The natural polyamine spermidine and spermine have been reported to ameliorate aging and aging-induced dementia. However, the mechanism is still confused. An aging model, the senescence accelerated mouse-8 (SAMP8), was used in this study. Novel object recognition and the open field test results showed that oral administration of spermidine, spermine and rapamycin increased discrimination index, modified number, inner squares distance and times. Spermidine and spermine increased the activity of SOD, and decreased the level of MDA in the aging brain. Spermidine and spermine phosphorylate AMPK and regulate autophagy proteins (LC3, Beclin 1 and p62). Spermidine and spermine balanced mitochondrial and maintain energy for neuron, with the regulation of MFN1, MFN2, DRP1, COX IV and ATP. In addition, western blot results (Bcl-2, Bax and Caspase-3, NLRP3, IL-18, IL-1β) showed that spermidine and spermine prevented apoptosis and inflammation, and elevate the expression of neurotrophic factors, including NGF, PSD95and PSD93 and BDNF in neurons of SAMP8 mice. These results indicated that the effect of spermidine and spermine on anti-aging is related with improving autophagy and mitochondrial function.

Reactive oxygen species (ROS)-based therapies have emerged as promising non-antibiotic strategies for treating infected wounds due to their broad-spectrum antimicrobial activity and low risk of resistance development. However, their clinical translation remains limited by low delivery efficiency, poor bacterial targeting, and uncontrolled post-treatment inflammation. To address these challenges, we reported a multifunctional bilayer microneedle patch (LFT/S-MN) designed for efficient bacterial eradication and enhanced wound healing. The patch integrates positively charged lactoferrin-based nanoparticles (LFT NPs) conjugated with a porphyrin sonosensitizer (TAPPFe) in the dissolvable base layer, and a pH-responsive tip layer loaded with the anti-inflammatory agent spermidine (SPD). Upon insertion into infected tissue, the released LFT NPs efficiently target and aggregate negatively charged bacteria, enabling robust ROS generation through synergistic chemodynamic and sonodynamic therapy (CDT/SDT) under ultrasound stimulation. Concurrently, the acidic wound microenvironment triggers the gradual degradation of the tip layer, allowing controlled SPD release for immunomodulation. In a murine P. aeruginosa -infected wound model, the LFT/S-MN patch significantly reduced bacterial load, suppressed inflammation, promoted collagen deposition and neovascularization, and accelerated wound closure. This work presents a promising non-antibiotic strategy for wound disinfection and regeneration through synergistic CDT/SDT and inflammation modulation.

The exploration of cost-effective methods for monitoring aquatic products freshness is significant. Metal nanoclusters (MNCs) show considerable potential in detecting food spoilage. A ligand modification strategy has been proposed as an effective approach for preparing high-performance MNCs. In this study, polyvinylpyrrolidone (PVP) and methylthioxanthine (MTU)-capped copper nanoclusters (PVP-MTU-CuNCs) with high quantum yield (43.76 %) were synthesized using an eco-friendly one-pot method. PVP improved the electron transfer and restricted the motion of MTU, enhancing photoluminescence, while its long-chain formed a physical barrier, effectively improving dispersibility and stability. PVP-MTU-CuNCs exhibited a satisfactory responsiveness to volatile basic nitrogens (VBNs), with detection limits of 0.34, 0.36, 0.33, 3.23, and 3.45 ppm for spermine, spermidine, histamine, cadaverine, and ammonia, respectively. An intelligent fluorescent label was developed for ammonia sensing and achieved visual semi-quantification and real-time tracking of the freshness of sea bass and shrimp, demonstrating its practical potential in assessing aquatic products freshness

The occurrence and progression of endometrial cancer are closely associated with metabolic reprogramming, in which polyamine metabolites play a critical role in tumor cell proliferation and invasion. In this study, we developed a label free surface enhanced Raman scattering (SERS) detection platform based on gold nanostars (AuNS), integrated with machine learning algorithms, to achieve highly sensitive detection and precise identification of polyamine metabolites related to endometrial cancer. In complex biological matrices such as serum, the platform yielded stable and reproducible spectral fingerprints, with a detection limit at the nanogram level. Furthermore, by constructing a polyamine metabolite spectral database and introducing machine learning models, both the classification accuracy and AUC values exceeded 95 %, enabling effective discrimination of different metabolic states and mixed systems. Taken together, the AuNS SERS strategy combined with machine learning provides a rapid, non-invasive, and intelligent detection tool for the early diagnosis and metabolic subtyping of endometrial cancer, with significant clinical application potential.

Oral ulceration is a prevalent mucosal disorder, and its healing process is frequently hampered by poor local drug retention and inadequate mucosal adhesion. However, the underlying pathological mechanisms remain incompletely elucidated. In this study, transcriptomic sequencing revealed that ferroptosis serves as a key driver of oral ulcer progression. To address the limitations of conventional formulations in the wet and dynamic oral environment, we constructed an asymmetrically adhesive, ferroptosis-targeting, spermidine (SPD)-functionalized Janus hydrogel microneedle system (MN-HTSO-C). This system was fabricated via chemical grafting and dynamic Schiff base crosslinking, enabling targeted drug delivery into mucosal tissues. By specifically delivering SPD, this system effectively inhibited ferroptosis, reduced reactive oxygen species (ROS) accumulation, reprogrammed the local immune microenvironment, and thereby promoted angiogenesis and epithelial regeneration. Our work not only identifies a novel pathological mechanism but also proposes an integrated therapeutic strategy that combines targeted delivery, immunomodulation, and ferroptosis inhibition, providing a new direction for the treatment of oral ulcers and other ferroptosis-related mucosal diseases.

N1,N12-diacetylspermine (DiAcSpm), a promising biomarker for cancer diagnosis, presents significant quantification challenges due to the structural homology within the polyamine family. To address this issue, we engineered a molecularly imprinted monolithic (MIM) column functionalized with biomimetic phosphorylcholine (PC) as functional monomer for the selective recognition of DiAcSpm in human urine. The zwitterionic polymer was synthesized via thermally initiated polymerization, with its morphology and pore architecture characterized through scanning electron microscopy (SEM) and brunauer-emmett-teller (BET) analysis. After optimizing capillary microextraction (CME) parameters, the MIM demonstrated a broad linear response (10–500 μM), a low detection limit (3.3 μM, S/N = 3), and high recoveries (76.8–91.2%) when coupled with HPLC-UV analysis. The biomimetic PC-based recognition significantly improved selectivity against key structural analogs, such as spermine, in complex biological matrices. This study underscores the potential of zwitterionic-based MIMs as a robust and efficient platform for the sensitive and selective monitoring of acetylated polyamines in clinical settings.

Protein-rich foods release biogenic amines (BAs) during prolonged storage, and these compounds serve as key biomarkers of food spoilage. Current separation-based methods for detecting BAs in food require sophisticated instrumentation and laborious sample pretreatment procedures. The array-sensing strategy, which mimics biological olfaction, provides a simple and rapid approach for the simultaneous detection of multiple BAs without the need for separation. Here, we constructed a multidimensional fluorescent sensor array using porphyrin-functionalized carbon dots (P-CDs) and their composites with metal ions (M-P-CDs), thereby incorporating the excitation wavelength as an additional signal dimension. This design reduces the number of physical sensors required while maintaining high discrimination capability, thereby overcoming a key limitation of conventional sensor arrays that depend solely on increasing the number of sensors. With the support of chemometric and machine learning methods, the sensor array successfully discriminated among ten BAs. We achieved quantitative detection of putrescine, histamine, and tyramine, determining their limits of detection (LODs) to be 2.37, 2.96, and 1.55 mg/L, respectively. The sensor array exhibited excellent stability (i.e., no significant fluorescence change over 30 days) and high selectivity against common interfering substances. Validation experiments on milk samples demonstrated the array’s ability to reliably discriminate specimens at different spoilage stages. This work establishes a novel multidimensional array-sensing strategy for detecting BAs, an approach that holds great potential for food safety applications. Graphical

Antibacterial hydrogel packaging often adheres poorly to hydrophobic prefabricated meat surfaces. Metal-polyphenol networks, combining metal ion antibacterial properties with polyphenol adhesion, offer promising preservation potential. In this study, tannic acid -Fe III metal polyphenol network-coated gallic acid -loaded ovalbumin/chitosan nanoparticles with adhesion and sustainable antibacterial properties were prepared and embedded into the gelatin hydrogel to elongate the shelf life of prefabricated meat. The composite hydrogel exhibited high adhesion, with an average adhesion force of 1.4-fold ∼1.9-fold larger than the other groups. It also exhibited good antibacterial properties, the colony-forming units (CFU) of 4 type of bacteria was reduced by 0.69, 0.34, 1.05 and 0.90 log 10 (CFU/mL). The composite hydrogel delayed pH, total volatile basic nitrogen (TVB-N), microorganism growth, and the marginally acceptable limit was delayed by 4 days compared with the other groups in applications. These results demonstrate the composite hydrogel's significant potential as a preservation material for prefabricated meat.

Intracellular polyamine metabolic homeostasis is closely linked to therapeutic outcomes in cancer. However, their intrinsic antitumor mechanisms have not been fully clarified. Herein, we demonstrate that the catabolism of spermine (Spm) and spermidine (Spd) mediated by polyamine oxidase directly generates two potent cytotoxic metabolites, namely acrolein and hydrogen peroxide. These metabolites act synergistically to trigger a marked elevation in intracellular reactive oxygen species (ROS), which in turn activate the caspase-1/GSDMD and caspase-3/GSDME signaling pathways, ultimately driving tumor cells into pyroptosis. Based on these findings, we develop a phenylboronic acid-functionalized sodium alginate-based hydrogel controlled-release system (Alg-PBA/Spd). This platform achieves sustained Spd release through coordination interactions and markedly inhibits tumor growth by inducing pyroptosis. Further studies demonstrate that the Alg-PBA/Spd hydrogel synergizes with radiotherapy (RT) to enhance radiosensitivity in an orthotopic breast cancer model and inhibit pulmonary metastasis. Moreover, this hydrogel enables co-delivery of αPD-L1 with sustained release, which significantly improves the response rate of immune checkpoint blockade therapy by inducing pyroptosis and remodeling the tumor immune microenvironment. Overall, this study not only elucidates a previously unrecognized mechanism underlying polyamine metabolism-driven pyroptosis but also offers a versatile platform to enhance the efficacy of RT and immunotherapy.

This study aims to investigate the role of exogenous spermidine (Spd) in mitigating the adverse effects of cadmium (Cd) stress on the growth and development of cucumber (Cucumis sativus). The cucumber cultivar “Xintaimici” was used as the experimental material, and a hydroponic experiment was carried out. Based on a baseline Cd concentration of 10 mg·L−1, Spd was supplemented at concentrations of 0.05, 0.1, 0.2, 0.4, and 0.5 mM, resulting in seven treatment groups: control group (CK), S0 group (Cd-only treatment, 10 mg·L−1Cd + 0 mM Spd), S1+ Cd group (10 mg·L−1Cd + 0.05 mM Spd), S2+ Cd group (10 mg·L−1Cd + 0.1 mM Spd), S3+ Cd group (10 mg·L−1Cd + 0.2 mM Spd), S4+ Cd group (10 mg·L−1Cd + 0.4 mM Spd), and S5+ Cd group (10 mg·L−1Cd + 0.5 mM Spd). This study analyzed the regulatory effects of Spd on the growth and development, antioxidant capacity and cadmium accumulation characteristics of cucumber seeds and seedlings. It was found that cadmium stress significantly inhibited their growth process and led to a decline in multiple physiological indicators. Under a Cd concentration of 10 mg·L−1, the application of 0.2 mM Spd significantly improved these parameters. During the seedling stage, the application of 0.2 mM Spd under Cd stress significantly enhanced the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX), as well as the content of soluble proteins, while significantly reducing malondialdehyde (MDA) levels. Cd content analysis revealed that 0.2 mM Spd promoted Cd accumulation in roots while suppressing its translocation to young leaves, thereby reducing Cd accumulation in aboveground tissues. Gene expression analysis demonstrated that this treatment significantly upregulated the expression levels of the phytochelatin synthase gene (CsPCS1) and the gene associated with reduced glutathione synthesis (CsGSHS). In conclusion, the exogenous application of 0.2 mM Spd effectively alleviates oxidative damage and osmotic stress induced by Cd stress in cucumber, promotes plant growth, and significantly enhances Cd tolerance.

Objective To explore potential serum biomarkers of children with Kashin-Beck Disease (KBD) and the metabolic pathways to which the biomarkers belong. Methods A two-stage metabolomic study was employed. The discovery cohort included 56 patients, 51 internal controls, and 50 external controls. The metabolites were determined by HPLC-(Q-TOF)-MS and confirmed by Human Metabolome Databases (HMDB) and Metlin databases. MetaboAnalyst 3.0 and the Kyoto Encyclopedia of Genes and Genomes (KEGG) database were used to analyze the metabolic pathways of the candidate metabolites. The use of HPLC-(Q-TRAP)-MS enabled quantitative detection of the target metabolites which were chosen using the discovery study and verified in another independent verification cohort of 31 patients, 41 internal controls, and 50 external controls. Results Eight candidate metabolites were identified out in the discovery study, namely kynurenic acid, N-α-acetylarginine, 6-hydroxymelatonin, sphinganine, ceramide, sphingosine-1P, spermidine, and glycine. These metabolites exist in sphingolipid, glutathione, and tryptophan metabolic pathways. In the second-stage study, five candidate metabolites were validated, including kynurenic acid, N-α-acetylarginine, sphinganine, spermidine, and sphingosine-1P. Except for spermidine, all substances exhibited low expression in the case group compared with the external control group, and the difference in levels of sphinganine, spermidine, and sphingosine-1P was statistically significant. Conclusion The direction of change of levels of sphinganine, spermidine, and sphingosine-1P in the two-stage study cohorts was completely consistent, and the differences were statistically significant. Therefore, these substances can be used as potential biomarkers of KBD. Furthermore, these results raise the possibility that sphingolipid metabolic pathways may be closely related to KBD.

Pseudomonas fluorescens is a prevalent spoilage bacterium in various chilled aquatic products, producing toxic biogenic amines (BAs) through the decarboxylation of amino acids via amino acid decarboxylases. Our previous studies revealed that P. fluorescens PF12 regulates BA production through its quorum sensing (QS) system, suggesting that disrupting QS could be a potential approach to control BA generation. In this study, we targeted the three-dimensional structure of the LuxI-type protein in P. fluorescens PF12 to screen for QS inhibitors (QSIs) through high-throughput screening from Seaweed Metabolite Database and Comprehensive Marine Natural Products Database. Nine compounds with high binding scores to the LuxI-type protein were identified. Considering cost-effectiveness and safety, phytol was selected for further investigation. Experimental results demonstrated that sub-inhibitory concentrations of phytol significantly suppressed BA production, siderophore production, motility, extracellular protease secretion, and biofilm formation in P. fluorescens PF12. Specifically, when the salmon fillets were stored for 288 h, the histamine content in the fillets treated with 25 mg/mL phytol decreased by 43.81 %. RT-qPCR revealed that the expression levels of key genes hisD and hisC related to histamine synthesis in the phytol treatment group were reduced by 7.68 ( p < 0.05) and 8.23 % ( p < 0.05), respectively, compared to the control group. Molecular docking analysis revealed that phytol interacts with key residues in the LuxI-type protein, thereby blocking the synthesis of acyl-homoserine lactones (AHLs) and downstream QS signaling. These findings indicate that phytol has great potential for controlling BA production in aquatic products and ensuring food safety.
