Extracts of Portulaca oleracea and Patrinia scabiosaefolia relieve ultraviolet B-induced skin injury in solar dermatitis mice via inhibiting IL-17/CCL2 pathway and oxidative stress

Li Bowei,Yang Jingyuan,Wang Zhixing,He Mingzhu,Chen Xujing,Chen Yixuan,Shen Baoying,Chen Junru,Yang Chunqi,Li Tian,Lai Chengcai,Gao Yue,Cai Hong

Introduction: Solar dermatitis, a condition triggered by excessive exposure to ultraviolet B (UVB) radiation, results in inflammatory skin damage marked by erythema, edema, and epidermal injury. Portulaca oleracea (PO) and Patrinia scabiosaefolia (PS) have been traditionally used in dermatological treatments, though their mechanistic pathways in UVB-induced skin injury are not fully understood. Methods: In this study, a mouse model of UVB-induced solar dermatitis was employed to evaluate the therapeutic potential of combined PO and PS (POPS) extracts. After UVB irradiation, POPS extracts were administered, and their bioactive compounds were identified through ultra-performance liquid chromatography-mass spectrometry (UPLC-MS). Network pharmacology, molecular docking, and pathway analysis were performed to identify key targets, focusing on the IL-17/CCL2 pathway and oxidative stress reduction. Results: Treatment with POPS extracts significantly diminished UVB-induced inflammation, erythema, and epidermal thickening in a dose-dependent manner. Network pharmacology and docking studies identified curvularin and olmesartan medoxomil as bioactive components with high affinity for IL-17 and IL-17RA targets, modulating the IL-17/CCL2 axis. In vivo experiments demonstrated that POPS extracts suppressed the expression of IL-17 and CCL2, reduced macrophage infiltration, and alleviated oxidative stress, effectively mitigating the symptoms of solar dermatitis. Conclusion: This study provides insight into the anti-inflammatory and protective properties of POPS extracts in solar dermatitis, highlighting their potential as a treatment through IL-17/CCL2 pathway modulation and oxidative stress reduction.

Selenium deficiency sensitizes the skin for UVB-induced oxidative damage and inflammation which involved the activation of p38 MAPK signaling

Xiaokang Zhu,Mingdong Jiang,Erqun Song,Xiaoji Jiang,Yang Song

Ultraviolet B (UVB) radiation causes oxidative damage and inflammation, and ultimately increases the risk of skin carcinogenesis. Selenium is an essential trace element, previous studies indicated selenium deficiency impairs tissue antioxidant capacity in different experimental models. However, the synergistic effect of selenium deficiency and UVB radiation on skin damage is not clear. In the current study, our data revealed selenium deficiency resulted in further increases of reactive oxygen species (ROS), thiobarbituric acid reactive substances (TBARS) and phosphorylated H2AX levels, decreases of GSH level and antioxidant enzyme activities in UVB-irradiated mice. Selenium deficiency also exacerbated UVB-induced cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), tumor necrosis factor-alpha (TNF-α), interleukin-1beta (IL-1β) and IL-6 mRNA expressions. Mechanism studies indicated that UVB-induced p38 signaling was further elevated in the skin of mice maintained with selenium deficiency diet, compared with those maintained with selenium adequate diet. Our investigation suggested that selenium deficiency diet weakens the antioxidant capacity of UVB-irradiated mice skin, which sensitizes to UV radiation-induced oxidative damage and inflammation.

Disguise as fluorescent powder: Ultraviolet-B persistent luminescence material without visible light for advanced information encryption and anti-counterfeiting applications

Lin Liu,Junpeng Shi,Yiang Li,Shanshan Peng,Hongyun Zhong,Liang Song,Yun Zhang

Persistent luminescent material (PLM) is promising candidate in anti-counterfeiting field. Specifically, Ultraviolet (UV) PLM without visible persistent luminescence (PersL) can realize information encryption with high concealment and security. However, UV-PLM has not yet been applied in anti-counterfeiting owing to the lack of invisible UV-PLM. Herein, we, for the first time, report a novel invisible ultraviolet-B (UVB) PLM and applied in advanced anti-counterfeiting and encryption field, exhibting its unique advantages. First, we prepared a novel series of UV-PLM, Mg 2 Ln 2 Ge 3 O 11 :Bi 3+ (Ln = Gd, La, Y, and Lu) and then realized the tuning of PersL from 350 to 310 nm via element substitution. Specifically, Mg 2 Lu 2 Ge 3 O 11 :Bi 3+ (MLG) can emit dim visible photoluminescence under 254 nm excitation and produce UVB-PersL without visible region after the excitation was ceased, which caused it appear like an ordinary fluorescent powder. Interestingly, the invisible UVB-PersL can be detected using a camera realizing the information encryption with high concealment. Moreover, owing to its unique optical properties, mixing MLG with different colors of fluorescent powder endows it with the ability to camouflage. Based on MLG, we designed a triple-mode information encryption method and demonstrated its unique advantages in the field of advanced anti-counterfeiting.

NaCl Induces Flavonoid Biosynthesis through a Putative Novel Pathway in Post-harvest Ginkgo Leaves

Ni Jun,Hao Juan,Jiang Zhifang,Zhan Xiaori,Dong Lixiang,Yang Xiuli,Sun Zhehang,Xu Wenya,Wang Zhikun,Xu Maojun

The flavonoids in the extracts of Ginkgo leaves have been shown to have great medical value: thus, a method to increase the flavonoid contents in these extracts is of significant importance for human health. In the present study, we investigated the changes in flavonoid contents and the corresponding gene expression levels in post-harvest Ginkgo leaves after various treatments. We found that both ultraviolet-B and NaCl treatment induced flavonoid accumulation. However, gene expression analysis showed that the increases in flavonoid contents were achieved by different pathways. Furthermore, post-harvest Ginkgo leaves responded differently to NaCl treatment compared with naturally grown leaves in both flavonoid contents and corresponding gene expression. In addition, combined treatment with ultraviolet-B and NaCl did not further increase the flavonoid contents compared with ultraviolet-B or NaCl treatment alone. Our results indicate the existence of a novel mechanism in response to NaCl treatment in post-harvest Ginkgo leaves, and provide a technique to increase flavonoid content in the pharmaceutical industry.

Panax ginseng C. A. Meyer Phenolic Acid Extract Alleviates Ultraviolet B-Irradiation-Induced Photoaging in a Hairless Mouse Skin Photodamage Model

Zhenzhuo Li,Rui Jiang,Jianzeng Liu,Xiaohao Xu,Liwei Sun,Daqing Zhao

Here, we evaluated the in vivo skin-protective effects of topical applications of Panax ginseng C. A. Meyer extract (PG2) and its phenolic acid- (PA-) based components against UVB-induced skin photoaging. PG2 or PA applied to skin of hairless mice after UVB-irradiation alleviated UVB-induced effects observed in untreated skin, such as increased transepidermal water loss (TEWL), increased epidermal thickness, and decreased stratum corneum water content without affecting body weight. Moreover, PG2 and PA treatments countered reduced mRNA-level expression of genes encoding filaggrin (FLG), transglutaminase-1 (TGM1), and hyaluronan synthases (HAS1, HAS2, and HAS3) caused by UVB exposure and reduced UVB-induced collagen fiber degradation by inhibiting the expression of matrix metalloproteinase genes encoding MMP-1, MMP-2, and MMP-9. Meanwhile, topical treatments reduced cyclooxygenase-2 (COX-2) mRNA-level expression in photodamaged skin, leading to the inhibition of interleukin-1 β (IL-1 β ) and interleukin-6 (IL-6) mRNA-level expression. Thus, ginseng phenolic acid-based preparations have potential value as topical treatments to protect skin against UVB-induced photoaging.

Degradation and Transformation Mechanisms of Zanthoxylum Alkylamides Exposed to UVB Light

Rui Wang,Chaolong Rao,Qiuyan Liu,Xiong Liu

Zanthoxylumoleoresin, a concentrated extract derived fromZanthoxylum bungeanum, is rich in non-volatile, intensely flavorful substances and amide compounds, such as hydroxy-α-sanshool, hydroxy-β-sanshool, and hydroxy-ε-sanshool. The production process ofZanthoxylumoleoresin remains unstandardized, and there is still a lack of research on the precise classification and quantification of its key chemical constituents, as well as the stability of these compounds when produced using different extraction methods. This study utilized preparative liquid chromatography to extract and purify amide compounds fromZanthoxylumoleoresin, successfully isolating three sanshools: hydroxy-α-sanshool, hydroxy-β-sanshool, and hydroxy-ε-sanshool. The stability of three these sanshools under UVB irradiation in different solvents was explored in UVB-simulated sunlight conditions to investigate the degradation or transformation mechanism ofZanthoxylumalkylamides under UVB irradiation. The findings indicate a rapid decrease in the hydroxy-α-sanshool content under UVB ultraviolet light, aligning with the second-order kinetics. This study revealed alterations in the contents of hydroxy-α-sanshool, hydroxy-β-sanshool, and hydroxy-ε-sanshool and the formation of a new compound following exposure to UVB light. This new compound, along with the three sanshools, possesses a uniformm/z264 and shares similar chemical structures. Further analysis also uncovered that these compounds are capable of undergoing isomerization reactions under UVB irradiation. This demonstrates that UVB irradiation of certain intensities can modify the concentrations and chemical structures of theseZanthoxylumalkylamides. These insights offer crucial guidance for future studies on the processing and preservation ofZanthoxylumalkylamides and their derivatives. Keywords:Zanthoxylumalkylamides;UVB light;degradation;transformation;mechanism

Development of ultraviolet-B long-lived persistent phosphors in Pr3+-doped garnets

Shao Yan,Yanjie Liang,Jingwei Liu,Dongxun Chen,Shihai Miao,Jianqiang Bi,Kangning Sun

Inorganic luminescent materials that emit in the ultraviolet-B (UVB) region (280–320 nm) have gained considerable attention in recent years because of the growing demands for applications in photochemistry and photomedicine. However, most of the available UVB phosphors are photoluminescent, for which continuous external excitation is essential. This common but very inconvenient luminescence form hinders the further development of UVB luminescence technology. Here, we report the design and development of a series of Pr3+-doped garnet-based UVB persistent phosphors by combining conduction band engineering, the vacuum referred binding energy (VRBE) diagram and persistent energy transfer control. The developed UVB persistent phosphors can not only be charged using a standard 254 nm UV lamp but also natural sunlight, leading to an intense and long-lasting UVB afterglow of more than 60 h. Besides, the persistent luminescence properties and trap charging and detrapping processes are comprehensively investigated using persistent luminescence excitation spectra and thermoluminescence experiments to uncover the possible luminescence mechanism in these phosphors. More importantly, the unique UVB persistent light emission from these garnet phosphors can be clearly imaged and recorded using a UVB camera in a bright indoor-lighting environment because of the zero-background noise from indoor ambient light, indicating that these UVB persistent phosphors have important application potential for optical tagging and optical data storage in a bright environment.

Enhancement of Ultraviolet B Irradiation with a Photoluminescent Composite Film and Its Application in Photochemical Microfluidic Synthesis

Jiaojiao Fan,Yuanzhi Zheng,Yingtian Yang,Le Du,Yujun Wang

Ultraviolet B (UVB) irradiation is of great importance for photochemical synthesis of steroidal intermediates, which could be efficiently provided by neither traditional mercury lamps nor novel light-emitting diodes. Herein, a photochemical microfluidic strategy with enhanced UVB irradiation was developed by the integration of novel photoluminescent composite films, which were readily prepared to shift the emission peak. The UVB emission intensity was increased by approximately 10 times for an 8 W lamp with this flexible and removable film. It allowed a productivity as high as that of a 100 W lamp without using any cooling devices, showing great potential to replace traditional medium-pressure mercury lamps for UVB-irradiated synthesis. In addition, a calculation method was developed to assess the effect of specific wavelengths based on the photon flux and photonic efficiency, which could provide theoretical guidance for photochemical microfluidic processes to refine the emission ranges.

Quercetin and Its Mixture Increase the Stress Resistance of Caenorhabditis elegans to UV-B

Shi-ming Li,Dan Liu,Yi-lin Liu,Bin Liu,Xing-huang Chen

Ultraviolet B (UV-B, 280–320 nm) radiation causes complex molecular reactions in cells, including DNA damage, oxidative stress, and apoptosis. This study designed a mixture consisting of quercetin, luteolin and lycopene and usedCaenorhabditis elegansas a model to study the resistance of these natural chemicals to UV-B. Specifically, we have confirmed that quercetin and its mixture can increase the resistance ofCaenorhabditis elegansto UV-B through lifespan test, reactive oxygen species level assay, germ cell apoptosis test, embryonic lethal test and RT-qPCR experiments. The results show that quercetin and its mixture prolonged the lifespan of UV-B-irradiatedCaenorhabditis elegansand reduced abnormal levels of reactive oxygen species, embryo death, and apoptosis induced by UV-B. The protective effect of quercetin and its mixture may be attributed to its down-regulation ofHUS-1,CEP-1,EGL-1andCED-13.Therefore, the results of this research could help the development of UV-B radiation protection agents. Keywords:Caenorhabditis elegans;UV-B;quercetin;luteolin;lycopene;apoptosis;stress resistance

Anti-Inflammatory Activity of Panax notoginseng Flower Saponins Quantified Using LC/MS/MS

Junchen Liu,Yuehang Wu,Wenrui Ma,Hongyan Zhang,Xianyao Meng,Huirong Zhang,Miaomiao Guo,Xiao Ling,Li Li

Panax notoginseng(Burk) F. H. Chen is a traditional Chinese medicinal and edible plant. However,Panax notoginsengflower (PNF) is rarely used. Therefore, the purpose of this study was to explore the main saponins and the anti-inflammatory bioactivity of PNF saponins (PNFS). We explored the regulation of cyclooxygenase 2 (COX–2), a key mediator of inflammatory pathways, in human keratinocyte cells treated with PNFS. A cell model of UVB-irradiation-induced inflammation was established to determine the influence of PNFS on inflammatory factors and their relationship with LL–37 expression. An enzyme-linked immunosorbent assay and Western blotting analysis were used to detect the production of inflammatory factors and LL37. Finally, liquid chromatography–tandem mass spectrometry was employed to quantify the main active components (ginsenosides Rb1, Rb2, Rb3, Rc, Rd, Re, Rg1, and notoginsenoside R1) in PNF. The results show that PNFS substantially inhibited COX–2 activity and downregulated the production of inflammatory factors, indicating that they can be used to reduce skin inflammation. PNFS also increased the expression of LL-37. The contents of ginsenosides Rb1, Rb2, Rb3, Rc, and Rd in PNF were much higher than those of Rg1, and notoginsenoside R1. This paper provides data in support of the application of PNF in cosmetics. Keywords:Panax notoginsengflower;UVB;inflammatory factor;antibacterial peptide LL-37;total saponin;LC/MS/MS

Structure characterization and protective effect against UVB irradiation of polysaccharides isolated from the plateau plant Gentiana dahurica Fisch

Wen Ji,Cuiyin Qian,Xiaopeng Su,Xiang Li,Zhenqing Zhang,Yonggui Ma,Mingjin Zhang,Duxin Li

Gentiana dahurica Fisch. ( G. dahurica ) is one of the legitimate sources of Qinjiao in Traditional Chinese Medicine (TCM) and grows on high-altitude plateaus. Plants develop unique biochemical accumulations to resist plateau conditions, especially the strong UV irradiation. Thus, this study aimed to investigate the polysaccharide of G. dahurica (GDP), its structure and its activity against UVB irradiation. Four GDPs were isolated and two of them were subjected to structural elucidation. The results suggested that GDP-1 has 53.5 % Ara and 30.8 % GalA as its main monosaccharides, with a molecular weight (Mw) of 23 kDa; the GDP-2 has 33.9 % Ara and 48.5 % GalA, with a Mw of 82 kDa. Methylation and NMR spectroscopy analysis revealed that GDP-1 contains →5)-α-Ara f -(1 → 5)-α-Ara f -(1 → 3,5)-α-Ara f -(1 → 3,4)-α-Gal p A-(6-OMe)-(1→ as the main chain, the branches of GalA (with esterification), and the terminal Ara; the GDP-2 contains →4)-α-Gal p A-(1 → 4)-α-Gal p A-(6-OMe)-(1 → 5)-α-Ara f -(1 → 3,5)-α-Ara f -(1→ as the main chain, the branches of →5)-α-Ara f -(1–5)-α-Ara f , and the terminal GalA. Both GDP-1 and GDP-2 exhibited concentration-dependent antioxidant activity against DPPH, ABTS and hydroxyl radicals. Moreover, GDPs significantly attenuated the decreases in viability and proliferation of HaCaT cells after UVB irradiation. They can scavenge reactive oxygen species (ROS) and improve the activities of endogenous antioxidant enzymes, including superoxide dismutase (SOD) and glutathione peroxidase (GSH). The potential mechanism explored by flow cytometry assays of cell apoptosis and cell cycle distribution suggested that GDPs exert protective effects against UVB irradiation by reducing ROS and attenuating S phase cell arrest. In brief, the GDP-1 and GDP-2 are α-1,3- and α-1,4- arabinogalacturonan, respectively. The high content of Ara could be attributed to biochemical accumulation in resisting to the plateau environment and to prevent UVB irradiation-related damage in cells. These findings provide insight into authentic medicinal herbs and the development of GDPs in the modern pharmaceutical and cosmetics industry.

Molecular characterization of a feruloyl-CoA 6′-hydroxylase involved in coumarin biosynthesis in Clematis terniflora DC

Minglei Tao,Shengzhi Liu,Yaohan Li,Amin Liu,Jingkui Tian,Yuchang Liu,Hongwei Fu,Wei Zhu

Coumarin is an important secondary metabolite that affects plant physiology. It is a lactone of cis- o -hydroxycinnamic acid and widely exists in medicinal plants . Clematis terniflora DC. is a plant belonging to Ranunculaceae and is rich in variety of coumarins. Feruloyl-CoA 6′-hydroxylase has been reported as a key enzyme in the formation of coumarin basic skeleton only in some common plants, however, its evidence in other species is still lacking especially for the biosynthesis of coumarins in C . terniflora . In the present study, we identified a feruloyl-CoA 6′-hydroxylase CtF6′H in C . terniflora , and functional characterization indicated that CtF6′H could hydroxylate feruloyl-CoA to 6-hydroxyferuloyl-CoA. Furthermore, the expression level of CtF6′H was differed among different tissues in C. terniflora , while under UV-B radiation, the level of CtF6′H was increased in the leaves. Biochemical characteristics and subcellular location showed that CtF6′H was mainly present in the cytosol. The crystal structure of CtF6′H was simulated by homology modeling to predict the potential residues affecting enzyme activity . This study provides the additional evidence of feruloyl-CoA 6′-hydroxylase in different plant species and enriches our understanding of biosynthetic mechanism of coumarin in C . terniflora .
