Magnetic field-assisted cascade effects of improving the quality of gels-based meat products: A mechanism from myofibrillar protein gelation

Jingwen Wang,Qiyuan Lu,Junming Gong,Fei Gao,Xinglian Xu,Huhu Wang

Physics-assisted processing technologies have huge potential in the meat processing industry. By modeling two essential procedures (pickling and preheating) of gels-based meat products, this work investigated the cascade effects of a new physical technology (magnetic field) on the conformational structures and gel properties of myofibrillar proteins (MPs). Samples were subjected with four magnetic field (MF)-assisted treatments (group A, both processes without MF; group B, pickling without MF combining with preheating with 4.5 mT MF; group C, pickling with 3.0 mT MF combining with preheating without MF; group D, pickling with 3.0 mT MF combining with preheating with 4.5 mT MF). The result showed that MF-assisted treatments significantly improved water holding capacity (WHC) of MP gels compared with group A (46.9%), reaching the maximum value of 52.1% in group D.   According to the low-field nuclear magnetic results, group D decreased the percentages of P 22 (6.97%) and increased the percentages of P 21 (93%), which showed that water molecules were more tightly bound to each other. Meanwhile, the unfolding of α-helix and the formation of random coil of MF-assisted treatments resulted in more exposure of internal groups, leading to the formation of a dense network. These findings would provide new insights to improve the quality of gels-based meat products via the MF.

Nose-to-brain delivery of gold nanozyme with cascade effect for bacterial meningitis therapy

Deng Shu-Yue,Zhou Xin-Yu,Zou Xiao-Peng,Tang Fang,Yang Dong,Sun Cai-Xia,Luo Jun,Ge Xing,Zhu Jia-Ying,Fang Tian-Ye,Yue Cai-Feng,Ju Yan-Min,Dai Jian-Jun

The presence of the blood–brain barrier limits the drug concentration in the brain, while low concentrations of antibiotics make it difficult to kill infecting bacteria and tends to induce drug resistance, making the clinical treatment of bacterial meningitis challenging. Herein, a nose-to-brain delivery strategy of small-sized nanozyme has been fabricated for combating bacterial meningitis, to overcome the low drug concentration and drug resistance. This strategy was achieved by a protein-supported Au nanozyme (ANZ). With a particle size of less than 10 nm, it possesses both glucose oxidase-like and peroxidase-like activities and can generate large amounts of reactive oxygen species through a cascade effect without the addition of external H 2 O 2 . Benefiting from the cascade catalytic amplification effect generated by its dual enzyme-like activities, ANZ shows significant broad-spectrum antibacterial activity without inducing bacterial resistance in vitro. Notably, small-sized ANZ exhibits higher brain entry efficiency and greater accumulation after intranasal administration compared to oral or intravenous administration. In a mouse model of bacterial meningitis, the mice treated with ANZ had lower bacterial loads in the brain and higher survival and clinical behavior scores compared to the classical antibiotic ceftriaxone. Additionally, the meningitis mice exhibited undamaged cognitive and behavioral abilities, indicating the excellent biocompatibility of ANZ. The above results demonstrate that nose-to-brain delivery of ANZ exhibits high intracerebral accumulation, strong antibacterial efficacy and does not lead to bacterial resistance. It holds broad prospects for the treatment of bacterial meningitis. Graphical abstract

Antibiotic-induced multi-trophic effects and their cascades in a sub-tropical freshwater ecosystem

Kai-Sheng Yao,Pei-Liang Zhou,Li-Juan Tan,Hua-Qiao Yang,Lu-Kai Qiao,Fang-Zhou Gao,You-Sheng Liu,Feng-Jiao Peng,Guang-Guo Ying,Paul J. Van den Brink

Antibiotics are commonly detected in aquatic ecosystems worldwide due to their extensive use and excretion by humans and animals, posing potential risks to the health of these ecosystems. This study aimed to assess the ecological effects of the antibiotic ciprofloxacin on both structural (microbes, phytoplankton, zooplankton, and macroinvertebrates) and functional endpoints of a sub-tropical freshwater ecosystem. Ciprofloxacin was applied at concentrations of 0, 0.5, 5, 50, and 500 µg/L for 21 consecutive days in outdoor mesocosms, followed by a five-week recovery period. Ciprofloxacin significantly affected the structure of microbial, phytoplankton, and zooplankton communities, with calculated NOECs of 5, 0.5, and 5 µg/L, respectively. Notably, the microbial community composition, particularly taxa within the phyla Cyanobacteria and Bacteroidetes, exhibited marked shifts. Among phytoplankton, the filamentous cyanobacterium Cylindrospermopsis sp. exhibited the largest negative response to ciprofloxacin, while Microcystis sp. displayed the largest increase in abundance. Ciprofloxacin exposure also indirectly led to significant increases in zooplankton populations belonging to Cladocera, Copepoda, and Rotifera. Significant effects of ciprofloxacin on physicochemical parameters related to carbon and nitrogen cycling were observed. Structural equation models revealed that ciprofloxacin induced both direct and indirect effects across multiple trophic levels through cascading effects, further impacting ecosystem-level endpoints. Overall, this study provides an understanding of the potential ecological risks posed by antibiotic pollution on freshwater ecosystems.

Assessing triclosan-induced direct and multi-trophic cascading effects in a sub-tropical freshwater ecosystem

Kai-Sheng Yao,Pei-Liang Zhou,Hua-Qiao Yang,Li-Juan Tan,Lu-Kai Qiao,Qing-Hua Zhao,Feng-Jiao Peng,Guang-Guo Ying,Paul J. Van den Brink

Triclosan, a synthetic broad-spectrum antimicrobial agent, is commonly used in personal care products, household goods, and industrial items. Its extensive use and subsequent release have led to frequent detections in aquatic ecosystems worldwide, potentially harming ecosystem health. Therefore, an outdoor mesocosm experiment was conducted to assess the effects of triclosan on community structure (microbes, phytoplankton, zooplankton, and macroinvertebrates) and ecosystem functioning in freshwater ecosystems under sub-tropical conditions. Triclosan was applied daily in pulses at nominal concentrations of 0.05, 0.5, 5, and 50 µg/L over a three-week treatment period, followed by a five-week recovery period. Triclosan significantly affected the structure of microbial (NOEC = 0.5 µg/L), phytoplankton (NOEC = 5 µg/L), and zooplankton (NOEC = 5 µg/L) communities. In contrast, no significant effects were observed on the macroinvertebrate community in response to triclosan exposure. Within the microbial community, the phyla Cyanobacteria and Bacteroidetes showed consistent responses to triclosan exposure, with taxa in the Cyanobiaceae family being the most adversely affected. Among phytoplankton taxa, the cyanobacteria Cylindrospermopsis sp. had the lowest consistent NOEC (<0.05 µg/L), while the lowest NOEC (0.5 µg/L) among zooplankton taxa was observed for the copepods nauplii and Mesocyclops leuckarti . Piecewise structural equation modelling revealed that triclosan exposure resulted in significant direct and indirect effects across multiple trophic levels, ultimately disrupting ecosystem functioning related to carbon and nitrogen cycling. Hence, our study highlights the threat posed by environmentally relevant concentrations of triclosan to both community structure and ecosystem functioning in sub-tropical freshwater ecosystems.

ECM-engineered electrospun fibers with an immune cascade effect for inhibiting tissue fibrosis

Ming Qian,Shun Li,Kun Xi,Jincheng Tang,Xiaofeng Shen,Yong Liu,Ran Guo,Nannan Zhang,Yong Gu,Yun Xu,Wenguo Cui,Liang Chen

Tissue regeneration/fibrosis after injury is intricately regulated by the immune cascade reaction and extracellular matrix (ECM). Dysregulated cascade signal could jeopardize tissue homeostasis leading to fibrosis. Bioactive scaffolds mimicking natural ECM microstructure and chemistry could regulate the cascade reaction to achieve tissue regeneration. The current study constructed an ECM-engineered micro/nanofibrous scaffold using self-assembled nanofibrous collagen and decorin (DCN)-loaded microfibers to regulate the immune cascade reaction. The ECM-engineered scaffold promoted anti-inflammatory and pro-regenerative effects, M2 polarization of macrophages, by nanofibrous collagen. The ECM-engineered scaffold could release DCN to inhibit inflammation-associated fibrous angiogenesis. Yet, to prevent excessive M2 activity leading to tissue fibrosis, controlled release of DCN was expected to elicit M1 activity and achieve M1/M2 balance in the repair process. Regulated cascade reaction guided favorable crosstalk between macrophages, endothelial cells and fibroblasts by proximity. Additionally, decorin could also antagonize TGF-β1 via TGF-β/Smad3 pathway to suppress fibrotic activity of fibroblasts. Hence, ECM-engineered scaffolds could exert effective regulation of the immune cascade reaction by microstructure and DCN release and achieve the balance between tissue fibrosis and regeneration. Statement of Significance With the incidence of up to 74.6%, failed back surgery syndrome (FBSS) has been a lingering issue in spine surgery, which poses a heavy socio-economic burden to society. Epidural fibrosis is believed to be responsible for the onset of FBSS. Current biomaterial-based strategies treating epidural fibrosis mainly rely on physical barriers and unidirectional suppression of inflammation. Regulation of the immune cascade reaction for inhibiting fibrosis has not been widely studied. Based on the simultaneous regulation of M1/M2 polarization and intercellular crosstalk, the ECM-engineered micro/nanofibrous scaffolds constructed in the current study could exert an immune cascade effect to coordinate tissue regeneration and inhibit fibrosis. This finding makes a significant contribution in the development of a treatment for epidural fibrosis and FBSS.

Metal-Organic Framework-Based Nanoheater with Photo-Triggered Cascade Effects for On-Demand Suppression of Cellular Thermoresistance and Synergistic Cancer Therapy

Yuanyuan Zhu,Yanyan Huang,Tian-Hao Yan,Jialuo Li,Yongming Li,Hannah F. Drake,Huifei Zhong,Yulong Jin,Rui Zhao,Hong-Cai Zhou

Nanomedicine with stable light-heat conversion and spatiotemporally controllable drug activation is crucial for the success of photothermal therapy (PTT). Herein, a metal–organic framework (MOF)-based nanoheater with light-triggered multi-responsiveness is engineered to in-situ and on-demand sensitize cancer cells to local hyperthermia. Well-dispersed platinum nanoparticles synthesized inside nanospaces of the MOF are employed as the near-infrared (NIR)-harvesting unit with stable and high light–heat conversion performance. A conformation switchable polymer shell is constructed as a secondary light-responding unit to gate the targeted activation of a molecular inhibitor against thermoresistance. By cascade transformation of light stimuli to downstream signals, the nanoheater enables inhibitor release to go with local heating at the same time restricted in lesion sites to maximize efficacy and minimize systemic toxicity. The efficient photothermal conversion and the blockage of cellular heat-protective pathways provide a dual-mode of action which selectively sensitizes cancer cells to hyperthermia in a spatiotemporally controlled manner. With NIR as the remote switch, the MOF-based nanosystem demonstrates localized and boosted PTT efficacy against cancer both in vitro and in vivo. These results present nanosized MOFs as tailorable and versatile platforms for synergistic and precise cancer therapy.
