Surface-enhanced Raman scattering (SERS) in conjunction with nanozymes has emerged as a powerful technique for bioanalysis due to their signal conversion and amplification effect. However, clinical applications continue to encounter challenges such as inadequate selectivity. Herein, we proposed a novel approach for the selective SERS detection of L-DOPA in human serum with dual enzyme-mimicking MOF-919(Fe–Cu). This material exhibits both catechol oxidase-like and peroxidase-like activities, enabling it to catalyze catecholic compounds to o -quinones and subsequently oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) molecules into SERS-active ox-TMB. Ultrasonic-assisted synthesis ensures uniform and dense loading of AgNPs within MOF-919(Fe–Cu), thereby enhancing SERS activity. A sensitive and selective method for SERS detection of L-DOPA was developed. The catechol oxidase-like activity of MOF-919(Fe–Cu) provides ideal selectivity and anti-interference to L-DOPA, while the cascade catalytic system transforms weak SERS signals of L-DOPA into amplified SERS signals of ox-TMB. The mechanism of dual enzyme-mimicking activity was further discussed. The detection process was optimized to achieve high sensitivity towards L-DOPA with a linear range 0.5–100 μmol/L and low limit of detection (LOD) of 0.16 μmol/L. Furthermore, it has been applied in serum sample of Parkinson's patients, demonstrating excellent recovery and low relative error compared to HPLC-UV. This approach offers an economical, efficient and accurate strategy for the detection of L-DOPA and provides valuable insights for the integration of selective multifunctional nanozyme and sensitive SERS detection method.

Multifunctional catalytic performance plays a crucial role in bio-applications through the diversity and durability of artificial nanozymes. An effective synergy with sufficient accessible active sites and high specific surface area is a challenge for composite catalysts, especially to avoid uncontrollable aggregation and structural instability. Here, we fabricated a Cu n O/Au heterostructure dendrimer on copper foam (Cu n O/Au HD/CF) as dual functional catalytic nanozyme to achieve enzyme mimic cascade reactions for efficient colorimetric analysis. A highly porous CF skeleton–based CuO nanowire array (CuO NWA) with a large specific surface area supported an efficient load capacity to assemble sufficient Cu n O/Au HD by electrodeposition. The bimetallic Au-Cu nanozyme successfully achieved an oxidase-like and peroxidase-like cascade catalysis by a target-responsive sensing mechanism. Due to the confirmed catalytic performance of selectivity, anti-interference ability, and reproducibility, a Cu n O/Au HD/CF-based quantitative analytical method was developed for glucose detection with a wide linear range and considerable detection limit of 8.4 μM. The robust nonenzymatic catalytic strategy for colorimetric detection not only confirmed the dual functional catalytic activity of Cu n O/Au HD/CF, but also showed great potential for applications in clinical diagnostics and biochemical analysis. The tumor microenvironment (TME) responsive multimodal theranostic nanoplatforms have attracted great attentions for precision medicine. To design well-defined cascade catalytic therapy and achieve the synergistic effect of different components in a nanoplatform, herein a spindle-shaped nanodrug termed as FeCu-GOx PNzyme-MTO was constructed by in-situ growth of bimetallic metal nanozyme (FeCu PNzyme), and simultaneous encapsulation of glucose oxidase (GOx) as well as fluorescent and chemotherapeutic mitoxanthrone (MTO). Interestingly, the obtained FeCu-GOx PNzyme is able to efficiently upregulate endogenous H 2 O 2 level and down-regulate acidity via inducing the catalytic decomposition of intratumoral glucose to gluconic acid and H 2 O 2 in TME, which is benefit to the cascade catalytic reactions for chemodynamic therapy (CDT). After efficient loading of MTO, such intelligent nanozyme triggers reactive oxygen species (ROS) generation in situ with tumor stimuli, self-augment ROS level upon intrinsic photothermal-conversion efficiency (53.05%) and glutathione (GSH) depletion, and specifically TME-responsive release of MTO to achieve high chemotherapeutic efficacy and biosafety. Both in vitro cellular assays and in vivo tumor-xenograft experiments proved that the tumor-specific cytotoxicity, biodegradability and potent antitumor efficiency of nanodrug, with co-contributions from highly cytotoxic ROS generated and precise drug delivery within tumors. This work thus presents the friendly construction of biocompatible FeCu-GOx PNzyme-MTO as effective nanodrug to provided a promising strategy for a new perspective on effective tumor therapy.

This work presents a generic strategy to create a series of metal mercaptides complexes via coordination self-assembly between transition metals (Mn, Cu, Co, Fe, and Ni) and cysteine (Cys) by forming the sulfur-metal bridges. This strategy involves dissolving metal chlorides and Cys into deep eutectic solvents (DES), followed by the precipitation of metal mercaptides complexes (such as Cys-Mn) by adding water as an antisolvent, where DES serves as the solvent, shape directing, and capping agent, thereby preventing the formation of other metal impurities. Interestingly, the prepared complexes possess both laccase and peroxidase-like properties, allowing the design of a technique for the detection of L-3,4-dihydroxyphenylalanine ( l -DOPA) and uric acid, respectively. The prepared Cys-Mn can linearly oxidize l -DOPA with its concentrations from 0.1 to 130 μM and the detection limit was calculated to be 75.5 nM. Additionally, the Cys-Mn can mimic the activity of peroxidase towards oxidization of o-phenylenediamine at neutral pH, allowing single-step and one-pot cascade reactions for visual and fluorometric measurements of uric acid (UA) that could work in the range of 0.2–500 μM UA with a detection limit of 0.06 μM and 0.054 μΜ, respectively. The assay was successful in detecting UA in serum and urine samples with relative standard deviation (RSD) ranging from 7.3% to 10.2% and 3.0%–8.5% respectively, suggesting that it may prove useful in medical diagnostic testing.

Biocatalytic transformations in living organisms, such as multi-enzyme catalytic cascades, proceed in different cellular membrane-compartmentalized organelles with high efficiency. Nevertheless, it remains challenging to mimicking biocatalytic cascade processes in natural systems. Herein, we demonstrate that multi-shelled metal-organic frameworks (MOFs) can be used as a hierarchical scaffold to spatially organize enzymes on nanoscale to enhance cascade catalytic efficiency. Encapsulating multi-enzymes with multi-shelled MOFs by epitaxial shell-by-shell overgrowth leads to 5.8~13.5-fold enhancements in catalytic efficiencies compared with free enzymes in solution. Importantly, multi-shelled MOFs can act as a multi-spatial-compartmental nanoreactor that allows physically compartmentalize multiple enzymes in a single MOF nanoparticle for operating incompatible tandem biocatalytic reaction in one pot. Additionally, we use nanoscale Fourier transform infrared (nano-FTIR) spectroscopy to resolve nanoscale heterogeneity of vibrational activity associated to enzymes encapsulated in multi-shelled MOFs. Furthermore, multi-shelled MOFs enable facile control of multi-enzyme positions according to specific tandem reaction routes, in which close positioning of enzyme-1-loaded and enzyme-2-loaded shells along the inner-to-outer shells could effectively facilitate mass transportation to promote efficient tandem biocatalytic reaction. This work is anticipated to shed new light on designing efficient multi-enzyme catalytic cascades to encourage applications in many chemical and pharmaceutical industrial processes.

Although Zeolitic Imidazolate Framework-8 (ZIF-8) shows significant promise in chemodynamic therapy of bacterial infections due to its large specific surface area and enzyme-like activity, it still faces a considerable gap compared to natural enzymes. The dependency on low pH and high concentrations of hydrogen peroxide ((H 2 O 2 ) is a major factor limiting the clinical progress of nanozymes. Single-atom nanozymes (SA-zyme), which exhibit superior catalytic performance, are expected to overcome this limitation. In this study, we used ZIF-8 as a template to prepare structurally regular molybdenum-based single-atom nanozymes (Mo-zyme) by coordinating molybdenum atoms with nitrogen atoms within the zeolitic imidazolate framework and evaporating the zinc element at high temperatures. The cascade catalytic performance of the nanodrugs was enhanced by loading glucose oxidase (GOx) and encapsulating it with a hyaluronic acid (HA) layer to form a composite (Mo/GOx@HA). Upon contact with hyaluronidase from bacteria in infected tissues, the cascade reaction is triggered, resulting in the degradation of the HA shell, and releasing the encapsulated GOx. Once exposed, GOx catalyzes the oxidation of glucose into gluconic acid, resulting in a localized decrease in pH and continuous production of H 2 O 2 . The combination of lowered pH and increased H 2 O 2 concentration significantly amplifies the catalytic activity of the Mo-zyme. This enhanced activity facilitates the in situ generation of hydroxyl radicals (•OH) on the bacterial surface, leading to effective and efficient bacterial eradication. Wound infection treatment has demonstrated that the as-prepared Mo/GOx@HA exhibits excellent antibacterial and anti-inflammatory activity. This work provided a promising enzymatic cascade reaction nanoplatform for the treatment of bacteria infected wounds.

Chemodynamic therapy (CDT) involving the highly toxic hydroxyl radical ( OH) has exhibited tremendous potentiality in combating bacterial infection. However, its antibacterial efficacy is still unsatisfactory due to the insufficient H 2 O 2 levels and near neutral pH at infection site. Herein, a glucose-fueled and H 2 O 2 -self-supplying OH nanogenerator (pFe 3 O 4 @GOx) based on cascade catalytic reactions is developed by immobilizing glucose oxidase (GOx) on the surface of PAA-coated Fe 3 O 4 (pFe 3 O 4 ). Magnetic pFe 3 O 4 can act as a horseradish peroxidase-like nanozyme, catalyzing the decomposition of H 2 O 2 into OH under acidic conditions for CDT. The immobilized GOx can continuously convert non-toxic glucose into gluconic acid and H 2 O 2 , and the former improves the catalytic activity of pFe 3 O 4 nanozymes by decreasing pH value. The self-supplying H 2 O 2 molecules effectively enhance the OH generation, resulting in the high antibacterial efficacy. In vitro studies demonstrate that the pFe 3 O 4 @GOx conducts well in reducing pH value and improving H 2 O 2 level for self-enhanced CDT. Moreover, the cascade catalytic reaction of pFe 3 O 4 and GOx effectively avoids strong toxicity caused by directly adding high concentrations of H 2 O 2 for CDT. It is worth mentioning that the pFe 3 O 4 @GOx performs highly efficient in vivo CDT of bacteria-infected wound via the localized long-term magnetic retention at infection site and causes minimal toxicity to normal tissues at therapeutic doses. Therefore, the developed glucose-fueled OH nanogenerators are a potential nano-antibacterial agent for the treatment of wound infections.

The bacterial resistance due to the abuse of conventional antibiotics is regarded as a major problem for bacterial-induced infections and chronic wound healing. There is an urgent need to explore alternative antimicrobial strategies and functional materials with excellent antibacterial efficacy. Herein, guanosine monophosphate (GMP) and glucose oxidase (GOD) were coordinated with copper ions to obtain a bi-functional hybrid nanoflower (Cu-GMP/GODNF) as a cascade catalyst for promoting antibacterial efficacy. Besides the efficient conversion of glucose to hydrogen peroxide, the produced gluconic acid by loading GOD can supply a compatible catalytic environment to substantially improve the peroxidase activity for generating more toxic reactive oxygen species (ROS). So, the glucose-powered cascade catalytic reaction effectively killed bacteria. Moreover, H2O2 self-supplied by glucose can reduce harmful side effects of exogenous H2O2. Meanwhile, the adhesion between the Cu-GMP/GODNF and the bacterial membrane can enhance the antibacterial efficacy. Therefore, the achieved bi-functional hybrid nanoflower exhibited high efficiency and biocompatibility for killing bacteria in diabetes-related infections.

Emerging natural-enzyme-based nanocatalytic tumor therapy depending on the high catalytic performance of natural enzymes has inspired great research interest in clinical applications. Nevertheless, the natural-enzyme-based catalytic therapy efficiency is seriously hampered by the low operational stability, poor delivery efficiency, and the short lifetime of enzymes. Herein, a bioreactor based on zeolitic imidazolate framework-8 (ZIF-8) was fabricated through one-pot embedding horseradish peroxidase (HRP) and glucose oxidase (Gox) strategy (ZIF-8@Gox/HRP) for synergistic cancer therapy. Notably, the obtained ZIF-8@Gox/HRP can efficiently consume endogenous glucose to generate gluconic acid and H2O2 for starving tumors, and subsequently, H2O2 was decomposed by encapsulated HRP to release high-toxic •OH radicals, inducing cancer cell apoptosis for oxidation therapy. Importantly, in vivo results showed that ZIF-8@Gox/HRP had an impressive tumor-suppression rate based on cascade catalytic reaction. Therefore, this work paves a new avenue to design smart enzyme-based platforms for safe and efficient cancer therapy.

The utilization of artificial multienzyme-catalyzed cascade reactions have greatly benefited biosensors. This study presents a novel nanoreactor consisting of manganese ions-doped hollow carbon nanospheres (Mn@HCNs), which can rapidly transform molecular information into an easily comprehensible colorimetric signal. The Mn@HCNs nanoreactor was utilized to detect acid phosphatase (ACP) within a range of 0.5–10 mU/mL, achieving a limit of detection (LOD) as low as 0.0851 mU/mL. The superiority of our nanoreactor over traditional ACP detection methods stems from its exceptional substrate affinity and the multi-enzymatic behaviors of Mn@HCNs. Especially, the specific POD-like activity (SA) value of Mn@HCNs was superior to most reported Fe 3 O 4 and other single-atom enzymes. Furthermore, this nanoreactor exhibited exceptional selectivity against protein and ion interference, and was furthered to detect ACP in clinical serum samples. The work may extend the development of artificial multienzyme-catalyzed cascade reactions for molecular biosensors and clinical application.

Background In vitro screening strategies based on the inhibition of α-glucosidase (GAA) activity have been widely used for the discovery of potential antidiabetic drugs, but they still face some challenges, such as poor enzyme stability, non-reusability and narrow range of applicability. To overcome these limitations, an in vitro screening method based on GAA@GOx@Cu-MOF reactor was developed in our previous study. However, the method was still not satisfactory enough in terms of construction cost, pH stability, organic solvent resistance and reusability. Thence, there is still a great need for the development of in vitro screening methods with lower cost and wider applicability. Results A colorimetric sensing strategy based on GAA/(Au–Au/IrO 2 )@Cu(PABA) cascade catalytic reactor, which constructed through simultaneous encapsulating Au–Au/IrO 2 nanozyme with glucose oxidase-mimicking and peroxidase-mimicking activities and GAA in Cu(PABA) carrier with peroxidase-mimicking activity, was innovatively developed for in vitro screening of GAA inhibitors in this work. It was found that the reactor not only exhibited excellent thermal stability, pH stability, organic solvent resistance, room temperature storage stability, and reusability, but also possessed cascade catalytic performance, with approximately 12.36-fold increased catalytic activity compared to the free system (GAA + Au–Au/IrO 2 ). Moreover, the in vitro GAA inhibitors screening method based on this reactor demonstrated considerable anti-interference performance and detection sensitivity, with a detection limit of 4.79 nM for acarbose. Meanwhile, the method owned good reliability and accuracy, and has been successfully applied to the in vitro screening of oleanolic acid derivatives as potential GAA inhibitors. Significance This method not only more effectively solved the shortcomings of poor stability, narrow scope of application, and non-reusability of natural enzymes in the classical method compared with our previous work, but also broaden the application scope of Au–Au/IrO 2 nanozyme with glucose oxidase and peroxidase mimicking activities, and Cu(PABA) carrier with peroxidase mimicking activity, which was expected to be a new generation candidate method for GAA inhibitor screening.

The diabetic chronic wound healing is extremely restricted by issues such as hyperglycemia, excessive exudate and reactive oxygen species (ROS), and bacterial infection, causing significant disability and fatality rate. Herein, the chitosan/silk fibroin nanofibers-based hierarchical 3D sponge (CSSF-P/AuGCs) with effective exudate transfer and wound microenvironment modulation are produced by integrating cascade reactor (AuGC) into sponge substrates with parallel-arranged microchannels. When applied to diabetic wounds, the uniformly parallel-arranged microchannels endow CSSF-P/AuGCs with exceptional exudate absorption capacity, keeping the wound clean and moist; additionally, AuGCs efficiently depletes glucose in wounds to generate H 2 O 2 , which is then converted into HClO via cascade catalytic reaction to eliminate bacterial infection and reduce inflammation. Experiments in vitro demonstrated that the antibacterial activity of CSSF-P/AuGCs against S. aureus and E. coli was 94.27 and 92.7 %, respectively. Experiments on animals indicated that CSSF-P/AuGC could cure wounds in 11 days, displaying superior wound-healing abilities when compared to the commercial medication Tegaderm™. This versatile CSSF-P/AuGCs dressing may be an attractive choice for expediting diabetic wound healing with little cytotoxicity, providing a novel therapeutic method for establishing a favorable pathological microenvironment for tissue repair.
