This study focused on developing an effective cell wall-breaking method forPhaffia rhodozyma, followed by utilizing subcritical fluid extraction to isolate, extract, and concentrate astaxanthin from the complex fermentation products ofP. rhodozyma. A comprehensive comparison of seven distinct methods for disrupting cell walls, including dimethyl sulfoxide treatment, lactic acid treatment, sodium hydroxide treatment,β-glucanase enzymatic digestion,β-mannanase enzymatic digestion, and a combined enzymatic treatment involving bothβ-mannanase andβ-glucanase was conducted. The results identified the lactic acid method as the most effective in disrupting the cell walls ofP. rhodozyma. The software, Design Expert, was used in the process of extracting astaxanthin from cell lysates using a subcritical extraction method. Through fitting analysis and response surface optimization analysis by Design Expert, the optimal extraction conditions were determined as follows: an extraction temperature of 41 °C, extraction frequency of two times, and extraction time of 46 min. These parameters facilitated the efficient extraction, concentration, and enrichment of astaxanthin fromP. rhodozyma, resulting in an astaxanthin concentration of 540.00 mg/L. This result can establish the foundation for its high-value applications.

Microbial carotenoids and lipids are two groups of versatile biomolecules widely used in the food, fuel, pharmaceutical, and cosmetic industries. Rhodotorula glutinis is one of the best-known oleaginous red yeasts that has the remarkable capability to produce an assortment of valuable carotenoids and lipids. Here, the effects of different concentrations of TiO 2 and H 2 O 2 treatments on carotenoid and lipid production in R. glutinis ZHK were investigated. After 120 h of cultivation, the total amount of carotenoids was significantly increased in the TiO 2 (100 mg/L: 23.165 μg/gdw, 750 mg/L: 19.65 μg/gdw) and H 2 O 2 (5 mM: 21.51 μg/gdw, 10 mM: 118.858 μg/gdw) treatments than the control (6.099 μg/gdw). In contrast, as compared to the control, the relative abundance of total lipids was significantly decreased in the TiO 2 (100 mg/L) and H 2 O 2 (10 mM) treatments, respectively. In response to TiO 2 and H 2 O 2 stresses, the increased carotenoids were probably attributed to the down-regulated lipid biosynthesis flux, while the decreased lipids might be ascribed to the down-regulated fatty acid biosynthesis flux. Our results presented herein not only provide an effective strategy for modulating carotenoid and lipid biosynthesis in R. glutinis ZHK but also reveal the molecular foundation to further enhance their bioproductivity by molecular manipulation.

Nine transformants of Rhodosporidium toruloides with significant changes in the carotenoid profile were obtained by Agrobacterium tumefaciens-mediated transformation, including a white, three red, and four yellow mutants. A red mutant A1-15-BRQ that showed a high torularhodin production was selected for culture condition optimization. Results indicated that the torularhodin yield was boosted with glucose as the carbon source, at a carbon/nitrogen ratio of 22, a loading volume of 75 mL, and 28 °C. The torularhodin yield of 21.3 mg/L consisting of 94.4% total carotenoids was obtained by Box–Behnken design experiments. The torularhodin yield was 17.0-fold higher than that of the wild type, with time shortened from 9 to 3 days. This study reports an effective strategy for improving torularhodin production and provides a candidate R. toruloides strain for highly selective production of torularhodin.

In the field of oil refining, the presence of excessive residual phosphorus in crude oil can significantly impact its quality, thereby emphasizing the necessity for compact and convenient testing equipment. This study primarily focuses on developing of self-powered biosensor (SPB) using immobilizing Choline Oxidase with a photoactive ternary nanocomposite complex (CHOx-BiOI-rGO-Fe 3 O 4 NPs-ITO) as the anode and utilizing a Pt electrode as the cathode. The successful preparation of the ternary composite photoelectrode for the anode was confirmed through a range of characterization techniques, including X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), Transmission electron microscopy (TEM), Scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), N 2 absorption/desorption, Dynamic light scattering (DLS), and Ultraviolet–visible diffuse reflection spectrometer (UV–vis DRS). The electrochemical and photoelectrochemical properties were assessed using an electrochemical workstation, revealing a significant enhancement photoelectrical responsiveness attributed to the formation of heterojunction structures. The SPB exhibited a remarkable linear relationship between the instantaneous photocurrent and phosphatidylcholine (PC) concentration, with a regression equation of I (μA) = 39.62071C (mM) + 3.47271. The linear range covered a concentration range of 0.01–10 mM, and the detection limit (S/N = 3) was determined to be 0.008 mM. It demonstrated excellent reproducibility and storage stability, positioning it a promising alternative to High-performance liquid chromatography (HPLC) for accurate quantification of PC content in rhodotorula glutinis oil. The standard recovery PC content ranged from 98.48% to 103.53%, with a relative standard deviation (RSD) ranging from 1.4% to 2.4%. This research presents a convenient and precise detection device that has the potential to address the issue of lagging detection in the oil refining process.

The livestock industry has undergone rapid expansion in recent years, resulting in an increased demand for feed protein. The production of plant and animal proteins cannot sustainably meet the expanding demand for protein, and the development of single cell protein (SCP) as a feed protein is a promising avenue. Molasses is a cheap and nutritious by-product of sugar production from sugarbeet and sugarcane, and is one of the common substrates available to microorganisms. In this study, R. glutinis As 2.703 was used to produce SCP by fermentation with inexpensive carbon source molasses and nitrogen source corn steep liquor instead of glucose and yeast extract, respectively. The results revealed that when the concentration of molasses in the substrate was 61 g/L, the biomass concentration reached 21.0 g/L, and the protein, lipid, carotenoid contents in dry cell were 30.4 %, 7.8 %, and 31.7 μg/g, respectively. SuperPro Designer was used to simulate the process and carry out the economic analysis for producing SCP from 100,000 tons of molasses annually. A plant of this size could produce 32,908 tons/year of SCP at cost of 988.2 USD/ton with payback period of 6.3 years. Sensitivity analysis showed that the selling price of SCP has the greatest impact on the economics of industrial production of SCP, and that by increasing its protein and carotenoid content, its value can be increased, and further increasing its selling price is the key to achieving industrialisation. This study provides new ideas to cope with the future protein feed crisis.

Catalytic hydrotreating of renewable oils is a promising way to get diesel alkanes, with the use of microbial lipids further facilitating scalable production of green diesel. The core issue of triglyceride hydrodeoxygenation (HDO) research is still to develop efficient catalysts under mild reaction conditions. Herein, we demonstrate that the heteropolyacid (HPA)-modified SiO2 supported Pd catalysts showed high activity for the HDO reaction of methyl stearate and soybean oil to diesel alkanes (C15∼C18, >90 wt % at 200 °C). Characterizations show that the Brønsted acid sites introduced by grafting heteropolyacid on SiO2 and metal sites of small Pd particles contribute to the high HDO activity of this bifunctional catalyst. Additionally, lipids produced by Rhodosporidium toruloides Y4 from glucose or corn stover were also transformed to about 75 wt % of diesel-range alkane products. This integration of microbial lipid production with chemical catalysis should facilitate the engineering of green diesel production from lignocellulosic biomass.
