The repellent effects of eight Chinese herbal essential oils on red imported fire ants, Solenopsis invicta Buren (Hymenoptera: Formicidae), and analysis of active components

Min Fan,Tongen Ye,Zitong Wang,Qunchen Li,Chunrui Li,Qingxing Shi,Hualong Qiu,Tao Ma,Xiaoge Nian,Yurong He,Desen Wang

The red imported fire ant (RIFA), Solenopsis invicta Buren, is an invasive pest that is extremely aggressive and thus poses a strong threat to human health, agricultural production, equipment and the environment. In this study, we evaluated the repellent effects of eight Chinese herbal essential oils (EOs) on RIFAs, and identified the main repellent components in the EOs. Results showed that all eight EOs had outstanding repellency to RIFAs, among which A. argyi and S. album EOs had the strongest repellent effects on RIFAs. GC-MS revealed 34 ingredients in A. argyi EO, among which sesquiterpenoids and their oxygenated derivatives accounted for the greatest proportion (50 %), followed by monoterpenes and their oxygenated derivatives (47.06 %) and unknown components (2.94 %). Dipentene, 1-caryophyllene, p-cymene, α-terpineol, and Δ-cadinene were the most abundant compounds. A total of 61 ingredients were identified in S. album EO, including monoterpenes and their oxygenated derivatives (40.98 %), sesquiterpenoids and their oxygenated derivatives (39.34 %), diterpenoids and their oxygenated derivatives (3.28 %), aromatic compounds (3.28 %), and unknown components (13.12 %). Among them, the most abundant compounds were diethyl phthalate, α-cetone, 2-phenylethanol and sandacanol. The GC-EAD results showed that RIFA workers exhibited strong electroantennographic responses to dipentene, α-terpineol and 1-caryophyllene from A. argyi EO, as well as to linalool, 2-phenylethanol and benzyl acetate from S. album EO. Furthermore, all of the above six ingredients had significant repellent effects against RIFAs based on behavioral experiments. In conclusion, A. argyi and S. album EOs have good repellent effects on RIFAs, and the active ingredients include dipentene, α-terpineol, 1-caryophyllene, linalool, 2-phenylethanol and benzyl acetate. Our findings provide new information for the development of green, safe, and efficient repellents against RIFAs.

Fumigant Activity of Sweet Orange Essential Oil Fractions Against Red Imported Fire Ants (Hymenoptera: Formicidae)

Hu Wei,Zhang Ning,Chen Hongli,Zhong Balian,Yang Aixue,Kuang Fan,Ouyang Zhigang,Chun Jiong

Sweet orange oil fractions were prepared by molecular distillation of cold-pressed orange oil from sample A (Citrus sinensis (L.) ‘Hamlin’ from America) and sample B (Citrus sinensis Osbeck ‘Newhall’ from China) respectively, and their fumigant activities against medium workers of red imported fire ants (Solenopsis invicta Buren) were investigated. The volatile composition of the orange oil fractions was identified and quantified using GC–MS. Fractions from sample A (A1, A2, and A3) contained 23, 37, and 48 chemical constituents, and fractions from sample B (B1, B2, and B3) contained 18, 29, and 26 chemical constituents, respectively. Monoterpenes were the most abundant components, accounting for 73.56% to 94.86% of total orange oil fractions, among which D-limonene (65.28–80.18%), β-pinene (1.71–5.58%), 3-carene (0.41–4.01%), β-phellandrene (0.58–2.10%), and linalool (0.31–2.20%) were major constituents. Fumigant bioassay indicated that all orange oil fractions exerted good fumigant toxicity against workers of fire ants at 3, 5, 10, and 20 mg/centrifuge tubes, and B1 had the strongest insecticidal potential, followed by A1, B2, A2, B3, and A3. The fractions composed of more high volatile molecules (A1 and B1) showed greater fumigant effects than others. Compounds linalool and D-limonene, which were the constituents of the orange oil, exhibited excellent fumigant toxicity against red imported fire ant workers. Linalool killed red imported fire ant workers completely at 5, 10, and 20 mg/tube after 8 h of treatment, and D-limonene induced >86% mortality at 8 h of exposure.

The Toxicity Differences of Fluralaner against the Red Imported Fire Ant (Solenopsis invicta) at Different Developmental Stages

Leyi Shao,Wei Wang,Xin Gong,Yinghao Yu,Junao Xue,Xinnian Zeng,Jiali Liu

The red imported fire ant (RIFA),Solenopsis invicta, is an invasive pest that causes damage to agricultural and ecological environments worldwide. Fluralaner is a new isoxazoline pesticide with the potential to become a control agent against RIFA. However, it is not clear whetherS. invictaresponds the same way to fluralaner at different reproductive stages. The present study firstly evaluated the toxicity of fluralaner toS. invictaat different developmental stages, finding that fourth instar larvae (LD50, 1744.23 mg/kg) and worker ants (LD50, 8.62 mg/kg) were differently susceptible to fluralaner, while the mortality rate of fourth instar larvae was significantly lower at the same concentration of 10 mg/L (5.56 ± 3.14%) than that of worker ants (62.22 ± 3.14%), demonstrating a greater tolerance to fluralaner. Subsequently, the metabolic responses of worker and larval ants to fluralaner stress (10 mg/L) were investigated using non-targeted metabolomics, which indicated that the amount of differential metabolites and the KEGG metabolic pathways enriched were different between workers and larvae when exposed to the same dose (10 mg/L) of fluralaner. Differential metabolites of larvae and worker ants under fluralaner stress were mainly concentrated in organic acids and their derivatives, lipids and lipid-like molecules, nucleosides, nucleotides, and analogues, combined with the enriched metabolic pathways, revealed that the differential metabolic responses of larvae and worker ants were mainly in energy metabolism, detoxification metabolism, and neurotransmitter ligands. Workers consumed more substrates in the arginine synthesis pathway (l-glutamic acid,l-aspartic acid, and fumaric acid) to provide energy for the detoxification (glutathione) of pesticides when exposed to fluralaner stress, and the high accumulation ofl-aspartic acid induced excitotoxicity in the worker ants. Larval ants consumed more arachidonic acid to synthesize PG D2, and changes in the metabolism of antioxidants such as catechins, hesperidin, andl-ascorbic acid suggested that larvae were more capable of scavenging the ROS response than worker ants. The results of non-targeted metabolomics successfully revealed differences in the sensitivity of larvae and workers to fluralaner agents, providing insights into the fluralaner control ofSolenopsis invicta. Keywords:contacting activity;fluralaner stress;invasive pest;metabolome;tolerance

Insecticidal and P450 mediate metabolism of fluralaner against red imported fire ant, Solenopsis invicta (Hymenoptera: Formicidae)

Ting Xiong,Si-quan Ling,Jia-li Liu,Xin-nian Zeng

The red imported fire ant ( Solenopsis invicta ), a worldwide invasive and polyphagous pest, and often nests in residential areas. Finding an alternative pesticide that is both effective on S. invicta and environmentally friendly is urgent and crucial. Fluralaner, a novel isoxazoline insecticide, has been proven to possess selective toxicity for insects versus mammals and has been safe for mammals and non-target organisms, suggesting its potential in pest management. However, little toxicity information is available for the controlment of S. invicta. In this article, we studied the toxicity of fluralaner against S. invicta systematically, and the roles of metabolism-related enzymes in the metabolism process of fluralaner. The toxicity results showed that the topical application and feeding application were all effective for S. invicta . Moreover, fluralaner can be transmitted among workers by contacting and feeding which leads to a toxic reaction among nestmates. By exploring the biochemistry change, we found cytochrome P450 monooxygenase ( P450 ) may be involved in the detoxification of fluralaner as well as carboxylesterase ( CarE ), but not glutathione S-transferase ( GST ). Synergism assays gave solid evidence in which piperonyl butoxide , an activity inhibitor of P450, increased the toxicity of fluralaner to S. invicta . Importantly, with the RNAi treatment, four of S.invicta P450 genes were significantly inhibited and showed more sensitivity to fluralaner at LC 50 concentration. Our result indicated that fluralaner could be a potential alternative pesticide in S. invicta control . And CYP9AS16 , CYP6AS161 , CYP6SQ20 , and CYP336A45 genes were closely associated with the metabolism process of fluralaner.

Using essential oils from Citrus paradisi as a fumigant for Solenopsis invicta workers and evaluating the oils’ effect on worker behavior

Zhang Ning,Liao Yihong,Xie Lianjie,Zhang Zhixiang,Hu Wei

The red imported fire ant is one of the world’s most devastating invasive species, adversely affecting humans, wildlife, crops, and livestock. To control infestations, chemical pesticides are deployed extensively around the world. However, their extensive use has led to negative effects on the environment and human health. Essential oils, which are safe and ecofriendly, can potentially be used as alternatives to chemical pesticides. In this study, grapefruit essential oils were used as fumigant agents to control red imported fire ants. The crude grapefruit oil (GO1) contained 28 compounds, and the concentrated grapefruit oil (GO2), which was refined from GO1 by vacuum distillation, contained 20 compounds. D-Limonene was the dominant constituent in both GO1 (70.1%) and GO2 (73.96%), and other important constituents included β-pinene, α-pinene, β-phellandrene, octanal, d-carvone, α-terpineol, and linalool. Both the essential oils and their individual constituents (α-pinene, α-terpineol, β-phellandrene, octanal, and d-carvone) showed strong lethal fumigant effects against workers. Workers were more susceptible to GO2 than GO1, and octanal was more toxic to workers as compared with the other four constituents. When antennas of workers were treated with the two oils or the five constituents, their walking and gripping abilities were significantly suppressed, and there was an obvious bending or breaking phenomenon on the sensilla of the antennas. Fumigant activity by grapefruit essential oils and their main compounds were associated with their effects on the walking and gripping behavior of workers, and this confirmed that grapefruit essential oil is a promising, ecofriendly, and safe fumigant for the control of red imported fire ants.

Exploring bioactive molecules with fumigation toxicity against the red imported fire ant Solenopsis invicta Buren from commercial essential oils by GC–MS combined with chemometrics

You Zhou,De Qiang Qin,Min Xing Zhang,Miao Sun,Lu Sheng Wan,Jing Sheng Chen

Metabolomics approaches are becoming a powerful analytical technique for exploring bioactive molecules in essential oils. The prevalence of imported red fire ants affected the environment, biodiversity of wildlife, infrastructure, and hypersensitivity reactions of people. Due to public health and environmental issues, essential oils as environmentally friendly alternatives to synthetic chemical insecticides in fire ant management have attracted great attention. However, there is a lack of an objective strategy suitable for identifying bioactive molecules that can guarantee the quality of essential oil biopesticides . Therefore, a metabolomics approach with four main steps, including bioassays, GC MS analysis of chemical constitution, construction of multivariate OPLS and univariate Pearson correlation statistical models, and feature selection, was proposed in this study to verify its potential to find bioactive molecules in essential oils that are effective against red imported fire ants. The fumigation mortalities of 20 commercial essential oils ranged from 0 to 100%. A total of 296 mass features were deconvoluted, and 121 of them were identified. Then, 52 potential bioactive molecules were selected by both VIP > 1.0 and P  < 0.05. Finally, 6 of these potential bioactive molecules, (+)− 2-bornanone,.beta.-pinene, eucalyptol , isoborneol, linalool , and L-.alpha.-terpineol, have shown good fumigation toxicity against minor and/or major red imported fire ant workers. This study has, in certain aspects, supported the feasibility of this metabolomics approach. It could be helpful for natural molecules to be alternatives to synthetic chemical insecticides, leading to more environmentally friendly and sustainable management policies for red imported fire ants.

Enhancement of tolerance against flonicamid in Solenopsis invicta (Hymenoptera: Formicidae) through overexpression of CYP6A14

Tian-yi Zhang,Chang-wei Gong,Jian Pu,An-chun Peng,Xu-yang Li,Yu-meng Wang,Xue-gui Wang

Solenopsis invicta is a main issue in southern China and is causing significant damage to the local ecological environment. The extensive use of insecticides has resulted in the development of tolerance in S. invicta . In our study, ten S. invicta colonies from Sichuan Province exhibited varying degrees of tolerance against flonicamid, with LC 50 values from 0.49 mg/L to 8.54 mg/L. The sensitivity of S. invicta to flonicamid significantly increased after treatment with the P450 enzyme inhibitor piperonyl butoxide (PBO). Additionally, the activity of P450 in S. invicta was significantly enhanced after being treated with flonicamid. Flonicamid induced the expression levels of CYP4aa1 , CYP9e2 , CYP4C1 , and CYP6A14 . The expression levels of these P450 genes were significantly higher in the tolerant colonies compared to the sensitive colonies, and the relative copy numbers of CYP6A14 in the tolerant colonies were 2.01–2.15 fold. RNAi feeding treatment effectively inhibited the expression of P450 genes, thereby reducing the tolerance of S. invicta against flonicamid. In addition, the overexpression of CYP6A14 in D. melanogaster resulted in reduced sensitivity to flonicamid. Our investigations revealed hydrophobic interactions between flonicamid and seven amino acid residues of CYP6A14, along with the formation of a hydrogen bond between Glu306 and flonicamid. Our findings suggest that flonicamid can effectively control S. invicta and P450 plays a pivotal role in the tolerance of S. invicta against flonicamid. The overexpression of CYP6A14 also increased tolerance to flonicamid.

Keratin-functionalized nanomesoporous silica enhances indoxacarb efficacy against red fire ants

Qun Zheng,Xiaofeng Xie,Suqing Huang,Jiao Ding,Nanhe Huang,Zhixiang Zhang

Feather keratin (FK) is a natural polymer material derived from discarded feathers with excellent modifiability and functionalization properties. However, the current applications in agriculture, especially in the development of nanopesticides targeting pest control, are still relatively limited. In this study, an insecticide controlled release system with pH and reduced glutathione (GSH) responsive properties was designed and prepared. The coating coverage was carried out by combining the carboxyl group of FK with aminated hollow mesoporous silica (HMS), which was loaded with indoxacarb (IDC) to obtain IDC@FK-HMS. IDC@FK-HMS had an average particle size of 178 nm, achieved a drug loading of 23.22 %, and exhibited superior thermal and photostability. Compared to IDC bait, IDC@FK-HMS bait has more significant insecticidal activity against red fire ants. The mortality of red fire ants fed with 8 mg/kg of bait for 72 h were 52.22 ± 1.92 % and 66.67 ± 3.34 %, respectively. In addition, IDC@FK-HMS had a more significant effect on the attack ability of red fire ants, which required more time to prey on yellow mealworms. IDC@FK-HMS was able to reduce the alertness and detoxification enzyme activity of red fire ants and enhance their sensitivity to insecticides, which in turn enhanced the insecticidal activity. FK-HMS encapsulation did not alter the toxic effects of IDC, both of which caused significant impacts on the midgut and acetylcholinesterase activity of red fire ants. The study enriched the avenues of application of FK in agriculture, especially in pest control, and provided new insights into the green and sustainable control of red fire ants.

A β-cyclodextrin-functionalized metal–organic framework enhances the insecticidal activity of indoxacarb by affecting amino acid metabolism in red imported fire ants

Liupeng Yang,Huiya Chen,Qun Zheng,Peiru Luo,Wenjuan Yan,Suqing Huang,Dongmei Cheng,Han Hong Xu,Zhixiang Zhang

The invasion area of red imported fire ants (RIFAs) in China continues to expand due to factors such as ecological changes and human activities, and the resulting damage is becoming increasingly severe. The development of smart nanopesticides that respond to environmental stimuli response with precisely controlled release has great potential in improving insecticidal effects. In this work, a smart pH and α-amylase dual stimuli-responsive pesticide delivery system was constructed by combining mono-(6-amino-6-deoxy)-β-cyclodextrin (β-CD-NH 2 ) with zeolitic imidazole skeleton-90 (ZIF-90) nanoparticles ( [email protected] ) loaded with indoxacarb. The average particle size of the prepared [email protected] was 249 nm, and the loading efficiency was 18.43%. The β-CD-NH 2 coating on the surface of nanoparticles reduced the photodegradation of indoxacarb and prevented the premature release of the active ingredient. In vitro release kinetics showed that [email protected] rapidly released IDC under acidic and amylase conditions, consistent with the oral and intestinal environment of RIFAs. The results of bioactivity tests showed that [email protected] enhanced the toxicity of IDC toward RIFAs by increasing feeding and damaging intestinal cells. The mortality rate of the [email protected] group was significantly higher than that of the IDC EC-treated group at 72 h. The nontargeted metabolomic results showed that [email protected] affected the metabolism of RIFAs by interfering with the metabolic pathways of six amino acids and one amino acid transport carrier, whereas IDC EC interfered with only two of these amino acid metabolic pathways. Overall, this work provides insight into the effective control of RIFA damage for sustainable pest management.

Bioactivity of the defensive chemicals of six millipede species in the superorder Juliformia against Solenopsis invicta Buren (Hymenoptera: Formicidae)

Delong Kong,Siquan Ling,Wanru Guo,Eduardo G.P. Fox,Jinzhu Xu,Jiamei Zhong,Desen Wang,Hualong Qiu,Weixin Liu

Millipedes are known to produce a diverse array of chemical defenses against natural predators, notably ants, positioning them as a promising source of natural products useful for pest ant control. A top invasive ant species of international scale is the red imported fire ant, Solenopsis invicta , for which there are numerous studies surveying alternative chemicals, as traditional insecticides have not been able to stop their spread. In this study, we characterized the composition of the defensive secretions from six quinone millipedes and evaluated the fumigant, contact, and repellent activities of some main components against S. invicta workers. In total, 27 components were detected in the defensive secretions of the six millipede species, with 2-methoxy-3-methyl-1,4-benzoquinone being the predominant compound overall. Nine compounds (1,4-benzoquinone, α -pinene, dimethyl trisulfide, β -pinene, 2-methyl-1,4-benzoquinone, 2-methoxy-3-methyl-1,4-benzoquinone, orcinol, 3,4-dimethoxyphenol, and 1-hydroxycyclohexyl phenyl ketone) were selected for bioassays. Among them, the three benzoquinones displayed significant fumigant and contact toxicity, as well as repellent effects against S. invicta . Dimethyl trisulfide exhibited fumigant and repellent activity, while 1-hydroxycyclohexyl phenyl ketone demonstrated just contact toxicity. These findings highlight the potential of millipede-derived compounds as natural agents for S. invicta control, possibly to direct a mixture of chemicals for use against fire ants' infestation in transported goods.

Effects of Gelsemium elegans extract on the red fire ant: disruption of peritrophic membrane integrity and alteration of gut microbial diversity, composition, and function

Zheng Qun,Yan Wenjuan,Zhu Shiqi,Miao Xiaoran,Wu Jian,Lin Zewei,Huang Suqing,Cheng Dongmei,Xu Hanhong,Zhang Zhixiang,Zhang Peiwen

Gelsemium elegans Benth. ( Loganiaceae ), also known as heartbreak herb, can be used in the manufacture of herbal medicines. Insecticidal activity has also been found and can be used to develop botanical insecticides. This study aimed to reveal the insecticidal mechanism of its extracts against red fire ants and provide strategies for the development of biopesticides and the promotion of green and sustainable agriculture. 16s rRNA, pathohistological, behavioral, and enzyme activity assays were performed to reveal its biological effects, including the effects on non-target organisms. Our results showed that red fire ants exposed to G. elegans extracts exhibited slowed growth, reduced feeding, and decreased aggressiveness. The midgut and its peritrophic membrane of red fire ant were significantly disrupted, the diversity of gut microbial community was reduced, and the balance of microbial composition was disturbed. Significant increases in functional abundance of xenobiotics biodegradation and metabolism pathway and P450s enzyme activity confirmed the toxic stress of G. elegans extract. Functional prediction of Kyoto Encyclopedia of Genes and Genomes pathway showed that the functional abundance of novobiocin biosynthesis, flavonoid biosynthesis, lysosome, proteasome, and wingless/integrated signaling pathways were significantly inhibited in the gut. Besides, G. elegans extracts induced an increase in acetylcholinesterase activity. These results revealed dysregulation of immune system and metabolic functions in red fire ants, as well as toxic effects of G. elegans extracts on physiological functions and nerves. These findings revealed the insecticidal mechanism of G. elegans and supported the development of eco-friendly insecticides for red fire ants.
