The influence of soluble dietary fiber (SDF) obtained from sweet potato dregs on the diversity and structure of gut microbiota in mice were explored. Thirty-two male Kunming (KM) mice were randomly divided into the following four treatment groups for a period of 30 days: control group (control diet, GC), SDF low dose (0.5 g/(kg.d), LSDF), middle dose (1.5 g/(kg.d), MSDF), and high dose (2.5 g/(kg.d), HSDF). High-throughput sequencing was used to profile the structure of g gut microbiota in the cecum samples. Different contents of SDF from sweet potato dregs showed different effects on the intestinal flora of mice, in terms of OUT , phylum, genus and species levels. MSDF group could reduce the relative abundance of Desulfovibrio , Corynebacterium_1, and Aerococcus (harmful bacteria, which could induce the inflammatory disorders); It could also increase the abundance of Lactobacillus and Faecalibaculum (beneficial bacteria, which maintain the intestinal flora balance and have anti-inflammatory effects). However, LSDF and HSDF groups showed the opposite trends to the MSDF group. It might be because that the intestinal environment formed by the intake of 1.5 g/(kg.d) of SDF from sweet potato dregs in mice was relatively suitable for the growth of beneficial bacteria (e.g. Lactobacillus spp. ), which is more conducive to the stability of intestinal flora. SDF from sweet potato dregs was effective on the intestinal flora of mice, and the MSDF group showed better effects than LSDF and HSDF.

Konjac glucomannan (KGM), a common food hydrocolloid widely used in the food industries, has broad biological activities, but its targeted effects on gut microbiota remain unclear. In this study, we used feces microbiota from 30 healthy volunteers, conducted an in vitro fermentation, employed multi-omics analysis to characterize the effect of KGM on the gut microbiota, and performed a stepwise culture system to investigate its microbial interactions. KGM suppressed the opportunistic pathogen (unclassified Enterobacteriaceae) and the accumulation of branched-chain fatty acids, p-cresol, sulfides, and indole. In contrast, KGM enriched the proliferation of some potentially beneficial bacteria ( Faecalibacterium and Parabacteroides ) and the accumulation of short-chain fatty acids, as well as volatile organic metabolites, such as butanoic acid propyl ester. Notably, F. prausnitzii and P. distasonis were specifically enriched, but neither could utilize KGM directly. The primary degrader, such as Bacteroides ovatus , can degrade KGM and promote the growth of F. prausnitzii and P. distasonis . Our findings offer novel insights into KGM-microbiota interactions while establishing a scientific foundation for developing KGM-based functional foods targeting gut health enhancement.

Processing techniques have been shown to alter the nutritional composition of rice bran (RB) and wheat bran (WB), influencing their ability to modify microbiota, but a systematic comparison is currently lacking. This study aimed to comprehensively investigate the effects of various processing methods, including physical modifications (grinding, extrusion, and steam explosion), enzymolysis (amylase, hemicellulose, and xylanase), and their combinations, on the modification of gut microbiota by RB and WB. Processing methods significantly altered the nutrient components of RB and WB as well as their capacity for gas and SCFAs production during in vitro fermentation. The RB23 (amylase treatment), RB31 (extrusion and hemicellulose treatment), and RB33 (steam explosion and xylanase treatment) samples exhibited the highest production of total SCFAs and butyric acid during in vitro fermentation. Overall, the microbiota responses to brans were conserved within individuals and the processing methods explained 21.6% of the variance. Fermentation of RB23, RB31, and RB33 samples was observed to enhance the richness of gut microbiota, decrease the Firmicutes/Bacteroidetes ratio, and stimulate the growth of Butyricicoccus and Faecallbacterium, while inhibiting the growth of Bilophila , Fusobacterium , and Escherichia-shigella . These findings may provide valuable insights for the processing of RB and WB.

Cadmium (Cd), a widely distributed environmental pollutant in agroecosystems, causes negative effects on crops and herbivores through bottom-up processes. The gut microbial community of an insect can play a critical role in response to metal stress. To understand how microbiota affect the stress responses of organisms to heavy metals in agroecosystems, we initially used 16S rRNA sequencing to characterize the larval gut microbiota of Chilo suppressalis , an important agricultural pest, exposed to a diet containing Cd. The species richness, diversity, and composition of the gut microbial community was then analyzed. Results revealed that while the richness (Chao1 and ACE) of gut microbiota in larvae exposed to Cd was not significantly affected, diversity (Shannon and Simpson) was reduced due to changes in species distribution and relative abundance. Overall, the most abundant genus was Enterococcus , while the abundance of the genera Micrococcaceae and Faecalibaculum in the control significantly superior to that in Cd-exposed pests. Phylogenetic investigation of microbial communities by the reconstruction of unobserved states (PICRUSt) showed that the intestinal microorganisms appear to participate in 34 pathways, especially those used in environmental information processing and the metabolism of the organism. This study suggests that the gut microbiota of C. suppressalis are significantly impacted by Cd exposure and highlights the importance of the gut microbiome in host stress responses and negative effects of Cd pollution in agroecosystems.

Grifola frondosa is a highly valued, edible and biologically active fungus in which the polysaccharide component plays a major role. To investigate the fundamental structure, in vitro digestion and fermentation characteristics of Grifola frondosa polysaccharides (GFP), GFP from the waste residue of Grifola frondosa after water extraction was re-extracted by alkaline solution. The carbohydrate and β-glucan contents of GFP were 91.61% and 60.57%, respectively, and were mainly composed of glucose. GFP can form triple supercoils and combine with Congo red, and the surface of GFP was smooth and dense, and the particles were in the shape of long column, sheet and ellipse. Analysis of carbohydrate, reducing sugars, and further chromatogram data also showed that GFP was not degraded by the simulated digestion process of the mouth, stomach and small intestine. Fecal fermentation of GFP significantly increased the abundance of Bacteroidota, and decreased Proteobacterias in both normal weight and obese subjects in vitro . The short-chain fatty acid (SCFA)-producing bacteria, Faecalibacterium , Erysipelotrichaceae_UCG_003 , Lactococcus were increased, and Dorea and Escherichia-Shigella were reduced after GFP treatment. Thus, the molecular of GFP was utilized and degraded by the gut microbiota and metabolized into SCFAs. This decreased pH, H 2 S, and NH 3, as well as increased CO 2 in the fermentation broth. In conclusion, GFP can modulate the composition of the gut microbiota and is a potentially functional product.

Antibiotics and microplastics (MPs) have become critical concerns worldwide because of their increasing amount and ecological risks to ecosystems. However, how MPs exposure affects the bioaccumulation and risks of antibiotics in waterfowls remains poorly understood. In this study, Muscovy ducks were exposed to single and combined contamination with polystyrene MPs and chlortetracycline (CTC) for 56 days, and the effects of MPs on CTC bioaccumulation and their risks in duck intestines were investigated. MPs exposure reduced the bioaccumulation of CTC in the intestine and liver of ducks and increased their fecal CTC excretion. MPs exposure caused severe oxidative stress, inflammatory response, and intestinal barrier damages. Microbiome analysis showed that MPs exposure induced microbiota dysbiosis by increasing the abundance of Streptococcus and Helicobacter , the increase of which may exacerbate intestinal damages. Co-exposure to MPs and CTC alleviated the intestinal damage by regulating the gut microbiome. Metagenomic sequencing revealed that the combined exposure to MPs and CTC increased the abundance of Prevotella , Faecalibacterium , and Megamonas and incidence of total antibiotic resistance genes (ARGs), especially tetracycline ARGs subtypes in the gut microbiota. The results obtained herein provide new insights into the potential risks of polystyrene MPs and antibiotics in waterfowls living in aquatic environments.

The intracellular polysaccharides of Aspergillus cristatus (IPSs) from Fuzhuan brick tea have been demonstrated to improve immune function linked to modulating the gut microbiota. Herein, to further investigate the efficacy of IPSs to maintain gut homeostasis, the protection of the purified fraction of IPSs (IPSs-2) on the mice with colitis induced by dextran sulfate sodium (DSS) and the underlying mechanisms were explored in this study. The results revealed that IPSs-2 alleviated the typical symptoms of colitis and suppressed the excessive inflammatory mediators, regulating the genes related to inflammatory responses in the colon at the mRNA level. Meanwhile, IPSs-2 treatment reinforced the intestinal barrier function by ameliorating the DSS-induced histological injury, facilitating the differentiation of goblet cells to enhance Mucin-2 generation, and enhancing the expression of tight junction proteins to alleviate colitis. In addition, IPSs protected against colitis by promoting the production of short-chain fatty acids (SCFAs), the activation of SCFAs receptors, and the leverage of the gut microbiota via the enrichment of Bacteroides, Parabacteroides, Faecalibacterium, Flavonifractor_plautii, and Butyricicoccus, linking with reducing inflammation and repairing intestinal barrier function. Overall, our research revealed the therapeutic potential of IPSs-2 as a prebiotic for attenuating inflammatory bowel disease and provided a rationale for future investigation.

Background: The gut microbiome is important for host nutrition and metabolism. Whether the gut microbiome under normal diet regulate human height remains to be addressed. Our study explored the possible relationship between gut microbiota, its metabolic products and the pathogenesis of idiopathic short stature disease (ISS) by comparing the gut microbiota between children with ISS and of normal height, and also the short-chain fatty acids (SCFAs) produced by the gut microbiota. Methods: The subjects of this study were 32 prepubescent children aged 4-8 years. The fecal microbial structure of the subjects was analyzed by 16S rRNA high-throughput sequencing technology. The concentrations of SCFAs in feces were determined by gas chromatography-mass spectrometry. Results: The richness of gut microbiota in ISS group was decreased, and the composition of gut microbiota was significantly different between ISS group and control group. The relative abundance of nine species including family Ruminococcaceae and genera Faecalibacterium and Eubacterium, in ISS group was significantly lower than that in control group (P<0.05). The relative abundance of 10 species, such as those belonging to genus Parabacteroides and genus Clostridium, in ISS group was significantly higher than that in control group (P<0.05). The concentration of total SCFAs and butyrate in ISS group was significantly lower than that in control group. The correlation analysis among different species, clinical indicators, and SCFAs showed that the relative abundance of family Ruminococcaceae and genera Faecalibacterium and Eubacterium was positively correlated with the standard deviation score of height. Furthermore, the concentrations of total SCFAs and butyrate were positively correlated with serum insulin-like growth factor 1 (IGF-1)-SDS. Disease prediction model constructed based on the bacteria who abundance differed between healthy children and ISS children exhibited high diagnostic value (AUC: 0.88).Conclusions: The composition of gut microbiota and the change in its metabolite levels may be related to ISS pathogenesis. Strains with increased or decreased specificity could be used as biomarkers to diagnose ISS.

Hyperuricemia is associated with various diseases. This study confirmed that intervention with Lacticaseibacillus rhamnosus JY027 ( L. rhamnosus JY027) reduced biochemical markers including uric acid , creatinine, and urea nitrogen in hyperuricemic mice. A combined analysis of gut microbiota , short-chain fatty acids (SCFAs), and metabolites based on the gut-kidney axis revealed that this bacterial intervention significantly enhanced the abundance of beneficial bacteria such as Bifidobacterium , Akkermansia , and Faecalibaculum and increased SCFA levels. The bacterial intervention also regulated the levels of guanosine , creatinine, arginine, choline , taurine and other differential metabolites in the kidneys of mice with hyperuricemia. This intervention ultimately reduced uric acid levels in mice through the modulation of multiple pathways, including arginine biosynthesis, taurine and hypotaurine metabolism, and glutathione metabolism. This study lays the foundation for exploring how L. rhamnosus JY027 alleviates hyperuricemia, providing theoretical support for developing probiotic preparations to mitigate hyperuricemia and their application.
