Effects of ultra-high pressure assisted extraction on the structure, antioxidant and hypolipidemic activities of Porphyra haitanensis polysaccharides

Mingjing Zheng,Xin Tian,Zhipeng Li,Tao Hong,Yanbing Zhu,Yuanfan Yang,Qingbiao Li,Hui Ni,Zedong Jiang

Porphyra haitanensis polysaccharides (PHP) has potential antioxidant and hypolipidemic activities, but still needs improvement. Therefore, ultra-high pressure (UHP) assisted extraction was applied to modify the structure, antioxidant, and hypolipidemic activities of PHP. UHP assisted extraction increased the total sugar, uronic acid, and 3,6-anhydro-ʟ-galactose contents of PHP, which increased by 15.85%–16.12%, 18.95%–24.32%, 20.54%–23.66% with 500–600 MPa UHP, respectively. Meanwhile, UHP modified PHP became more rough and porous than native PHP. Besides, UHP assisted extracted PHP showed better in vitro antioxidant and hypolipidemic abilities. Especially, 500–600 MPa UHP increased 72.43%–86.42% 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacities, 12.32%–12.82% pancreatic lipase inhibitory ability, and 14.93%–15.23% glycocholate binding abilities of PHP ( p < 0.05). Moreover, UHP assisted extracted PHP greatly decreased the lipid droplet and triglyceride contents of 3T3-L1 adipocytes ( p < 0.05). Our findings can provide the theoretical basis for the high value utilization of Porphyra haitanensis and its polysaccharides with UHP modification.

Green and efficient removal of heavy metals from Porphyra haitanensis using natural deep eutectic solvents

Xi Yang,Yuan-Yuan Zang,Sha Yang,Zhi-Gang Chen

BACKGROUND Porphyra haitanensis now faces serious heavy metal pollution problems. Natural deep eutectic solvents (NADESs) have been recognized as a novel class of sustainable solvents, which can be used for heavy metal removal. In this study, 28 kinds of NADESs were prepared and investigated as eluent in the removal of lead (Pb), cadmium (Cd), chromium (Cr), arsenic (As), and copper (Cu) from P. haitanensis for the first time, and the adsorption mechanism of NADESs was also studied. RESULTS The removals were greatly improved by NADESs compared with control where the removal rates of Pb, Cd, Cr, As and Cu were 17.4–87.54%, 57.54–100%, 9.8–48.59%, 21.32–78.24% and 11.68–79.73%, respectively. The optimal condition was 10% water content and solid–liquid ratio of 1:20. Moreover, the addition of 20% natural surfactant arabic gum can further increase the heavy metals removal rates of NADESs. The adsorption mechanism experiments showed that the pseudo second-order model and the Freundlich adsorption model can better explain the adsorption mechanism of NADESs on heavy metals removal. CONCLUSION Taken together, a green and efficient method for removing heavy metals from P. haitanensis was established. © 2020 Society of Chemical Industry

Partially Acid-Hydrolyzed Porphyran Improved Dextran Sulfate Sodium-Induced Acute Colitis by Modulation of Gut Microbiota and Enhancing the Mucosal Barrier

Biao Yu,Min Wang,Bo Teng,Suresh Veeraperumal,Peter Chi-Keung Cheung,Saiyi Zhong,Kit-Leong Cheong

The pharmacological values of marine algal polysaccharides on gut health are being recognized in recent research. However, the protective effect of degraded polysaccharides from Porphyra haitanensis (PHP-D) on the colonic mucosal barrier damaged in ulcerative colitis is poorly understood. The purpose of this study was to investigate how PHP-D could maintain the integrity of colonic mucosal layer mediated by microbiota in a dextran sulfate sodium (DSS)-induced colitis mouse model. Structural analysis revealed that PHP-D had a typical porphyran structure having a backbone of alternating (1 → 3)-linked β-d-galactopyranose units linked to either (1 → 4)-3,6-anhydro-α-l-galactopyranose units or (1 → 4)-linked α-l-galactose-6-sulfate units. An in vivo study demonstrated that PHP-D treatment reduced the severity of DSS-induced ulcerative colitis. 16S rRNA phylogenetic sequencing revealed that PHP-D affected the diversity of gut microbiota with an increase of Bacteroides, Muribaculum, and Lactobacillus species. Similarly, PHP-D increased levels of short-chain fatty acids. Furthermore, PHP-D restored mucus thickness and improved the expression of tight junction proteins. This work demonstrates that PHP-D is capable of enhancing a colonic mucosal barrier. These outcomes offer unique perspectives on the potential application of P. haitanensis as a promising natural product for the management of ulcerative colitis.

Structural characterization of Porphyra haitanensis extracts and their effects on structure, physicochemical properties, and in vitro digestibility of corn starch

Manling Yu,Hongmei Yin,Jingjing Huang,Xixi Cai,Jiulin Wu,Xiaodan Shi,Shaoyun Wang

Porphyra haitanensis with many bioactive components has great prospects for application in the functional food, pharmaceutical, and cosmeceutical industries. In this study, two sequentially extracted fractions (PhE1 & PhE2) were isolated and structurally characterized. The results showed that PhE1 was mainly composed of protein (36.82%) and neutral sugar (32.43%), while PhE2 mainly contained neutral sugar (58.70%). Both had galactose as main monosaccharides, PhE2 also contained a small amount of glucose. To widen the application of PhEs in starchy foods, they were added to corn starch (CS) system and co-gelatinized. Rheological tests indicated that PhEs could effectively affect gel strength of CS. The addition of PhEs effectively inhibited the content of leached amylose in CS gelatinization system except those at an addition of 5% PhE1 and 5% PhE2. FT-IR data suggested that PhE1 could improve the short-range order, but PhE2 showed no significant effects. SEM images showed that the pore wall of the composites thickened, and the network structure became relatively ordered. In vitro tests, PhEs significantly reduced the contents of rapidly digestible starch, and PhE1 presented better effects. Therefore, PhEs may be used as potential materials to delay the digestion of CS and related products.

Copper nanoclusters @ nitrogen-doped carbon quantum dots-based ratiometric fluorescence probe for lead (II) ions detection in porphyra

Wenting Li,Xuetao Hu,Qian Li,Yongqiang Shi,Xiaodong Zhai,Yiwei Xu,Zhihua Li,Xiaowei Huang,Xin Wang,Jiyong Shi,Xiaobo Zou,Shifei Kang

A novel ratiometric fluorescence probe was proposed for detecting lead (II) ions (Pb 2+ ) in porphyra, the approach was based on copper nanoclusters and nitrogen-doped carbon quantum dots (CuNCs-CNQDs). In this probe, the CuNCs delivered the response signal, the fluorescence of which was enhanced by Pb 2+ due to the aggregation-induced emission enhancement (AIEE) between Pb 2+ and CuNCs. The CNQDs provided the self-calibration signal, whose fluorescence remained almost unchanged in coexistence with Pb 2+ . According to the change of fluorescence intensity ratio between the fluorophores, CuNCs–CNQDs nanohybrid was used as ratiometric probes for the sensitive detection of Pb 2+ in the range of 0.010–2.5 mg L −1 , with a detection limit of 0.0031 mg L −1 . Finally, the probe was successfully applied to detect Pb 2+ in porphyra with relative standard deviations (RSDs) lower than 5%. This study provides a straightforward, stable, and sensitive approach for detecting Pb 2+ in porphyra.

Purification, characterization and antioxidant activity of polysaccharides from Porphyra haitanensis

Yun-Tao Wu,Yun-Feng Huo,Lin Xu,Ying-Ying Xu,Xiao-Ling Wang,Tao Zhou

In order to study the antioxidant activity of Porphyra haitanensis polysaccharides and the effect of their structure on the antioxidant activity, the Porphyra haitanensis polysaccharide (PHP), prepared by hot water extraction and alcohol precipitation, was separated and purified by Cellulose DEAE-52 ion exchange and Sephadex G-100 column chromatography. Three purified components including PHP1, PHP2 and PHP3 were obtained, and chemical composition analysis and structural characterization were performed. The in vitro free radical scavenging activity of the purified polysaccharide fractions and their ability to relieve oxidative stress in macrophage RAW264.7 were investigated. The results indicated that all the three fractions possessed appreciable DPPH radical, superoxide anion radical, and hydroxyl radical scavenging ability and reducing power. They were also found to effectively reduce the MDA content and ROS level, and to improve the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) in H 2 O 2 -treated RAW264.7 cells. Among the three polysaccharides, PHP3 possessed the most potent activity. Porphyra haitanensis polysaccharides have potential to develop as natural non-toxic antioxidants and may find application as the ingredients of functional foods.

Effects of Porphyra yezoensis Polysaccharide with Different Molecular Weights on the Adhesion and Endocytosis of Nanocalcium Oxalate Monohydrate in Repairing Damaged HK-2 Cells

Hui Zhang,Xin-Yuan Sun,Jian-Ming Ouyang

The adhesion and endocytosis of renal epithelial cells to urinary microcrystals are closely related to kidney stone formation; however, the mechanism of cell state changes that affect adhesion and endocytosis remains unclear. In this study, a damaged cell model was established using oxalate to impair human kidney proximal tubular epithelial cells (HK-2). Then, we used four degraded Porphyra yezoensis polysaccharides (PYPs), namely, PYP1, PYP2, PYP3, and PYP4, with molecular weight of 576.2, 49.5, 12.6, and 4.02 kDa, respectively, to repair the damaged cells. After repairing the damaged HK-2 cells by PYPs, the cell morphology gradually recovered to near normal; the cell migration speed increased; the phosphatidylserine eversion ratio and osteopontin expression decreased; and the intracellular adenosine triphosphate was promoted. The adhesion and endocytosis of calcium oxalate monohydrate (COM) crystals were closely related to cell state. Normal cells could endocytose crystals more than the damaged cells, whereas damaged cells could adhere to crystals more than the normal cells. The cells repaired by PYPs inhibited the adhesion of nano-COM crystals and promoted the endocytosis of the adherent crystals, thereby reducing the adhesion of crystals on the cell surface under both effects. As the molecular weight of PYP decreased, the ability of PYP to repair cells, inhibit crystal adhesion, and promote endocytosis of cells was enhanced; that is, PYP4, which had the lowest molecular weight, exhibited the best biological activity. Therefore, PYPs, especially PYP4, may become an optional green drug to inhibit the formation and recurrence of calcium oxalate stones.

Porphyran from Porphyra haitanensis Enhances Intestinal Barrier Function and Regulates Gut Microbiota Composition

Sathuvan Malairaj,Suresh Veeraperumal,Wanzi Yao,Mugesh Subramanian,Karsoon Tan,Saiyi Zhong,Kit-Leong Cheong

In this study, the effects of a homogenous porphyran fromPorphyra haitanensis(PHP) on the intestinal barrier and gut microbiota were investigated. The results showed that oral administration of PHP resulted in a higher luminal moisture content and a lower pH environment for the growth of beneficial bacteria in the colon of mice. PHP significantly increased the production of total short-chain fatty acids during the fermentation process. PHP made the intestinal epithelial cells of mice arrange more tidily and tightly with a significant increase in mucosal thickness. PHP also increased the amount of mucin-producing goblet cells and the expression of mucin in the colon, which maintained the structure and function of the intestinal mucosal barrier. Moreover, PHP up-regulated the expression of tight junctions including ZO-1 and occludin, improving the intestinal physical barrier function. The results of 16S rRNA sequencing showed that PHP regulated the composition of gut microbiota in mice, increasing the richness and diversity of gut microbiota and the ratio of Firmicutes to Bacteroidetes. This study revealed that the intake of PHP is beneficial for the gastrointestinal tract and PHP could be a potential source of prebiotics in the functional food and pharmaceutical industries. Keywords:Porphyra haitanensis;porphyran;intestinal barrier;gut microbiota

Phosphorus-functionalized Porphyra toward uranium removal from water

Wenxuan Sui,Yanan Chen,Jiayu Zhao,Kairuo Zhu

The high-value utilization of marine biomass has garnered significant attention in recent years, particularly the application of biomass-based adsorbents in water environment remediation. In this study, we innovatively combined ball-milling treatment with phosphorylation modification to construct a phosphorus-functionalized Porphyra (P- Porphyra ) adsorbent, which can be applied for uranium adsorption from water. Systematic characterization and batch adsorption experiments revealed that P- Porphyra exhibited optimal uranium adsorption at pH 4.0, with the adsorption process fitted the pseudo-second-order kinetic model and Langmuir isotherm well. Under optimal conditions, the maximum adsorption capacity reached 262.76 mg·g −1 , almost unaffected by common ions in natural waterbody. Fourier-transform infrared (FTIR) spectroscopy and X-ray photoelectron spectroscopy (XPS) analyses demonstrated that the adsorption mechanism primarily relied on the complexation between phosphate groups and uranium ions, enabling efficient uranium enrichment. P- Prophyra exhibited the fast uranium adsorption in the real seawater, and the adsorption capacity reached to be 0.90 mg·g −1 . This work proposes a green, low-cost, and scalable strategy for converting Porphyra into phosphorylated biomass materials towards uranium enrichment from water, also offering significant potential for sustainable marine resource utilization.

Pyropia yezoensis porphyran alleviates metabolic disorders via modulating gut microbiota in high-sucrose-fed Drosophila melanogaster

Dan He,Qifang Wu,Chenying Lu,Jinlan Wu,Peichao Chen,Mingjiang Wu,Jong-il Choi,Haibin Tong

BACKGROUND Prebiotics, such as algal polysaccharides, can be used to manage metabolic diseases by modulating gut microbiota. However, the effect of Pyropia yezoensis porphyran (PYP), a red algal polysaccharide, on gut microbiota has not been reported. Thus, the objective of this study was to determine effects of PYP on metabolic disorders caused by high sucrose (HS) and underlying mechanisms involved in such effects. RESULTS Biochemical analysis demonstrated that an HS diet increased triglyceride and circulating sugar contents (metabolic abnormalities) in Drosophila larvae. It also increased the relative abundance of harmful microbiota within the larvae as identified by 16S ribosomal DNA analysis. PYP supplementation at 25 and 50 g kg −1 equivalently reduced metabolic abnormalities in the HS group. Therefore, 25 g kg −1 PYP was selected to investigate its effects on the metabolic pathway and gut microbiota of larvae in the HS group. The activity of PYP in ameliorating metabolic abnormalities by reverse transcription quantitative real-time polymerase chain reaction analysis was consistent with the expression trend of key factors involved in metabolism regulation. PYP reduced the relative abundance of bacteria causing metabolic abnormalities, such as Escherichia–Shigella and Fusobacterium , but increased the relative abundance of beneficial bacteria such as Bacillus and Akkermansia . However, PYP had no effect on triglyceride and circulating sugar contents in HS-fed larvae treated with a mixture of antibiotics designed to remove gut microbiota. CONCLUSION PYP exhibits anti-metabolic disorder activity by modulating gut microbiota, thereby supporting the development of PYP as a functional prebiotic derived from red algae food. Resveratrol-loaded ovalbumin/Porphyra haitanensis polysaccharide composite nanoparticles: Fabrication, characterization and antitumor activity

Ying-Ying Xu,Yun-Feng Huo,Lin Xu,Yu-Zhu Zhu,Yun-Tao Wu,Xiao-Yi Wei,Tao Zhou

Resveratrol (RES) possesses a variety of health-promoting effects. However, the application of RES in food and pharmaceutical industries is limited due to the poor water solubility and bioavailability, and low photostability. In order to overcome the drawback of RES, in the present work, RES-loaded nanoparticles were prepared using ovalbumin (OVA) and Porphyra haitanensis polysaccharide (PHP) as encapsulating materials. Response surface methodology was employed to optimize the fabrication of OVA-PHP-RES nanoparticles (OPR). It was found that OVA-PHP possessed superior effects on both stability of the dispersion and RES protection to those of OVA alone. In vitro simulated digestion tests demonstrated that RES in OPR was released slower in stomach and more rapidly in intestine when compared to OVA-RES nanoparticles (OR). In addition, OPR was found to effectively inhibit the growth of two tumor cell lines (HeLa and HepG2), indicating that OPR nanoparticles could deliver effectively RES in biological system, thereby improving its bioavailability. The finding in this study demonstrated OVA-PHP system could improve the stability and bioavailability of RES.

Overexpression of Rboh enhances inorganic carbon acquisition through coordinating with carbonic anhydrase in Pyropia yezoensis

Zhizhuo Shao,Menglin Guo,Hong Wang,Wenhui Gu,Xiujun Xie,Guangce Wang

Pyropia yezoensis is an important intertidal economic macroalgae, which is periodically affected by various stresses, such as the limitation of inorganic carbon (C i ) deficiency. Under such environment, the redox homeostasis within the cells of Pyropia yezoensis is seriously affected, and the reactive oxygen species (ROS) signal transduction system would be activated to regulate the photosynthetic activity. Therefore, how P. yezoensis manage ROS to maintain effective photosynthetic carbon fixation has aroused great interest. Here, we characterize transformants overexpressing respiratory burst oxidase homolog ( Rboh ), an important gene that can actively produce ROS, at the levels of cellular physiology, biochemistry, and transcriptomics. Our data indicated the expression of Rboh significantly increased, accompanied by a significant upregulated expression of alpha-type carbonic anhydrase 3 (α CA3 ) and increased extracellular carbonic anhydrase activity in the Rboh overexpressing strains. Interestingly, compared with the wild type, the photosynthetic activity of transgenic strains was significantly higher under the low C i and high light condition, implying that the ROS signal triggered by overexpression of Rboh was involved in regulating the C i absorption and utilization in P. yezoensis when the C i source was limited. In summary, this study provided evidence supporting the correlation between the ROS production and the Ci utilization under stress environments in P. yezoensis .
