Microbiota regulation by different Akebia trifoliata fruit juices upon human fecal fermentation in vitro

Sun Yuhao,Hu Yaru,Hu Danqi,Xiao Zhuqian,Wang Hongpeng,Huang Jun,Mao Jianwei

Three different Akebia trifoliata fruit juices were prepared, and their effects on modulation of human fecal microbiota were elucidated through an anaerobic fermentation in vitro. Results indicated that the introduction of inoculatedly-fermented Akebia trifoliata fruit juice promoted short-chain fatty acids productivity. Fecal microbiota analysis demonstrated up-regulations for abundances of Limosilactobacillus , Megamonas , Bifidobacterium , and Escherichia_Shigella , and down-regulations for numbers of Bacteroides , Prevotella_9 , Parasutterella , and Sutterella . Correlation analysis confirmed relationships among sample components, short-chain fatty acids productivity, and microbial abundances, suggesting that sugars and organic acids stimulated growth of Actinobacteriota and suppressed proliferation of Proteobacteria , thus uncovering the underlying mechanism for the better ability of inoculatedly-fermented Akebia trifoliata fruit juice to regulate microbiota structure. Besides, clusters of orthologous groups of proteins analysis indicated that metabolite biosynthesis, energy metabolism, homeostasis maintenance and other physiological functions were ameliorated.

In vitro dynamic digestion properties and fecal fermentation of Dictyophora indusiata polysaccharide: Structural characterization and gut microbiota

Lin Wang,Zhikun Zeng,Yaqing Lin,Baodong Zheng,Yi Zhang,Lei Pan

The in vitro dynamic digestive model more realistically simulates the human digestive system compared to static digestive model. In this study, the dynamic in vitro stomach-intestine digestive system and fecal fermentation was used to investigate the dynamic digestion properties and fermentation properties of Dictyophora indusiata polysaccharide. The results showed that there were no significant changes in molecular weight, functional groups and surface morphology after the in vitro dynamic simulated digestion, indicating that D. indusiata polysaccharide maintained a relatively stable structure during the dynamic in vitro salivary-gastrointestinal digestion. In addition, D. indusiata polysaccharide improved the abundance of beneficial bacteria, including Blautia , Coprobacter and Fusicatenibacter . It is remarkable that D. indusiata polysaccharide significantly increased the level of acetic acid and propionic acid. In conclusion, these results suggested that D. indusiata polysaccharide was a potential source of prebiotics, which provides a basis for the development of D. indusiata polysaccharide in the food or medical field.

In vitro digestion and fecal fermentation of Tremella fuciformis exopolysaccharides from basidiospore-derived submerged fermentation

Yan Fang,Qi Zhang,Caiyuan Yu,Xiaoqi Xu,Peng Lei,Hong Xu,Sha Li

Tremella fuciformis polysaccharides (TFPS) belong to natural bioactive macromolecule with both edible and medicinal value, possessing much bioactivities such as anti-tumor, antioxidant, antidiabetics, and immunomodulatory. Currently, the production of TFPS through submerged fermentation (TFPS-1) is gradually replacing the polysaccharides extracted from the fruiting body due to improved fermentation efficiency and reduced separation costs. However, it is still unclear about the effect of TFPS-1 on gastrointestinal digestion and gut microbial fermentation, which is directly related to the function of its biological activity. This study aimed to illustrate the effect of TFPS-1 on digestive process through in vitro digestion and fecal fermentation. TFPS-1 was indigestible during the simulated gastrointestinal tract. But TFPS-1 can be digested by intestinal flora leading to alterations in the total polysaccharides content, molecular weight, and apparent viscosity. Moreover, TFPS-1 regulated the composition of gut microbial, lowering the proportion of Firmicutes to Bacteroidetes and enhancing the abundances of Parabacteroides and Bacteroides. The change of intestinal flora produced more short chain fatty acids and lowered the pH. The KEGG metabolic pathway analysis indicated that TFPS-1 enriched lipid metabolism, glycan biosynthesis and metabolism, and biosynthesis of other secondary metabolites. This study demonstrated that T. fuciformis fermented polysaccharide can be a potential prebiotic to optimize intestinal flora homeostasis and maintain host intestinal health.

Lactobacillus reuteri Biofilms Inhibit Pathogens and Regulate Microbiota in In Vitro Fecal Fermentation

Meng-Xin Hu,Fei He,Ya-Xin Guo,Li-Zhen Mo,Xuan Zhu

Bacteria colonizing the gastrointestinal tract generally grow well in biofilms. In recent years, probiotic biofilms have been considered the most promising fourth-generation probiotics. However, the research into the functions of probiotic biofilms is just starting. In this study, Lactobacillus reuteri DSM 17938 biofilms formed on electrospun cellulose acetate nanofibrous scaffolds were contrasted with planktonic cells. Pathogen inhibition analysis of Escherichia coli, Staphylococcus aureus, and Listeria monocytogenes suggested a significant distinction between the planktonic and biofilm groups. In human fecal fermentation, L. reuteri remodeled the microbiota by decreasing the relative abundances of Proteobacteria, Escherichia–Shigella, and Desulfovibrio and increasing the relative abundances of Phascolarctobacterium, Bacteroides, and Lactobacillus. Moreover, L. reuteri biofilms played more positive roles in microbiota modulation and short-chain fatty acid production than planktonic L. reuteri. These findings provide an understanding of the beneficial effects of probiotic biofilms, laying a foundation for the application of probiotic biofilms as a health promoter.

Cypermethrin adsorption by Lactiplantibacillus plantarum and its behavior in a simulated fecal fermentation model

Zhang Mengmei,Chen Yuan,Lai Jinghui,Wang Xingjie,Hu Kaidi,Li Jianlong,Li Qin,He Li,Chen Shujuan,Liu Aiping,Ao Xiaolin,Yang Yong,Liu Shuliang

Abstract  The presence of cypermethrin in the environment and food poses a significant threat to human health. Lactic acid bacteria have shown promise as effective absorbents for xenobiotics and well behaved in wide range of applications. This study aimed to characterize the biosorption behavior of cypermethrin by Lactiplantibacillus plantarum RS60, focusing on cellular components, functional groups, kinetics, and isotherms. Results indicated that RS60 exopolysaccharides played a crucial role removing cypermethrin, with the cell wall and protoplast contributing 71.50% and 30.29% to the overall removal, respectively. Notably, peptidoglycans exhibited a high affinity for cypermethrin binding. The presence of various cellular surface groups including –OH, –NH, –CH 3 , –CH 2 , –CH, –P = O, and –CO was responsible for the efficient removal of pollutants. Additionally, the biosorption process demonstrated a good fit with pseudo-second-order and Langmuir–Freundlich isotherm. The biosorption of cypermethrin by L. plantarum RS60 involved complex chemical and physical interactions, as well as intraparticle diffusion and film diffusion. RS60 also effectively reduced cypermethrin residues in a fecal fermentation model, highlighting its potential in mitigating cypermethrin exposure in humans and animals. These findings provided valuable insights into the mechanisms underlying cypermethrin biosorption by lactic acid bacteria and supported the advancement of their application in environmental and health-related contexts. Key points • Cypermethrin adsorption by L. plantarum was clarified. • Cell wall and protoplast showed cypermethrin binding ability. • L. plantarum can reduce cypermethrin in a fecal fermentation model.

Simulated digestion, dynamic changes during fecal fermentation and effects on gut microbiota of Avicennia marina (Forssk.) Vierh. fruit non-starch polysaccharides

Qingxia Yuan,Kunling Lv,Jinwen Huang,Shujing Sun,Ziyu Fang,Hongjie Tan,Hong Li,Dan Chen,Longyan Zhao,Chenghai Gao,Yonghong Liu

Grey mangrove ( Avicennia marina (Forssk.) Vierh.) fruit is a traditional folk medicine and health food consumed in many countries. In this study, its polysaccharides (AMFPs) were obtained and analyzed by chemical and instrumental methods, with the results indicating that AMFPs consisted of galactose, galacturonic acid, arabinose, and rhamnose in a molar ratio of 4.99:3.15:5.38:1.15. The dynamic changes in AMFPs during the digestion and fecal fermentation processes were then investigated. The results confirmed that AMFPs were not depolymerized by gastric acid and various digestive enzymes. During fermentation, 56.05 % of the AMFPs were utilized by gut microbiota. Galacturonic acid, galactose, and arabinose from AMFPs, were mostly consumed by gut microbiota. AMFPs obviously decreased harmful bacteria and increased some beneficial microbiota, including Megasphaera , Mistuokella , Prevotella , and Megamonas . Furthermore, AMFPs obviously increased the levels of various short-chain fatty acids. These findings suggest that AMFPs have potential prebiotic applications for improving gut health.

In Vitro Digestion and Fecal Fermentation of Arecanut Polysaccharides: Effects on Gut Microbiota and Metabolites

Xiaolong Ji,Ke Jiang,Yuqing Liu,Chenyu Zhao,Jun Du,Liang Chen,Zhigang Zhu,Xiaoqiong Li

Recent studies have increasingly emphasized the regulatory potential of plant-derived polysaccharides on gut microbial composition and metabolic function. Despite this growing interest, investigations focusing specifically on the simulated digestion and fermentation properties of arecanut polysaccharide (PAP1b) remain limited. In this work, we employed the standardized INFOGEST 2.0 protocol to mimic the oral, gastric, and intestinal digestion of PAP1b, followed by 48 h anaerobic fermentation using pooled human fecal samples from healthy adult donors. PAP1b treatment led to a progressive decrease in pH and a substantial elevation in SCFAs levels, notably acetic, propionic, and butyric acids. Simultaneously, PAP1b significantly promoted the growth of SCFA-producing microbial taxa, particularly members of theFirmicutesphylum such asLachnospiraceae,Lachnoclostridium,Bilophila, andPhascolarctobacterium, while markedly suppressingBacteroidotapopulations. Metabolomic analysis further indicated that PAP1b intake enhanced bile acid metabolism, suggesting its potential as a prebiotic candidate for improving intestinal health.

Fecal fermentation characteristics of Rheum tanguticum polysaccharide and its effect on the modulation of gut microbial composition

Wu Ding-Tao,Yuan Qin,Feng Kang-Lin,Zhang Jinming,Gan Ren-You,Zou Liang,Wang Shengpeng

Background Rheum tanguticum is utilized as one of the well known traditional Chinese medicine for the treatment of gastrointestinal diseases. Recently, R. tanguticum polysaccharides (RP) have received increasing attention due to their diversely pharmacological activities. Usually, the pharmacological activities of polysaccharides are closely correlated to their metabolic properties from the stomach to the intestine. However, the digestive behavior and fecal fermentation characteristics of RP are unknown, which need to be fully investigated. Methods In this study, an in vitro simulated gastrointestinal model was carried out for the investigation of the digestive behavior and fecal fermentation characteristics of RP. The possible changes in physicochemical properties of RP, such as molecular weight, monosaccharide composition, reducing sugar released, chemical composition, pH value, and short chain fatty acids, were determined during in vitro simulated digestion and human fecal fermentation, and its effect on the modulation of gut microbial composition was also evaluated. Results The results revealed that RP was indigestible under the in vitro simulated digestion conditions according to its stabilities in physicochemical properties. Conversely, the indigestible RP (RPI) could be notably utilized by colonic microbiota in human feces after the in vitro fermentation, especially, at the initial fermentation stage (0–6 h). The fecal fermentation characteristics of RPI were revealed. Results showed that the content of reducing sugars obviously increased from 0.177 to 0.778 mg/mL at the initial stage of fermentation, and its molecular weight notably declined from 2.588 × 10 5 to 0.828 × 10 5  Da at the end stage of fermentation. Notably, the utilization of arabinose and galactose in RPI by colonic bacteria was faster than that of galacturonic acid. Besides, RPI could obviously modulate gut microbial composition via promoting the relative abundances of several beneficial bacteria, such as genera Bacteroides , Bifidobacterium , and Megamonas , resulting in the promoted production of several short-chain fatty acids, such as acetic, propionic, and butyric acids. Conclusions Results from this study showed that RP was indigestible in the human upper gastrointestinal tract in vitro, but could be easily utilized by colonic microbiota in human feces at the initial stage of fermentation. RP could be used as potential prebiotics for the improvement of intestinal health.

The synergistic effects of Lactobacillus casei HDS-01-konjac glucomannan synbiotic: probiotic potential and in vitro fecal fermentation

Shan Yu,Yingying Ning,Jiaju Xu,Ruoxi Yang,Dan Zhao

The study used the mannanase-producing Lactobacillus casei HDS-01 as the probiotic component and combined it with konjac powder, which contains konjac glucomannan (KGM), to create a synergistic synbiotic. The aim was to investigate its probiotic potential and the changes in gut microbiota and short-chain fatty acids (SCFAs). The results indicated that KGM selectively stimulated the growth of L. casei HDS-01, confirming their synergistic effect. The survival rate of L. casei HDS-01 at pH levels of 2.5–5.5, bile salt concentration of 0–0.35 g/L, and NaCl concentration of 1–8 g/L reached more than 55 %, suggesting that L. casei HDS-01 can survive in the gastrointestinal tract. Furthermore, the colonisation ability of L. casei HDS-01 in the intestinal tract was significantly enhanced by KGM. The antidigestive property of konjac oligo-glucomannan (KOGM) in gastric juice was better than that of inulin, with 94 % of KOGM expected to reach the jejunum of the small intestine after entering the human digestive system. In vitro fecal fermentation results indicated that the L. casei HDS-01-KGM synbiotic effectively promote the proliferation of gut SCFAs-producing bacteria (2.43 × 10 8  CFU/mL) and increase SCFAs production. The contents of acetic acid (7.68 ± 0.04 μg/mL), propionic acid (6.72 ± 0.03 μg/mL) and butyric acid (4.37 ± 0.06 μg/mL) were higher than those of the other treatment groups. Meanwhile, L. casei HDS-01-KGM synbiotic could enhance the population of beneficial bacteria, such as Pediococcus while significantly reducing harmful bacteria, such as Shigella . These results demonstrate the synergistic effect of L. casei HDS-01-KGM synbiotic and establish the foundation for its application in the food industry

In vitro digestion and fecal fermentation of encapsulated starch constructed via layer-by-layer coating of calcium alginate

Congli Cui,Mingju Li,Xiaoyang He,Na Ji,Yang Qin,Lei Dai,Liu Xiong,Qingjie Sun

This research prepared functional-resistant starch capsules by constructing multiple calcium alginate shells around calcium alginate/starch beads. Scanning electron microscopy revealed a dense structure for each layer of the calcium alginate shell, tightly encapsulating the calcium alginate/starch beads. The rapidly digestive starch content of three-layer calcium alginate shelled starch capsules decreased to 0.50 ± 1.14%, while the resistant starch content increased to 78.25 ± 1.55%. The estimated glycemic index (eGI) experiment suggested that the two-layer shell capsules (eGI: 59.91) and Three-layer shell capsules (eGI: 47.75) were classified as low-glycemic index foods . Furthermore, calcium alginate/corn starch multilayer capsules showed reduced microbial gas production and increased concentrations of propionic acid , isobutyric acid, and valeric acid compared to lactulose in vitro fecal fermentation experiment. Moreover, three-layer capsules and beads promoted a relative abundance of Bacteroidetes but inhibited the reproduction of Proteobacteria and Actinobacteria compared with lactulose and corn starch. These results suggest that constructing multi-layered sodium alginate shells for starch beads can effectively increase the digestive resistance of starch within the capsules and control its fermentation in the colon. This study will have a profound impact on the development of RS1-type resistant starch and be beneficial in the design and production of functional foods for prevention and assistance in the treatment of chronic diseases such as diabetes and obesity.

Effect of in vitro simulated digestion and fecal fermentation on Boletus auripes polysaccharide characteristics and intestinal flora

Qingying Luo,Xuejiao Li,Hongyu Li,Keyang Kong,Cheng Li,Zhengfeng Fang,Bin Hu,Caixia Wang,Saiyan Chen,Wenjuan Wu,Xiaolin Li,Yuntao Liu,Zhen Zeng

Boletus auripes is edible and medicinal boletus mushrooms rich in diverse nutrients and bioactive compounds, of which indigestible dietary polysaccharides are the most abundant compounds involved the regulation of gut microbes. However, the physicochemical, digestive, and fermentation characteristics of Boletus auripes polysaccharide (BAP) are not well studied. This study aimed to investigate the influence of different digestive stages on BAP's physicochemical characteristics and biological activities, and its effect on intestinal flora. We found that mannose (0.23 %), glucose (0.31 %), galactose (0.17 %), and fucose (0.19 %) were the main monosaccharides of BAP, with a high-molecular-weight (Mw) and a low-Mw fraction of 2084.83 and 62.93 kDa, respectively. During the course of digestion, there were slight alterations in the chemical composition, monosaccharide composition, and Mw of BAP. Despite these changes, the fundamental structural features of BAP remained largely unaffected. Moreover, the antioxidant and hypoglycemic activities of BAP were weakened under simulated saliva-gastrointestinal digestion. However, gut microbiota decomposed and utilized BAP to generate various short-chain fatty acids during fermentation, which decreased the pH of fecal cultures. Meanwhile, BAP modulated the gut microbiota composition and increased the relative abundance of Bacteroidetes . These findings suggest that BAP have potential for maintaining intestinal health and protecting against interrelated diseases.

Bioaccessibility and transformation pathways of phenolic compounds in processed mulberry (Morus alba L.) leaves after in vitro gastrointestinal digestion and faecal fermentation

Yanfang Yu,Bing Zhang,Yuhui Xia,Hongyan Li,Xuping Shi,Junwen Wang,Zeyuan Deng

The changes of phenolic compounds from the freeze-dried, hot-air dried, roasted and steamed mulberry ( Morus alba L.) leaves during the in vitro digestion were investigated. Caffeoylquinic acids, quercitrin glycosides and kaempferol glycosides were the main compounds. After simulated gastrointestinal digestion, the bioaccessibility of phenolics was much higher in roasted samples than those in hot-air, dried and steamed samples. Approximately 50%–80% of the bioaccessible phenolics were dialyzable depending on the different processing methods. The freeze-dried samples contributed to the highest amount of bioaccessible phenolics, followed by hot-air dried and roasted ones. All the initial phenolics degraded rapidly and were not detected after 24 h of faecal fermentation, except that at 0.5 h the amount of isoquercitrin and astragalin were increased by 156.48% and 177.44%, respectively. Consequently, quercetin, kaempferol, dihydrocaffeic acid, 3-(4-hydroxyphenyl) propionic acid and 3-methoxyphenyl-acetic acid were formed at different time. Transformation pathways for phenolics in the faecal fermentation were proposed.
