Integrating Inertial Microfluidics with SERS Bioprobe for Efficient Enrichment and Accurate Identification of Tumor Cells in Gastric Fluid and Ascites

Jiahao Zhang,Lei Xu,Yue Hu,Li Sun,Yujiao Xie,Xinyu Miao,Aochi Liu,Zhiwei Hou,Yixing Gou,Aiguo Wu,Jie Lin

Gastric cancer (GC) is a disease with high mortality rates and remains a central focus in medical research. Efficient enrichment, separation, and precise diagnosis of human gastric cancer (HGC) cells from biological samples are essential for early detection and treatment. However, the similarity in size of white blood cells (WBCs) and HGC cells in gastric fluid has posed vital challenges in the rapid identification and precise diagnosis of GC. To address this issue, a smart system that combines enrichment and detection of HGC cells using inertial microfluidic chips and a label-free surface-enhanced Raman spectroscopy (SERS) bioprobe has been developed. The HGC cells in simulated gastric fluid and ascites are rapidly separated and enriched by a spiral microfluidic chip. Silver bioprobes with excellent SERS performance are prepared to collect the label-free SERS spectra of HGC cells, normal gastric cells, and WBCs. SERS spectral data analysis and model prediction are performed by machine-learning-assisted principal component analysis-linear discriminant analysis (PCA-LDA). The result shows that HGC cells can be well identified with an accuracy as high as 96%. The microfluidic-SERS system proposed in this study can effectively separate and accurately identify tumor cells in body fluids, providing a new method of precision gastric cancer detection.

Harnessing X-Ray Energy-Dependent Attenuation of Bismuth-Based Nanoprobes for Accurate Diagnosis of Liver Fibrosis

Shiman Wu,Xianfu Meng,Xingwu Jiang,Yelin Wu,Shaojie Zhai,Xiaoshuang Wang,Yanyan Liu,Jiawen Zhang,Xinxin Zhao,Yan Zhou,Wenbo Bu,Zhenwei Yao

Timely detection of liver fibrosis by X-ray computed tomography (CT) can prevent its progression to fatal liver diseases. However, it remains quite challenging because conventional CT can only identify the difference in density instead of X-ray attenuation characteristics. Spectral CT can generate monochromatic imaging to specify X-ray attenuation characteristics of the scanned matter. Herein, an X-ray energy-dependent attenuation strategy originated from bismuth (Bi)-based nanoprobes (BiF 3 @PDA@HA) is proposed for the accurate diagnosis of liver fibrosis. Bi element in BiF 3 @PDA@HA can exhibit characteristic attenuation depending on different levels of X-ray energy via spectral CT, and that is challenging for conventional CT. In this study, selectively accumulating BiF 3 @PDA@HA nanoprobes in the hepatic fibrosis areas can significantly elevate CT value for 40 Hounsfield units on 70 keV monochromatic images, successfully differentiating from healthy livers and achieving the diagnosis of liver fibrosis. Furthermore, the enhancement produced by the BiF 3 @PDA@HA nanoprobes in vivo increases as the monochromatic energy decreases from 70 to 40 keV, optimizing the conspicuity of the diseased areas. As a proof of concept, the strategically designed nanoprobes with energy-dependent attenuation characteristics not only expand the scope of CT application, but also hold excellent potential for precise imaging-based disease diagnosis.

A highly sensitive living probe derived from nanoparticle-remodeled neutrophils for precision tumor imaging diagnosis

Qiansai Qiu,Ya Wen,Haiqing Dong,Aijun Shen,Xingxing Zheng,Yongyong Li,Feng Feng

Timely and precise diagnosis of malignant tumors is of great value to patients’ treatment and prognosis. Although nanotechnology-based molecular imaging represents a major advancement in the field of tumor imaging diagnosis, it is restricted by the rapid blood clearance of nanoparticles and the diverse physiological barriers in vivo; hence, its further application is greatly hindered. Cell carriers, ascribed to their natural biological properties, are thus gaining increasing attention for addressing such issues. Here, taking full advantage of the inflammation-homing capability of neutrophils, we constructed a highly sensitive cell probe in which reduced bovine-serum albumin (BSA) nanoparticles, loaded with imaging agents (gadolinium (Gd) and BODIPY), were covalently fixed onto the cellular surface by 5-thio-2-nitrobenzoate (TNB)-mediated fast and efficient disulfide–thiol exchange. Impressively, the remodeling process exerted a negligible effect on the neutrophils’ biological profiles with regard to cell viability, morphology, and cell-surface protein markers. Compared with nanoparticle-based imaging agents in a lung-cancer xenograft model, the living neutrophil probe demonstrated faster targeting and stronger accumulation in the tumor site, as revealed by fluorescence and magnetic-resonance (MR) imaging. These results indicate the great potential of neutrophil-based living probe for precision tumor-diagnosis applications.

Interfacial DNA Framework-Enhanced Background-to-Signal Transition for Ultrasensitive and Specific Micro-RNA Detection

Tongtong Guo,Yuanhang Xiang,Hao Lu,Minmin Huang,Fengfei Liu,Min Fang,Jia Liu,Yujin Tang,Xinchun Li,Fan Yang

Interfacial DNA self-assembly is fundamental to solid nucleic acid biosensors, whereas how to improve the signal-to-noise ratio has always been a challenge, especially in the charge-based electrochemical DNA sensors because of the large noise from the negatively charged DNA capture probes. Here, we report a DNA framework-reversed signal-gain strategy through background-to-signal transition for ultrasensitive and highly specific electrical detection of microRNAs (miRNAs) in blood. By using a model of enzyme-catalyzed deposition of conductive molecules (polyaniline) targeting to DNA, we observed the highest signal contribution per unit area by the highly charged three-dimensional (3D) tetrahedral DNA framework probe, relative to the modest of two-dimensional (2D) polyA probe and the lowest of one-dimensional (1D) single-stranded (ss)DNA probe, suggesting the positive correlation of background DNA charge with signal enhancement. Using such an effective signal-transition design, the DNA framework-based electrochemical sensor achieves ultrasensitive miRNAs detection with sensitivity up to 0.29 fM (at least 10-fold higher than that with 1D ssDNA or 2D polyA probes) and high specificity with single-base resolution. More importantly, this high-performance sensor allows for a generalized sandwich detection of tumor-associated miRNAs in the complex matrices (multiple cell lysates and blood serum) and further distinguishes the tumor patients (e.g., breast, lung, and liver cancer) from the normal individuals. These advantages signify the promise of this miRNA sensor as a versatile tool in precision diagnosis.

Active-matrix extended-gate field-effect transistor array for simultaneous detection of multiple metabolites

Chaochao Shen,Xin Xi,Dongqing Wu,Xiaojun Guo,Yuezeng Su,Ruili Liu

With the deepening understanding of diseases, increasing attention has been paid to personalized healthcare and precise diagnosis, which usually depend on the simultaneous monitoring of multiple metabolites, therefore requiring biological sensing systems to possess high sensitivity, specificity, throughput, and instant monitoring capabilities. In this work, we demonstrated the active-matrix extended-gate field-effect transistor (AMEGFET) array that can perform instant analysis of various metabolites in small amounts of body fluids collected during routine physiological activities. The extended gate electrodes of the AMEGFETs comprise ordered mesoporous carbon fibers loaded with both oxidoreductase enzymes for specific metabolites and platinum nanoparticles. By selecting customized electrode combinations, the AMEGFET array can monitor the concentrations of metabolites closely associated with chronic diseases and lifestyles, such as glucose, uric acid, cholesterol, ethanol, and lactate. The switch function of AMEGFET not only simplifies the readout circuitry for large-scale arrays but also avoids the mutual interferences among sensing units. The high flexibility and scalability make the AMEGFET array widely applicable in establishing high-throughput sensing platforms for biomarkers, providing highly efficient technical support for proactive health and intelligent healthcare.
