung tissue damage. In turn, damaged airway further facilitates bacterial colonization and elevated mucus production. To thoroughly disintegrate the stubborn triad of “thickened mucus & dense biofilm & excessive inflammation” and address drug resistance, tailored multilayer nanoparticles (NPVC/PBIP NPs) were developed. NPVC/PBIP NPs were engineered through self-assembly of vanillin-chitosan amphiphilic polymer loading polymyxin B-linoleic acid ion pairs in. Then polyaspartic acid and N -acetylcysteine-ε-poly- l -lysine were coated by layer-by-layer on the surface of vanillin-chitosan NPs via electrostatic interactions. As expected, the NAC units on NPVC/PBIP NPs effectively thinned human clinical sputum and porcine sputum, resulting in rapid sputum penetration followed by biofilm permeation. NPVC/PBIP NPs achieved over 99 % eradication of mature biofilms in vitro . Furthermore, they effectively inhibited virulence factors production and bacteria re-adhesion (biofilm reformation) while exhibiting superior anti-inflammatory and antioxidant activities. In a chronic pulmonary infection model, NPVC/PBIP NPs remarkably thinned airway mucus, reduced bacterial burden by 99.7 %, alleviated inflammatory cell infiltration, and minimized lung tissue damage. In summary, the NPVC/PBIP NPs represent a novel and promising strategy to manage MDR P. aeruginosa biofilms associated infections by disintegrating the stubborn triad of “thickened mucus & dense biofilm & excessive inflammation”.

Mucilage-capped silver nanoparticles for electrochemical quantification of methyl red in drinking water samples

Moustafa Zahran,Shereen Gamal,Magdy A.-H. Zahran,Magdi Abdel Azzem

Herein, we report a new electrochemical sensor based on silver nanoparticles (AgNPs) to quantify methyl red (MR) in drinking water. Firstly, AgNPs were greenly synthesized using mucilage natural polymer for the reduction of Ag + and subsequent stabilization of the formed AgNPs. The formation of AgNPs was confirmed by monitoring their characteristic absorption peak at a wavelength of 425 nm and characteristic oxidation potential at 0.035 V using ultraviolet–visible spectroscopy and square wave voltammetry (SWV), respectively. Transmission electron microscopy also confirmed the synthesis of spherical AgNPs with a size range of 10 to 30 nm, while scanning electron microscopy showed the morphology of glassy carbon electrode (GCE) after modification with AgNPs. Additionally, the electrocatalytic activity of AgNPs/GCE was approved by cyclic voltammetry, electrochemical impedance spectroscopy, and SWV. The highly electrocatalytic mucilage-AgNPs were employed as a GCE modifier to quantify MR under optimized conditions of scan rate (0.1 V/s), amplitude (0.025 V), and pH (6) using SWV. The sensor showed a limit of detection of 1.5 picomolar and high selectivity in the presence of some possible interfering dyes in drinking water sources. Additionally, after spiking the drinking water samples with MR, the sensor showed acceptable recovery values.

Hydrophilic and Electroneutral Nanoparticles to Overcome Mucus Trapping and Enhance Oral Delivery of Insulin

Xinyi Tan,Na Yin,Zixu Liu,Rong Sun,Jingxin Gou,Tian Yin,Yu Zhang,Haibing He,Xing Tang

The oral delivery of macromolecules using nanoparticles is limited by secreted mucus, resulting in low contact or internalization via intestinal cells and, thus, both mucus trapping and further low cellular uptake need to be overcome. Here, hydrophilic and electroneutral nanoparticles were developed to overcome mucus trapping and enhance the oral delivery of macromolecules. Mesoporous silica nanoparticles (MSNs) were synthesized and modified with a hydrophilic block polymer (poly(lactic acid)–methoxy poly(ethylene glycol), PLA–PEG), and then an overall electroneutrality and promoted cellular uptake were achieved by sequential modification with cell-penetrating peptides (CPPs). Reduced hydrophobic and electrostatic interactions of MSN@PLA–PEG-CPP with mucus decreased mucus trapping by 36.0%, increased the cellular uptake of MSN@PLA–PEG-CPP by 2.3-folds in mucous conditions via active heparan sulfate proteoglycan receptor (HSPG)-mediated and caveolae-mediated endocytosis and electrostatic interactions. Furthermore, insulin, a model macromolecular drug, was successfully loaded into the nanoparticles (INS@MSN@PLA–PEG-CPP). Compared with insulin solution, in vitro cellular uptake in mucous conditions and in vivo pharmacodynamic effects were significantly increased by 9.1- and 14.2-folds, respectively. As well, all nanoparticles with or without insulin loading presented negligible in vitro and in vivo toxicity. Herein, hydrophilic and electroneutral nanoparticles with sequential PEG and CPP modification could promote cellular uptake against mucus trapping and finally show good prospects for oral insulin delivery.

Natural okra gum as functional binder enables highly stable Lithium–Selenium batteries

Yang Xia,Jun Tong,Chengwei Lu,Xinping He,Yongping Gan,Hui Huang,Jun Zhang,Xinhui Xia,Wenkui Zhang,Zhen Xiao,Ruyi Fang

Lithium–selenium (Li–Se) batteries are garnered significant attention due to their high theoretical specific capacity, high volumetric energy density and moderate output voltage. Nevertheless, Li–Se batteries are still restricted by severe volume expansion and “shuttle effect” of polyselenides, resulting in deteriorated electrochemical performance . Binders are a crucial component of Li–Se batteries, playing a key role in addressing the mentioned issues. Herein, a non-toxic, bio-friendly, hydro-soluble binder of okra gum (OG) extracted from okra pods is proposed to ameliorate the electrochemical performance of Li–Se batteries. The as-prepared OG binders are rich in branched-chain polysaccharides (glucoside) and polar functional groups (–C–O–C–, –COOH), which not only have a good interpenetrating binding network to ensure the mechanical stability of Se cathode, but also have excellent interfacial chemical binding force for immobilizing polyselenides. Consequently, the OG binder endows Li–Se batteries with high initial discharge capacity (607 mA h g −1  at 0.2C), good cycling stability (218 mA h g −1 after 100 cycles at 0.2C), and remarkable rate performance (364 mA h g −1  at 1C), all of which are superior to Li–Se batteries with PVDF binder. Our study demonstrates the feasibility of natural mucilage as functional binder for Li–Se batteries and paves a research path in developing natural plant-based binders to stabilize electrode materials for reliable high-energy batteries.

A Self-Cleaning Mucus-like and Hierarchical Ciliary Bionic Surface for Marine Antifouling

Xiaoting Ren,Mingshuai Guo,Lili Xue,Qingyang Zeng,Xianze Gao,Yonglei Xin,Likun Xu,Li Li

The flexible mucus film and hydrophilic hierarchical micro-/nanoprotuberance structure on the skin surfaces of marine organisms contribute to an excellent intrinsic antifouling performance. Inspired by this fact, a self-cleaning mucus-like and hierarchical ciliary bionic antifouling surface (SMCAS) is designed for marine antifouling based on the electrostatic flocking technology. The results of scanning electron microscopy (SEM) show that the bioinspired SMCAS has a hierarchical micro-/nanociliary structure and the micrometer array of polyamide (PA) microfibers is covered with the nanometer carbon nanotube/polyvinyl alcohol (CNT/PVA) hydrogel particles. The biomasses of the two marine microoganisms, Nitzschia closterium f. minutissima and Marinobacter lipolyticus SM19 (T), attached on the SMCAS are investigated using a UV–visible spectrophotometer (UV–vis) and SEM. The results reveal that the microoganisms attached on the SMCAS are significantly lower number in comparison with the organic silicon coating. In addition, the SMCAS has a greater ability of stain removal in the dyed petroleum ether washing test. Therefore, the SMCAS demonstrates an extraordinary antifouling performance due to the synergetic effect of the mucus-like hydrogel film and the hierarchical micro-/nanostructured micropillars.

Modified β-CD-Cu ion complex and yam mucilage assisted batch foam fractionation for separating puerarin from Ge-gen (Radix puerariae)

Wei Liu,Zhaoliang Wu,Yanji Wang,Huijie Zheng,Hao Yin

At present, the large scale application of batch foam fractionation is still in confrontation with many problems and difficulties, especially for the enrichment of non-surface-active materials. It is difficult to select a suitable surfactant as the collector when a non-surface-active material can only form a stable association with the expensive surfactants or has not any interactions with the commercially available surfactants. The objective of this work was to investigate the feasibility of batch foam fractionation for recovering such a non-surface-active material, puerarin. Yam mucilage, an abundant byproduct from the manufacture of yam starch, was used to generate a stable foam phase based on its good foamability. According to the properties of puerarin and yam mucilage, β -cyclodextrin ( β -CD) was selected as a basic material to establish an association between them. The results of fluorescence sensitization experiments indicated that puerarin could form an equimolar inclusion complex with β -CD and the inclusion constant was 405.8. Subsequently, β -CD was modified by introducing carbon disulfide and copper (Cu) ion. Then, puerarin could be adsorbed on the gas-liquid interface in the form of ternary complex with modified β -CD-Cu ion complex and glycoprotein molecules (see Fig. 7). Batch foam fractionation was carried out by using the crude extracting solution as the initial feeding solution, in which the concentration of puerarin was as high as 4.6   g/L. Under the suitable conditions of temperature 50   °C, pH 6.0, loading volume of feed solution 400   mL, the pore diameter of gas distributor 0.450   mm, additive volume of yam mucilage 30   mL and volumetric air flow rate 100   mL/min, the enrichment ratio and the percent recovery of puerarin were 4.3 and 82.3%, respectively. This strategy could be generalized to the separation of various non-surface-active materials due to the low cost and the diversiform cavity dimension of cyclodextrins.

Synthesis of CSK-DEX-PLGA Nanoparticles for the Oral Delivery of Exenatide to Improve Its Mucus Penetration and Intestinal Absorption

Yina Song,Yanan Shi,Liping Zhang,Haiyan Hu,Chunyan Zhang,Miaomiao Yin,Liuxiang Chu,Xiuju Yan,Mingyu Zhao,Xuemei Zhang,Hongjie Mu,Kaoxiang Sun

The oral absorption of exenatide, a drug for type 2 diabetes treatment, can be improved by using nanoparticles (NPs) for its delivery. To improve the mucus penetration and intestinal absorption of exenatide, we designed a block copolymer, CSKSSDYQC-dextran-poly(lactic-co-glycolic acid) (CSK-DEX-PLGA), and used it for the preparation of exenatide-loaded NPs. The functionalized exenatide-loaded NPs composed of CSK-DEX-PLGA were able to target intestinal epithelial cells and reduce the mucus-blocking effect of the intestine. Moreover, the CSK modification of DEX-PLGA was found to significantly promote the absorption efficiency of NPs in the small intestine based on in vitro ligation of the intestinal rings and an examination of different intestinal absorption sites. Compared to DEX-PLGA-NPs (DPs), the absorption of CSK-DEX-PLGA-NPs (CDPs) was increased in the villi, allowing the drug to act on gobletlike Caco-2 cells through clathrin-, caveolin-, and gap-mediated endocytosis. Furthermore, the enhanced transport ability of CDPs was observed in a study on Caco-2/HT-29-MTX cocultured cells. CDPs exhibited a prolonged hypoglycemic response with a relative bioavailability of 9.2% in diabetic rats after oral administration. In conclusion, CDPs can target small intestinal goblet cells and have a beneficial effect on the oral administration of macromolecular peptides as a nanometer-sized carrier.

Mucus Mimic Hydrogel Coating for Lubricous, Antibiofouling, and Anti-Inflammatory Urinary Catheters

Sijia Zheng,Ying Liu,Jun Yao,Ruiying Zhu,Xing Yu,Zhihai Cao

Silicone is a common elastomer used in indwelling urinary catheters, and catheters are widely used in various medical applications due to their exceptional biocompatibility, hypoallergenic properties, and flexibility. However, silicones exhibit hydrophobic characteristics, lack inherent biolubrication, and are susceptible to nonspecific biosubstance adsorption, resulting in complications including but not limited to tissue trauma, postoperative pain, and urinary tract infections (UTIs). The development of effective surface designs for biomedical catheters to mitigate invasive damage and UITs has been a longstanding challenge. Herein, we present a novel approach to prepare a mucus mimic hydrogel coating. A thin layer of hydrogel containing xylitol is fabricated via photopolymerization. The surface modification technique and the interface-initiated hydrogel polymerization method ensure robust interfacial coherence. The resultant coating exhibits a low friction coefficient (CoF ≈ 0.1) for urinary catheter applications. Benefiting from the hydration layer and the antifouling of the xylitol unit, the xylitol hydrogel-coated surfaces (pAAAMXA) demonstrate outstanding antibiofouling properties against proteins (98.9% reduction relative to pristine polydimethylsiloxane (PDMS)). Furthermore, the pAAAMXA shows general adhesion resistance against bacteria primarily responsible for UITs (Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), Methicillin-resistant strains of Staphylococcus aureus (MRSA), and Staphylococcus epidermidis (S. epidermidis)) without compromising biotoxicity (cell viability 98%). In vivo, catheters coated with the mucus mimic hydrogel displayed excellent biocompatibility, resistance to adhesion of bio substance, and anti-inflammatory characteristics. This work describes a promising alternative to conventional silicone catheters, offering potential for clinical interventional procedures with minimized complications.

Palm kernel oil mucilage as a source of bio-based cationic surfactants for effective control of water surface pollution: Synthesis, surface activity and petroleum capacities

Mohamed A. Abo-Riya,Hamdy A. Zahran,E.A.A. Hendawy,Ahmed H. Tantawy,Hany I. Mohamed

Water contamination is a serious environmental issue with great negative impacts on the health of living organisms. Petroleum films from oil slicks represent one of the major water pollutants. The utilization of bio-based surfactants derived from natural sources/wastes can be an eco-friendly solution to address this problem. In such direction, a new series of palm kernel oil mucilage (PKOM)-based cationic surfactants ( 4a–e ) has been synthesized through an efficient procedure and the structures were characterized spectroscopically. Firstly, the free fatty acids mixture was extracted from PKOM, reduced with LiAlH 4 to the corresponding alcohols mixture and esterified with chloroacetic acid to afford fatty chlorides mixture. Parallelly, tertiary amine-tethering various aryl sulfonamides ( 3a–e ) were prepared from the reaction of sulfonyl chlorides with a primary amine-tertiary amine hybrid compound. Finally, the target cationic surfactants were obtained via quaternization of the tertiary- N in 3a–e with the fatty chlorides. The surface activity and thermodynamic parameters of the synthesized bio-based surfactants have been determined. Moreover, the ability of these compounds to collect or disperse petroleum thin films in different water types was evaluated. Interestingly, almost all compounds exhibited strong abilities to collect or disperse these films from water even at high salinity percent (9 % NaCl).

Mutation of CESA1 phosphorylation site influences pectin synthesis and methylesterification with a role in seed development

Shuangxi Zhang,Huachun Sheng,Yue Ma,Yanping Wei,Dan Liu,Yanhua Dou,Huiying Cui,Boyou Liang,Johannes Liesche,Jisheng Li,Shaolin Chen

Cell wall biogenesis is required for the production of seeds of higher plants. However, little is known about regulatory mechanisms underlying cell wall biogenesis during seed formation. Here we show a role for the phosphorylation of Arabidopsis cellulose synthase 1 (AtCESA1) in modulating pectin synthesis and methylesterification in seed coat mucilage. A phosphor-null mutant of AtCESA1 on T166 ( AtCESA1 T166A ) was constructed and introduced into a null mutant of AtCESA1 ( Atcesa1-1 ). The resulting transgenic lines showed a slight but significant decrease in cellulose contents in mature seeds. Defects in cellulosic ray architecture along with reduced levels of non-adherent and adherent mucilage were observed on the seeds of the AtCESA1 T166A mutant. Reduced mucilage pectin synthesis was also reflected by a decrease in the level of uronic acid. Meanwhile, an increase in the degree of pectin methylesterification was also observed in the seed coat mucilage of AtCESA1 T166A mutant. Change in seed development was further reflected by a delayed germination and about 50% increase in the accumulation of proanthocyanidins, which is known to bind pectin and inhibit seed germination as revealed by previous studies. Taken together, the results suggest a role of AtCESA1 phosphorylation on T166 in modulating mucilage pectin synthesis and methylesterification as well as cellulose synthesis with a role in seed development.

Enhancing Urolithin A Yield by Limosilactobacillus fermentum FUA033 in Human Gut Microbiota Across Three Urolithin-Metabotype Mice

Ziyan Hua,Xinzhu Gong,Kexin Chi,Yajian Su,Qianyan Tan,Rui Tang,Wenjun Zhang,Shu Liu,Yaowei Fang

Ellagic acid (EA), a polyphenol found in various fruits and nuts, is converted by the gut microbiota into urolithins, including Uro-A, known for its notable bioactivities. This study utilized fecal microbiota transplantation (FMT) to establish three distinct urolithin metabotypes (UMs) in pseudo-germ-free (PGF) mice, categorized as UM-A, UM-B, and UM-0 based on their metabolic outputs. Limosilactobacillus fermentum FUA033 was administered to the FMT UMs mice, resulting in a marked increase in Uro-A production across all metabotypes. In Group-0, Uro-A concentrations increased from undetectable levels to 15.97 ± 2.21 μM, while in Group-B, levels rose from 4.13 ± 2.49 μM to 19.56 ± 1.77 μM. Additionally, L. fermentum FUA033 significantly impacted gut microbiota diversity and composition, with a notable rise in Lachnospiraceae , a group crucial for Uro-A synthesis. These results underscore the potential of probiotics in modulating urolithin metabotypes and highlight an innovative strategy to improve gut health and metabolic functions using next-generation probiotics.

In situ monitoring of the structural change of microemulsions in simulated gastrointestinal conditions by SAXS and FRET

Xia Lv,Shuguang Zhang,Huipeng Ma,Peipei Dong,Xiaodong Ma,Ming Xu,Yan Tian,Zeyao Tang,Jinyong Peng,Haibo Chen,Jianbin Zhang

Microemulsions are promising drug delivery systems for the oral administration of poorly water-soluble drugs. However, the evolution of microemulsions in the gastrointestinal tract is still poorly characterized, especially the structural change of microemulsions under the effect of lipase and mucus. To better understand the fate of microemulsions in the gastrointestinal tract, we applied small-angle X-ray scattering (SAXS) and fluorescence resonance energy transfer (FRET) to monitor the structural change of microemulsions under the effect of lipolysis and mucus. First, the effect of lipolysis on microemulsions was studied by SAXS, which found the generation of liquid crystalline phases. Meanwhile, FRET spectra indicated micelles with smaller particle sizes were generated during lipolysis, which could be affected by CaCl 2 , bile salts and lecithin. Then, the effect of mucus on the structural change of lipolysed microemulsions was studied. The results of SAXS and FRET indicated that the liquid crystalline phases disappeared, and more micelles were generated. In summary, we studied the structural change of microemulsions in simulated gastrointestinal conditions by SAXS and FRET, and successfully monitored the appearance and disappearance of the liquid crystalline phases and micelles.
