Effects of baking factors and recipes on the quality of butter cookies and the formation of advanced glycation end products (AGEs) and 5-hydroxymethylfurfural (HMF)

Huiyu Hu,Yuting Wang,Mingyue Shen,Yousheng Huang,Chang Li,Shaoping Nie,Mingyong Xie

Maillard reaction during food processing contributes to the formation of some unpleasant heat-induced toxicants including advanced glycation end products (AGEs) and 5-hydroxymethylfurfural (HMF), which have been linked to various health risks. The effects of baking factors and recipes, such as baking temperature (130°C–180 °C) and time (8 min–15 min), sucrose levels (0 g–20 g), butter levels (0 g–20 g) and egg liquid levels (0 g–12 g) on the formation of free N ε -(carboxymethyl)lysine (CML), free N ε -(carboxyethyl)lysine (CEL), protein-bound CML, protein-bound CEL, HMF, glyoxal (GO), methylglyoxal (MGO), 3-deoxyglucosone (3-DG) and on the sensory qualities were investigated in butter cookies. The results suggested that the levels of AGEs initially increased and then followed by decrease as baking temperature and time increased, HMF is very sensitive to baking temperature and time and grows sharply. The changes of protein-bound AGEs are lagging behind that of free AGEs. The proportions of sucrose, butter and egg liquid in butter cookies were positively correlated with AGEs, with sucrose greatly promoting on the formation of HMF and 3-DG. In addition, the high level of sucrose and butter in cookies is preferred by panelists, especially in terms of appearance, taste and smell.

Identification of volatile and flavor metabolites in three varieties of broccoli sprouts

Yu Xia,Ming-Yue Li,Syed Abdul Wadood,Han-Jun Hong,Yi Liu,Yu-Xuan Luo,Yi-Yan Wang,Hong-Yan Liu,Ren-You Gan

Broccoli sprouts are promising functional food sources and their taste and flavor play a pivotal role in the acceptance of consumers. In this study, the flavor profiles of three varieties of broccoli sprouts, namely Bi Lv, You Xiu, and Lv Hua, were comprehensively characterized using HS-SPME-GC/MS analysis. A total of 364 volatile and flavor components across 15 chemical classes were successfully identified. The results revealed a majority of volatile metabolites exhibiting upregulation during the germination process, leading to an enhancement in taste intensity after germination, particularly for umami and sweet tastes, which was associated with an increase in associated amino acids and sugar content. Although the total glucosinolate content in broccoli sprouts has decreased compared to seeds, it remains the primary contributor to the bitterness of broccoli sprouts. The present study elaborated on the flavor contribution of broccoli sprouts, supporting the cultivation and consumption of them as a nutritious food.

Evaluation of the matrix effect of pH value and sugar content on the analysis of pesticides in tropical fruits by UPLC-MS/MS

Jing Zhao,Jingjing Pu,Xiaopeng Wu,Boyu Chen,Yan He,Yue Zhang,Bingjun Han

Matrix effect (ME) is a critical factor that can significantly compromise the accuracy of pesticide residue analyses performed using ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS). In this study, five pesticides of different types, namely difenoconazole (triazoles), chlorpyrifos (organophosphates), thiram (dithiocarbamates), carbaryl (carbamates), and pyraclostrobin (strobilurins), were selected to investigate the UPLC-MS/MS MEs in five typical tropical fruits. The MEs and possible mechanisms of the main influence factors, i.e., pH value and sugar content, were studied for the pesticide residues in the tropical fruits. The proposed method was validated in terms of linearity, limit of detection (LOD), recovery, and precision. In the range of 0.005–0.5 μg mL −1 , the coefficients of determination (r) were better than 0.999, while the LODs and recoveries ranged from 0.16 to 4.4 μg kg −1 and from 75% to 120%, respectively. Results show that among the five typical tropical fruits, the ME was largest in longan, followed by that in banana; pineapple and guava showed lower MEs than those in banana, whereas papaya had the lowest. Chlorpyrifos has the largest ME, followed by thiram and difenoconazole, and the other pesticides have very small MEs. Moreover, the MEs can be reduced by adjusting the pH value and sugar content of the sample matrix. This study can be applied to assess the influence of MEs on the analysis results.

Enhanced fermentable sugar production in lignocellulosic biorefinery by exploring a novel corn stover and configuring high-solid pretreatment conditions

Hossain M. Zabed,Suely Akter,Mudasir A. Dar,Jamila A. Tuly,Mukesh Kumar Aswathi,Junhua Yun,Jia Li,Xianghui Qi

This study aimed to produce enhanced fermentable sugars from a novel stover system through the bioprocessing of its soluble sugars and insoluble carbohydrates. The pretreatment conditions were optimized for this high sugar-containing stover (HSS) to control inhibitor formation and obtain enhanced fermentable sugar concentrations. The optimum temperature, acid loading, and reaction time for the pretreatment were 155 °C, 0.5%, and 30 min, respectively, providing up to 97.15% sugar yield and 76.51 g/L total sugars at 10% solid-load. Sugar concentration further increased to 126.9 g/L at 20% solid-load, generating 3.89 g/L acetate, 0.92 g/L 5-hydroxymethyl furfural, 0.82 g/L furfural, and 3.75 g/L total phenolics as inhibitors. To determine the effects of soluble sugars in HSS on fermentable sugar yield and inhibitor formation, sugar-removed HSS was further studied under the optimum conditions. Although prior removal of sugars exhibited a reduction in inhibitor generation, it also decreased total fermentable sugar production to 115.45 g/L.

Biochemical component changes of Curcuma longa - Black tea triggered by kombucha fermentation using metabolomics analysis

Song Hu,Wen-Jing Ma,Li-Juan Fu,Xiao-Yun He,Guo-Hong Wang,Jian-Wei Fu,Min-He Yang

This study explores the biochemical indices across three distinct experimental groups: the Curcuma longa L. fermentation Group (CF), the Curcuma longa L. and black tea fermentation Group (CT), and the black tea fermentation Group (KF). After fermentation, the detection of basic indexes revealed that the CT group had the highest total sugar content and gluconic acid levels, at 4.48 mg/mL and 3.20 mg/mL, respectively. Additionally, the KF group exhibited the highest levels of sucrose, polyphenols, and total flavonoids, measuring 28.67 mg/mL, 236.76 μg/mL, and 0.20 mg/mL, respectively. Ethanol content was no more than 0.16% vol in all groups. A qualitative analysis of the aforementioned biochemical parameters revealed the presence of 8 carbohydrates and analogs, 8 sesquiterpenoids, 6 flavonoids, 6 organic acids, 5 alkaloids, and 21 glycosides. We further identified two unique pathways pertinent to fermentation: glucuronic acid interconversion pathway and the cyanogenic amino acid metabolism pathway. Key metabolites influencing kombucha include α-ketoglutaric acid, D-xylitol, ribitol, dhurrin, and lotaustralin. Underscoring the significance of Curcuma longa L. and black tea fermentation. These results elucidate the differential expression of metabolites and their regulatory mechanisms.

Discrimination of Radix Astragali from Different Growth Patterns, Origins, Species, and Growth Years by an H1-NMR Spectrogram of Polysaccharide Analysis Combined with Chemical Pattern Recognition and Determination of Its Polysaccharide Content and Immunological Activity

Yali Guo,Bing Wang,Lifei Gu,Guo Yin,Shuhong Wang,Meifang Li,Lijun Wang,Xie-An Yu,Tiejie Wang

The fraud phenomenon is currently widespread in the traditional Chinese medicine Radix Astragali (RA) market, especially where high-quality RA is substituted with low-quality RA. In this case, focused on polysaccharides from RA, the classification models were established for discrimination of RA from different growth patterns, origins, species, and growth years. 1H Nuclear Magnetic Resonance (H1-NMR) was used to establish the spectroscopy of polysaccharides from RA, which were used to distinguish RA via chemical pattern recognition methods. Specifically, orthogonal partial least squares discriminant analysis (OPLS-DA) and linear discriminant analysis (LDA) were used to successfully establish the classification models for RA from different growth patterns, origins, species, and growth years. The satisfactory parameters and high accuracy of internal and external verification of each model exhibited the reliable and good prediction ability of the developed models. In addition, the polysaccharide content and immunological activity were also tested, which was evaluated by the phagocytic activity of RAW 264.7. And the result showed that growth patterns and origins significantly affected the quality of RA. However, there was no significant difference in the aspects of origins and growth years. Accordingly, the developed strategy combined with chemical information, biological activity, and multivariate statistical method can provide new insight for the quality evaluation of traditional Chinese medicine. Keywords:Radix Astragali;immunomodulatory activity;polysaccharide content;chemical pattern recognition;quality evaluation

Valorization of cell wall polysaccharides extracted from Liubao brick tea residues: Chemical, structural, and hypoglycemic properties

Li Wang,En-Zhong Chi,Xin-Huai Zhao

BACKGROUND Tea dregs, typically generated during the production of instant tea or tea beverages, have conventionally been regarded as waste material and routinely discarded. Nevertheless, contemporary research endeavors are concentrating on discovering efficient methods for utilizing the potential of this discarded resource. RESULTS In this study, we employed a sequential extraction method using chemical chelating agents to extract and isolate four distinct cell wall polysaccharides, designated as CWTPS-1 through CWTPS-4, from the tea dregs of Liubao brick tea. A comprehensive investigation into their physicochemical, structural, and hypoglycemic properties was conducted. The analysis of chemical composition and physicochemical characteristics revealed that all four CWTPSs were characterized as acidic polysaccharides, albeit with varying chemical compositions and physicochemical attributes. Specifically, the xyloglucan fractions, CWTPS-3 and CWTPS-4, were found to be rich in glucose and xylose, displaying a more uniform molecular weight distribution, greater structural stability, and a more irregular surface compared to the others. Moreover, they exhibited a higher diversity of monosaccharide residues. Importantly, our research unveiled that all four CWTPSs exhibited the capacity to modulate key glucose-regulated and antioxidant enzyme activities within HepG2 cells via the IRS-1-PI3K/Akt signaling pathway, thereby ameliorating cellular insulin resistance. Furthermore, our correlation analysis highlighted significant associations between monosaccharide composition and neutral sugar content with the observed hypoglycemic activity of CWTPSs. CONCLUSIONS This study highlights the potential of utilizing tea dregs as a valuable resource, making a significant contribution to the advancement of the tea industry. Furthermore, CWTPS-4 exhibits promising prospects for further development as a functional food ingredient or additive. This article is protected by copyright. All rights reserved.

Maleic anhydride-modified xylanase and its application to the clarification of fruits juices

Yang Zhao,Luyue Zhang,Shiyu Zhang,Xing Zheng,Mingzhu Zheng,Jingsheng Liu

At presently, the catalytic activity of xylanase is sub-optimal, and the required reaction conditions are harsh. To improve its catalytic activity and stability, xylanase (XY) was chemically modified with maleic anhydride (MA). The enzymatic properties of this maleic anhydride-modified xylanase (MA-XY) were then evaluated and analyzed spectroscopically. The results showed that the thermal stability, use of organic solvents, storage stability and the pH range of 3.0 to 9.0 for MA-XY were better than that for XY alone. The kinetic parameters of the enzyme (K m values) decreased from 40.63 to 30.23 mg/mL. Spectroscopic analysis showed that XY had been modified by the acylation reaction to become a tertiary structure. An assay based on clarifying fruit juices showed that the clarification capacity and reducing sugar content using MA-XY increased compared with those using XY. Overall, this study provides a theoretical basis for improving the application of XY in the food industry.

Effects of pre- and post-treatment with ethephon on gum formation of peach gummosis caused by Lasiodiplodia theobromae

Z. Li,W. Zhu,Y.-C. Fan,J.-L. Ye,G.-H. Li

Peach gummosis, caused by Botryosphaeria spp. fungi, is the process of gum accumulation and exudation in plants. Ethephon (2-chloroethylphosphonic acid) has profound effects on plants, including enhanced production of secondary metabolites and regulation of plant diseases. This study investigates the effects of application of ethephon before and after inoculation with Lasiodiplodia theobromae on gum formation. Gum formation was promoted by ethephon treatment prior to pathogen inoculation, but inhibited by ethephon applied after the pathogen. The inhibitory effect was counteracted by 1-methylcyclopropane, which is an ethylene signal inhibitor. 1-methylcyclopropane also promoted gum formation. Exposure of three isolates of Botryosphaeria to ethephon inhibited mycelial growth. Both treatment methods increased the sugar content at 12 and 24 h post-inoculation (hpi). However, the sucrose, glucose and fructose contents were significantly higher in shoots with ethephon post-treatment (application of ethephon after the pathogen inoculation) than those in shoots with ethephon pre-treatment (application of ethephon prior to pathogen inoculation) at 48 and 72 hpi. The expression of two putative senescence-related genes, SEN2 and SEN4 , were significantly enhanced in pre- and post-treated shoots with ethephon at 24, 48 and 72 hpi. Ethephon application also up-regulated expression of the pathogenesis-related protein PR4 while down-regulating PR1a and PR10. The results show that ethephon has a dual function in regulating gum formation by affecting both the peach shoots and the pathogen.

Effects of organic and bacterial fertilizer application on cabbage characteristic and rhizosphere soil microorganisms

Caixia Sun,Liping Chen,Yuhong Liu,Weiran Zheng,Yumei Hua,Qiaoyan Zhang

The increasing demand for high quality fertilizer, which is being driven by human nutrition demand and agricultural soil protection, necessitates sustainable alternatives to be used as vegetable production. Thus, utilizing biochar (BC), bacterial fertilizer and their combination with organic fertilizer (OF) could be a sustainable approach. In our research work, OF, BC, liquid bacterial fertilizer (LB), and solid bacterial fertilizer (SB) were used to investigate different fertilizer combinations. Greenhouse field experiment was conducted to compare different fertilizer application and their effects on cabbage biomass, cabbage characteristics and soil physical and chemical properties. The results showed that combined application OF, BC, and LB promoted cabbage growth and resulted in the highest yield of 16.6 kg/m 2 . The vitamin C content was highest at 48.4 mg/kg with the combined application OF, BC, and SB treatment. The combined application OF, BC, and SB achieved the best overall comprehensive evaluation for cabbage biomass weight, vitamin C content, and total sugar content. The combined application OF and a bacterial fertilizer improved soil properties, while the addition of BC reduced the soil nutrient content. Gas chromatography-ion mobility spectrometry (GC-IMS) analysis showed that volatile organic compounds from cabbages were mainly esters, alcohols, acids, acetates, aldehydes, and ketones. These results provide a comprehensive understanding of different fertilizer applications and their effects on cabbage quality and rhizosphere soil.

Fingerprint profiling for quality evaluation and the related biological activity analysis of polysaccharides from Liuweizhiji Gegen-Sangshen beverage

Wei Shulin,Li Mingxing,Zhao Long,Wang Tiangang,Wu Ke,Yang Jiayue,Tang Mingyun,Zhao Yueshui,Shen Jing,Du Fukuan,Chen Yu,Deng Shuai,Xiao Zhangang,Wei Mei,Li Zhi,Wu Xu

Introduction: Liuweizhiji Gegen-Sangshen beverage (LGS) is popular in China, which has been used for alleviating alcohol-mediated discomfort and preventing alcoholic liver disease (ALD). This beverage is consisted of six herbal components that are known as functional foods and fruits. LGS is rich in polysaccharides, however, the activity and quality evaluation of LGS-derived polysaccharides remain unexplored. The purpose of this study is thus to establish a comprehensive quality control methodology for the assessment of LGS polysaccharides (LGSP) and to further explore the anti-oxidant, anti-inflammatory as well as prebiotic effect of LGSP.Methods: LGSP was extracted, followed by analysis of molecular weight distribution, monosaccharide content and structural characterization via integrating the application of high-performance size exclusion chromatography (HPSEC), 1-phenyl-3-methyl-5-pyrazolone-HPLC (PMP-HPLC), fourier transform infrared spectroscopy (FT-IR) as well as nuclear magnetic resonance spectroscopy (NMR) techniques. The anti-oxidation activity of LGSP was determined by DPPH, ABTS, hydroxyl radical scavenging capacity and total antioxidant capacity. The anti-inflammation of LGSP were assessed on the RAW 264.7 cells. The effect of LGSP on growth of Lactobacillus, Bifidobacterium bifidum and Bifidobacterium adolescentis was evaluated. Results: The results demonstrated that LGSP had two molecular weight distribution peaks, with the average molecular weights of (6.569 ± 0.12) × 104 Da and (4.641 ± 0.30) × 104 Da. LGSP was composed of 8 monosaccharides, with galacturonic acid, glucose rhamnose and galactose representing the highest molar ratios. Homogalacturonic acid (HG) type and rhamnosegalacturonic acid glycans I (RG-I) type and α-1,4-glucan were present in LGSP. LGSP concentration in LGS was 17.94 ± 0.28 mg/mL. Furthermore, fingerprint analysis combined with composition quantification of 10 batches of LGSP demonstrated that there was a high similarity among batches. Notably, LGSP exhibited anti-oxidant effect and inhibited expressions of pro-inflammatory factors (TNF-α and IL-6) in LPS-stimulated RAW 264.7 cells. In addition, LGSP remarkably promoted the proliferation of probiotics Lactobacillus, Bifidobacterium bifidum and Bifidobacterium adolescentis, showing good prebiotic activity.Discussion: The results of present study would be of help to gain the understanding of structure–activity relationship of LGSP, provide a reference for quality evaluation of bioactive LGSP, and facilitate development of unique health and functional products in the future.

Direct transformation of xylan-type hemicelluloses to furfural via SnCl4 catalysts in aqueous and biphasic systems

Wenju Wang,Junli Ren,Huiling Li,Aojie Deng,Runcang Sun

Direct catalytic transformation of xylan-type hemicelluloses to furfural in the aqueous system and the biphasic system were comparatively investigated under mild conditions. Screening of several promising chlorides for conversion of beech xylan in the aqueous system revealed the Lewis acid SnCl 4 was the most effective catalyst. Comparing to the single aqueous system, the bio-based 2-methyltetrahydrofuran (2-MTHF)/H 2 O biphasic system was more conducive to the synthesis of furfural, in which the highest furfural yield of 78.1% was achieved by using SnCl 4 as catalysts under the optimized reaction conditions (150   °C, 120   min). Additionally, the influences of xylan-type hemicelluloses with different chemical and structural features from beech, corncob and bagasse on the furfural production were studied. It was found that furfural yield to some extent was determined by the xylose content in hemicelluloses and also had relationships with the molecular weight of hemicelluloses and the degree of crystallization.
