Strain-specific endophytes modulate physiological and metabolic responses of Perilla frutescens to cadmium stress

Gen Li,Simiao Wang,Yijin Wang,Yibang Zhang,Qing Dang,Yashuo Tian,Longfei Li,Lijuan Wang,Li Han

Perilla frutescens is a medicinal plant that bioactive compounds such as perillaldehyde and luteolin, but cadmium (Cd) contamination threatens its safe cultivation and medicinal quality. While dark septate endophytes (DSEs) are known to increase plant stress tolerance, their strain-specific roles in regulating Cd uptake and secondary metabolite biosynthesis remain poorly understood. This study aimed to investigate three DSE strains, DSE6 ( Paraphoma radicina ), DSE7 ( P. chlamydocopiosa ), and MD3-1 ( Acrocalymma aquatica ), to determine their effects on growth, physiological traits, Cd accumulation, and key metabolite production in P. frutescens under 0, 1, 5, and 10 mg/kg Cd stress. DSE6 and DSE7 significantly reduced root Cd content and improved photosynthetic pigments, antioxidant activity, and biomass under moderate Cd levels. Notably, MD3-1 did not reduce Cd uptake but increased luteolin content by 22.67 % under high Cd stress, suggesting a decoupling between Cd accumulation and metabolite production. All three strains increased perillaldehyde content, with DSE7 showing the greatest increase (262 %) at 5 mg/kg Cd. These findings demonstrate that DSEs regulate Cd tolerance and medicinal quality via distinct mechanisms, and that internal metabolic tolerance may sustain bioactive compound synthesis under heavy metal stress. This highlights their potential as microbial agents for cultivating medicinal plants in contaminated soils.

Facile green and one-pot synthesis of purple perilla derived carbon quantum dot as a fluorescent sensor for silver ion

Xinyi Zhao,Sen Liao,Lumin Wang,Qi Liu,Xiaoqing Chen

In this work, biomass-derived carbon quantum dots (CQDs) with excellent water solubility, strong fluorescence and favorable biocompatibility were synthesized via one-step hydrothermal treatment of purple perilla for the first time. The functional group composition , morphology, and pH stability of the synthesized CQDs were systematically investigated. And based on fluorescence quenching of CQDs, the as-prepared CQDs were innovatively developed as an effective “signal-off” fluorescent probe for selective and sensitive detection of silver ion (Ag + ) with two linear ranges of 0–10 and 10–3000 nM, and a detection limit 1.4 nM. The specificity and selectivity of this fluorescent probe were also verified through challenging the detection by using similarmetallic cations or in real water samples. In addition, the as-prepared CQDs exhibit a low cytotoxicity and a good biocompatibility, revealing its potential bioimaging applications in living cells.

Inhibitory Effects and Mechanisms of Perilla Essential Oil and Perillaldehyde against Chestnut Pathogen Botryosphaeria dothidea

Qi Zeng,Lu Wang,Sha Long,Wanrong Dong,Yaoyao Li,Yuxin Chen,Gao Zhou

Botryosphaeria dothidea, a notorious plant pathogen, is responsible for causing chestnut rot during postharvest storage. This research aimed to assess the antifungal properties of perilla essential oil (PEO) and perillaldehyde (PAE) againstB. dothidea. PEO’s and PAE’s inhibitory effects onB. dothideawere investigated using an agar dilution method, a fumigation method, and an in vivo assay in chestnuts and shell buckets. Based on the results of gas chromatography-mass spectrometry, it was confirmed that the main component of PEO was elemicin. The antifungal mechanism of PEO and PAE againstB. dothideawas investigated by conducting staining experiments of the fungal cell wall and cell membrane. PEO and PAE strongly inhibit the mycelial growth ofB. dothideain a dose-dependent manner. The inhibitory mechanism is mainly related to the destruction of the integrity of the fungal cell wall and plasma membrane. Notably, PEO retains its antifungal efficacy againstB. dothideain chestnuts, effectively prolonging their storage life. These findings indicate that PEO and PAE are nontoxic, eco-friendly botanical fungicides, holding promise for controlling postharvest chestnut rot.

A modified microwave hydrodistillation and simultaneous extraction in a rotating state to obtain essential oil, rosmarinic acid, and polysaccharides with sucrose stearate as an additive from Perilla frutescens

Xinyu Yang,Tingting Liu,Mengxia Wei,Ru Zhao,Huiyan Gu,Jialei Li,Fengli Chen,Lei Yang

An improved approach was developed to obtain essential oil, rosmarinic acid , and polysaccharides with sucrose stearate as a green additive from Perilla frutescens (L.) Britt. Leaves using microwave-induced hydrodistillation and simultaneous extraction (MHDE). In this process, we realize the rotation of the material through a rotary motor so that the material is uniformly irradiated by microwaves. The influencing factors on MHDE were systematically optimized by single factor and central composite design. Under the obtained optimization conditions, the yields of essential oil, rosmarinic acid, and polysaccharides were 0.64% ± 0.03%, 0.18% ± 0.01%, and 4.35% ± 0.19%, respectively. GC-MS analysis results showed that the main component of essential oil prepared by MHDE was perillaldehyde , which accounted for 51.91% of the total essential oil, followed by caryophyllene , accounting for 15.79%. However, the ratio of perillaldehyde in the essential oil obtained by traditional hydrodistillation and extraction (HDE) was only 40.69%, and the proportion of caryophyllene was only 12.50%. MHDE uses sucrose stearate with a long hydrocarbon chain structure as an additive, which reduces the nonterpene aliphatics in the obtained essential oil.

Optimization of Enzymatic Hydrolysis of Perilla Meal Protein for Hydrolysate with High Hydrolysis Degree and Antioxidant Activity

Henghui Zhang,Zhijun Zhang,Dongliang He,Shuying Li,Yongping Xu

Botanical oils are staple consumer goods globally, but as a by-product of oil crops, meal is of low utilization value and prone to causing environmental problems. The development of proteins in meal into bioactive peptides, such asPerillapeptide, through biotechnology can not only solve environmental problems, but also create more valuable nutritional additives. In the present work, the hydrolysis process ofPerillameal protein suitable for industrial application was optimized with the response surface methodology (RSM) on the basis of single-factor experiments. Alcalase was firstly selected as the best-performing among four proteases. Then, based on Alcalase, the optimal hydrolysis conditions were as follows: enzyme concentration of 7%, hydrolysis temperature of 61.4 °C, liquid-solid ratio of 22.33:1 (mL/g) and hydrolysis time of 4 h. Under these conditions, the degree of hydrolysis (DH) ofPerillameal protein was 26.23 ± 0.83% and the DPPH scavenging capacity of hydrolysate was 94.15 ± 1.12%. The soluble peptide or protein concentration ofPerillameal protein hydrolysate rose up to 5.24 ± 0.05 mg/mL, the ideal yield of which was estimated to be 17.9%. SDS-PAGE indicated that a large proportion of new bands in hydrolysate with small molecular weights appeared, which was different from the originalPerillameal protein. The present data contributed to further, more specific research on the separation, purification and identification of antioxidant peptide from the hydrolysate ofPerillameal protein. The results showed that the hydrolysis ofPerillameal protein could yield peptides with high antioxidant activity and potential applications as natural antioxidants in the food industry. Keywords:Perillameal protein;hydrolysate;response surface methodology;antioxidant activity;peptide

Protein enabled foam fractionation of anthocyanins from purple perilla leaves: Mechanisms, performance, and optimization

Yanfei Li,Chunquan Zhao,Nan Hu,Yang Jing,Lin Chen,Yunqing Guo,Zhijun Zhang

Anthocyanins (ACNs) are the primary bioactive compounds of purple perilla leaves with a variety of biological activities. The scale separation of perilla ACNs is crucial for their practical application. This study presents the development of a green and highly efficient foam fractionation technology employing a protein-based frother and collector. The protein utilized, namely perilla seed meal protein (PSMP), is derived from perilla seed meal (PSM), which is normally discarded as an industrial waste during seed oil extraction. PSMP demonstrated notable foamability and foam stability. The interaction or collection mechanism between PSMP and ACNs was comprehensively investigated by SEM-EDS, FTIR, and fluorescence spectroscopy , revealing a hydrophobic interaction between PSMP and ACNs. Furthermore, the impacts of PSMP concentration, pH, and gas velocity on the foam fractionation of ACNs were thoroughly examined. Response surface methodology (RSM) was used to optimize the operating conditions for ACNs separation. Remarkable results were achieved under the optimum conditions, including a high recovery percentage of 95.13% and an impressive enrichment ratio of 27.56. The composition and antioxidation of isolated ACNs product were characterized, revealing its excellent antioxidant activities . This highlights its potential applications in drugs, food, and cosmetics. This work not only highlights an efficient foam fractionation technology for ACNs separation, but also offers valuable insights into the separation of other active plant components.

Extraction, preparative monomer separation and antibacterial activity of total polyphenols from Perilla frutescens

Yana Zhao,Huizhen Li,Zhijun Zhang,Zhiqing Ren,Fuhan Yang

Polyphenols exhibit potential functional activities, especially rosmarinic acid (RosA) and caffeic acid (CafA). In this study, two different methods, ultrasonic-assisted ethanol extraction (60%) and ultrasound-assisted cellulase (≥15 000 Ug−1, 2%) hydrolysis, were used for the extraction of the total phenolics from 44 species of Perilla frutescens. The Folin–Ciocalteu method of detection showed that the content of the total phenolics extracted by cellulase hydrolysis was the highest and attained up to 28.00 mgGAE per gextracts for ZB1. Continuously, the extracts were purified using XDA-8 macroporous resin and medium-pressure liquid chromatography (MPLC), and the content of the total phenolics improved to 66.62 mgGAE per gextract. A high-performance liquid chromatography (HPLC) assay showed that the total polyphenols were mainly composed of gallic acid, caffeic acid, rosmarinic acid, luteolin and apigenin. Besides, a sequential XDA-8 macroporous resin combined with high-speed counter-current chromatography (HSCCC)/MPLC system was established for the simultaneous isolation and preparation of RosA (purity 98.29%) and CafA (purity 97.01%) from the extracts. Furthermore, the antibacterial activities of the total polyphenols were evaluated by the disc diffusion method and scanning electron microscopy (SEM) observation. The results verified that the total polyphenols had effective antibacterial activity on three kinds of bacteria including E. coli, S. aureus, and B. subtilis in a concentration-dependent manner. All of these results demonstrated that the ultrasound-assisted cellulase hydrolysis extraction of the total polyphenols and the proposed three-step separation of RosA and CafA gave high yields and good purity, and they exhibited effective antibacterial ability.

Rapid discrimination and quantification of chemotypes in Perillae folium using FT-NIR spectroscopy and GC–MS combined with chemometrics

Dai-xin Yu,Cheng Qu,Jia-yi Xu,Jia-yu Lu,Di-di Wu,Qi-nan Wu

Perillae Folium (PF) is a well-known food and herb containing different chemotypes, which affect its quality. Herein, a method was proposed to classify and quantify PF chemotypes using gas chromatography–mass spectrometry (GC–MS) and Fourier transform-near infrared spectroscopy (FT-NIR). GC–MS results revealed that PF contains several chemotypes, including perilla ketone (PK) type, α -asarone (PP-as) type, and dillapiole (PP-dm) type, with the PK type being the predominant chemotype. Based on FT-NIR data, different chemotypes were accurately classified. The random forest algorithm achieved >90 % accuracy in chemotype classification. Furthermore, the main components of perilla ketone and isoegomaketone in PF were successfully quantified using partial least squares regression models, with prediction to deviation values of 3.76 and 2.59, respectively. This method provides valuable insights and references for the quality supervision of PF and other foods.

Perilla juice and ginger juice reduced warmed-over flavor (WOF) in surimi gels: Due to the inhibition of the formation of the WOF compounds and the masking of the WOF

Meiqi Dang,Wenrong Li,Juan You,Shanbai Xiong,Yueqi An

The effects and reasons of perilla juice (PJ) and ginger juice (GJ) on the reduction of “warmed-over flavor” (WOF) in surimi gels were revealed by detecting odor profiles and protein and lipid oxidation degrees of surimi gels, concentrations and odor activity values (OAVs) of WOF compounds. Adding PJ and GJ to surimi gels significantly reduced the WOF and improved the fish fragrance odor, but sodium ascorbate (SA) only weakened the WOF. The (E,E)-2,4-heptadienal's OAVs in the PJ and GJ groups were decreased by >50% compared with the control check (CK) and SA groups. Meanwhile, surimi gels added with PJ and GJ presented lower lipid and protein oxidation degrees. The verification test indicated that PJ and GJ's aroma had a masking effect on the WOF. In conclusion, PJ and GJ reduced the WOF in surimi gels by preventing WOF compounds' production and masking the WOF with their distinct aroma.

Mechanisms of slow-release antibacterial properties in chitosan‑titanium dioxide stabilized perilla essential oil Pickering emulsions: Focusing on oil-water interfacial behaviors

Hengheng Wang,Geoffrey I.N. Waterhouse,Huan Xiang,Dongxiao Sun-Waterhouse,Yongqiang Zhao,Shengjun Chen,Yanyan Wu,Yueqi Wang

Perilla essential oil (PLEO) offers benefits for food preservation and healthcare, yet its instability restricts its applications. In this study, chitosan (CS) and TiO 2 used to prepare composite particles. TiO 2 , after being modified with sodium laurate (SL), was successfully introduced at 0.1 %–3 % into the CS matrix. The resulting CS-SL-TiO 2 composite particles can be formed by intertwining and rearranging through intramolecular and intermolecular interactions, and form an O/W interface with stability and viscoelasticity. The Pickering emulsions stabilized by these particles exhibit non-Newtonian pseudoplastic behavior, shear-thinning properties, and slow-release characteristics, along with antibacterial activity. Emulsions with 0.5 % and 1 % CS-SL-TiO 2 composites demonstrated superior antibacterial effects against Escherichia coli and Staphylococcus aureus . The study revealed that all emulsions undergo Fickian diffusion and a sustained release of PLEO, with the Ritger-Peppas model best describing this release mechanism. The slow-release behaviors positively correlates with interfacial pressure, composite particle size, composite particle potential, composite contact angle, emulsion particle size and emulsion potential, but negatively correlates with diffusion rate, penetration rate, release kinetics and release rate. The findings lay groundwork for developing slow-release antimicrobial emulsions within polysaccharide matrices, showcasing promise for antimicrobial packaging solutions and enhanced food preservation techniques.

Study of the optimization and kinetics of the surfactant-induced ultrasonic-assisted extraction of perilla seed oil: Free radical scavenging capacity and physicochemical and functional characteristics

Zhanjun Li,Yunwei Liu,Yantao Liang,Honggang Wang,Fengjian Yang

In this paper, we study the technology of tea saponin-induced ultrasonic-assisted extraction of perilla seed oil. Based on single-factor experimental data, the Plackett‒Burman design (PBD) cooperated with the Box–Behnken design (BBD) to optimize the influencing factors. The optimal process conditions after optimization were as follows: ultrasonic power of 90 W, ultrasonic time of 16 min, tea saponin dose of 0.3%, extraction temperature of 40 °C, ethanol volume fraction of 80% and liquid material ratio of 10:1 mL/g. In this situation, the actual average oil yield was 39.10 ± 2.13%, which was highly consistent with the expected theoretical value of 39.94%. Based on a comparative analysis, the first-order kinetic model was more suitable for fitting the experimental data and interpreting the mechanism. The efficiency, unsaturation degree, free radical scavenging ability and physical and chemical properties of oil obtained using UAE and microstructures of defatted cake were superior to those of SE. Therefore, the development of UAE induced by tea saponin technology markedly improves the yield of perilla seed oil and has great potential to extract effective ingredients from perilla seed, which provides a foundation for the efficient utilization of perilla resources and other oil crops.
