Simple theranostics nanoagent for precision suppression of tumor growth and Metastasis: A traditional fermented product having a novel functional breakthrough

Pengmin Liu,Rong Dai,Juan Li,Lin Chen,Qi Liu,Yuhang Wang,Shichao Meng,Binyue Zhang,Jinxuan Li,Weiwei Kang,Ziliang Zheng,Ruiping Zhang

Biomass-based nanoagents constitute a focal point in the clinical development of tumor nanotheranostics, characterized by their high biocompatibility, biodegradability, controllability, and sustainability. However, achieving satisfactory nanoagents with biological safety and high theranostic efficacy for clinical translation remains a significant challenge. Herein, based on the design concept of medicine food homology, we developed a simple, mass-produced theranostics nanoagent (HSAVMn@CM NA) using Shanxi aged vinegar (SAV) as the raw material, enabling second near-infrared fluorescence/photoacoustic (NIR-II FL/PA) imaging-guided photo/catalytic/chemodynamic synergistic tumor therapy. A novel strategy involving the hydrolysis of SAV (HSAV) via the hydrolysis-acidification method has been developed to enhance NIR-II FL imaging performance. The HSAVMn@CM NAs have improved tumor-targeted efficiency through biomimetic modification of tumor membranes. Guided by NIR-II FL/PA dual-modal imaging, the HSAVMn@CM NAs achieve targeted photothermal/photodynamic synergistic phototherapy. Notably, the employment of a single laser source for simultaneous photothermal/photodynamic therapy has led to an enhancement in the efficacy of phototherapy. Moreover, the HSAVMn@CM NAs effectively chelated Zn 2+ ions within the tumor microenvironment, to inhibit matrix metalloproteinase 2/9 (MMP 2/9) activity, thereby synergistically suppressing primary tumor growth and metastasis. Taken together, this research provides a novel design strategy for biomass-based nanoagents with potential clinical translation in precise tumor theranostics.

Magnetic Targeting Enhanced Theranostic Strategy Based on Multimodal Imaging for Selective Ablation of Cancer

Zhiwei Li,Shengnan Yin,Liang Cheng,Kai Yang,Yonggang Li,Zhuang Liu

The booming development of nanomedicine offers great opportunities for cancer diagnostics and therapeutics. Herein, a magnetic targeting-enhanced cancer theranostic strategy using a multifunctional magnetic-plasmonic nano-agent is developed, and a highly effective in vivo tumor photothermal therapy, which is carefully planed based on magnetic resonance (MR)/photoacoustic (PA) multimodal imaging, is realized. By applying an external magnetic field (MF) focused on the targeted tumor, a magnetic targeting mediated enhanced permeability and retention (MT-EPR) effect is observed. While MR scanning provides tumor localization and reveals time-dependent tumor homing of nanoparticles for therapeutic planning, photoacoustic imaging with higher spatial resolution allows noninvasive fine tumor margin delineation and vivid visualization of three dimensional distributions of theranostic nanoparticles inside the tumor. Utilizing the near-infrared (NIR) plasmonic absorbance of those nanoparticles, selective photothermal tumor ablation, whose efficacy is predicted by real-time infrared thermal imaging intra-therapeutically, is carried out and then monitored by MR imaging for post-treatment prognosis. Overall, this study illustrates the concept of imaging-guided MF-targeted photothermal therapy based on a multifunctional nano-agent, aiming at optimizing therapeutic planning to achieve the most efficient cancer therapy.

A Real-Time Cell Death Self-Reporting Theranostic Agent for Dynamic Optimization of Photodynamic Therapy

Wei Bian,Qiyue Wang,Cui He,Pan Tao,Juanjuan Zheng,Yulu Zhang,Jing Li,Fangyuan Li,Hongyan Jia,Daishun Ling

The therapeutic efficiency of photodynamic therapy (PDT) hinges on the drug-light interval (DLI), yet conventional approaches relying on photosensitizer accumulation often lead to suboptimal irradiation and adverse side effects. Here, a real-time cell death self-reporting photodynamic theranostic nanoagent (CDPN) is presented that dynamically monitors extracellular potassium ion ([K⁺] ex ) fluctuations as direct indicators of tumor cell death. By exploiting [K⁺] ex dyshomeostasis associated with apoptosis and necrosis, CDPN combines a photosensitizer and a potassium-sensitive fluorophore within mesoporous silica nanoparticles, encapsulated by a K⁺-selective membrane for enhanced specificity. In vitro and in vivo studies validate that [K⁺] ex dynamics closely correlate with cell death, enabling precise evaluation of PDT efficacy and data-driven optimization of the DLI. Using a breast cancer model, CDPN-guided adjustments identify optimized DLI conditions, achieving significantly improved therapeutic outcomes. This study introduces a new paradigm for PDT, establishing a real-time, adaptable strategy for guiding treatment parameters and advancing precision oncology.

MIL-101 (Fe) @Ag Rapid Synergistic Antimicrobial and Biosafety Evaluation of Nanomaterials

Xi Li,Huiying Zheng,Jiehan Chen,Mengyuan Xu,Yan Bai,Tiantian Liu

Metal-organic frameworks (MOFs), which have become popular in recent years as excellent carriers of drugs and biomimetic materials, have provided new research ideas for fighting pathogenic bacterial infections. Although various antimicrobial metal ions can be added to MOFs with physical methods, such as impregnation, to inhibit bacterial multiplication, this is inefficient and has many problems, such as an uneven distribution of antimicrobial ions in the MOF and the need for the simultaneous addition of large doses of metal ions. Here, we report on the use of MIL-101(Fe)@Ag with efficient metal-ion release and strong antimicrobial efficiency for co-sterilization. Fe-based MIL-101(Fe) was synthesized, and then Ag+was uniformly introduced into the MOF by the substitution of Ag+for Fe3+. Scanning electron microscopy, powder X-ray diffraction (PXRD) Fourier transform infrared spectroscopy, and thermogravimetric analysis were used to investigate the synthesized MIL-101(Fe)@Ag. The characteristic peaks of MIL-101(Fe) and silver ions could be clearly seen in the PXRD pattern. Comparing the diffraction peaks of the simulated PXRD patterns clearly showed that MIL-101(Fe) was successfully constructed and silver ions were successfully loaded into MIL-101(Fe) to synthesize an MOF with a bimetallic structure, that is, the target product MIL-101(Fe)@Ag. The antibacterial mechanism of the MOF material was also investigated. MIL-101(Fe)@Ag exhibited low cytotoxicity, so it has potential applications in the biological field. Overall, MIL-101(Fe)@Ag is an easily fabricated structurally engineered nanocomposite with broad-spectrum bactericidal activity.Keywords:metal-organic framework;Ag nanoparticles;hybrid nanoagents;antibacterial activity

GSH-responsive sequential mitochondria-targeting nanoagents for photothermal-enhanced chemodynamic therapy

Huaming He,Fang Zhao,Wukun Zhong,Yuling Yang,Yuhong Lin,Yihang Ding,Jiajia Yang,Chunhua Lu,Xiankun Tu

Mitochondria-targeting nanoparticles have proven to be promising platforms for cancer therapy. However, precise mitochondria targeting in the complex tumor environment is still challenging and requires more ingenious design. Herein, we developed the glutathione (GSH)-responsive nanoagents to target mitochondria in tumor cells precisely for photothermal-enhanced chemodynamic therapy (CDT). The nanoagents were designed by preparing the cubic shape of CuFeS2 nanoparticles that sequentially modified with mitochondrial-targeting peptide (KLAK) and integrin-targeting peptide (RGD), which KLAK and RGD were covalently conjugated via a disulfide bond. After being endocytosed into integrin-overexpressed tumor cells, disulfide bond was broken and RGD was separated from the nanoagents in the presence of overexpressed GSH. Then, the exposure of KLAK on the surface further aid the rustling nanoagents to target mitochondria. Under NIR laser irradiation, the surrounding temperature of the nanoagents would rise due to their excellent photothermal effect, resulting in the improvement of the Fenton reaction from the CuFeS2. It was well demonstrated that this photothermal-enhanced chemodynamic therapy nanoplatform could effectively damage the mitochondrial function and further induce the apoptosis of tumor cells. As a therapeutic platform with both tumor/mitochondria-targeting and PTT-enhanced CDT, our study has provided a new path in the field of cancer therapy.

Chemotherapy-Sensitized In Situ Vaccination for Malignant Osteosarcoma Enabled by Bioinspired Calcium Phosphonate Nanoagents

Yangyun Wang,Yanxian Wu,Liubing Li,Chunjie Ma,Shaodian Zhang,Subin Lin,Leshuai W. Zhang,Yong Wang,Mingyuan Gao

How to effectively treat malignant osteosarcoma remains clinically challenging. Programmed delivery of chemotherapeutic agents and immunostimulants may offer a universal strategy for killing osteosarcoma cells while simultaneously eliciting in situ antitumor immunity. However, targeted chemoimmunotherapy lacks a reliable delivery system. To address this issue, we herein developed a bioinspired calcium phosphonate nanoagent that was synthesized by chemical reactions between Ca2+ and phosphonate residue from zoledronic acid using bovine serum albumin as a scaffold. In addition, methotrexate combination with a phosphorothioate CpG immunomodulator was also loaded for pH-responsive delivery to enable synergistic chemoimmunotherapy of osteosarcoma. The calcium phosphonate nanoagents were found to effectively accumulate in osteosarcoma for nearly 1 week, which is favorable for exerting the vaccination effects in situ by maturing dendritic cells and priming CD8+ T cells to suppress the osteosarcoma progression and pulmonary metastasis through controlled release of the three loaded agents in the acidic tumor microenvironment. The current study may thus offer a reliable delivery platform for achieving targeted chemotherapy-induced in situ antitumor immunity.

Reactive oxygen species-activated self-amplifying prodrug nanoagent for tumor-specific Cu-chelate chemotherapy and cascaded photodynamic therapy

Jie Chen,Xiaomin Tan,Yanjuan Huang,Congjun Xu,Zishan Zeng,Ting Shan,Zilin Guan,Xiaoyu Xu,Zeqian Huang,Chunshun Zhao

Disulfiram (DSF), an effective FDA-approved anti-alcoholism drug, shows potent antitumor activity by producing Cu(DTC) 2 , a chelate of its metabolite diethyldithiocarbamate (DTC) and copper. However, the rapid metabolism and unselective distribution of DSF and the insufficient endogenous copper severely restrict enough bioactive Cu(DTC) 2 generation in tumor tissues to achieve satisfactory antitumor effect. Moreover, directly Cu(DTC) 2 administration also suffers from serious systemic toxicity. Herein, a reactive oxygen species (ROS)-activatable self-amplifying prodrug nanoagent (HA-DQ@MOF) was developed for the stable co-delivery of DTC prodrug and Cu-quenched photosensitizer , aiming to achieve tumor-specific dual-activation of highly-toxic Cu(DTC) 2 -mediated chemotherapy and cascaded photodynamic therapy (PDT). The ROS-cleavable hyaluronic acid-conjugated DTC prodrug (HA-DQ) was decorated on Cu 2+ and photosensitizer Zn-TCPP coordinated MOF (PDT-shielded state) to construct HA-DQ@MOF. HA-DQ@MOF could specifically activated in ROS-overexpressed tumor cells to rapidly release DTC, while remaining relatively stable in normal cells. The free DTC immediately grabbed Cu 2+ from MOF to in situ generate highly-cytotoxic Cu(DTC) 2 chelate, accompanied by MOF dissociation to restore the PDT effect of Zn-TCPP. Importantly, ROS produced by PDT could in turn trigger more DTC release, which further promoted Zn-TCPP liberation, forming a self-amplifying prodrug/photosensitizer activation positive feedback loop . Experimental results confirmed the dual-activated and combined tumor-killing effect of Cu(DTC) 2 -mediated chemotherapy and Zn-TCPP-based PDT with little systemic toxicity. This work provides a dual-activated “low toxic-to-toxic” transformable treatment pattern for tumor-specific chemo-photodynamic therapy.

Tumor acidity-activatable photothermal/Fenton nanoagent for synergistic therapy

Zhe Yang,Li Zhang,Jielin Wei,Ruiqi Li,Qi Xu,Han Hu,Zushun Xu,Jinghua Ren,Chun-Yuen Wong

Intracellular formation of therapeutic agents has become one of the effective ways for cancer-specific treatment. Herein, a tumor acidity-activatable photothermal/Fenton nanoagent (denoted as CoPy) was constructed based on oxidized zeolitic imidazolate framework-67 (oxZIF-67) nanosheet and pyrrole (Py) monomer for synergistic therapy. The CoPy showed negligible toxicity to normal cell models RAW264.7 and 3T3 cell lines, and could be degraded by ascorbic acid in normal physiological conditions. However, once uptaken by 4T1 cells, the acidic pH led to the release of Co 3+ , which served as a strong oxidant to induce the polymerization of Py to form polypyrrole (PPy) for site-specific photothermal therapy (PTT). Most appealingly, the PPy could chelate the generated Co 2+ in the polymerization process to initiate the Fenton-like reaction, which was more capable to produce highly toxic hydroxyl radical (•OH) for chemodynamic therapy (CDT) compared to the free Co 2+ ones. In vitro and in vivo experiments demonstrated that all functionalities on CoPy worked collaboratively, and 78% of tumors were inhibited through cooperative PTT/CDT. Such a novel therapeutic nanoagent with tumor selectivity opens new opportunities for combinational treatment paradigms.

Metal–Organic Framework-Based Nanoagents for Effective Tumor Therapy by Dual Dynamics-Amplified Oxidative Stress

Jiajie Chen,Yitong Wang,Huicong Niu,Yuwei Wang,Aijun Wu,Chaoqin Shu,Yufang Zhu,Yuhai Bian,Kaili Lin

Overproduction of reactive oxygen species (ROS) within tumors can cause oxidative stress on tumor cells to induce death, which has motivated us to develop ROS-mediated tumor therapies, such as typical photodynamic therapy (PDT) and Fenton reaction-mediated chemodynamic therapy (CDT). However, these therapeutic modalities suffer from compromised treatment efficacy owing to their limited generation of highly reactive ROS in a tumor microenvironment (TME). In this work, a nanoscale iron-based metal–organic framework, MIL-101(Fe), is synthesized as a Fenton nanocatalyst to perform the catalytic conversion of hydroxyl radicals (·OH) from hydrogen peroxide (H2O2) under the acidic environment and as a biocompatible and biodegradable nanocarrier to deliver a 5,10,15,20-tetrakis(4-carboxyphenyl)porphyrin (TCPP) photosensitizer for light-activated singlet oxygen (1O2) generation. By coupling such chemodynamic/photodynamic effects, the photosensitizer-integrated nanoagents (MIL-101(Fe)@TCPP) could enable more ROS production within tumors to induce amplified oxidative damage for tumor-specific synergistic therapy. In vitro results show that MIL-101(Fe)@TCPP nanoagents achieve the acid-responsive CDT and effective PDT, and synergistic CDT/PDT provides an enhanced therapeutic effect. Ultimately, based on such synergistic therapy, MIL-101(Fe)@TCPP nanoagents cause a significant tumor growth inhibition in vivo without severe side effects, showing great potential for anti-tumor application.

An Ultra-Small Osmium Clusters-Loaded Organosilica-Based Antibacterial Nanoagent for Efficient Infected-Wound Healing

Jinyong Wu,Wenjuan Xiao,Shiyu Xu,Lihua Zhao,Zhongzhen Ren,Dechao Niu

Recently, nanomaterials with natural enzymatic activity and photothermal effect offer an alternative pathway to combat bacterial infections and the abuse of antibiotics. Unfortunately, the development of these reported antibacterial nanoagents is always hampered by insufficient enzymatic activity and low photothermal conversion efficiency. To address these issues, herein, novel ultra-small osmium clusters-loaded F127-organosilica micelles (Os@FOMs) are developed as antibacterial nanoagents for efficient infected-wound healing. The preparation of nanoagents mainly involves the confined sol-gel transition of thiol-containing silane coupling agent in the hydrophobic region of F127 micelles and subsequent self-reduction process of osmium salt in the thiols/disulfides-doped organosilica framework. As a promising candidate for bacterial-infected wound healing, Os@FOMs exhibit not only high photothermal conversion efficiency of 70% under 808 nm near infrared (NIR) irradiation, but also display peroxidase (POD)-like activity in acidic conditions, which together contribute excellent antibacterial ability in acidic bacterial infectious microenvironment. On the other hand, Os@FOMs possess good catalase-like effect in neutral or weak alkaline conditions, thus protecting normal tissues from the POD-like protocol with exogenous H 2 O 2 damage for efficient wound healing. It is highly expected that the new noble metal@organosilica-based nanoagents present a promising paradigm to fight antibiotic-resistant pathogens.

Single-molecular phosphorus phthalocyanine-based near-infrared-II nanoagent for photothermal antitumor therapy

Li-Na Zhou,Houhe Pan,Jing-Lan Kan,Qun Guan,Yang Zhou,Yu-Bin Dong

As one of the noninvasive cancer treatments, photothermal therapy (PTT) has drawn intense attention recently. In this context, an important task is to explore novel and versatile nanoscale photothermal agents (PTAs), especially those with strong NIR-II light absorption, high photothermal conversion efficiency, good photostability and biocompatibility. Phthalocyanines (Pcs), as the second-generation photosensitizers, are a promising class of candidates for PTT due to their strong NIR absorption and high photothermal conversion efficiency. However, the poor water solubility severely limited their application as PTAs in tumor treatment. Herein, we report a molecular phosphorus phthalocyanine (P-Pc)-based nanoagent via incorporation of human serum albumin (HSA) under mild conditions. The obtained nanoscale P-Pc-HSA possesses excellent photothermal conversion efficiency (64.7%) upon 1064 nm light irradiation, furthermore, it can be a highly efficient NIR-II antitumor nanoagent via photothermal treatment (PTT), which is fully evidenced by the in vitro and in vivo experiments.

Synergistic cytotoxicity of low-energy ultrasound and innovative mesoporous silica-based sensitive nanoagents

Zhao Yang,Zhu Yingchun

Low-energy ultrasound (LEUS) shows distinct potential as a safe therapeutic strategy for cancer treatment. Herein, mesoporous silica nanoparticles with closed-end cavities as sensitive nanoagents are prepared for effective cancer cell killing, when synergistically combined with mild LEUS (1 MHz, ≤1.0 W cm −2 ). The closed-end cavities can entrap gas bubbles, and provide a large number of cavitation nucleation sites, which could lead to drastically amplify ultrasonic cavitation effect by responding to the mild LEUS (1 MHz, ≤1.0 W cm −2 ). Significant killing effect against cancer cells is observed, when cells are treated by synergetic combination of mild LEUS and the nanoagents with closed-end cavities, showing distinct dose dependency on the nanoagents and irradiation intensity. Nevertheless, the killing effect is disappeared when the closed-end cavities are destructed. Moreover, no obvious cytotoxicity is observed when either the nanoagents or the LEUS is applied alone. The research may open up application opportunities of mild low-energy ultrasound for cancer therapy.
