Richard Osae,Maurice Tibiru Apaliya,Emmanuel Kwaw,Michelle T.R. Chisepo,Otu Phyllis Naa Yarley,Eric Aforo Antiri,Raphael N. Alolga

This work aimed to determine the effects of five drying methods on the quality and essential oil composition of Ghanaian ginger. To this end, slices of fresh ginger rhizomes were dried using the following dryers; relative humidity convective (RHC) dryer, freeze (F) dryer, infrared (IR) dryer, microwave (MC) dryer and pulsed vacuum (PV) dryer. The antioxidant activities, total phenolic content (TPC), sensory characteristics and moisture content of the dried slices were assessed. The effects of the various drying techniques on the chemical composition of the essential oil components were determined using gas chromatography-mass spectrometry (GC–MS). With respect to the sensory attributes of the various dried ginger samples, the RHC-dried samples attained the highest scores for aroma, flavor and general acceptability while the F-dried samples scored the highest marks for appearance (i.e., colour). The RHC-dried samples had the lowest moisture content (5.45 %) followed by the F-dried (6.36 %), IR-dried (8.06 %), MC-dried (9.15 %) and PV-dried (10.07 %) samples. The F-dried samples retained the highest TPC (110.36 mg Gallic acid equivalent (GAE)/g db) and exhibited the highest antioxidant activities as evidenced by the outcomes of 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and ferric reducing antioxidant power capacity (FRAP) tests. The chemical profiles of the essential oil extracted from the various dried ginger samples were distinct and unique. The essential oil obtained from the F-dried samples was observed to have the highest compounds (24). The number of compounds identified in the oil extracted from the other dried samples were : RHC drying, 23; IR drying, 16; MC drying, 15; PV drying, 3. In general, the samples subjected to F and RHC drying gave the best outcomes for all the quality parameters assessed and are therefore recommended for large-scale industrial application.

Essential Oil Composition Anti-inflammatory Activities and Distribution of Fruits of Alpinia oxyphylla Miq.

Chunlian Li,Xiaobin Lin,Qiuyang Cai,Weicheng Zhang,Xianyi Wu,Zekai Tan,Lewen Yao,Shiyuan Huang,Yanghe Yuan,Lanyue Zhang

The perennial herb Alpinia oxyphylla Miq. (Zingiberaceae) is an important traditional medicinal plant in China. The present study evaluated the chemical composition of A. oxyphylla essential oil (AOEO) using GC-MS. The AOEO contained 55 compounds, including nootkatone (20.12%), valencene (12.26%), and ethyl-p-methoxycinnamate (5.00%). Previous studies have shown that AOEO possesses significant antioxidant and antibacterial activities. However, the anti-inflammatory effect of AOEO on tissue plasminogen activator (TPA)-induced ear edema is yet to be reported. In this study, TPA-induced ear edema was used to investigate the anti-inflammatory activity of AOEO. Hematoxylin and eosin staining of mouse tissue sections indicated that AOEO reduced the degree of edema, and immunohistochemistry experiments revealed that AOEO inhibited the expression of tumor necrosis factors, such as cyclooxygenase 2 (COX-2), interleukin-6 (IL-6), and nuclear factor-kappa B (NF-κB). A. oxyphylla is a commonly used traditional Chinese medicine with good biological activity. Analysis of the species potential geographical distribution and diversity indicated that the species can be rationally protected and exploited. Based on the chemical composition and biological activity of AOEO, further research and development should effectively promote the development of medicine, food, and cosmetics industries. The potential distribution of A. oxyphylla under current climatic conditions was predicted using DIVA-GIS and geographic information from herbarium specimens and other documents. The species predicted distribution spanned more than 12 provinces in China and was the most extensive in Hainan. Under current climatic conditions, A. oxyphylla is suitable for growing and expanding its population in the warm, humid regions of southern China.

The influence of the chemical composition of essential oils of Clausena lansium seeds on the growth of Candida strains

Ma Yinzheng,Wang Yuanxiao,Zhou Xia,Yang Heng,Zhang Huixin,Chen Wenhan,Zhang Haiying,Zhang Yunxia,He Xiaowen

Clausena lansium (Lour.) Skeels seeds have been shown to have diverse beneficial medical value due to their unique active components. This study analysed the composition of essential oils (EOs) of C. lansium seeds and investigated their potential antifungal effects against Candida strains . A total of forty-six components were identified in all samples by gas chromatography-mass spectrometry (GC–MS). The main components were sabinene, β -phellandrene and 4-terpineol. Thirteen EOs of C. lansium seeds were classified into three clusters based on their components. Cluster analysis showed that the difference between the tropics and subtropics was the greatest. These EOs and the three main chemicals showed different antifungal activities against five Candida species ( C. albicans , C. tropicalis , C. glabrata , C. krusei and C. parapsilosis ). The antifungal activity against C. glabrata and C. krusei was higher than that against other Candida strains. EOs of C. lansium seeds displayed noteworthy antifungal activity against both sensitive and fluconazole-resistant strains, with inhibition zone diameters in the range of 9.4–23.4 mm. Comprehensive analysis illustrated the importance of sabinene, β -phellandrene and 4-terpineol to antifungal activity, and there may be some synergistic effects with other components. These results represent the first report about the correlation between the chemical composition of EOs of C. lansium seeds and antifungal activity. Taken together, the results obtained provide scientific evidence for the traditional use of C. lansium seeds waste.

Antifungal activity of 40 plant essential oil components against Diaporthe fusicola from postharvest kiwifruits and their possible action mode

Shuzhen Yang,Min He,Dongmei Li,John Shi,Litao Peng,Liu Jinjing

The antifungal activity of essential oils is closely related to their components and contents. The study aimed to explore the structural activity of essential oil (EO) components against Diaporthe fusicola and modes of action. The results showed most of the tested compounds showed inhibitory effects on the growth of D. fusicola and carvacrol was the strongest one with half maximal effective concentration (EC 50 ) of 11.21 μL/L. The chemical structures of the tested compounds had great influences on their antifungal activity, in which the presence of CHO group, C C group or Ar-OH group in any EO components significantly increase the antifungal potency, but the increment of carbon chain length of saturated fatty aldehyde decreased antifungal activity. In order to analyze the antifungal action mode of EO components, the correlations between EC 50 and oxidation-reduction potential (ORP), cell-membrane permeability and respiration inhibition of the tested compounds were further evaluated. The results showed antifungal activities of these EO components seemed to be accomplished by increase of cell membrane permeability and inhibition of the respiration of D. fusicola . The levels of increased membrane permeability, inhibited respiration and ORP of chemicals were all significantly correlated with EC 50 . This results indicated the action mode of these EO components against D. fusicola may be associated with the increment of cell membrane permeability and respiration inhibition via disrupting redox homeostasis in fungal cells, and oxidation-reduction property could be used as a secondary indicator for screening the antifungal agent from EO components.

Encapsulation of essential oil components with methyl-β-cyclodextrin using ultrasonication: Solubility, characterization, DPPH and antibacterial assay

Subramanian Siva,Changzhu Li,Haiying Cui,Venkatasamy Meenatchi,Lin Lin

Essential oils derived from medicinal plants are prosperous sources of active components having high biological potential. Cuminaldehye and isoeugenol, are hydrophobic essential oil components (EOC), are showing drastic limitations in their applications by low water solubility and the respective volatility. Methyl- β -cyclodextrin inclusion complexes (M β CD-ICs) were prepared in aqueous solution and in solid state with the EOC via the ultrasonication method, an energy saving, high efficiency and eco-friend technique, aim to extend their aqueous solubility and biological properties. UV–Vis absorption, fluorescence, thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), proton nuclear magnetic resonance ( 1 H NMR) spectroscopy, Fourier transform infrared (FT-IR) spectroscopy, powder X-ray diffraction (PXRD) and computational simulations confirmed the formation of EOC/M β CD-ICs. Result of solubility studies proved the enhanced solubilization of EOC in the presence of M β CD in aqueous and double reciprocal profiles substantiated the guest/host stoichiometry of 1:1. TGA and DSC studies indicated the improved stability of EOC in M β CD-ICs. The efficiency of ICs in terms of the antioxidant activity was verified and the IC displayed higher antioxidant activity compared to that of free EOC, as determined by free radical scavenging assay. Finally, the antibacterial effect of EOC/M β CD-ICs against gram-positive Staphylococcus aureus and gram-negative Escherichia coli bacteria was demonstrated. Overall results not only revealed the potential of M β CD on the bioavailability, solubility and stability, but also that the intensification caused by the IC may be greater that the antioxidant and antibacterial effects of the selected EOC for this study.

Ultrasound-assisted extraction and dispersive liquid–liquid microextraction coupled with gas chromatography-mass spectrometry for the sensitive determination of essential oil components in lavender

Ping Chen,Bing Liu,Xin Liu,Jihong Fu

In this research, a novel method based on ultrasound-assisted extraction and dispersive liquid–liquid microextraction (UAE-DLLME) coupled with gas chromatography-mass spectrometry (GC-MS) has been successfully developed for the determination of essential oil constituents in lavender. The effective parameters of UAE-DLLME, including the type of extraction solvent, type and volume of preconcentration solvent, type and volume of disperser solvent, extraction temperature, and extraction time, were studied and optimized in detail. The optimal analytical conditions were as follows: 2 mL 50% ethanol as the extraction solvent, 150 μL methanol as the disperser solvent, 40 μL CCl4 as the preconcentration solvent, an extraction time of 20 min and an extraction temperature of 50 °C. Under the optimum conditions, a total of 13 samples of three varieties of lavender were analyzed and 64 compounds were identified. The precision of UAE-DLLME-GC-MS was evaluated and the relative standard deviation (RSD) was below 9% (n = 5). Good linearity with correlation coefficients of >0.9982 was obtained; limits of detection ranging from 1.75 to 7.50 ng indicated high sensitivity. Principal component analysis (PCA) was used to distinguish different varieties of lavender. This is the first application of UAE-DLLME for the analysis of essential oil constituents in lavender. The results showed that UAE-DLLME-GC-MS is a fast, sensitive, simple and small sample consumption method for the determination of essential oils in plant materials.
