Hydrogels with outstanding mechanical performance, self-healing capacity, and special functionality are highly desirable for their practical applications. However, it remains a great challenge to achieve such hydrogels by a facile approach. Here, we report a new type of nanocomposite hydrogels by in situ copolymerization of acrylic acid (AA) and 2-acrylamido-2-methylpropane sulfonic acid (AMPS) using alumina nanoparticles (Al2O3NPs) as the cross-linkers. The obtained hydrogels are highly stretchable and compressible, which could sustain large-scale extension (>1700%) or compression (90%) without failure, and exhibit tensile and compressive strength up to 660 kPa and 8.3 MPa, respectively. Furthermore, this kind of hydrogel also display considerable self-healing capacity due to their noncovalent cross-linking mechanism, as well as the hydrogen-bonding interactions between polymer chains. More interestingly, it was found that the resultant gels possess a long-lasting antifogging property that could prevent the formation of fog on the glass plate above hot water for at least 90 min. It is expected that this novel type of hydrogel would show great promise for various applications, including soft robots, artificial muscles, and optical devices. Keywords:nanocomposite hydrogels;Al2O3;high-strength;self-healing;antifogging

Fracture healing in patients with type 2 diabetes mellitus (T2D) is markedly impaired, characterized by a prolonged inflammation phase and defective osteoblast differentiation at the fracture site. In this study, we identified aberrant cellular glycolysis at T2D fracture sites, with bone marrow mesenchymal stem cells (BMSCs) exhibiting suppressed glycolysis and macrophages displaying enhanced glycolysis, mediated by the dysregulation of hypoxia-inducible factor-1α (HIF-1α). To rectify these metabolic imbalances, we developed a multifunctional nanocomposite PN@MHV hydrogel. Myricitrin, a flavonoid glycoside, forms the MHV hydrogel by cross-linking with HA-PBA and PVA via hydrogen bonds, and upregulates glycolysis through HIF-1α, thus promoting osteoblast differentiation under high glucose environment. To further regulate the inflammatory microenvironment, we incorporated nanoparticles loaded with PX-478, a HIF-1α specific inhibitor, into the hydrogel, with folic acid covalently modified to target proinflammatory M1 macrophages. This PN@MHV hydrogel bidirectionally regulated glycolysis via HIF-1α, enhancing osteoblast differentiation while attenuating macrophage-mediated inflammation. Comprehensive in vitro and in vivo experiments in a T2D fracture mouse model confirmed the hydrogel's ability to improve the inflammatory microenvironment and accelerate bone healing. Our findings underscore the therapeutic potential of targeting cellular glycolysis as a promising approach for enhancing fracture healing in diabetic patients.

Hydrogel actuators that deform in response to external stimuli have promising applications in diverse fields. It is desirable to develop robust hydrogels with multiple and synergistic stimuli responsiveness. This work reports on robust poly(NIPAM-co-DMAPMA)/clay hydrogel bilayers that undergo reversible and repeatable curling/uncurling in response to pH and/or temperature changes. The bilayers are fabricated by sequential synthesis of thermoresponsive N-isopropylacrylamide (NIPAM) and N-[3-(dimethylamino)propyl] methacrylamide (DMAPMA) monomers, which produces robust interface bonding between the layers. Different NIPAM/DMAPMA ratios were used between two gel layers to manipulate the asymmetric responsiveness, thus leading to curling/uncurling of bilayers upon exposure to stimuli. The hydrogel bilayers are fabricated into soft manipulators to grasp and release target objects by pH and temperature changes, and as artificial muscle to lift and move a hanging weight by synergistic pH and temperature stimuli. This work provides a new type of asymmetric responsive hydrogel bilayer with potential applications in biomimetic actuators.

Multi-responsive hydrogels with high strength have great significance for potential applications in smart soft devices. However, it remains a challenge to incorporate multiple responsive moieties with energy dissipation mechanisms. Herein, multi-responsive nanocomposite hydrogels with high compressive strength and toughness were synthesized via in situ copolymerization of N-isopropylacrylamide (NIPAM) and acryloyloxyethyltrimethyl ammonium chloride (DAC) in an aqueous dispersion of exfoliated LAPONITE® RDS with a minute amount of N,N′-methylenebisacrylamide (MBAA) as a crosslinker. The combined use of clay and MBAA is demonstrated to be favorable for the high strength and toughness, and helped in avoiding precipitation of clay nanosheets, which otherwise occurred upon addition of cationic DAC. The effect of the NIPAM/DAC molar ratio, MBAA and clay contents on the properties of the hydrogels has been systematically investigated. Compression tests showed a compressive strength up to 6.2 MPa, with fracture strain higher than 90%. The presence of ionic DAC moieties in the hydrogels rendered a very high swelling ratio up to 40 (g g−1). These hydrogels were responsive to temperature changes due to the presence of NIPAM units, with the transition temperature (Ttrans) dependent on the molar ratio of NIPAM and DAC monomers. The internal electrostatic repulsion of the NIPAM/DAC copolymer network changed upon exposure to solutions with different pH and/or ion strength. Cyclic swelling–shrinking was demonstrated by shuttling the gels between pure water and 0.1 mol L−1 NaCl solution.

A facile method is developed to fabricate nanocomposite double-network (DN) gels with excellent mechanical properties, which do not fracture upon loading up to 78 MPa and a strain above 0.98, by compositing of carbon nanotubes (CNTs) without organic modification. Investigations of swelling behaviors, and compressive and tensile properties indicate that equilibrium swelling ratio, compressive modulus and stress, fracture stress, Young’s modulus, and yield stress are significantly improved in the presence of CNTs. Scanning electron microscopy (SEM) reveals that the pore size of nanocomposite DN gels is decreased and some embedded micro-network structures are observed on the fracture surface in comparison to DN gels without CNTs, which leads to the enhancement of mechanical properties. The compressive loading-unloading behaviors show that the area of hysteresis loop, dissipated energy, for the first compressive cycle, increases with addition of CNTs, which is much higher than that for the successive cycles. Furthermore, the energy dissipation mechanism, similar to the Mullins effect observed in filled rubbers, is demonstrated for better understanding the nanocomposite DN polymer gels with CNTs.Keywords:nanocomposite hydrogels;double network;CNTs;mechanical strength

Osteosarcoma (OS) is the most prevalent primary malignancy of bone. Inhibition of OS recurrence and subsequent bone formation after surgery remains a challenge for decades. Innovative biomaterials with the dual function of tumor therapy and bone regeneration have emerged as a promising strategy for OS treatment. We developed a bioactive nanoparticle (MDA-NPs) composed of magnesium oxide nanoparticle (M-NPs) core and 2-Aminoethyl methacrylate (2-AM) grafted polydopamine (PDAM) shell. The phosphonate modified methacrylamide chitosan (CMP) and polyacrylamide (PAM) were prepared to form a pre-gel, and then a series of MDA-NPs nanocomposite with 0 mg/mL, 0.5 mg/mL, 2.5 mg/mL, 5 mg/mL, and 10 mg/mL were incorporated and coded as [email protected] , 0.5NP/ [email protected] , 2.5NP/ [email protected] , 5NP/ [email protected] , 10NP/ [email protected] , respectively. We found that 5NP/ [email protected] hydrogel showed a balanced photothermal effect, mechanical property, and osteogenesis for MDA-NPs can be used as a co-crosslinker, photothermal agent, and Mg 2+ reservoir. Human OS cells (143B) could be efficiently inhibited by 5NP/ [email protected] hydrogel with near-infrared (NIR) laser irradiation, achieving complete suppression of tumor recurrence in vitro and in vivo . The released Mg 2+ efficiently promotes the osteogenic activities of mouse embryo osteoblast precursor cells (MC3T3-E1) and 5NP/ [email protected] hydrogels exhibit highly effective bone repair performance in a critical cranial defect rat model. In conclusion, the 5NP/ [email protected] hydrogel demonstrated excellent dual functions of suppressing OS recurrence and repairing bone defects. This novel multifunctional bioactive hydrogel provides an encouraging idea of synergistic postoperative treatment of OS.

Development of combination photothermal–chemotherapy platform is of great interest for enhancing antitumor efficacy and inhibiting tumor recurrence , which supports selective and dose-controlled delivery of heat and anticancer drugs to tumor. Here, an injectable nanocomposite hydrogel incorporating PEGylated gold nanorods (GNRs) and paclitaxel-loaded chitosan polymeric micelles (PTX-M) is developed in pursuit of improved local tumor control. After intratumoral injection, both GNRs and PTX-M can be simultaneously delivered and immobilized in the tumor tissue by the thermo-sensitive hydrogel matrix. Exposure to the laser irradiation induces the GNR-mediated photothermal damage confined to the tumor with sparing the surrounding normal tissue. Synergistically, the co-delivered PTX-M shows prolonged tumor retention with the sustained release of anticancer drug to efficiently kill the residual tumor cells that evade the photothermal ablation due to the heterogeneous heating in the tumor region. This combination photothermal–chemotherapy presents superior effects on suppressing the tumor recurrence and prolonging the survival in the Heps-bearing mice, compared to the photothermal therapy alone.

We reported a series of tough nanocomposite hydrogels with good antifouling properties based on quaternized hybranched polyethylenimine (HPEI) nanoparticles. The quaternized HPEI nanoparticles acted as both cross-linkers to produce tough hydrogels and antibacterial agents to prevent fouling attachments. Compared with the polymer hydrogel, these nanocomposite hydrogels exhibited significantly improved mechanical properties. The mechanical strength of the nanocomposite hydrogels had a strong dependence with proportion of the quaternized HPEI nanoparticles. Furthermore, the anti-algae/bacterial assay indicated the nanocomposite hydrogels showed better antimicrobial and anti-algae performances due to the broad spectrum antimicrobial activity of Quaternary Ammonium Salts (QAS) functional groups.

The emergence of eutectic gallium–indium (EGaIn) liquid metal (LM) alloys as a soft multifunctional nanofiller presents an opportunity for the fabrication of hydrogel-based strain sensors with advanced multifunctional properties. However, developing a facile and efficient approach to synthesize nanocomposite conductive hydrogels that exhibit excellent stretchability, conductivity, self-adhesion, and antibacterial properties remains a significant challenge. In this study, we introduce a semi-interpenetrating network design strategy to synthesize a high-performance nanocomposite hydrogel [liquid metal/silver nanowires/sodium lignosulfonate/polyacrylamide] [LM/AgNWs/SL/pAM] (LASM). This hydrogel consists of a single polyacrylamide (pAM) network combined with a semi-interpenetrating network formed by silver nanowires (AgNWs) and LM nanoparticles. The semi-interpenetrating network is primarily cross-linked through hydrogen bonds, electrostatic interactions, and metal coordination. The resulting conductive hydrogels demonstrate superior stretchable properties (tensile stress: 120.28 kPa; tensile strain: 373.15%), impressive conductivity (0.64 S/m), high antifatigue performance, self-adhesive characteristics (Ti: 25.40 kPa; Al: 20.66 kPa), and notable antibacterial activity, all achieved through the construction of a hybrid chemical and physical cross-linking network. Leveraging these attributes, the nanocomposite hydrogel was assembled into a flexible sensor capable of distinguishing an extensive range of human movements, from large scale motions to subtle joint bending with remarkable stability and sensitivity. Furthermore, the LASM strain sensor can function as an adaptable writing keyboard that accurately recognizes English letters (“a”, “p”, “e”, “L,” and “HUT”) in real time when written on its surface. This multifunctional 3D nanocomposite conductive hydrogel holds great potential for applications in wearable electronics.

Hydrogels are appealing biomaterials for applications in regenerative medicine due to their tunable physical and bioactive properties. Meanwhile, therapeutic metal ions, such as magnesium ion (Mg 2+ ), not only regulate the cellular behaviors but also stimulate local bone formation and healing. However, the effective delivery and tailored release of Mg 2+ remains a challenge, with few reports on hydrogels being used for Mg 2+ delivery. Bisphosphonate exhibits a variety of specific bioactivities and excellent binding affinity to multivalent cations such as Mg 2+ . Herein, we describe a nanocomposite hydrogel based on hyaluronic acid and self-assembled bisphosphonate-magnesium (BP-Mg) nanoparticles . These nanoparticles bearing acrylate groups on the surface not only function as effective multivalent crosslinkers to strengthen the hydrogel network structure, but also promote the mineralization of hydrogels and mediate sustained release of Mg 2+ . The released Mg 2+ ions facilitate stem cell adhesion and spreading on the hydrogel substrates in the absence of cell adhesion ligands, and promote osteogenesis of the seeded hMSCs in vitro . Furthermore, the acellular porous hydrogels alone can support in situ bone regeneration without using exogenous cells and inductive agents, thereby greatly simplifying the approaches of bone regeneration therapy. Statement of Significance In this study, we developed a novel bioactive nanocomposite hydrogel based on hyaluronic acid and self-assembled bisphosphonate-magnesium (BP-Mg) nanoparticles. Such hydrogels are stabilized by the multivalent crosslinking domains formed by the aggregation of Ac-BP-Mg NPs, and therefore show enhanced mechanical properties , improved capacity for mineralization, and controlled release kinetics of Mg 2+ . Moreover, the released Mg 2+ can enhance cell adhesion and spreading, and further promote the osteogenic differentiation of hMSCs. Owing to these unique properties, these acellular hydrogels alone can well facilitate the in vivo bone regeneration at the intended sites. We believe that the strategy reported in this work opens up a new route to develop biopolymer-based nanocomposite hydrogels with enhanced physical and biological functionalities for regenerative medicine.

The management of chronic diabetic wounds remains a significant challenge due to persistent bacterial infections and impaired angiogenesis. Herein, we reported a nanocomposite hydrogel (M/P-SNO/G) incorporated with M/P-SNO nanoparticles engineered by supramolecular assembly of the photosensitizing mono-carboxyl corrole (MCC) and S-nitrosothiol-modified polyethylene glycol (mPEG-SNO) for synergistic photothermal therapy (PTT)/nitric oxide (NO) treatment of diabetic wounds. The strong π-π interaction among aggregated MCC in M/P-SNO enhances the optical absorption and photothermal ability, thereby facilitating the precise release of NO upon laser irradiation. The hydrogel matrix, composed of oxidized hyaluronic acid and carboxymethyl chitosan crosslinked by Schiff-base, demonstrates good injectability and self-healing characteristics, providing an ideal environment for wound repair. As expected, M/P-SNO/G exhibits a desirable photothermal performance and a controlled laser-responsive NO release, realizing enhanced bactericidal effect and anti-biofilm ability in vitro . In a full-thickness skin defect model on diabetic mice, M/P-SNO/G has proven effective in bacteria clearance and angiogenesis, significantly accelerating wound healing. This study presents a feasible supramolecular strategy to develop diabetic wound dressings with synergistic PTT/NO treatment. Statement of significance Developing advanced dressings that simultaneously eliminate bacteria and accelerate wound recovery is essential for treating diabetic wounds. This study developed a nanocomposite hydrogel (M/P-SNO/G) featuring the synergistic effect of photothermal therapy (PTT) and nitric oxide (NO) treatment to accelerate infected diabetic wound healing. M/P-SNO nanoparticles within the hydrogel are self-assembled through the hydrophobic photosensitizing mono-carboxyl corrole (MCC) and the hydrophilic NO-releasing polymer (mPEG-SNO), where highly aggregated MCC molecules ensure superior photothermal performance. Meanwhile, the temperature increase induced by the photothermal effect activates NO release from the hydrogel. Under 660 nm laser irradiation, M/P-SNO/G demonstrates a PTT/NO synergy to effectively inhibit bacterial proliferation and promote angiogenesis, offering significant benefits in diabetic wound repair and further expanding the biomedical applications of corroles.

Gradient materials exist widely in natural living organisms, affording fascinating biological and mechanical properties. However, the synthetic gradient hydrogels are usually mechanically weak or only have relatively simple gradient structures. Here, we report on tough nanocomposite hydrogels with designable gradient network structure and mechanical properties by a facile post-photoregulation strategy. Poly(1-vinylimidazole-co-methacrylic acid) hydrogels containing gold nanorods (AuNRs) are in a glassy state and show typical yielding and forced elastic deformation at room temperature. The gel slightly contracts its volume when the temperature is above the glass-transition temperature that results in a collapse of the chain segments and formation of denser intra- and interchain hydrogen bonds. Consequently, the mechanical properties of the gels are enhanced, when the temperature returns to room temperature. The mechanical performances of hydrogels can also be locally tuned by near-infrared light irradiation due to the photothermal effect of AuNRs. Hydrogels with arbitrary two-dimensional gradients can be facilely developed by site-specific photoirradiation. The treated and untreated regions with different stiffness and yielding stress possess construct behaviors in stretching or twisting deformations. A locally reinforced hydrogel with the kirigami structure becomes notch-insensitive and exhibits improved strength and stretchability because the treated regions ahead the cuts have better resistance to crack advancement. These tough hydrogels with programmable gradient structure and mechanics should find applications as structural elements, biological devices, etc.
