Tissue distribution and enhanced in vivo anti-hyperlipidemic-antioxidant effects of perillaldehyde-loaded liposomal nanoformulation against Poloxamer 407-induced hyperlipidemia

Emmanuel Omari-Siaw,Qilong Wang,Congyong Sun,Zengquan Gu,Yuan Zhu,Xia Cao,Caleb Kesse Firempong,Rita Agyare,Ximing Xu,Jiangnan Yu

An optimized perillaldehyde-loaded liposomal nanoformulation (PAH-LNF) was successfully applied to improve the pharmacological effect of perillaldehyde (PAH) in poloxamer 407-induced hyperlipidemia . Oral administration of PAH-LNF (240   mg/kg per body weight) in rats significantly enhanced solubility and relative bioavailability (270.7%) compared to the free PAH with about 2.7-, 1.5-, 1.3-, 1.3- and 1.5-fold increase in AUC, T 1/2 , MRT, C max and T max , respectively. Tissue distribution study also revealed the accumulation of PAH in the liver, lungs, spleen, kidney, brain and heart in order of decreasing affinity. Moreover, a significant decrease in serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) with simultaneous increase in high-density lipoprotein cholesterol (HDL-C) level was observed in the chemically-induced hyperlipidemic mice which further confirmed PAH’s anti-hyperlipidemic properties. Additionally, PAH-LNF also significantly increased the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) with a concurrent decrease in malondialdehyde (MDA) to affirm the antioxidant and hepatoprotective effects of PAH. Thus, liposomal nanoformulation promises to be a useful drug delivery system for the development of PAH.

A novel gelatin/chitosan-based “sandwich” antibacterial nanofiber film loaded with perillaldehyde for the preservation of chilled chicken

Xinxia Wang,Zaitian Wang,Zhilan Sun,Daoying Wang,Fang Liu

Quality deterioration caused by microorganisms is a crucial problem in food industry. Herein, to enhance the antibacterial effect and extend the storage life of chilled chicken, a gelatin/chitosan (GC)-based “sandwich” nanofiber film was prepared by sandwiching GP2 (gelatin nanofibers loaded with 2 % ( v /v) perillaldehyde) between two layers of GC nanofibers. The diameter of GP nanofibers was positively correlated with perillaldehyde concentration. However, the thermal stability of GP nanofibers was negatively correlated. The thermal degradation temperature (Tm) of GP2 was the lowest. Fortunately, GC nanofibers improved Tm of GP2 from 98.91 °C to 111.60 °C. Besides, FTIR absorption bands at 1636 and 1442 cm −1 indicated that perillaldehyde was successfully embedded. Moreover, poor water resistance of gelatin nanofibers was also improved. Specifically, benefiting from the synergy between perillaldehyde and chitosan, the shelf-life of chilled chicken was efficiently prolonged (over 10 days). Thus, this “sandwich” nanofiber film shows promise for food packaging.

Perillaldehyde pretreatment alleviates cerebral ischemia-reperfusion injury by improving mitochondrial structure and function via the Nrf2/Keap1/Trx2 axis

Ji Liu,Ying Han,Zhiyun Wu,Manli Chen,Wenwen Wu,Zijun Zhao,Jinjin Yuan,Zhijian Zheng,Qiang Lin,Nan Liu,Hongbin Chen

Background Perilladehyde, an extract of perillae in the Labiatae family, can produce significant anti-inflammatory and antioxidant effects. Although literature evidences the favorable effect of perillaldehyde on ischemic stroke, the exact mechanism remains blurred. Purpose This study attempted to explore the impact of perillaldehyde on cerebral ischemia-reperfusion injury and the related action mechanism. Methods The rat tMCAO and neuronal OGD/R models were established to simulate cerebral ischemia-reperfusion injury. Lentiviruses were used to interfere with the expression of Nrf2 and Trx2 in neurons. The effects and action mechanisms of perillaldehyde were explored by various experimental methods, including chromatin immunoprecipitation assay, Western Blot, flow cytometry, dual-luciferase reporter gene assay, transmission electron microscopy, MRI, RNA-seq, and immunofluorescence staining. Results Perillaldehyde pretreatment effectively mitigated the tMCAO-induced brain injury in rats by reducing cerebral infarction, improving neuromotor function, and attenuating cell apoptosis in the ischemic penumbra. In vitro, perillaldehyde pretreatment alleviated cell death and excessive oxidative stress, preserved the mitochondrial membrane integrity, enhanced mitochondrial energy metabolism, and facilitated the restoration of mitochondrial ultrastructure after OGD/R. The mechanism probe revealed that perillaldehyde activated the Nrf2/Keap1/Trx2 signaling axis, thus promoting the transcription of Trx2 and improving mitochondrial structure and function. The aforementioned impacts of perillaldehyde were somewhat counteracted by disrupting the expression of Nrf2 and Trx2, suggesting that the neuroprotection of perillaldehyde partially involves the activation of the Nrf2/Keap1/Trx2 axis. Conclusions This study firstly demonstrates the existence of the Nrf2/Keap1/Trx2 signaling axis in cerebral ischemia-reperfusion injury and evidences that perillaldehyde pretreatment can promote the restoration of neuronal mitochondrial structure and function by activating the Nrf2/Keap1/Trx2 axis after cerebral ischemia-reperfusion injury. These findings signify that perillaldehyde holds great promises for clinical management of ischemic stroke.

Strain-specific endophytes modulate physiological and metabolic responses of Perilla frutescens to cadmium stress

Gen Li,Simiao Wang,Yijin Wang,Yibang Zhang,Qing Dang,Yashuo Tian,Longfei Li,Lijuan Wang,Li Han

Perilla frutescens is a medicinal plant that bioactive compounds such as perillaldehyde and luteolin, but cadmium (Cd) contamination threatens its safe cultivation and medicinal quality. While dark septate endophytes (DSEs) are known to increase plant stress tolerance, their strain-specific roles in regulating Cd uptake and secondary metabolite biosynthesis remain poorly understood. This study aimed to investigate three DSE strains, DSE6 ( Paraphoma radicina ), DSE7 ( P. chlamydocopiosa ), and MD3-1 ( Acrocalymma aquatica ), to determine their effects on growth, physiological traits, Cd accumulation, and key metabolite production in P. frutescens under 0, 1, 5, and 10 mg/kg Cd stress. DSE6 and DSE7 significantly reduced root Cd content and improved photosynthetic pigments, antioxidant activity, and biomass under moderate Cd levels. Notably, MD3-1 did not reduce Cd uptake but increased luteolin content by 22.67 % under high Cd stress, suggesting a decoupling between Cd accumulation and metabolite production. All three strains increased perillaldehyde content, with DSE7 showing the greatest increase (262 %) at 5 mg/kg Cd. These findings demonstrate that DSEs regulate Cd tolerance and medicinal quality via distinct mechanisms, and that internal metabolic tolerance may sustain bioactive compound synthesis under heavy metal stress. This highlights their potential as microbial agents for cultivating medicinal plants in contaminated soils.

Perillaldehyde controls citrus green mold by inhibiting the ribosome biogenesis of Penicillium digitatum and improving citrus disease resistance

Shuqi Liu,Yuxin Jiang,Jiancheng Sun,Fan Yang,Yuqing Wang,Yongqing Lu,Weiqiang Lai,Chao-an Long

Perillaldehyde (PAE) is a green and safe active ingredient of natural plants. The inhibitory activity of PAE against the citrus postharvest pathogen Penicillium digitatum and its ability to control green mold were evaluated by in vitro and in vivo experiments. The results revealed that the minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) of PAE against P. digitatum were 0.5 and 1 μL/mL, respectively. The results of conductivity, cell activity staining, and calcium fluoride white staining revealed that PAE damaged the membrane and cell wall integrity of P. digitatum . PAE promoted ROS accumulation in P. digitatum , as shown by 2',7'-Dichlorofluorescein diacetate (DCHF-DA) staining, which may induce cell death. To further investigate the molecular regulatory network of PAE in P. digitatum , we performed an RNA-sequencing analysis. The results displayed that differentially expressed genes were significantly enriched in the biogenetic pathway of ribosomes, and all of these genes were downregulated, suggesting that PAE treatment interfered with ribosome biogenesis in P. digitatum and inhibited the normal life processes of cells. Furthermore, PAE treatment reduced the incidence and severity of citrus green mold, and the overall control effect of the PAE treatment group with 5 × MFC was greater than that with 10 × MFC. Therefore, we determined the activity of enzymes related to fruit disease resistance. The results revealed that PAE treatment significantly increased the activities of CAT, POD, and PAL in citrus. These findings demonstrate that PAE prevents green mold disease by increasing the activity of citrus disease-resistant enzymes.

A modified microwave hydrodistillation and simultaneous extraction in a rotating state to obtain essential oil, rosmarinic acid, and polysaccharides with sucrose stearate as an additive from Perilla frutescens

Xinyu Yang,Tingting Liu,Mengxia Wei,Ru Zhao,Huiyan Gu,Jialei Li,Fengli Chen,Lei Yang

An improved approach was developed to obtain essential oil, rosmarinic acid , and polysaccharides with sucrose stearate as a green additive from Perilla frutescens (L.) Britt. Leaves using microwave-induced hydrodistillation and simultaneous extraction (MHDE). In this process, we realize the rotation of the material through a rotary motor so that the material is uniformly irradiated by microwaves. The influencing factors on MHDE were systematically optimized by single factor and central composite design. Under the obtained optimization conditions, the yields of essential oil, rosmarinic acid, and polysaccharides were 0.64% ± 0.03%, 0.18% ± 0.01%, and 4.35% ± 0.19%, respectively. GC-MS analysis results showed that the main component of essential oil prepared by MHDE was perillaldehyde , which accounted for 51.91% of the total essential oil, followed by caryophyllene , accounting for 15.79%. However, the ratio of perillaldehyde in the essential oil obtained by traditional hydrodistillation and extraction (HDE) was only 40.69%, and the proportion of caryophyllene was only 12.50%. MHDE uses sucrose stearate with a long hydrocarbon chain structure as an additive, which reduces the nonterpene aliphatics in the obtained essential oil.

Inhibitory effect and action mechanism of perillaldehyde on the Fusarium graminearum in postharvest fresh ginger

Xiuling Meng,Weijie Wu,Ben Niu,Ruiling Liu,Huizhi Chen,Haiyan Gao,Hangjun Chen

Perillaldehyde , the predominant antimicrobial compound found in Perilla , offers a safe and highly efficient solution for the preservation of fruits and vegetables. This study delved into the inhibitory potential of perillaldehyde against Fusarium graminearum , the primary postharvest pathogen affecting ginger. The current research encompassed both in vitro and in vivo investigations. Our results revealed that the minimum inhibitory concentration of perillaldehyde required to combat F. graminearum was 240 µL·L −1 . At this concentration, perillaldehyde exhibited remarkable efficacy in reducing ginger disease rates, with a staggering 99.9% inhibition in mycelium growth. Furthermore, perillaldehyde disrupted the integrity of the fungal cell membrane, hence elevated outer membrane permeability . This resulted in the leakage of proteins, sugars, nucleic acids , and an increase in electrical conductivity within F. graminearum . Notably, perillaldehyde treatment led to a substantial increase in malondialdehyde content in F. graminearum , which was 2.5 times higher than the control group. It also significantly reduced ergosterol content and ATPase activity by 87% and 85%, respectively. The presence of reactive oxygen species (ROS) increased, exacerbating membrane peroxidation damage. Electron microscopy unveiled the transformative impact of perillaldehyde on F. graminearum . Perillaldehyde induced extensive shrinkage, morphological thinning, spatial structure folding, and the leakage of essential cell nutrients, cell wall membranes, and organelle damage. In summary, perillaldehyde's mode of action disrupts the energy supply and metabolism of mycelia, ultimately inducing apoptosis and proving to be a potent inhibitor of F. graminearum . Its application in preserving fresh ginger effectively extended the nutritional quality of the product, demonstrating its potential as an efficient preservative. This biological preservation technique offers convenience over traditional physical methods and addresses concerns related to chemical residue, thereby opening new avenues for ginger green preservation technology.

Perillaldehyde Alleviates Spinal Cord Ischemia-Reperfusion Injury Via Activating the Nrf2 Pathway

Wenjun Zheng,Bing Liu,Enyi Shi

ABSTRACT Background Spinal Cord ischemia-reperfusion injury (SCII) is one of the most destructive complications in thoracic-abdominal aortic surgery, which can cause physical abnormalities, paralysis and even brain death. Evidence has shown that perillaldehyde (PAH) can ameliorate rat's cerebra ischemia-reperfusion injury. However, the effect of PAH on SCII remains unknown. Methods The current study established SCII rat models and oxygen and glucose deprivation/reoxygenation-induced BV2 microglia models to explore whether PAH could alleviate SCII symptoms and to investigate underlying mechanism. Results SCII rats underwent severe neurologic motor dysfunction and histopathologic injury compared with the normal rats, which are exhibited by loss of motor neurons and decrease of nissl bodies . Treatment with PAH significantly ameliorated motor dysfunction and neuron damage. PAH downregulated the expression of NLR family pyrin domain containing 3 , cleaved/pro caspase-1, interleukin-1β and interleukin-18 in spinal cord tissues of SCII rats. Besides, the contents of oxidative stress-related factors superoxide dismutase, manganese-dependent superoxide dismutase, catalase and glutathione peroxidase were significantly increased and malondialdehyde content was decreased after PAH treatment. PAH treatment upregulated the expression of nuclear factor-E2-related factor 2 and heme oxygenase-1 in spinal cord tissues of SCII rats. Our in vitro study confirmed that PAH inhibited microglial activation by activating the nuclear factor-E2-related factor 2/heme oxygenase-1 pathway, exhibited by alleviated inflammation and oxidative stress . Conclusions This study elucidates that PAH has the potential value for treating SCII, which provides an experimental basis for clinical trials in the future.
