Red blood cell membrane-derived phototherapeutic nanodiscs: A new platform for enhanced phototherapy

Zhuge Deli,Li Li,Sun Xueying,Liang Hui,Jin Chenjie,Lu Ailing,Gao Wenli,Zhong Yutong,Li Wenlu,Chen Sihao,Wang Fang,Yan Linzhi,Meng Weiyang,Lin Xiaoji,Zhang Xufei,Liu Yong,Li Yuanfeng,Zhao Yingzheng,Zhang Xiang,Chen Mengchun,Chen Yijie

Phototherapy holds great potential for treating cancer and infections but faces limitations related to photosensitizer accumulation, tissue penetration, and diminished photo-conversion efficiency, particularly in deep-seated tumors and infections. Here, a biomimetic phototherapeutic nanodisc platform consisting of erythrocyte membranes and the photosensitizer IR780 is developed. With the advantages of the ultra-small size and exceptional biosafety of cell membrane-derived nanodiscs, this platform facilitates efficient accumulation and deep tissue penetration at disease sites. Upon near-infrared (NIR) irradiation, IR780 delivered via the nanodisc exhibits enhanced photothermal conversion efficiency, markedly inhibiting tumor growth in an orthotopic 4T1 breast cancer model and reducing bacterial load in a methicillin-resistant Staphylococcus aureus (MRSA) skin infection model. Furthermore, the nanodisc platform demonstrates outstanding biocompatibility in mice. In conclusion, this nanodisc system significantly extends the functional potential of cellular nanodiscs, presenting a promising strategy to address the challenges of photosensitizer delivery and biosafety in phototherapeutic applications.

Erythrocyte membrane-coated gold nanocages for targeted photothermal and chemical cancer therapy

Dao-Ming Zhu,Wei Xie,Yu-Sha Xiao,Meng Suo,Ming-Hui Zan,Qing-Quan Liao,Xue-Jia Hu,Li-Ben Chen,Bei Chen,Wen-Tao Wu,Li-Wei Ji,Hui-Ming Huang,Shi-Shang Guo,Xing-Zhong Zhao,Quan-Yan Liu,Wei Liu

Recently, red blood cell (RBC) membrane-coated nanoparticles have attracted much attention because of their excellent immune escapability; meanwhile, gold nanocages (AuNs) have been extensively used for cancer therapy due to their photothermal effect and drug delivery capability. The combination of the RBC membrane coating and AuNs may provide an effective approach for targeted cancer therapy. However, few reports have shown the utilization of combining these two technologies. Here, we design erythrocyte membrane-coated gold nanocages for targeted photothermal and chemical cancer therapy. First, anti-EpCam antibodies were used to modify the RBC membranes to target 4T1 cancer cells. Second, the antitumor drug paclitaxel (PTX) was encapsulated into AuNs. Then, the AuNs were coated with the modified RBC membranes. These new nanoparticles were termed EpCam–RPAuNs. We characterized the capability of the EpCam–RPAuNs for selective tumor targeting via exposure to near-infrared irradiation. The experimental results demonstrate that EpCam–RPAuNs can effectively generate hyperthermia and precisely deliver the antitumor drug PTX to targeted cells. We also validated the biocompatibility of the EpCam–RAuNs in vitro. By combining the molecularly modified targeting RBC membrane and AuNs, our approach provides a new way to design biomimetic nanoparticles to enhance the surface functionality of nanoparticles. We believe that EpCam–RPAuNs can be potentially applied for cancer diagnoses and therapies.

Zwitterionic choline phosphate conjugated folate-poly (ethylene glycol): a general decoration of erythrocyte membrane-coated nanoparticles for enhanced tumor-targeting drug delivery

Yuyue Zhang,Yuemin Wang,Qiangwei Xin,Mingjing Li,Peng Yu,Jun Luo,Xinyuan Xu,Xingyu Chen,Jianshu Li

Erythrocyte membrane nanosystems have become one of the important research directions of disease treatment, especially for tumor treatment, and can enhance the long circulation time of anti-cancer drugs in vivo, and penetrate and accumulate in the tumor site effectively. However, erythrocyte membranes lack targeting properties and it is necessary to provide tumor-targeting function by modifying erythrocyte membranes. In this study, we report on a novel modification method of an erythrocyte membrane nanosystem to target tumors. Specifically, the tumor-targeting molecule folate-poly (ethylene glycol) (FA-PEG) was modified with a zwitterionic 2-(methyl acryloyoxy) ethyl choline phosphate (MCP) by the Michael addition reaction to obtain MCP-modified FA-PEG (MCP-PEG-FA). Based on the strong “N–P” tetravalent electrostatic interaction between MCP and phosphatidyl choline on the erythrocyte membranes, MCP-PEG-FA can be modified on the erythrocyte membrane encapsulated doxorubicin (DOX) loaded poly(lactic-co-glycolic acid) (PLGA) nanosystem to form a tumor-targeting erythrocyte membrane nanosystem (FA-RBC@PLGA-DOX). The results show that MCP-PEG-FA was synthesized and successfully bonded to the erythrocyte membrane nanosystem, and the FA-RBC@PLGA-DOX nanosystem had a better tumor-targeting function and tumor killing effect compared with those of the nanosystems without FA ligand modification. The universal modification method of erythrocyte membranes is successfully provided and can be applied to the treatment of various diseases.

Erythrocyte Membrane-Wrapped pH Sensitive Polymeric Nanoparticles for Non-Small Cell Lung Cancer Therapy

Lipeng Gao,Hao Wang,Lijuan Nan,Ting Peng,Lei Sun,Jinge Zhou,Ye Xiao,Jing Wang,Jihong Sun,Weiyue Lu,Lin Zhang,Zhiqiang Yan,Lei Yu,Yiting Wang

The application of nano drug delivery systems (NDDSs) may enhance the effectiveness of chemotherapeutic drugs in vivo. However, the short blood circulation time and poor drug release profile in vivo are still two problems with them. Herein, by using red blood cell membrane (RBCm) wrapping and pH sensitive technology, we prepared RBCm wrapped pH sensitive poly(l-γ-glutamylcarbocistein)-paclitaxel (PGSC–PTX) nanoparticles (PGSC–PTX@RBCm NPs), to prolong the circulation time in blood and release PTX timely and adequately in acidic tumor environment. The PGSC–PTX NPs and PGSC–PTX@RBCm NPs showed spherical morphology with average sizes about 50 and 100 nm, respectively. The cytotoxicity of PGSC–PTX@RBCm NPs was considerably decreased compared with that of PGSC–PTX NPs. PTX release from PGSC–PTX and PGSC–PTX@RBCm NPs at pH 6.5 was remarkably higher than those at pH 7.4, respectively. The PGSC–PTX@RBCm NPs exhibited remarkably decreased uptake by macrophages than PGSC–PTX NPs. The area under the curve within 72 h (AUC0–72h) for is significantly higher than PGSC–PTX NPs. The PGSC–PTX@RBCm NPs also showed significantly stronger growth-inhibiting effect on tumor than PGSC–PTX NPs. These results indicated that PGSC–PTX@RBCm NPs have acidic drug release sensitivity, the characteristics of long circulation, and remarkable tumor growth inhibiting effect. This study may provide an effective strategy for improving the antitumor effect of NDDS.

Galactose-modified erythrocyte membrane fusion liposomes enable the targeted delivery of drug nanoparticles to the liver

Jiayu Song,Huanhuan Zhang,Xiaohui Zhang,Meiying Liu,Dan Peng,Yuan Ren,Yan Sun,Yunlan Li

The safe and efficient delivery of chemicals and biologics remains crucial for liver disease therapy. In this study, we developed a targeted drug delivery system utilizing a galactose-modified erythrocyte membrane coating technique and drug liposome nanoparticles, which were further optimized using orthogonal experiments and response surface analysis. The specificity, precision, accuracy, and stability exhibited satisfactory performance in bioanalytical analysis. Specifically, targeting ligands (Gal-DSPE-PEG3400) were efficiently inserted into red blood cell (RBC) membranes using a facile insertion method. When Gal-DSPE-PEG3400-RBC was fused with fenofibrate liposome nanoparticles (FNB-Lip) by co-extrusion, the resulting galactose-modified erythrocyte membrane fusion liposome nanoparticles (Gal-RBC-FNB-Lip) showed long-term stability, excellent biocompatibility, prolonged retention time, and superior liver accumulation and therapeutic efficacy. These qualities make it suitable for effective drug delivery. The findings of this study will provide a fundamental basis for research and development of liver-targeted drugs and offer novel insights into the treatment of clinical liver diseases.

Enhanced biocompatibility of silk sericin/caffeic acid nanoparticles by red blood cell membranes cloaking

Chunru Wang,Qingyang Lu,Yingjie Xiang,Yulan Yin,Junyao Li,Yalu Liu,Xiaochen Wu

Caffeic acid (CA) is an antioxidant phenolic compound that enriched in coffee beans, however, its administration often restrains by the instability and low solubility. Nanoparticle encapsulation is an effective approach to improve the therapeutic activity of CA . For example, silk sericin (SS), a natural biomaterial finds applications in food, cosmetics and biomedical fields, is proved here to be an appropriate encapsulation agent for CA, and a SS/CA composite nanoparticle has been fabricated. To further improve the biocompatibility of SS/CA, a red blood cell membranes (RM) cloaking strategy is adopted. The as-formed SS/CA/RM preserves the antioxidant activity of CA, and shows satisfactory biocompatibility especially under high concentration. Hope this can provide a potential appropriative strategy to adjust the chemical stability of insoluble drugs and to improve their biocompatibility.

Boron nitride nanosheets elicit significant hemolytic activity via destruction of red blood cell membranes

Xuejie Xie,Zhenyu Hou,Guangxin Duan,Shitong Zhang,Hong Zhou,Zaixing Yang,Ruhong Zhou

Boron nitride (BN) nanosheets have emerged as promising nanomaterials in a wide range of biomedical applications. Despite extensive studies on these bio-nano interfacial systems, the underlying molecular mechanisms remain elusive. In this study, we used hemolysis assays and morphology observations to demonstrate for the first time that BN nanosheets can cause damages to the red-blood-cell membranes, leading to significant hemolysis. Further molecular dynamics simulations revealed that BN nanosheets can penetrate into the cell membrane and also extract considerable amount of phospholipid molecules directly from the lipid bilayer. The potential of mean force calculations then showed that their penetration effect was thermodynamically favorable due to the strong attractive van der Waals interactions between BN nanosheets and phospholipids. Overall, these findings provided valuable insights into the interaction of BN nanosheets with cell membranes at the atomic level, which can help future de novo design of BN-based nanodevices with better biocompatibility.

Preparation and evaluation of long circulating erythrocyte membrane-cloaked anti-cancer drug delivery system

Chu Yuqi,Zhang Jinfeng,Pan Hao,Shi Jinyan,Wang Jinglei,Chen Lijiang

Recently, biomimetic hybrid drug delivery systems, especially erythrocyte membrane-based drug delivery systems, have been utilized to achieve high bioavailability, and biocompatibility, in the meantime, to reduce immunogenicity and effectively evade phagocytosis of the host immune system. Here, we developed a novel drug delivery system of red blood cell membrane-derived vesicles (RDVs) cloaked poly (acrylic acid)-cystamine hydrochloride-D-α-tocopherol succinate (PAAssVES) nanoparticles. The PAAssVES nanoparticles were prepared via emulsification and solvent volatilization method, followed by loading of the model anti-cancer drug, sorafenib (SFN). Then RDVs and SFN-PAAssVES nanoparticles were uniformly mixed and co-extruded through polycarbonate membrane. The prepared RDV-coated nanoparticles (RDV-NPs) had good stability, with a zeta potential of − 10.7 mV and particle size of 113.5 nm. MTT assay was used to analyze the effects of RDV-NPs on cell viability in two kinds of gastric cancer cell lines BGC-823 and MKN-45. The results showed that RDV-NPs significantly decreased cell viability. In vitro drug release investigation showed that RDV-NPs had good sustained release properties and the cumulative release was 71.5% in 72 h. In pharmacokinetic studies, SD male rats’ intravenous injection with RDV-NP solution showed a more smooth plasma concentration-time profile. Compared with free SFN treatment and SFN-PAAssVES group, RDV-NPs enhanced the AUC by about 4.1-fold and 2.0-fold. The MRT and t 1/2 of RDV-NPs were increased to 23.670 ± 2.347 h and 24.450 ± 2.652 h. Our study demonstrated the promise of using RDV-NPs as a long circulating anti-cancer drug delivery system. Graphical abstract

Synthetic nanoparticles camouflaged with biomimetic erythrocyte membranes for reduced reticuloendothelial system uptake

Lang Rao,Jun-Hua Xu,Bo Cai,Huiqin Liu,Ming Li,Yan Jia,Liang Xiao,Shi-Shang Guo,Wei Liu,Xing-Zhong Zhao

Suppression of the reticuloendothelial system (RES) uptake is one of the most challenging tasks in nanomedicine. Coating stratagems using polymers, such as poly(ethylene glycol) (PEG), have led to great success in this respect. Nevertheless, recent observations of immunological response toward these synthetic polymers have triggered a search for better alternatives. In this work, natural red blood cell (RBC) membranes are camouflaged on the surface of Fe3O4 nanoparticles for reducing the RES uptake. In vitro macrophage uptake, in vivo biodistribution and pharmacokinetic studies demonstrate that the RBC membrane is a superior alternative to the current gold standard PEG for nanoparticle 'stealth'. Furthermore, we systematically investigate the in vivo potential toxicity of RBC membrane-coated nanoparticles by blood biochemistry, whole blood panel examination and histology analysis based on animal models. The combination of synthetic nanoparticles and natural cell membranes embodies a novel and biomimetic nanomaterial design strategy and presents a compelling property of functional materials for a broad range of biomedical applications.

Red blood cell membrane-coated functionalized Cu-doped metal organic framework nanoformulations as a biomimetic platform for improved chemo-/chemodynamic/photothermal synergistic therapy

Luping Ren,Yanfang Sun,Junhao Zhang,Lei Nie,Amin Shavandi,Khaydar E. Yunusov,Uladzislau E. Aharodnikau,Sergey O. Solomevich,Guohua Jiang

Nanoformulations for combining chemotherapy, chemodynamic therapy, and photothermal therapy have enormous potential in tumor treatment. Coating nanoformulations with cell membranes endows them with homologous cellular mimicry, enabling nanoformulations to acquire new functions and properties, including homologous targeting and long circulation in vivo , and can enhance internalization by homologous cancer cells. Herein, we fused multifunctional biomimetic nanoformulations based on Cu-doped zeolitic imidazolate framework-8 (ZIF-8). Hydroxycamptothecin (HCPT), a clinical anti-tumor drug, was encapsulated into ZIF-8, which was subsequently coated with polydopamine (PDA) and red blood cell membrane. The as-fabricated biomimetic nanoformulations showed an enhanced cell uptake in vitro and the potential to prolong blood circulation in vivo , producing effective synergistic chemotherapy, chemodynamic therapy, and photothermal therapy under the 808 nm laser irradiation. Together, the biomimetic nanoformulations showed a prolonged blood circulation and evasion of immune recognition in vivo to provide a bio-inspired strategy which may have the potential for the multi-synergistic therapy of breast cancer.

All-stage targeted red blood cell membrane-coated docetaxel nanocrystals for glioma treatment

Yuan Ding,Qianzhu Xu,Zhilan Chai,Sunyi Wu,Weixia Xu,Jun Wang,Jianfen Zhou,Zimiao Luo,Yu Liu,Cao Xie,Linwei Lu,Weiyue Lu

Challenges for glioma treatment with nanomedicines include physio-anatomical barriers (the blood-brain barrier and blood-brain tumor barrier), low drug loading capacity, and limited circulation time. Here, a red blood cell membrane-coated docetaxel drug nanocrystal (pV-RBCm-NC(DTX)), modified with pHA-VAP (pV) for all-stage targeting of glioma, was designed. The NC(DTX) core exhibited a high drug loading capacity but low in vivo stability, and the RBCm coating significantly enhanced the stability and prolonged in vivo circulation. Moreover, the Y-shaped targeting ligand pV was modified by a mild avidin-biotin interaction, which endowed RBCm-NC(DTX) with superior barrier-crossing ability and therapeutic efficacy. The integration of nanocrystal technology, cell membrane coating, and the avidin-biotin insertion method into this active targeting biomimetic formulation represents a promising drug delivery strategy for glioma.

Cryptotanshinone-loaded biomimetic pH-sensitive liposomes for the treatment of liver fibrosis

Kai Liu,Bolin Zhao,Zihao Sun,Nini Luo,Xiaofang Li

Liver fibrosis is characterized by the aberrant accumulation of extracellular matrix (ECM) components, primarily due to the persistent activation of hepatic stellate cells (HSCs). It is well-known that chronic liver inflammation from various etiologies leads to hepatocellular damage, which activates HSCs, initiating and exacerbating fibrosis. This presents a significant challenge for therapeutic interventions aimed at restoring ECM balance. Here, we present CPT-pH@RLs, a novel drug delivery system consisting of pH-sensitive liposomes modified with erythrocyte membranes (RBCm), designed to deliver cryptotanshinone to the liver. The results of cellular uptake and real-time in vivo imaging indicate that the modification of RBCm reduces the uptake of CPT-pH@RLs by macrophages and prolongs the in vivo cycling time of these particles. CPT-pH@RLs effectively reduce liver inflammation by inhibiting secretion of interleukin-1β (IL-1β). Moreover, they improve liver function, decrease ECM deposition, and alleviate liver fibrosis. Our findings demonstrate that CPT-pH@RLs represent a safe and effective passive targeting drug delivery system for anti-inflammatory and ECM normalization therapies, offering a promising approach for the reversal of liver fibrosis.
