Anthocyanins (ACNs) are the primary bioactive compounds of purple perilla leaves with a variety of biological activities. The scale separation of perilla ACNs is crucial for their practical application. This study presents the development of a green and highly efficient foam fractionation technology employing a protein-based frother and collector. The protein utilized, namely perilla seed meal protein (PSMP), is derived from perilla seed meal (PSM), which is normally discarded as an industrial waste during seed oil extraction. PSMP demonstrated notable foamability and foam stability. The interaction or collection mechanism between PSMP and ACNs was comprehensively investigated by SEM-EDS, FTIR, and fluorescence spectroscopy , revealing a hydrophobic interaction between PSMP and ACNs. Furthermore, the impacts of PSMP concentration, pH, and gas velocity on the foam fractionation of ACNs were thoroughly examined. Response surface methodology (RSM) was used to optimize the operating conditions for ACNs separation. Remarkable results were achieved under the optimum conditions, including a high recovery percentage of 95.13% and an impressive enrichment ratio of 27.56. The composition and antioxidation of isolated ACNs product were characterized, revealing its excellent antioxidant activities . This highlights its potential applications in drugs, food, and cosmetics. This work not only highlights an efficient foam fractionation technology for ACNs separation, but also offers valuable insights into the separation of other active plant components.

Rosmarinic acid (RA), a prominent phytochemical, is abundant in perilla leaves and is sought after for its beneficial properties. A highly effective, economically applicable and environmentally safe method has been successfully developed for extracting and separating RA from RA extract. Response surface methodology (RSM) was used to optimize the ultrasound-assisted extraction conditions of Perilla leaves. The optimum extraction conditions for the ultrasound-assisted extraction of RA were obtained as liquid to material ratio of 49 mL/g, a temperature of 44 °C, and an extraction time of 31 min. Under these conditions, the yield of RA was 0.87 %. Aqueous two-phase flotation (ATPF) is an innovative method that demonstrates exceptional efficiency in isolating biomolecules characterized by surface-active properties. Our research innovates by employing perilla seed meal protein (PSMP) as an environmentally sustainable carrier for the ATPF process to isolate RA, overcoming its deficiency in surface activity. Utilizing a dual-phase system consisting of polyethylene glycol and (NH 4 ) 2 SO 4 , we conducted a comprehensive analysis of various factors influencing the separation efficiency. Under the optimized conditions comprising 30 % (NH 4 ) 2 SO 4 , 8 % PEG-800, pH 4.0, PSMP carrier concentration 101 mg/L, gas velocity 54 mL/min, and operation time 7 min, we attained outstanding results with an E % of 97.89 % and a K p of 98.45. The RA obtained through this method displayed potent antioxidant activity, suggesting its utility in various applications. This research delineates an efficient ATPF protocol for RA isolation and offers valuable insights for the extraction of other botanical compounds.

Perilla leaves are a common traditional medicinal and edible crop, in order to further develop its medicinal value, a neutral polysaccharide was extracted and analyzed on its structure and studied on sulfation modification. After purification by different column chromatography, the optimal process for sulfation modification of PLP-2-1 was determined, and S-PLP-2-1 was prepared. The potential structural framework and molecular weight of PLP-2-1 were determined by HPLC, methylation analysis, and NMR techniques, while sulfated modified S-PLP-2-1 maintained a high degree of structural similarity with it. Particle size, Zeta potential, infrared spectroscopy, and NMR analysis confirmed the presence of sulfate groups in S-PLP-2-1, while thermogravimetric and rheological analyses provided support for the development and utilization of the two polysaccharides. After determined the safe concentration range in vitro, the antioxidant and hypoglycemic activities of PLP-2-1 and S-PLP-2-1 were measured. Besides, the effects on the levels of TC, TG, HDL-C, and LDL-C in OA induced high-fat HepG2 cells of both polysaccharides were determined. The results showed that the various activities of S-PLP-2-1 were significantly improved after sulfation modification. Therefore, PLP-2-1 and S-PLP-2-1 may be potential drugs for anti-glycolipid metabolism disorders.

Perillae Folium (PF) is a well-known food and herb containing different chemotypes, which affect its quality. Herein, a method was proposed to classify and quantify PF chemotypes using gas chromatography–mass spectrometry (GC–MS) and Fourier transform-near infrared spectroscopy (FT-NIR). GC–MS results revealed that PF contains several chemotypes, including perilla ketone (PK) type, α -asarone (PP-as) type, and dillapiole (PP-dm) type, with the PK type being the predominant chemotype. Based on FT-NIR data, different chemotypes were accurately classified. The random forest algorithm achieved >90 % accuracy in chemotype classification. Furthermore, the main components of perilla ketone and isoegomaketone in PF were successfully quantified using partial least squares regression models, with prediction to deviation values of 3.76 and 2.59, respectively. This method provides valuable insights and references for the quality supervision of PF and other foods.

Fenoxaprop-P-ethyl (FE) can effectively control weeds in rice fields, but it has been found to cause phytotoxicity in rice. In this study, the phytotoxicity of FE was mitigated by perilla leaf essential oil (PEO) in rice seedlings. The injury recovery rates (IRRs) for shoot length and fresh weight treated with 800 mg/L of PEO were 101.51% and 99.05%, respectively. Moreover, the damage ofs-metolachlor and pretilachlor was also alleviated when co-applied with 800 mg/L PEO; the IRR ofs-metolachlor phytotoxicity was 26.07% and 27.34%, respectively, and the IRR of pretilachlor phytotoxicity was 127.27% and 124.39%, respectively. However, PEO had no significant effect on the phytotoxicity of pinoxaden, mesotrione, penoxsulam, mesosulfuron-methyl, and nicosulfuron. The results of GC–MS analysis showed that a total of 23 components were detected in PEO, among which linalool (36.49%), linalyl formate (26.96%), α-terpineol (10.63%), 2-hexanoylfuran (5.81%), geranyl acetate (4.13%), and neryl acetate (2.30%) were the primary components. Among them, 2-hexanoylfuran was the most effective component to alleviate FE damage, for which the IRR of shoot length and fresh weight was 73.17% and 73.02%, respectively, followed by the geranyl acetate, for which the IRR was 72.32% and 60.56%, respectively, and neryl acetate, for which the IRR was 65.28% and 58.11%, respectively. Furthermore, the application of 50 mg/L of 2-hexanofuran significantly improved the tolerance of shoot length and fresh weight to FE stress by factors of 5.32 and 5.35, respectively. This research demonstrates that PEO and 2-hexanoylfuran have the potential to serve as natural safeners to reduce phytotoxicity.

Perillae Folium (PF), the leaf of Perilla frutescens (L.) Britt is extensively used as culinary vegetable and medicinal herb in many countries. Purple PF ( P. frutescens var. arguta ) and green PF ( P. frutescens var. frutescens ) are two of the main varieties. In this study, UHPLC-Q-TOF/MS assisted with feature-based molecular networking (FBMN) was applied for chemical characterization. In total, 82 metabolites, mainly phenolic acid derivatives were identified from PF. With the help of FBMN, five organic acid glucosides together with three N -phenylpropenoyl-L-amino acids (NPAs) were identified in PF using FBMN for the first time. Multivariate statistical methods were utilized for comprehensive comparison of purple and green PF profiles. 12 compounds with their relative contents varied significantly between purple and green PF were screened out. Overall, the present study offers valuable insights for chemical elucidation of PF which would be helpful for comprehensive utilization of a PF resource.
