Structure Differences of Water Soluble Polysaccharides in Astragalus membranaceus Induced by Origin and Their Bioactivity

Zhili Sheng,Junmei Liu,Bao Yang

Astragalus membranaceusis a functional food with multiple bioactivities. It presents differentiated health benefits due to origins. Polysaccharides (APS) are the leading bioactive macromolecules ofA. membranaceus, which are highly related to its health benefits. However, the effect of origin on the structural characteristics of APSs remains unclear. In this work, polysaccharides from four origins were isolated and identified by NMR. The results showed APSs of four origins had identical monosaccharide composition and glycosidic linkage. Rhamnogalacturonan II pectins and α-(1→4)-glucan were the dominant polysaccharides. However, the level of methyl ester in pectins varied to a large extent. The molecular weight profiles of APSs were also different. Inner Mongolia APS had the largest percentage of 20–40 kDa polysaccharides. Molecular weight and methyl ester level were two important parameters determining the difference of APSs from four origins. These results were helpful to recognize the origin-related quality ofA. membranaceus. Keywords:glycosidic linkage;monosaccharide composition;NMR;pectin

Characterization and immunological activity of polysaccharides from Potentilla chinensis

Yuanhao Qiu,Zahra Batool,Rui Liu,Guoqing Sui,Bulei Sheng,Xinhua Zheng,Dan Xu

Three novel polysaccharides (PCPW, PCPS1 and PCPS2) were isolated from Potentilla chinensis and subjected to structural analysis by using spectral and physicochemical methods. The molecular weights of PCPW, PCPS1 and PCPS2 were calculated to be 4.45 × 10 3  Da, 1.18 × 10 4  Da and 4.23 × 10 4  Da, respectively. Analysis of monosaccharides composition confirmed that PCPW was composed of mannose, rhamnose, glucose, galactose and arabinose, while the two acidic polysaccharides PCPS1 and PCPS2 were consisted of six monosaccharides, including mannose, rhamnose, galacturonic acid, glucose, galactose and arabinose respectively. In addition, the main linkages of glycosidic bonds of PCPS2 were 1, 4-linked-rha, 1, 4-linked-man,1, 4-linked-galA and 1, 6-linked-man. Immunological tests indicated that both PCPW and PCPS2 could increase NO production of RAW264.7 cells, and promote splenocyte proliferation. All three polysaccharides proved to be activators of NF-κB. Overall, three polysaccharides showed a good immunological activity and pose great potential as a novel food or drug additive.

Effects of Hydrolysis Condition and Detection Method on the Monosaccharide Composition Analysis of Polysaccharides from Natural Sources

Meijuan Zhao,Fengyan Kuang,Yingyue Zhang,Guangping Lv

Monosaccharide composition analysis is essential to the structural characterization and research into the biological activity of polysaccharides. In this study, a systematic comparison was performed among commonly used monosaccharide composition analysis methods, including colorimetric and chromatographic methods. These were tested on 16 aldoses, ketoses, alditols, amino sugars, and uronic acids. Furthermore, the effect of hydrolysis methods was also investigated. The results showed that the phenol sulfuric acid method is greatly affected by the type of monosaccharide that is used as the reference substance. The determination of uronic acid using sulfuric acid carbazole is less affected by neutral sugars than that method using m-hydroxybiphenyl. The high-performance thin-layer chromatography (HPTLC) method can simultaneously analyze multiple samples and accurately determine the type of uronic acid. High-performance liquid chromatography (HPLC) can provide a good qualitative and quantitative analysis of aldose, amino sugars, and uronic acids, while gas chromatography–mass spectrometry (GC-MS) can detect aldose, ketose, and alditols. Fructose was detected in a large amount in inulin andCodonopsis pilosulaafter one-step hydrolysis, while it was totally destroyed in two-step hydrolysis. The release of galacturonic acid significantly increased after two-step hydrolysis in pectin andLycium barbarum, which indicated that one-step hydrolysis is not enough for acidic polysaccharides. The results of this study are beneficial for selecting appropriate hydrolysis and analysis methods in order to accurately analyze the monosaccharide compositions of natural polysaccharides. Keywords:polysaccharide;monosaccharide composition;hydrolysis condition;detection method

Multi-fingerprint analysis for interpretation of the quality differences in polysaccharides during Dendrobium huoshanense traditional processing

Baojie Zhu,Cunwu Chen,Jiaxian He,Shaoping Li,Jing Zhao

The interpretation of the chemical quality differences during Dendrobium huoshanense ( D. huoshanense ) traditional processing into ‘Fengdou’ is critical for scientifically evaluating the medicinal value, optimizing the processing methods, and ensuring the stability and predictability of its efficacy. In this study, the quality differences of polysaccharides during the processing of D. huoshanense were conducted utilizing high performance size exclusion chromatography coupled with multi-angle laser light scattering and refractive index detector (HPSEC-MALLS-RID), nuclear magnetic resonance (NMR) and saccharide mapping based on polysaccharides analysis by using carbohydrate gel electrophoresis (PACE) and high-performance anion exchange chromatography-pulsed amperometric detection (HPAEC-PAD). The results showed that the concentration of non-starch D. huoshanense polysaccharides (NDHP) rose and exhibited greater stability during the processing. The molecular weight (Mw), molecular weight distribution, cumulative molar mass, radius of gyration (Rz), and monosaccharide composition molar ratio of NDHP did not have significant changes during the processing, and these parameters would be affected by the presence of starch. The NMR investigation indicated that the primary glycosidic linkages of β -1,4-Man p and β -1,4-Glc p , together with the acetyl group substitutions at O-2 and O-3 of 1,4-linked Man p , were unaltered. Additionally, the PACE and HPAEC-PAD profiles of acidic and enzymatic hydrolysates of NDHP were similar. These findings contribute to a deeper understanding of the changes in bioactive polysaccharides during the processing of D. huoshanense into ‘Fengdou’, providing theoretical support for improving its processing techniques.

Characterization and immunological activity of polysaccharides from two types of Dendrobium devonianum with different appearance

Wen Cao,Baojie Zhu,Xuan Zhang,Jurun Zhao,Shaoping Li,Jing Zhao

Dendrobium devonianum , a precious species of Dendrobium , is also called “ Zi-pi Shi-hu ” in Chinese due to its purple-skinned appearance. In this study, polysaccharides from green (GDP) and purple (PDP) skin D. devonianum were isolated by homogenate assisted extraction, ethanol precipitation and ultrafiltration. Their difference in physicochemical properties and immunological activities were investigated and compared. The results suggested that both GDP and PDP mainly contained glycosidic linkages of β -1,4-Man p and β -1,4-Glc p , but their molecular weights , 2.044 × 10 5 (GDP) vs 3.563 × 10 5 (PDP), and compositional monosaccharides of mannose to glucose ratio, 16.46: 1.00 (GDP) vs 7.61: 1.00 (PDP), were significantly different. Especially, GDP showed stronger immunological potentiation activity in vitro . Although the extraction yield of GDP was lower than that of PDP, considering the production of raw material and immunological activity of its polysaccharides, green skin D. devonianum should be a good alternative of purple one. These findings are helpful to provide evidence for improving the cultivation and production of D. devonianum .

Revealing the architecture and solution properties of polysaccharide fractions from Macrolepiota albuminosa (Berk.) Pegler

Yu-Xiao Wang,Yue Xin,Jun-Yi Yin,Xiao-Jun Huang,Jun-Qiao Wang,Jie-Lun Hu,Fang Geng,Shao-Ping Nie

Macrolepiota albuminosa (Berk.) Pegler is abundant in active polysaccharides , but little is known about their structures and solution properties. In this study, water-extracted polysaccharides from M. albuminosa (MAWP) were purified into three fractions with structural heterogeneity, which was attributed to the diversity in molecular weight, monosaccharide composition and linkage patterns, further affecting their solution properties. Methylation and NMR analysis revealed MAWP-60p and MAWP-70 were a 3- O -methylated glucomannogalactan and a previously unreported glucomannogalactan, whereas MAWP-80 was elucidated as a branched galactoglucan. Besides, three fractions exhibited random coil conformation in aqueous solution, while MAWP-60p had the highest viscosity due to its highest molecular weight, mean square radius of gyration ( R g ) and O -methyl group attached to the backbone. The molecular weight, monosaccharide composition and glycosidic linkages might be the major contributors to the flexibility, molecular size and stereochemistry of mushroom polysaccharide chains.

Comparison of the Antioxidant Activities and Polysaccharide Characterization of Fresh and Dry Dendrobium officinale Kimura et Migo

Wang Zhang,Xinjie Liu,Xun Sun,Rongchun Han,Nianjun Yu,Juan Liang,An Zhou

It is generally believed that freshDendrobium officinale(FDO) has more significant pharmacological activity than driedDendrobium officinale(DDO); however, the difference has not been clearly shown. Our study compared their antioxidant properties both in vitro and in vivo, and the molecular weight arrangement and monosaccharide composition of the freshDendrobium officinalepolysaccharides (FDOPs) and the driedDendrobium officinalepolysaccharides (DDOPs) were analyzed by HPLC-GPC and GC-MS. The results showed that the FDO and its polysaccharides had more significant effects on scavenging DPPH, ABTS, and hydroxyl radicals than the DDO. In addition, both the FDO and DDO significantly reduced lipid peroxidation levels and increased the SOD, T-AOC, CAT, and GSH levels in mice with acute liver damage caused by CCl4, while the FDO and its polysaccharides were more effective. Histopathological analysis further verified the protective effect of the Dendrobium polysaccharides on CCl4-induced liver injury. The determination of the polysaccharides revealed that the polysaccharide and mannose contents of the FDO were significantly higher than their dried counterparts, and the homogeneous arrangement of the polysaccharides in the FDO was degraded into three polysaccharide fragments of different molecular weights in the DDO. Overall, our data identified differences in the antioxidant activities of the FDO and DDO, as well as the reasons for these differences. Keywords:Dendrobium officinale;antioxidant activity;polysaccharide;mannose;monosaccharide composition

Structural elucidation and antioxidant activity of a novel heteroglycan from Tricholoma Lobayense

Shomaila Mehmood,Li-Yuan Zhou,Xiao-Fang Wang,Xiao-Du Cheng,Fan-Ju Meng,Ya Wang,Yong-Ming Lu,Yan Chen

A novel heteroglycan TLH-G was successfully isolated from Tricholoma lobayense cultivated in south China. TLH-G was composed of five monosaccharides, namely, mannose, rhamnose, glucuronic acid, glucose and galactose, and consisted of →1)-β-D-Glcp-(6→ and →1)-α-D-Galp-(3→ motifs. TLH-G had excellent scavenging activities towards 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals. Structure-activity relationship analysis demonstrated that glycosidic bond types, monosaccharide composition, and branching degree were more important for the antioxidant activity of polysaccharides from Tricholoma lobayense than other structural factors. All the above results showed that the growth environment played a crucial role in the structure and antioxidant activity of polysaccharides from Tricholoma lobayense.

Chemical characteristic and bioactivity of hemicellulose-based polysaccharides isolated from Salvia miltiorrhiza

Kui Zhao,Bo Li,Dongmei He,Can Zhao,Zhengjun Shi,Binbin Dong,Duo Pan,Rahul Rangrao Patil,Zhuyun Yan,Zhanhu Guo

Salvia miltiorrhiza roots (SMRs), the main component of cell wall from the residual waste extraction, differ depending on the forming ways of monosaccharides . The extraction from 8% sodium hydroxide solution (H-8) was characterized by gel permeation chromatography (GPC), monosaccharide composition, Fourier transform infrared spectroscopy (FT-IR), and nuclear magnetic resonance (NMR) spectroscopy. The structure model of hemicellulose-based polysaccharides (HBPs) was derived by combining one-dimensional and two-dimensional NMR. Monosaccharides difference and correlation were performed by partial least square analysis (PLS). Seven H-8s exhibited optimal inhibitory activities, which varied based on different sources of Danshen. The backbone structure indicated that 4-β-D-Xylp served as the main chain connected by 3-α-L-Araf or 5-α-L-Araf-1, 4-β-D-Galp, and β-D-Glcp branch, as well as α-L-Rhap, α-D-GalpA and α-D-GlcpA fragments. The variation of HBPs in terms of the structure and bioactivity of SMRs correlated with different cultivation sites can be a new approach to optimize and utilize the medical materials by chemical and biological aspects of natural macromolecules .

A Novel Method for the Analysis of Soluble Carbohydrates from Cistanche deserticola

Guo Yuanheng,Zhao Bing

Objective: A comprehensive monosaccharide quantifying method was developed in this article. Monosaccharides (including glucose, arabinose, galacturonic acid, galactose, mannose, rhamnose and xylose) were analyzed by RP-HPLC-DAD method. Meanwhile, mannitol and fructose were calculated based on peak areas obtained by HPIEC-RID and corresponding data obtained by RP-HPLC-DAD. Monosaccharide compositions of CDSC were analyzed, nine kinds of monosacchardes (glucose, arabinose, galctose, galacturonic acid, mannose, rhamnose, xylose, fructose and mannitol) in were quantified. Background: Cistanche deserticola Y. C. Ma, is a perennial holoparasite and mainly distributed in the desert region of northwestern China. It reported that C. deserticola soluble carbohydrate (CDSC) could prompt lymphocyte proliferation, modulate immunity, inhibit type 2 diabetes, anti-inflammation. For these reasons, it is very appealing to researchers to have a convenient method for monosaccharides analysis of the CDSC. However, previous researches have neglected the faultiness in monosaccharide composition analysis. Therefore, in this study we will focus on establishing a quantifying method of monosaccharides possibly found in C. deserticola. Methods: Soluble carbohydrates were hydrolyzed with trifluoroacetic acid, monosaccharides were labeled with 1-phenyl-3-methyl-5-pyrazolone (PMP), separated by HPLC system equipped with C18 column and detected with DAD (RP-HPLC-DAD). RP-HPLC-DAD method was used for quantitative analysis of glucose, arabinose, galacturonic acid, galactose, mannose, rhamnose and xylose. Meanwhile, hydrolyzates were separated by HPIEC system equipped with aminex HPX-87H ion exclusion column and detected by refractive index detector (HPIEC-RID). The quantity of mannitol and fructose can be calculated based on peak areas obtained by HPIEC-RID and corresponding data obtained by RP-HPLC-DAD. Results: Utilizing this method, monosaccharide compositions of CDSC with different molecular weight (CDCSA, CDCS-B, CDCS-C and CDCS-D) were analyzed, nine kinds of monosacchardes (glucose, arabinose, galctose, galacturonic acid, mannose, rhamnose, xylose, fructose and mannitol) in were found. Conclusion: Fructose and mannitol, which are abundant present in C. deserticola, cannot be analysis by normally PMP modify method, therefore, a comprehensive quantitative analysis method was developed in this research. Galacturonic acid was found out in macro-molecular polysaccharides from C.deserticola, which was not reported before. Galactose and galacturonic acid mainly exist in macro-molecular polysaccharide. Fructose and mannitol mainly exist in mini-molecule or exist in free form.

Yin Zhenhua,Zhang Wei,Zhang Juanjuan,Kang Wenyi

In this study, Ligustrum lucidum flowers as raw material, the extraction, isolation and coagulatory activity of polysaccharides were carried out for the first time. The crude polysaccharide was obtained by hot water extraction and ethanol precipitation, and preliminarily purified by Sevage method and D101 macroporous resin. Then the polysaccharide was further purified by DEAE-52 cellulose and Sephadex G-100 column chromatography, respectively. The structural characteristics were detected by LC, GC, FT-IR and NMR. Furthermore, the coagulatory activity of the polysaccharides were investigated by APTT, TT, PT and FIB assays in vitro. The results demonstrated that four polysaccharides were isolated from flowers of L. lucidum, named as LLP-1a, LLP-1b, LLP-2 and LLP-3, and the yields were 0.039, 0.0054, 0.0055 and 0.017%, respectively based on the weight of the dried flowers. The four polysaccharides components were free of nucleic acids and proteins, and their average molecular weights were 25,912, 64,919, 3,940,246 and 2,975,091 g/mol, respectively. The monosaccharide compositions of LLp-1a were l-rhamnose, l-arabinose, d-xylose, d-glucose and d-galactose (molar ratio of 3.16: 2.46: 1.00: 7.27: 4.22). Only d-galactose was detected from LLp-1b. LLp-2 was composed of l-arabinose, d-glucose and d-galactose (molar ratio of 1.28:1.32:1.00). LLp-3 was composed of l-rhamnose, l-arabinose, d-xylose, d-glucose and d-galactose (molar ratio of 5.85: 2.21: 2.23: 1.00: 2.25). Coagulation assays indicated that LLp-1a and LLp-3 had good anticoagulant effect in vitro, while LLp-1b showed procoagulant activity.
