Bienzymatic Cascade Combining a Peroxygenase with an Oxidase for the Synthesis of Aromatic Aldehydes from Benzyl Alcohols

Yunjian Ma,Zongquan Li,Hao Zhang,Vincent Kam Wai Wong,Frank Hollmann,Yonghua Wang

Aromatic aldehydes are important aromatic compounds for the flavour and fragrance industry. In this study, a parallel cascade combining aryl alcohol oxidase fromPleurotus eryngii(PeAAOx) and unspecific peroxygenase from the basidiomyceteAgrocybe aegerita(AaeUPO) to convert aromatic primary alcohols into high-value aromatic aldehydes is proposed. Key influencing factors in the process of enzyme cascade catalysis, such as enzyme dosage, pH and temperature, were investigated. The universality ofPeAAOx coupled withAaeUPO cascade catalysis for the synthesis of aromatic aldehyde flavour compounds from aromatic primary alcohols was evaluated. In a partially optimised system (comprising 30 μMPeAAOx, 2 μMAaeUPO at pH 7 and 40 °C) up to 84% conversion of 50 mM veratryl alcohol into veratryl aldehyde was achieved in a self-sufficient aerobic reaction. Promising turnover numbers of 2800 and 21,000 forPeAAOx andAaeUPO, respectively, point towards practical applicability. Keywords:aromatic aldehydes;flavour compounds;cascade catalysis;PeAAOx;AaeUPO

Interface Engineering p-n Heterostructured Core–Shell Mesoporous Particles for Cascade Catalysis Promoted Gas Sensing

Lingxiao Xue,Jiahao Cui,Ruiying Li,Wenhe Xie,Keyu Chen,Hongxiu Yu,Limin Wu,Tianjun Ni,Qin Yue,Yonghui Deng

Cells have greatly inspired advancements in chemical processes, including leveraging the idea of cascade catalysis to drive thermodynamically unfavorable reactions and mimicking the compartmentalized architecture to design novel nanostructures. Here, a single-particle cascade catalysis promoted gas sensing platform is inspired to be designed (denoted as CoSnO 3 @mCeO 2 ) by positionally assembling n-type mesoporous CeO 2 catalytic shell on p-type CoSnO 3 gas sensitive core. Uniform CoSn(OH) 6 @mCe(OH) x core–shell particles with tailored mesostructures, tunable large mesopores, and adjustable shell thicknesses are first constructed. After thermal treatment, CoSnO 3 @mCeO 2 particles are obtained, which serve as cascade catalysis enhanced sensitive layer for fabricating gas sensors with independent catalytic and sensing control. As a proof-of-concept, the CoSnO 3 @mCeO 2 gas sensors exhibit nearly three times higher acetone sensitivity ( R g /R a = 26.81–50 ppm) than individual CoSnO 3 sensors with an ultralow limit of detection of 5.22 ppb. The enhanced sensitivity is achieved through a tandem catalytic reforming-oxidation sensing procedure, which begins with the primary catalytic reforming of acetone to acetic acid in the mCeO 2 shell, followed by the secondary sensing reaction of acetic acid in the CoSnO 3 core. The design concept of cascade catalysis promotes gas sensor can serve as a paradigm for developing single-particle functional nanodevices for various applications.

Dual enzyme-mimicking bimetallic MOF for selective SERS detection of L-DOPA in human serum based on cascade catalytic reaction

Shuojiang Liu,Ying Chen,Xingliang Cheng,Gongke Li,Yuling Hu

Surface-enhanced Raman scattering (SERS) in conjunction with nanozymes has emerged as a powerful technique for bioanalysis due to their signal conversion and amplification effect. However, clinical applications continue to encounter challenges such as inadequate selectivity. Herein, we proposed a novel approach for the selective SERS detection of L-DOPA in human serum with dual enzyme-mimicking MOF-919(Fe–Cu). This material exhibits both catechol oxidase-like and peroxidase-like activities, enabling it to catalyze catecholic compounds to o -quinones and subsequently oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) molecules into SERS-active ox-TMB. Ultrasonic-assisted synthesis ensures uniform and dense loading of AgNPs within MOF-919(Fe–Cu), thereby enhancing SERS activity. A sensitive and selective method for SERS detection of L-DOPA was developed. The catechol oxidase-like activity of MOF-919(Fe–Cu) provides ideal selectivity and anti-interference to L-DOPA, while the cascade catalytic system transforms weak SERS signals of L-DOPA into amplified SERS signals of ox-TMB. The mechanism of dual enzyme-mimicking activity was further discussed. The detection process was optimized to achieve high sensitivity towards L-DOPA with a linear range 0.5–100 μmol/L and low limit of detection (LOD) of 0.16 μmol/L. Furthermore, it has been applied in serum sample of Parkinson's patients, demonstrating excellent recovery and low relative error compared to HPLC-UV. This approach offers an economical, efficient and accurate strategy for the detection of L-DOPA and provides valuable insights for the integration of selective multifunctional nanozyme and sensitive SERS detection method.

Enhanced catalytic activity and stability of lactate dehydrogenase for cascade catalysis of D-PLA by rational design

Yufeng Zhou,Xiaolong Sun,Jiahuan Hu,Yingjie Miao,Xiangyu Zi,Xi Luo,Yongqian Fu

Serving as a vital medical intermediate and an environmentally-friendly preservative, D-PLA exhibits substantial potential across various industries. In this report, the urgent need for efficient production motivated us to achieve the rational design of lactate dehydrogenase and enhance catalytic efficiency . Surprisingly, the enzymatic properties revealed that a mutant enzyme , Lr LDH T247I/D249A/F306W/A214Y ( Lr LDH-M 1 ), had a viable catalytic advantage. It demonstrated a 3.3-fold increase in specific enzyme activity and approximately a 2.08-fold improvement of K cat . Correspondingly, molecular docking analysis provided a supporting explanation for the lower K m and higher K cat /K m of the mutant enzyme. Thermostability analysis exhibited increased half-lives and the deactivation rate constants decreased at different temperatures (1.47–2.26-fold). In addition, the mutant showed excellent resistance abilities in harsh environments, particularly under acidic conditions. Then, a two-bacterium ( E. coli /pET28a- lrldh -M 1 and E. coli /pET28a- ladd ) coupled catalytic system was developed and realized a significant conversion rate (77.7%) of D-phenyllactic acid, using 10 g/L L-phenylalanine as the substrate in a two-step cascade reaction.

Self-Cascade of the Cu/Ce-MOF-808 Nanozyme for One-Step Colorimetric Sensing

Yuan Qin,Huimei Liang,Jia Wu,Zheyang Chi,Shulin Zhao,Fanggui Ye

Natural enzyme cascades, including the same and different kinds of enzymes, have been widely used in biosensing. However, it is still a challenge for nanozymes, the mimics of natural enzymes, to achieve biosensing with different types of enzyme-like activities based on cascade reactions, such as hydrolase and oxidoreductase. Considering the characteristics of the multienzyme-like activity of some nanozymes, as a proof of concept, this study demonstrates a Cu/Ce-MOF-808 nanozyme as a self-cascade catalyst with hydrolase-like and catechol oxidase-like activities. Taking organophosphorus pesticides as an example, we constructed a self-cascade catalytic reaction for the one-step colorimetric detection of isocarbophos. Specifically, the hydrolase-like activity of Cu/Ce-MOF-808 can hydrolyze isocarbophos to produce colorless isopropyl salicylate, which is further oxidized by the catechol oxidase-like activity of Cu/Ce-MOF-808 and complexed with 4-aminoantipyrine (4-AAP) to produce red quinoneimine under neutral conditions with illumination. Thus, a colorimetric strategy for sensitive isocarbophos analysis is developed, with a linear range of 0.90–400.00 μg·mL–1 and a detection limit of 0.42 μg·mL–1. This research presents a new approach to the design and synthesis of nanozymes and has great application potential in the field of nanozyme cascade biosensing.

CunO/Au heterostructure dendrimer anchored on Cu foam as dual functional catalytic nanozyme for glucose sensing by enzyme mimic cascade reaction

Gao Tiemei,Lian Huiting,Cao Xuegong,Liu Bin,Wei Xiaofeng

Multifunctional catalytic performance plays a crucial role in bio-applications through the diversity and durability of artificial nanozymes. An effective synergy with sufficient accessible active sites and high specific surface area is a challenge for composite catalysts, especially to avoid uncontrollable aggregation and structural instability. Here, we fabricated a Cu n O/Au heterostructure dendrimer on copper foam (Cu n O/Au HD/CF) as dual functional catalytic nanozyme to achieve enzyme mimic cascade reactions for efficient colorimetric analysis. A highly porous CF skeleton–based CuO nanowire array (CuO NWA) with a large specific surface area supported an efficient load capacity to assemble sufficient Cu n O/Au HD by electrodeposition. The bimetallic Au-Cu nanozyme successfully achieved an oxidase-like and peroxidase-like cascade catalysis by a target-responsive sensing mechanism. Due to the confirmed catalytic performance of selectivity, anti-interference ability, and reproducibility, a Cu n O/Au HD/CF-based quantitative analytical method was developed for glucose detection with a wide linear range and considerable detection limit of 8.4 μM. The robust nonenzymatic catalytic strategy for colorimetric detection not only confirmed the dual functional catalytic activity of Cu n O/Au HD/CF, but also showed great potential for applications in clinical diagnostics and biochemical analysis. Inbuilt self-cascade catalysis of the bimetal-confined structural nanozyme CoNi@CNTs-N/GO for increased bienzymatic activity and H2O2-free smartphone-based visual assay of total antioxidant capacity in foods

Hailong Gong,Xiaofeng Hou,Binrong Li,Siyu Shen,Chen Wang,Qinqin Ma,Tingting Liu,Xuedong Wang

The level of total antioxidant capacity (TAC) reflects the overall ability of food to resist oxidative damage, maintain quality and nutritional stability. In this work, we pioneered a Co Ni alloy-confined N-doped carbon nanozyme (CoNi@CNT-N/GO) with a self-cascade catalysis. Compared to other nanozymes, the synergistic effect of OXD and POD realized self-sustained generation of H 2 O 2 , eliminating the need of exogenous addition, and further decomposition of H 2 O 2 into ·OH and oxidizes colorless 3,3′,5,5′- tetramethylbenzidine (TMB) to blue oxTMB as an efficient catalyst. The integration of multiple components and the built-in unique mechanism enhance bienzymatic activity. A “Thing Identify” APP was utilized to construct a smartphone-based visualization platform, demonstrating satisfactory linearity (0.01–1.2 mM) and low detection limit (3.3 μM) in TAC detection of real-world foods. This platform yielded data comparable to those from commercially colorimetric kits. Overall, it proposes a novel idea for engineering multi-functional and non-additional H 2 O 2 nanozymes in on-site food-quality monitoring.

Proficient photothermal-enhanced cascade catalytic nanoplatform for multi-responsive release and synergistic tumor therapy

Wenquan Zhu,Li Fan,Tingxing Hou,Tao Xu,Jiahuan Liu,Defu Cai,Chang Liu,Ke Jiang,Yufang Wang,Wenli Tao,Lipin Wang,Wenbao Wang,Likun Liu,Cuiyan Han

The chemotherapy effect is generally affected by the premature release of nanoplatform and multi-drug resistance. In light of the advantages of photothermal therapy (PTT) and cascade catalytic therapy, we developed a multi-sensitive drug delivery system (MHC-Au-P) by grafting the gold NPs onto the opening of mesoporous hollow carbon nanoparticles (MHC) using gold sulfur bonds. MHC had excellent photothermal conversion efficiency and GSH consumption function, serving as a carrier for highly loading doxorubicin (DOX). The gold NPs were used due to their diverse functions, including smart switches to block the release of harmful substances, PTT agents to enhance the thermogenic effect of MHC, and nanozymes with GOx-like activity. When physical conditions are normal, gold-sulfur bond, which serves as the link between MHC carrier and Au-NPs, was stable, but the stability will decrease when glutathione (GSH) concentration in tumor cytoplasm was high. The DOX loaded D@MHC-Au-P can be rapidly decomposed into DOX@MHC and gold NPs at high GSH concentration after reaching the tumor microenvironment. The separated Au-NPs was capable of catalyzing the oxidation of glucose to H 2 O 2 and glucuronic acid, providing raw materials and optimizing conditions for the POD-like enzyme catalysis of MHC. Under near-infrared irradiation, the release rate of drugs was further accelerated, GSH consumption rate, the catalytic rate of Au-NPs and MHC were further improved. The synergistic effect of D@MHC-Au-P for PTT-enhanced cascade catalytic therapy and chemotherapy was realized in vivo and in vitro with a combination index of 0.62. Therefore, D@MHC-Au-P can be used as a smart nanocarrier for photothermal-enhanced synergistic cascade catalysis and chemotherapy.

Efficient nitrate electroreduction to ammonia via synergistic cascade catalysis at Cu/Fe2O3 hetero-interfaces

Xiang Zhang,Weihang Li,Jin Zhang,Haoshen Zhou,Miao Zhong

Electrochemical nitrate (NO 3 − ) reduction offers a promising route for ammonia (NH 3 ) synthesis from industrial wastewater using renewable energy. However, achieving selective and active NO 3 − to NH 3 conversion at low potentials remains challenging due to complex multi-electron transfer processes and competing reactions. Herein, we tackle this challenge by developing a cascade catalysis approach using synergistic active sites at Cu-Fe 2 O 3 interfaces, significantly reducing the NO 3 − to NH 3 at a low onset potential to about +0.4 V RHE . Specifically, Cu optimizes *NO 3 adsorption, facilitating NO 3 − to nitrite (NO 2 − ) conversion, while adjacent Fe species in Fe 2 O 3 promote the subsequent NO 2 − reduction to NH 3 with favorable *NO 2 adsorption. Electrochemical operating experiments, in situ Raman spectroscopy, and in situ infrared spectroscopy consolidate this improved onset potential and reduction kinetics via cascade catalysis. An NH 3 partial current density of ~423 mA cm −2 and an NH 3 Faradaic efficiency (FE NH3 ) of 99.4% were achieved at −0.6 V RHE , with a maximum NH 3 production rate of 2.71 mmol h −1 cm −2 at −0.8 V RHE . Remarkably, the half-cell energy efficiency exceeded 35% at −0.27 V RHE (80% i R corrected), maintaining an FE NH3 above 90% across a wide range of NO 3 − concentrations (0.05−1 mol L −1 ). Using 15 N isotopic tracing, we confirmed NO 3 − as the sole nitrogen source and attained a 98% NO 3 − removal efficiency. The catalyst exhibit stability over 106-h of continuous operation without noticeable degradation. This work highlights distinctive active sites in Cu-Fe 2 O 3 for promoting the cascade NO 3 − to NO 2 − and NO 2 − to NH 3 electrolysis at industrial relevant current densities.

Metal–Organic Framework-Based Cascade Catalysis-Enabled Fluorescent ELISA with Higher Enzymatic Stability for Zearalenone Detection in Maize

Chaochao Chen,Liang Luo,Jianzhong Shen,Zhanhui Wang,Yantong Pan

The enzyme-linked immunosorbent assay (ELISA) stands as one of the most frequently employed rapid detection techniques for both chemical and biological contaminants. Horseradish peroxidase (HRP) and hydrogen peroxide (H2O2), serving as signal generators, play indispensable roles throughout the entire ELISA process. However, HRP and H2O2 exhibit high sensitivity to elevated temperatures, hindering the broader utilization and transportation of ELISA. Hereby, being inspired by the cascade catalysis of glucose by glucose oxidase (GOx) and HRP as well as the advantageous properties of metal–organic frameworks (MOFs), we developed a cascade catalysis-based competitive fluorescent ELISA (CCF-ELISA) for mycotoxin detection in maize using zearalenone (ZEN) as a model. The results revealed the remarkable protective effects of MOFs on enzymatic activities. Based on upon characteristics, the built CCF-ELISA enabled the detection of ZEN with a half-maximal inhibitory concentration of 0.54 ng/mL and allowed a detection range from 0.19 to 1.51 ng/mL. When applied to naturally contaminated maize samples, the outcomes obtained from the CCF-ELISA showed a high level of comparability with high-performance liquid chromatography–tandem mass spectrometry, demonstrating the promising potential of the developed CCF-ELISA in the rapid detection for food security.

Polylysine-modified MXene nanosheets with highly loaded glucose oxidase as cascade nanoreactor for glucose decomposition and electrochemical sensing

Minying Wu,Qian Zhang,Yuanyuan Fang,Chao Deng,Fangzhou Zhou,Yi Zhang,Xingdong Wang,Yi Tang,Yajun Wang

Two-dimensional (2D) nanoreactors with cascade catalytic activity for glucose oxidation and hydrogen peroxide decomposition are prepared via immobilizing glucose oxidase (GOx) on Ti 3 C 2 MXene nanosheets. Amino-rich polypeptide, poly- l -lysine (PLL), is applied to modify the ultra-thin Ti 3 C 2 MXene nanosheets with a compatible surface for GOx immobilization. The PLL-modified Ti 3 C 2 nanosheets possess a positively charged surface and show an excellent GOx loading capacity as high as 50 wt% of the Ti 3 C 2 nanosheets. The physically adsorbed enzymes are then cross-linked with the amine groups in the PLL chains to form a robust GOx-PLL network covered on the MXene nanosheets. The GOx-conjugated Ti 3 C 2 -PLL (Ti 3 C 2 -PLL-GOx) nanosheets showed superior enzymatic activities than the activities of GOx immobilized on an inert porous silica substrate, largely because that the Ti 3 C 2 nanosheets can catalyze the decomposition of the toxic intermediate H 2 O 2 generated from the glucose oxidation. Given the excellent electrical conductivity of Ti 3 C 2 MXene, the Ti 3 C 2 -PLL-GOx nanosheets are further deposited on glassy carbon electrode to construct a high-performance biosensor with a glucose detection limit of 2.6 μM.

Engineering Single-Atom Iron Nanozymes with Radiation-Enhanced Self-Cascade Catalysis and Self-Supplied H2O2 for Radio-enzymatic Therapy

Xianyu Zhu,Jiabin Wu,Ruixue Liu,Huandong Xiang,Wenqi Zhang,Qingchao Chang,Shanshan Wang,Rui Jiang,Feng Zhao,Qiqiang Li,Liang Huang,Liang Yan,Yuliang Zhao

Single-atom nanozymes (SAzymes), with individually isolated metal atom as active sites, have shown tremendous potential as enzyme-based drugs for enzymatic therapy. However, using SAzymes in tumor theranostics remains challenging because of deficient enzymatic activity and insufficient endogenous H2O2. We develop an external-field-enhanced catalysis by an atom-level engineered FeN4-centered nanozyme (FeN4-SAzyme) for radio-enzymatic therapy. This FeN4-SAzyme exhibits peroxidase-like activity capable of catalyzing H2O2 into hydroxyl radicals and converting single-site FeII species to FeIII for subsequent glutathione oxidase-like activity. Density functional theory calculations are used to rationalize the origin of the single-site self-cascade enzymatic activity. Importantly, using X-rays can improve the overall single-site cascade enzymatic reaction process via promoting the conversion frequency of FeII/FeIII. As a H2O2 producer, natural glucose oxidase is further decorated onto the surface of FeN4-SAzyme to yield the final construct GOD@FeN4-SAzyme. The resulting GOD@FeN4-SAzyme not only supplies in situ H2O2 to continuously produce highly toxic hydroxyl radicals but also induces the localized deposition of radiation dose, subsequently inducing intensive apoptosis and ferroptosis in vitro. Such a synergistic effect of radiotherapy and self-cascade enzymatic therapy allows for improved tumor growth inhibition with minimal side effects in vivo. Collectively, this work demonstrates the introduction of external fields to enhance enzyme-like performance of nanozymes without changing their properties and highlights a robust therapeutic capable of self-supplying H2O2 and amplifying self-cascade reactions to address the limitations of enzymatic treatment.
