Fabrication of an Ag-based SERS nanotag for histamine quantitative detection

Tingting Zhang,Gongying Wang,Juan Bao,Chang Liu,Weihua Li,Zhe Kong,Xiuyun Sun,Jiansheng Li,Rui Lu

A crucial issue in analytical science and physiology is the detection of histamine with high sensitivity, specificity and credibility, which served as an important neurotransmitter in biofluids. Despite the high sensitivity of surface-enhanced Raman spectroscopy (SERS) at the level of single molecule, there are still challenges in providing high sensitivity for histamine with a small cross section. For the selective detection of histamine using SERS, a highly sensitive sandwich structure substrate combining Fe 3 O 4 and an Ag-based SERS nanotag was developed. The Fe 3 O 4 @SiO 2 –COOH served as a capture component for enriching histamine. Upon functionalized Ag nanoparticles with glycine (Gly) and (3-Aminopheyonyl) boronic acid (APBA), they were then used to connect with histamine and serve as a SERS nanotag, respectively. A linear relationship between the Raman intensity and the histamine concentration was observed over the range 10 −4 -10 −8  M with a limit of detection of 7.24 × 10 −9  M. This methodology also exhibited good selectivity in the presence of other neurotransmitters. With our new approach, histamine can be detected sensitively and reliably in fish samples, which indicates the potential prospect of an effective method for analyzing histamine in complex specimens.

Raman Nanotags-Guided Intraoperative Sentinel Lymph Nodes Precise Location with Minimal Invasion

Binge Deng,Yaohui Wang,Yifan Wu,Wenjin Yin,Jinsong Lu,Jian Ye

The accurate positioning of sentinel lymph node (SLN) by tracers during surgery is an important prerequisite for SLN biopsy. A major problem of traditional tracers in SLN biopsy is the short surgery window due to the fast diffusion of tracers through the lymphatics, resulting in a misjudgment between SLN and second echelon lymph node (2 nd LN). Here, a nontoxic Raman nanoparticle tracer, termed gap-enhanced Raman tags (GERTs), for the accurate intraoperative positioning of SLNs with a sufficient surgical time window is designed. In white New Zealand rabbit models, GERTs enable precise identification of SLNs within 10 min, as well as provide the surgeon with a more than 4 h time window to differentiate SLN and 2 nd LN. In addition, the ultrahigh sensitivity of GERTs (detection limit is 0.5 × 10 −12 m ) allows detection of labeled SLNs before surgery, thereby providing preoperative positioning information for minimally invasive surgery. Comprehensive biosafety evaluations carried out in the context of the Food and Drug Administration and International Standard Organization demonstrate no significant toxicity of GERTs, which supports a promising clinical translation opportunity of GERTs for precise SLN identification in breast cancer.

Engineering a SERS Sensing Nanoplatform with Self-Sterilization for Undifferentiated and Rapid Detection of Bacteria

Jun Cao,Wei Zhu,Ji Zhou,Bai-Chuan Zhao,Yao-Yu Pan,Yong Ye,Ai-Guo Shen

The development of a convenient, sensitive, rapid and self-sterilizing biosensor for microbial detection is important for the prevention and control of foodborne diseases. Herein, we designed a surface-enhanced Raman scattering (SERS) sensing nanoplatform based on a capture–enrichment–enhancement strategy to detect bacteria. The gold−Azo@silver−cetyltrimethylammonium bromide (Au−Azo@Ag−CTAB) SERS nanotags were obtained by optimizing the synthesis process conditions. The results showed that the modification of CTAB enabled the nanotags to bind to different bacteria electrostatically. This SERS sensing nanoplatform was demonstrated to be fast (15 min), accurate and sensitive (limit of detection (LOD): 300 and 400 CFU/mL forE. coliandS. aureus, respectively). Of note, the excellent endogenous antibacterial activity of CTAB allowed the complete inactivation of bacteria after the assay process, thus effectively avoiding secondary contamination.

Dual-Mode Lateral Flow Immunoassay Based on “Pompon Mum”-Like Fe3O4@MoS2@Pt Nanotags for Sensitive Detection of Viral Pathogens

Meimei Xu,Shuai Zhao,Chenglong Lin,Yanyan Li,Weida Zhang,Yusi Peng,Rui Xiao,Zhengren Huang,Yong Yang

Lateral flow immunoassay (LFIA) has been widely used for the early diagnosis of diseases. However, conventional colorimetric LFIA possesses limited sensitivity, and the single-mode readout signal is easily affected by the external environment, leading to insufficient accuracy. Herein, multifunctional Fe3O4@MoS2@Pt nanotags with a unique “pompon mum”-like structure were triumphantly prepared, exhibiting excellent peroxidase (POD)-like activity, photothermal properties, and magnetic separation capability. Furthermore, the Fe3O4@MoS2@Pt nanotags were used to establish dual-mode LFIA (dLFIA) for the first time, enabling the catalytic colorimetric and photothermal dual-mode detection of severe acute respiratory syndrome coronavirus 2 nucleocapsid protein (SARS-CoV-2 NP) and influenza A (H1N1). The calculated limits of detection (cLODs) of SARS-CoV-2 NP and H1N1 were 80 and 20 ng/mL in catalytic colorimetric mode and 10 and 8 ng/mL in photothermal mode, respectively, demonstrating about 100 times more sensitive than the commercial colloidal Au-LFIA strips (1 ng/mL for SARS-CoV-2 NP; 1 μg/mL for H1N1). The recovery rates of dLFIA in simulated nose swab samples were 95.2–103.8% with a coefficient of variance of 2.3–10.1%. These results indicated that the proposed dLFIA platform showed great potential for the rapid diagnosis of respiratory viruses.

Raman Ink for Steganography

Yuqing Gu,Chang He,Fugang Liu,Jian Ye

Steganography, also known as “invisible” communication, refers to the technique of hiding information into another medium such as video, audio, image, and text. Surface-enhanced Raman scattering (SERS) nanotags are well suited to information encoding owing to their nanoscale dimensions, fingerprint optical spectral features, and remarkable multiplexing capability. Herein, Raman ink, fabricated by doping a new type of SERS nanotags (gap-enhanced Raman tags, GERTs) into commercial ink, is demonstrated as security ink for multiplexing steganography. A stego-text is written using two types of Raman ink containing different GERTs with diverse Raman signals, imaged via a confocal Raman system, and processed by the classical least squares method to extract the hidden message. The Raman ink generates distinct spectral profiles with low background from pure ink upon near-infrared laser irradiation. A multiplexing combination of seven kinds of messages extracted from the written stego-text is demonstrated, which adds enhanced safety and flexibility to information encoding. In addition, Raman ink exhibits good photostability and long-term stability. Therefore, GERTs-based Raman ink is promising for steganographic use in information security.

Simultaneous Electrochemical Multiplexed Immunoassay of Biomarkers Based on Multifunctionalized Graphene Nanotags

Qunfang Li,Dianping Tang,Fangming Lou,Ximei Yang,Guonan Chen

Graphical Multitasker: A sensitive and feasible multiplexed immunoassay protocol for simultaneous electrochemical determination of biomarkers is presented. The assay uses multifunctionalized graphene nanotags on a cyclodextrin-modified immunosensor. A sensitive and feasible multiplexed immunoassay protocol for simultaneous electrochemical determination of carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) was developed using multifunctionalized graphene nanotags on a cyclodextrin-modified immunosensor. The sensor was fabricated via the guest–host chemistry between the immobilized cyclodextrin and anti-CEA/anti-AFP antibodies. Two nanotags including cyclodextrin-thionine-graphene and cyclodextrin-ferrocene-graphene nanostructures were synthesized based on a wet chemistry method. The synthesized nanotags were employed for the labelling of horseradish peroxidase (HRP)–anti-AFP and HRP–anti-CEA conjugates, respectively. With a sandwich-type assay format, the signal was monitored through the labelling of HRP toward catalytic reduction of H 2 O 2 at various peak potentials in the presence of thionine and ferrocene, respectively. Under optimal conditions, the multiplexed immunoassay enabled the simultaneous determination of CEA and AFP with wide working ranges of 0.001–60 ng mL −1 for AFP and 0.003–40 ng mL −1 for CEA. The detection limits were 0.5 pg ml −1 for AFP and 0.8 pg ml −1 for CEA (at 3 s B ). No obvious nonspecific adsorption and cross-talk were observed during a series of analyses to detect target analytes. Intra- and inter-assay coefficients of variation (CV) were below 9.5 %. Importantly, the methodology was also evaluated for the analysis of AFP and CEA in the clinical human serum specimens, receiving a good relationship between the developed immunoassay and a commercialized electrochemiluminescence enzyme-linked immunoassay.

An electrochemical sensing strategy based on a three dimensional ordered macroporous polyaniline–platinum platform and a mercury(II) ion-mediated DNAzyme functionalized nanolabel

Rong Tian,Xiaojun Chen,Nan Jiang,Ning Hao,Lin Xu,Cheng Yao

A highly sensitive electrochemical sensor for the detection of mercury(II) ions (Hg2+) was developed by using a three dimensional ordered macroporous polyaniline–platinum (3DOM PANI–Pt) composite film as a sensing platform. 3DOM PANI–Pt was realized by electrochemical co-deposition of Pt nanoparticles and PANI into the sacrificed silica template through the redox reaction involving the monomer aniline (AN) and PtCl62−. The G-rich oligonucleotide strand functionalized Au nanoparticle was employed as the Hg2+-mediated structural switch and to enhance the sensitivity. In the absence of Hg2+, the oligonucleotide strand formed an intramolecular duplex where the G-rich sequence was partially caged. In the presence of Hg2+, the release of the G-rich sequence was observed due to the stabilization of T–Hg2+–T, which formed an active G-quadruplex DNAzyme and catalyzed the reduction of H2O2. Under the optimal conditions, the current signal of H2O2 increased with increasing Hg2+ concentration in the dynamic range from 10−13 to 10−6 M, and the detection limit up to 8.7 × 10−14 M was seen. Further, the sensor was successfully utilized for the determination of Hg2+ in an authentic aquatic sample, with an acceptable accuracy compared to the method commercially available. In this paper, we proposed a general and simple strategy because only one oligonucleotide strand was required for both Hg2+ recognition and signal amplification, potentially allowing the detection of other metal ions or trace pollutants in environmental matrices by simply employing various DNA or aptamer probes.

Resonant Strategy in Designing NIR-II SERS Nanotags: A Quantitative Study

Haoqi He,Yumin Zhang,Shuo Zhu,Jian Ye,Li Lin

Surface-enhanced Raman scattering (SERS) nanotags have garnered much attention for in vivo and intraoperative imaging due to their high sensitivity and intrinsic fingerprint spectrum. Recently, in vivo bioimaging at the second biological near-infrared (NIR-II) window has demonstrated significant advances with better spatial resolution and deeper tissue penetration. The development of SERS nanotags at the NIR-II window are therefore highly desired. The resonant strategy, i.e., the use of resonant substrates and resonant molecules, can be helpful in fabricating bright SERS nanotags at the NIR-II window; however, this strategy has not been demonstrated with the lack of quantitative assessments. Here we report a quantitative study on the Raman enhancements of the resonant strategy in preparing NIR-II SERS nanotags. By comparing the resonant and nonresonant plasmonic nanorod substrates, the resonant substrates have been shown to provide an enhancement factor of up to 4 orders of magnitude. When using the resonant Raman reporter molecules, SERS intensities can be further increased by 25–546 times. This work demonstrates the potential of the resonant strategy in fabricating ultrasensitive NIR-II SERS nanotags and provides insights into the design of related plasmonic devices.

Surface-enhanced Raman scattering nanotag of tunable ZIF-8 shell-encapsulated magnetic core-plasmonic satellites for disentangling chemical enhancement from electromagnetic enhancement

Hao Chen,ZhaoGuo Wu,AiWu Zhao,Jin Wang

To enhance the stability of Raman reporters, these reporters were trapped in a metal organic framework (MOF) exoskeleton that was grown and compressed on Fe3O4@Au core–satellites, producing recyclable surface-enhanced Raman scattering (SERS) nanotags. Furthermore, encapsulation of Raman reporters in the assembled MOF-based nanocomposites was divided into two types of patterns, pre-enrichment and post-enrichment, in order to disentangle chemical enhancement of charge transfer (CT) from electromagnetic enhancement (EM) in SERS. Hence, to demonstrate the effect of encapsulation, a typical non-thiolated Raman reporter, for example crystal violet (CV) trapped in a core–satellite nanoassembly-based zeolitic imidazolate framework (ZIF-8) shell, was selected. The results suggest that stability and Raman intensity are remarkably improved. Moreover, the pattern of incorporation of CV into the ZIF-8 shell with tunable shell thickness can contribute to the disentangling of CT effects from EM effects.

Investigating SERS performance of controllable fabricated Ag periodic nanostructures with morphologies of nanoclaws and nanocones in detecting SARS-CoV-2 S protein

Yansheng Liu,Yongquan Wei,Guofu Wang,Xiaobo Jia,Jin Zhou,Hongqi Li,Haixin Chang,Zhaoxu Li

Surface-enhanced Raman scattering (SERS) spectroscopy plays an important role in enabling high-precision detection and non-invasive diagnostics. Periodic metallic nanostructures with strong electromagnetic fields and tunable localized surface plasmon resonance can achieve high SERS sensitivity, but fabricating such nanostructures remains a challenge. In this work, the anodic aluminum oxide (AAO) nanocavity template-assisted process of fabricating periodic nanostructures with modulating morphologies from Ag nanoclaws (AgNCWs) to Ag nanocones (AgNCEs) is proposed. The morphologies-related SERS performances of nanostructures are analyzed by using rhodamine 6 g (R6G) as a probe molecule and the optimized AgNCEs possess a limit of detection (LOD) of 1 × 10 −9 M. A sandwich strategy is applied to detect SARS-CoV-2 S protein in phosphate-buffered saline (PBS) and pharyngeal swab solution (PSS). The ACE2-modified AgNCEs (ACE2/AgNCEs) structure is applied to bind to the SARS-CoV-2 S protein. The 4-MBA and SARS-CoV-2 S antibody functionalized Au nanoparticles (AuNPs) are used as nanotags. By forming the ACE2/AgNCEs-SARS-CoV-2 S protein-nanotags sandwich structure, characteristic peak intensities of 1580 cm −1 are used as the report signal, and the LOD for SARS-CoV-2 S protein in PBS and PSS solution are 10 fg/mL.

Gap-enhanced resonance Raman tags for live-cell imaging

Yuqing Gu,Xinyuan Bi,Jian Ye

Surface-enhanced Raman scattering (SERS) nanotags are widely used in the biomedical field including live-cell imaging due to the high specificity from their fingerprint spectrum and the multiplexing capability from the ultra-narrow linewidth. However, long-term live-cell Raman imaging is limited due to the photodamage from a relatively long exposure time and a high laser power, which are needed for acquiring detectable Raman signals. In this work, we attempt to resolve this issue by developing ultrabright gap-enhanced resonance Raman tags (GERRTs), consisting of a petal-like gold core and a silver shell with the near-infrared resonant reporter of IR-780 embedded in between, for long-term and high-speed live-cell imaging. GERRTs exhibit an ultrahigh Raman intensity down to a single-nanoparticle level in aqueous solution and the solid state upon 785 nm excitation, allowing for high-resolution time-lapse live-cell Raman imaging with an exposure time of 1 ms per pixel and a laser power of 50 μW. Under these measurement conditions, we can possibly capture dynamic cellular processes with a high temporal resolution, and track living cells for long periods of time owing to the reduced photodamage to cells. These nanotags open new opportunities for ultrasensitive, low-phototoxic, and long-term live-cell imaging.

Silver Nanolabels-Assisted Ion-Exchange Reaction with CdTe Quantum Dots Mediated Exciton Trapping for Signal-On Photoelectrochemical Immunoassay of Mycotoxins

Youxiu Lin,Qian Zhou,Dianping Tang,Reinhard Niessner,Huanghao Yang,Dietmar Knopp

Mycotoxins, highly toxic secondary metabolites produced by many invading species of filamentous fungi, contaminate different agricultural commodities under favorable temperature and humidity conditions. Herein, we successfully devised a novel signal-on photoelectrochemical immunosensing platform for the quantitative monitoring of mycotoxins (aflatoxin B1, AFB1, used as a model) in foodstuffs on the basis of silver nanolabels-assisted ion-exchange reaction with CdTe quantum dots (QDs) mediated hole-trapping. Initially, a competitive-type immunoreaction was carried out on a high-binding microplate by using silver nanoparticle (AgNP)-labeled AFB1–bovine serum albumin (AFB1–BSA) conjugates as the tags. Then, the carried AgNPs with AFB1–BSA were dissolved by acid to release numerous silver ions, which could induce ion-exchange reaction with the CdTe QDs immobilized on the electrode, thus resulting in formation of surface exciton trapping. Relative to pure CdTe QDs, the formed exciton trapping decreased the photocurrent of the modified electrode. In contrast, the detectable photocurrent increased with the increase of target AFB1 in a dynamic working range from 10 pg mL–1 to 15 ng mL–1 at a low limit of detection (LOD) of 3.0 pg mL–1 under optimal conditions. In addition, the as-prepared photoelectrochemical immunosensing platform also displayed high specificity, good reproducibility, and acceptable method accuracy for detecting naturally contaminated/spiked blank peanut samples with consistent results obtained from the referenced enzyme-linked immunosorbent assay (ELISA) method.
