Parameter Effects and Kinetics of Ultrasound Assisted Ionic Liquid Mediated Hydro-distillation and Essential Oil Composition of Flowers of Paeonia suffruticosa Andr. ‘Jitsugetsu Nishiki’ from Central China

Gaoming Lei,Chaoying Song,Haiyu Shi,Yihao Xing,Xue Song,Guojin Liang

Parameter effects and kinetics of ultrasound assisted ionic liquid mediated hydro-distillation (UILHD) were investigated. An ionic liquid 1-ethyl-3-methylimidazolium bromide ([C2C1im]Br) was employed both as a substrate under sonication and as an additive during distillation. Paeonia suffruticosa Andr. ‘Jitsugetsu Nishiki’, a widely cultivated ornamental plant in China, was selected as a case study. The optimal condition of UILHD was [C2C1im]Br concentration of 10 %, ultrasonic power of 400 W and pretreatment duration of 20 min. Under such a condition, the extraction equilibrium could be basically reached at 35 min of distillation. The kinetics obeyed pseudo-first-order equation. Calculated values of maximum essential oil yield and first-order rate constant were 1.53 g and 0.117 min-1, respectively. The main essential oil components of flowers of P. suffruticosa Andr. ‘Jitsugetsu Nishiki’ were 1,3,5-trimethoxybenzene (16.6 %), tricosane (8.7 %), nerol (8.6 %), nonadecane (6.4 %) and nonanal (5.5 %). The most abundant chemical class was non-terpene hydrocarbons (44.6 %) followed by non-terpene oxygenated compounds (31.2 %) and oxygenated terpenes (22.2 %). UILHD proved to be a promising technique for efficient isolation of essential oils.

Fang-Xiang Xu,Jing-Yu Zhang,Jing Jin,Zu-Guang Li,Yuan-Bin She,Maw-Rong Lee

In the past decade, natural deep eutectic solvents (NADESs) as green and sustainable extraction solvents with great potential for the efficient extraction of bioactive compounds from the plants are emerging. In this study, a microwave-assisted technology is used to prepare natural deep eutectic solvents. And natural deep eutectic solvents as pretreatment solvents coupled with microwave-assisted hydrodistillation (MAHD) for isolating essential oil (EO) derived from turmeric (Curcuma longa L.) is investigated. To improve the essential oil yield of turmeric (Curcuma longa L.) as a target, various factors affecting extraction efficiency including the type and amount of natural deep eutectic solvents, pretreatment time, pretreatment temperature and hydrodistillation (HD) time are discussed and optimized through central composite design (CCD) of the response surface methodology (RSM). The optimal conditions are as follows: natural deep eutectic solvent composed of choline chloride and oxalic acid (molar ratio with 1:1) as a pretreatment solvent, an amount of 60 g, a pretreatment time of 5 min, a pretreatment temperature of 84 ºC, a hydrodistillation time of 76 min. Under the optimum conditions, the highest essential oil yield of 0.85% is achieved. Additionally, the essential oil is analyzed by using gas chromatography-mass spectrometry (GC-MS), with a total of 49 compounds being identified. Through combining natural deep eutectic solvents with a microwave-assisted hydrodistillation technique, this work provides an eco-friendly extraction way of isolating essential oil, which boosts development in the monitoring other spice quality field.

Xiaojin Peng,Xinyu Yang,Huiyan Gu,Lei Yang,Hongfei Gao

A solvent-free microwave-assisted method and the screw extrusion of raw materials are combined and successfully applied to obtain the essential oil of fresh needles of Pinus pumila (Pall.) Regel. Single-factor screening and the Box-Behnken design method were applied to optimize the extraction of essential oils, and the yield of essential oil under the optimal conditions was 12.00 ± 0.60 mL/kg, which was highly consistent with the results predicted by the Box-Behnken design model. The process developed offers advantages in terms of the yield of essential oil and a reduced extraction time (31 min in comparison to 4 h for conventional hydrodistillation). The microstructures, chemical structures and thermal stability of needle samples before and after extraction were also investigated. The main components found in the essential oil extracted from the fresh needles of P . pumila were (+)-3-carene (22.75 %), δ-cadinene (12.12 %), terpinolene (12.11 %) and β-bisabolene (9.12 %). Furthermore, the semi-inhibition concentrations (IC 50 ) of acetylcholinesterase inhibition activity of the essential oil, terpinolene, (−)- trans -caryophyllene and (+)-3-carene were determined to be 0.76 ± 0.04, 0.55 ± 0.03, 0.45 ± 0.02 and 0.64 ± 0.03 mg/mL, respectively. In general, P . pumila needle essential oil showed notable inhibitory ability to acetylcholinesterase, and the components terpinolene, (−)- trans -caryophyllene, and (+)-3-carene play major roles in the process of essential oils inhibiting acetylcholinesterase. Additionally, the acetylcholinesterase inhibition activities of all the tested samples were mostly related to the corresponding concentrations. The acetylcholinesterase inhibition activity of kojic acid was also determined in parallel experiments as a positive control.

Qingfen Zhang,Chenglin Li,Shengcheng Mi,Xiaojie Xu,Zhiqiang He,Lin Wang,Zhonghua Tang,Qi Lu

The primary purpose of this study is to develop a highly efficient extraction method to meet the growing demand for Kaempferia galanga essential oil (KGEO) in various industries. In this study, an innovative approach for extracting essential oil from fresh K. galanga rhizomes using salt solution-enhanced homogenization pretreatment and microwave extraction (HP-SOME) was proposed. A kinetic study was conducted on the extraction process of essential oil. The key factors were screened using the Plackett–Burman Design and optimized using the Box-Behnken Design. Under the optimization parameters: sodium chloride concentration of 1.5 %, microwave power of 475 W, and microwave time of 23 min, the actual yield was 5.26 ± 0.10 %. The HP-SOME method exhibited a significant increase of 58.91 % in essential oil yield compared to that of the hydrodistillation method (3.31 ± 0.10 %). In addition, the composition of the KGEO was determined using Fourier transform infrared spectroscopy, Raman spectroscopy, and gas chromatography-mass spectrometry. The KGEO obtained by HP-SOME had a 6.26 % increase in the main component ethyl p-methoxycinnamate and a 4.13 % increase in ethyl cinnamate compared to that obtained by the HD method. Investigations into antimicrobial activity revealed that KGEO extracted by the HP-SOME method exhibited superior and more comprehensive antimicrobial characteristics. In conclusion, the HP-SOME method demonstrated remarkable capabilities in obtaining KGEO with notable efficiency, yield, and quality. This method holds great promise in the field of essential oil extraction.

Microwave Hydrodistillation Based on Deep Eutectic Solvent for Extraction and Analysis of Essential Oil from Three Amomum Species Using Gas Chromatography–Mass Spectrometry

Yu Guo-Wei,Cheng Qiang,Nie Jing,Wang Xia-Jun,Wang Peng,Li Zu-Guang,Lee Maw-Rong

Abstract Deep eutectic solvent has been gaining much attention in recent years due to its various novel properties. The present study develops a novel microwave-assisted hydrodistillation method based on pretreatment using deep eutectic solvent, for the extraction of essential oil (EO) from dry fruits of Amomum kravanh , Amomum tsaoko, and Amomum villosum , respectively. The process is optimized by single-factor experiments taking the improvement in the essential oil yield of A. kravanh as the target. The results indicate that using the reaction product of choline chloride and ethylene glycol as the deep eutectic solvent with 7:1 of mass ratio to fruit powder, the optimal conditions are (1) pretreatment stage: 500 W of microwave power, 50 °C of temperature, and 7 min of duration; (2) fast heating stage: 600 W of microwave power, 110 °C of temperature, and 5 min of duration; (3) hydrodistillation stage: 300 W of microwave power, 110 °C of temperature, and 30 min of duration. Under these conditions, the EO yields of A. kravanh , A. tsaoko, and A. villosum are 3.64, 2.16, and 1.62%, respectively. The EOs are analyzed by gas chromatography–mass spectrometry, with totally 63 compounds identified. In most cases, the new method brings about more identified compounds than the traditional methods. Moreover, the relative contents of hydrocarbons in most essential oils are improved by applying the new method. Thus, the approach based on deep eutectic solvent has been proved superior in the enhancement of essential oil production. Graphical abstract

An eco-friendly and effective method for isolating essential oil from fresh Kaempferia galanga L.: Process optimization based on entropy weight method and response surface methodology

Qingfen Zhang,Dan Ge,Shengcheng Mi,Qi Lu

The bioactive properties of essential oil are strongly associated with their chemical composition. In this work, a novel method of salt-enhanced homogenization pretreatment coupled with solvent-free microwave extraction (HP-SSFME) was developed for extracting essential oil (KGEO) from fresh Kaempferia galanga rhizomes, which significantly improved the extraction efficiency. Gas chromatography was employed to measure the contents of the main active components, ethyl p -methoxycinnamate (EPMC) and ethyl cinnamate (EC) in the essential oil. The extraction process was optimized by the Box-Behnken Design using the comprehensive scores of the yields of EPMC, EC, and KGEO obtained by the entropy weight method. The optimal parameters were determined as follows: amount of sodium chloride was 1.60 %, homogenization speed of 15000 r/min, homogenization time of 4 min, microwave irradiation power of 300 W, microwave irradiation time of 23 min. Under these conditions, the KGEO yield was 5.10 %, EPMC yield was 3.81 %, EC yield was 0.89 %. Compared with the conventional methods, the HP-SSFME method not only significantly increased the extraction yield of KGEO but also remarkably enhanced the yields of EPMC and EC. Additionally, the HP-SSFME method has low CO 2 emission and energy consumption, highlighting its environmental friendliness. Moreover, in terms of antibacterial activity, the HP-SSFME outperformed other methods, demonstrating a more powerful and extensive inhibitory effect against a range of bacteria. This study provides a valuable reference for the efficient extraction and quality control of essential oils from Kaempferia galanga rhizomes.

Ultrasonic pretreatment combined with microwave-assisted hydrodistillation for the situ extraction of essential oil from Pinus koraiensis seed scales induced by tea saponin: Functional activity, composition, thermal stability and material characterization

Zhanjun Li,Yibin Xu,Yunwei Liu,Mingru Kong,Ke Ma,Jiarong Wang,Honggang Wang,Yuhong Zhao

In this work, in situ extraction of essential oil from Pinus koraiensis seed scales using tea saponin-induced ultrasonic pretreatment-microwave-assisted hydrodistillation (UP-MAHD) was studied. By fitting the extraction kinetic curves with different factors, a first-order kinetic model was determined to be more suitable. The key factors were screened and optimized via Plackett Burman Design (PBD) and BOX-Behnken Design (BBD). Under the optimization parameters of a liquid material ratio of 14 mL/g, power of 100 W, time of 15 min, temperature of 50 °C, and tea saponin dose of 0.4% for the ultrasonication and then microwave at 458 W for 18 min, the actual yield was 8.02 μL/g± 0.35% and was highly consistent with the theoretical yield of 8.21 μL/g. UP-MAHD-treated Pinus koraiensis seed scales essential oil (PKSSEO) had good inhibition functional activity of α-amylase . In addition, the chemical composition of the PKSSEO were determined using Gas chromatography mass spectrometry (GC MS) and Fourier transform infrared spectroscopy (FT IR), thermal gravimetry (TG), and differential scanning calorimetry (DSC), and its spatial structure characterization (SSC) obtained via UP-MAHD treatment was enhanced. In summary, this extraction method efficiently extracted PKSSEO with a lower effect on its activity and composition and could have solid future development prospects. Our study provided effective theoretical data support for the reuse and development of abandoned Pinus koraiensis seed scales resources.

Natural deep eutectic solvents combined with COSMO-RS model for efficient extraction of essential oil from Fructus Aurantii: Theoretical screening, process optimization, biological activity and mechanism

Hua Du,Yaodeng Wang,Yi Wu,Wenhui Wu,Bixing Gao,Jiaqi Yang,Jing Leng,Min Yang

Traditional extraction methods of essential oil suffer from limitations such as low extraction efficiency or the use of toxic solvents, which fail to meet the principles of green chemistry. To address these issues, this study established a high-efficiency, eco-friendly method for extracting essential oils from Fructus Aurantii by integrating natural deep eutectic solvents (NADES) with the COSMO-RS tool. We initially screened 18 types of NADESs through theoretical analysis (COSMO-RS) and experimental validation, ultimately identifying the optimal extraction solvent (Gly-Laa, molar ratio 1:3). Subsequently, the extraction process was optimized using a Box-Behnken design (BBD) approach. The results revealed that the optimal conditions were 10 % water content, a 5:1 (g/g) liquid/solid ratio, and 7 h extraction time, respectively—achieved a maximum yield of 2.82 %. This represents a 4.7-fold increase compared to steam distillation (0.6 %) and a 1.76-fold enhancement over petroleum ether extraction (1.6 %). Furthermore, the extraction mechanism was investigated through MD simulations and RDG analysis. The results indicated that NADES enhanced its interactions with limonene via hydrogen bonding (average bond length: 0.14–0.22 nm) and van der Waals forces. Finally, the biological activities of the extracted essential oil were analyzed and compared. The results revealed that the NADES-extracted essential oil exhibited superior antioxidant capacity (DPPH and ABTS scavenging rates >80 %), significant antibacterial efficacy against pathogens such as Staphylococcus aureus (inhibition zone diameter >10 mm), and demonstrated notable anti-tumor activity. In summary, the combination of NADES and COSMO-RS model is a sustainable and efficient essential oil extraction strategy.
