Amino acids (AAs) are abundant in seminal fluid and play an important role in the reproduction process. However, the functional significance of AAs in seminal is not clear. This study was designed to profile the AAs in seminal fluid of asthenozoospermia (AS) and oligospermia (OL) patients and evaluate its potential diagnostic value. To determine the true function of AAs in seminal fluid, an ultra high performance liquid chromatography-mass spectrometry (UPLC-MS/MS) was developed and validated. The seminal fluid samples collected from 76 AS patients, 58 OL patients, and 76 healthy subjects were determined by UPLC-MS/MS and analyzed by fisher discriminant and receiver operating characteristic (ROC). The results showed that, the AAs levels were dramatically decreased in AS and OL patients compared with healthy subjects. What’s more there were significant variation in the AAs profile of AS and OL patients. Specifically, some polar AAs decreased, while nonpolar AAs increased in OL patients. Based on the level of AAs, 93.4 % of original and 90.9 % of cross-validated grouped cases were correctly classified between AS patients, OL patients, and healthy subjects. The further ROC analysis shows that glutamic acid, tyrosine, aspartic acid, proline, and tryptophan have the diagnostic value (P < 0.01) in the three groups. Moreover, aspartic acid and glutamic acid exhibit a high correlation (Correlation Coefficient >0.9) in AS, OL, and healthy subjects. In conclusion, there are significant variation in AAs profile in seminal plasma of AS and OL patients with high accuracy in fisher classification. The associations between the levels of AAs in seminal fluid of AS, OL, and healthy subjects may have clinical applications in providing valuable diagnostic indicator.

Background Gold nanoclusters (AuNCs) obtained by electroreduction have excellent electrochemiluminescence (ECL) properties, and its ECL intensity is regulated by the valence state. In addition, their ECL signals can be rapidly quenched by reactive oxygen species (ROS). Based on this observation, a sensitive ROS biosensor was designed based on valence regulation of AuNCs. Excessive ROS in seminal plasma can lead to male infertility, and the short half-life and instability of ROS pose a challenge for their detection. Since valence regulation can be done quickly and is very sensitive, this ECL biosensor holds promise to address this issue. Results The ECL mechanism of AuNCs and the quenching mechanism of AuNCs by ROS were explored, mainly because ROS change the valence state of AuNCs. The ECL signals of the biosensor have a linear relationship with logarithm of the target concentration in the range of 1.0 × 10 −8 to 1.0 × 10 −3 M and 1.0 × 10 −3 to 1.0 × 10 −1 M, with a detection limit of 0.75 × 10 −10 M (S/N = 3). The biosensor enables rapid one-step detection of ROS and has the advantage of being stable and reusable. More notably, the results of 57 real samples showed that the biosensor can be used to accurately assess the concentration of seminal plasma ROS, guiding the monitoring of sperm quality and the diagnosis of male infertility. Significance Compared with the traditional strategy of applying AuNCs only as a luminescent body, this strategy of regulating the valence state of AuNCs to achieve sensitive and rapid detection broadens the application of AuNCs in the field of analysis. Compared with other ROS detection strategies, the one-step immediate detection method effectively avoids the inaccuracy caused by the short half-life and natural dissipation of ROS, and is expected to improve the accuracy and efficiency of clinical diagnosis.

This paper presents the successful homogeneous processing of Y 2 O 3 green compacts by pressure-assisted alcoholic slip casting (PASC) method for the preparation of high-quality Y 2 O 3 transparent ceramics. The optimal Y 2 O 3 alcoholic slurry with 1.5 wt% polyethyleneimine as the dispersant and the solid load of 30.69 vol% was employed for PASC. Compared to dry-pressing green compact, PASC-prepared green compact exhibits high homogeneity and sinterability with less microstructural defects, such as large pores or particle agglomeration . Based on these advantages, we successfully prepared transparent Y 2 O 3 ceramic by simply vacuum sintering with low porosity, excellent optical homogeneity, and high in-line transmittance of 80.4% at the wavelength of 600 nm.

Excessive reactive oxygen species (ROS) in seminal plasma can trigger male infertility. Therefore, the development of simple and rapid ROS detection methods is urgently needed, particularly for the early self-screening of preconception couples. Herein, a gold nanobipyramid (Au NBP)-based colorimetric hydrogel for convenient and fast ROS detection is described. In the hydrogel, Au NBP is etched efficiently by ROS under the synergistic effect of Fe 2+ and I − , which finally causes color variations. Besides, agarose gel with the function of molecular sieve enables the separation of biomacromolecules, improving the interference resistance of the system and the stability of Au NBP. This chemical sensor can complete all the tests within 20 min, covering two detection range of 10–125 μM at relative low H 2 O 2 concentration and 125–1000 μM at relative high H 2 O 2 concentration, with the detection limits of 1.76 μM and 12.10 μM (S/N = 3) respectively. Furthermore, via visual observation of the color variations, it allows the initial interpretation of ROS concentration without any additional equipment. We applied this device to the detection of ROS in clinical seminal plasma samples and obtained promising results, demonstrating its potential for rapid and convenient detection in clinical applications.

Male infertility affects 10–15% of couples globally, with azoospermia — complete absence of sperm — accounting for 15% of cases. Traditional diagnostic methods for azoospermia are subjective and variable. This study presents a novel, noninvasive, and accurate diagnostic method using surface-enhanced Raman spectroscopy (SERS) combined with machine learning to analyze seminal plasma exosomes. Semen samples from healthy controls ( n = 3 2 ) and azoospermic patients ( n = 2 2 ) were collected, and their exosomal SERS spectra were obtained. Machine learning algorithms were employed to distinguish between the SERS profiles of healthy and azoospermic samples, achieving an impressive sensitivity of 99.61% and a specificity of 99.58%, thereby highlighting significant spectral differences. This integrated SERS and machine learning approach offers a sensitive, label-free, and objective diagnostic tool for early detection and monitoring of azoospermia, potentially enhancing clinical outcomes and patient management.

The harmful effects of microplastics (MPs) on male fertility are receiving more and more attention. However, the impact of low-dose MPs exposure on the reproductive function of male mice is still unclear. In this study, we exposed male mice to low-dose MPs (25–30 μg/kg body weight/day) or low-dose MPs combined with high-fat diet (HFD) feeding. Our results showed that low-dose MPs exposure or HFD feeding significantly reduced sperm quality and the number of offspring born, while low-dose MPs exposure combined with HFD feeding further enhanced the above effects. The combination of low-dose MPs exposure and HFD feeding resulted in a notable elevation of inflammatory level within the prostate of mice and induced apoptosis of prostate epithelium and a decrease in nutrients (zinc, citrate) in seminal plasma fluid. Our findings in this study could provide valuable clues for better understanding the influence of low-dose MPs exposure on the reproductive system under metabolic disorders and facilitate the development of the prevention of reproductive toxicity caused by MPs exposure.

STUDY QUESTIONWhat is the significance and mechanism of human seminal plasma extracellular vesicles (EVs) in regulating human sperm functions?SUMMARY ANSWEREV increases the intracellular Ca2+ concentrations [Ca2+]i via extracellular Ca2+ influx by activating CatSper channels, and subsequently modulate human sperm motility, especially hyperactivated motility, which is attributed to both protein and non-protein components in EV.WHAT IS KNOWN ALREADYEVs are functional regulators of human sperm function, and EV cargoes from normal and asthenozoospermic seminal plasma are different. Pre-fusion of EV with sperm in the acidic and non-physiological sucrose buffer solution could elevate [Ca2+]i in human sperm. CatSper, a principle Ca2+ channel in human sperm, is responsible for the [Ca2+]i regulation when sperm respond to diverse extracellular stimuli. However, the role of CatSper in EV-evoked calcium signaling and its potential physiological significance remain unclear.STUDY DESIGN, SIZE, DURATIONEV isolated from the seminal plasma of normal and asthenozoospermic semen were utilized to investigate the mechanism by which EV regulates calcium signal in human sperm, including the involvement of CatSper and the responsible cargoes in EV. In addition, the clinical application potential of EV and EV protein-derived peptides were also evaluated. This is a laboratory study that went on for more than 5 years and involved more than 200 separate experiments.PARTICIPANTS/MATERIALS, SETTING, METHODSSemen donors were recruited in accordance with the Institutional Ethics Committee on human subjects of the Affiliated Hospital of Nantong University and Jiangxi Maternal and Child Health Hospital. The Flow NanoAnalyzer, western blotting, and transmission electron microscope were used to systematically characterize seminal plasma EV. Sperm [Ca2+]i responses were examined by fluorimetric measurement. The whole-cell patch-clamp technique was performed to record CatSper currents. Sperm motility parameters were assessed by computer-assisted sperm analysis. Sperm hyperactivation was also evaluated by examining their penetration ability in viscous methylcellulose media. Protein and non-protein components in EV were analyzed by liquid chromatography-mass spectrum. The levels of prostaglandins, reactive oxygen species, malonaldehyde, and DNA integrity were detected by commercial kits.MAIN RESULTS AND THE ROLE OF CHANCEEV increased [Ca2+]i via an extracellular Ca2+ influx, which could be suppressed by a CatSper inhibitor. Also, EV potentiated CatSper currents in human sperm. Furthermore, the EV-in [Ca2+]i increase and CatSper currents were absent in a CatSper-deficient sperm, confirming the crucial role of CatSper in EV induced Ca2+ signaling in human sperm. Both proteins and non-protein components of EV contributed to the increase of [Ca2+]i, which were important for the effects of EV on human sperm. Consequently, EV and its cargos promoted sperm hyperactivated motility. In addition, seminal plasma EV protein-derived peptides, such as NAT1-derived peptide (N-P) and THBS-1-derived peptide (T-P), could activate the sperm calcium signal and enhance sperm function. Interestingly, EV derived from asthenozoospermic semen caused a lower increase of [Ca2+]i than that isolated from normal seminal plasma (N-EV), and N-EV significantly improved sperm motility and function in both asthenozoospermic samples and frozen-thawed sperm.LARGE SCALE DATAN/A.LIMITATIONS, REASONS FOR CAUTIONThis was an in vitro study and caution must be taken when extrapolating the physiological relevance to in vivo regulation of sperm.WIDER IMPLICATIONS OF THE FINDINGSOur findings demonstrate that the CatSper-mediated-Ca2+ signaling is involved in EV-modulated sperm function under near physiological conditions, and EV and their derivates are a novel CatSper and sperm function regulators with potential for clinical application. They may be developed to improve sperm motility resulting from low [Ca2+]i response and/or freezing and thawing.STUDY FUNDING/COMPETING INTEREST(S)This research was supported by the National Natural Science Foundation of China (32271167), the Social Development Project of Jiangsu Province (BE2022765), the Nantong Social and People's Livelihood Science and Technology Plan (MS22022087), the Basic Science Research Program of Nantong (JC22022086), and the Jiangsu Innovation and Entrepreneurship Talent Plan (JSSCRC2021543).
