Antibacterial composite paper with corn stalk-based carbon spheres immobilized AgNPs

Qimeng Jiang,Bichong Luo,Zhengguo Wu,Xiaoying Wang

Silver nanoparticles (AgNPs) have been widely used for sterilization due to their broad-spectrum bactericidal properties. However, there exist the problems of premature releasing and accumulative toxicity when free AgNPs are applied. This study proposed a one-pot hydrothermal strategy to synthesize carbon spheres immobilized silver nanoparticles (AgNPs@CS). The synthesis involves with silver ammonia solution as Ag precursor, and corn stalk as green reducing agent and carbon precursor. Furthermore, AgNPs@CS was anchored by cellulose nanofibers (CNF) to obtain the antibacterial composite paper. The obtained CNF/AgNPs@CS paper exhibited superior antibacterial properties against E. coli and S. aureus . Notably, the accumulative release rate of AgNPs from AgNPs@CS was 10.2% in 9 days, while that from CNF/AgNPs@CS paper was only 6.7% due to the anchoring effect of both CS and CNF, which was low for avoiding the cumulative toxicity problem. In addition, the mechanical and barrier properties of CNF/AgNPs@CS paper were also improved by 29.4% (tensile index), 2.7% (tear index), 7.4% (burst index), 10% (folding endurance), 0.8% (water vapor transmission) and 9.4% (oxygen transmission rate), respectively. Therefore, the composite paper has potential application as a medical antibacterial material.

Eco-Friendly Bio-Hydrogels Based on Antheraea Pernyi Silk Gland Protein for Cell and Drug Delivery

Jia Li,Bo-Xiang Wang,De-Hong Cheng,Yan-Hua Lu

The Antheraea Pernyi silk gland protein originates from natural organisms and synthesized by tussah silk glands and has widely potential biomaterial applications due to the superior biocompatibility. This study investigates the Antheraea Pernyi silk gland protein-based drug-loaded bio-hydrogels for bioengineered tissue fabricated by using an eco-friendly method without the harsh extracting process and the usage of toxic chemicals. The drug-loaded bio-hydrogels exhibited a porous structure and interconnected pore walls. The swelling ratio and water absorption of drug-loaded bio-hydrogels were, respectively, above 95% and 1.5 × 103%. The cumulative release of drug loaded hydrogels all reached more than 90% within 4 h, and this indicates the potential of drug-loaded hydrogels as future drug-carrying biomaterials. RSC96 Schwann cells cultured on drug-loaded hydrogels for 72 h under cell culture medium show no toxic effects and more pro-proliferative effects. The results suggest the suitability of drug-loaded bio-hydrogels as natural biopolymer for the potential in vitro RSC96 cell culture platform and other biomaterial applications.

A one-dimensional bacterial cellulose nano-whiskers-based binary-drug delivery system for the cancer treatment

Xiaoli Liu,Xiaofang Qian,Zirui Yu,Xingxing Zheng,Yang Qiao,Chen Chen,Wenping Li,Wenjing Li,Jiazhi Yang,Jing Zhu

It's currently a challenge to design a drug delivery system for chemotherapy with high drug contents and minimal side effects. Herein, we constructed a novel one-dimensional binary-drug delivery system for cancer treatment. In this drug delivery system, drugs (doxorubicin (DOX) and resveratrol (RES)) self-assemble on bacterial cellulose nano-whiskers (BCW) and are subsequently encapsulated by polydopamine (PDA) with high encapsulation efficiencies (DOX: 81.53 %, RES: 70.32 %) and high drug loading efficiencies (DOX: 51.54 %, RES: 36.93 %). The cumulative release efficiencies can reach 89.27 % for DOX and 80.05 % for RES in acidic medium within 96 h. The BCW/(DOX + RES)/PDA can enter tumor cells easily through endocytosis and presents significant anti-cancer effects. Furthermore, the released-RES plays a protective role in normal cells through up-regulation of antioxidant enzymes activities and scavenging of reactive oxygen species. Taken together, the one-dimensional BCW/(DOX + RES)/PDA binary-drug delivery system can be used for the anticancer treatment along with slight side effects.

Hierarchical antibiotic delivery system based on calcium phosphate cement/montmorillonite-gentamicin sulfate with drug release pathways

Lei Chen,Xiuying Lin,Min Wei,Bo Zhang,Yani Sun,Xi Chen,Shitong Zhang,Hao Zhang,Jieyu Zhang,Xiaojiao Yu,Binghua Yao,Kang Zhao,Yufei Tang,Quanchang Tan,Zixiang Wu

Antibiotic-loaded calcium phosphate cement (CPC) has emerged as a promising biomaterial for drug delivery in orthopedics. However, there are problems such as the burst release of antibiotics, low cumulative release ratio, inappropriate release cycle, inferior mechanical strength, and poor anti-collapse properties. In this research, montmorillonite-gentamicin (MMT-GS) was fabricated by solution intercalation method and served as the drug release pathways in CPC to avoid burst release of GS, achieving promoted cumulative release ratios and a release cycle matched the time of inflammatory response. The results indicated that the highest cumulative release ratio and release concentration of GS in CPC/MMT-GS was 94.1 ± 2.8 % and 1183.05 μg/mL, and the release cycle was up to 504 h. In addition, the hierarchical GS delivery system was divided into three stages, and the kinetics followed the Korsmeyer-Peppas model, the zero-order model, and the diffusion-dissolution model, respectively. Meanwhile, the compressive strength of CPC/MMT-GS was up to 51.33 ± 3.62 MPa. Antibacterial results demonstrated that CPC/MMT-GS exhibited excellent in vitro long-lasting antibacterial properties to E. coli and S. aureus . Furthermore, CPC/MMT-GS promoted osteoblast proliferation and exhibited excellent in vivo histocompatibility. Therefore, CPC/MMT-GS has favorable application prospects in the treatment of bone defects with bacterial infections and inflammatory reactions.
