Overuse and abuse of morphine (MOP), one of the main components of pericarpium papaveris, have attracted increasing attention in the medical field owing to its pharmacological and toxicological activity. Herein, we proposed a new electrochemical nano-biosensor for MOP detection based on surface-confined building of Au@Pt-centered and multi-G-quadruplex/hemin wire-surrounded electroactive super-nanostructures. The center Au@Pt was flower-shaped and irregularly protruded, allowing substantial loading of multiple G-quadruplex wire/hemin complexes on its surface to accomplish the assembly of electroactive super-nanostructures. Interestingly, as the super-nanostructures were closely confined on the electrode surface, a significantly amplified electrochemical signal was thus obtained in the absence of MOP. In contrast, the introduction of target MOP can induce an intense competitive effect and strongly destroy the assembly process, resulting in the reduction of the electrochemical response that is correlated with the logarithmic concentration of MOP. Under optimal conditions, the electrochemical nano-biosensor is capable of highly sensitive detection of MOP in a dynamic concentration range from 1 ppt to 500 ppb. The limit of detection is achieved as low as 0.69 ppt, and the practical application was confirmed by examining MOP from chafing dish condiments. We expect the electrochemical platform utilizing this unique nanoarchitecture to provide rational guidelines to design high-performance analytical tools.

Ochratoxin A (OTA), which is prone to contaminating food products, poses a significant threat to human health. To accurately detect and provide timely early warnings for OTA contamination (COTA) in food, a novel four-modal nano-biosensor detection system based on MXenes was developed in this work. MXenes is a new family of two-dimensional materials including vanadium carbide (V 2 C) MXenes, which has great potential in the field of biosensing and detection due to its large specific surface area, rich surface functional groups, and good chemical stability. By in-situ chemical etching, metal ion intercalation, and synergistic physical exfoliation techniques, V 2 C nano-materials (V 2 C-NMS) with excellent fluorescence quenching characteristics were synthesized, and V 2 C-NMS@ssDNA with excellent peroxidase like activity was constructed by functionalizing V 2 C-NMS with adapter. Based on this, a fluorescence/colorimetric biosensor for OTA was constructed, and the fluorescence/colorimetric signal output by the sensor had good linear relationship with COTA, with the detection limit (LOD) as low as 6.77 pg mL −1 . Furthermore, a fully connected artificial neural network (FCANN) was developed based on a series of RGB values obtained from the fluorescence/colorimetric mode of the biosensor, and the fluorescence/colorimetry channel of the FCANN could accurately predict the COTA of the sample on-site or remotely in just a few seconds, with the LOD as low as 7.10 pg mL −1 . Importantly, the four-mode method performed well in real sample detection, with recovery rates ranging from 95.33% to 105.79%, and the detection results of the four modalities could be mutually verified.

Optical glucose detection holds considerable promise for continuous in vivo glucose monitoring with wireless transdermal transmission and long-lasting activity. To construct a new class of optical glucose nanobiosensors with high sensitivity and selectivity at physiological conditions, the first generation of fluorescent poly(amido amine) (G1.0 PAMAM), serving as the optical code, was introduced into glucose-sensitive poly(N-isopropylacrylamide-(2-dimethylamino)ethyl methacrylate-3-acrylamidephenylboronic acid) copolymer microgels via a facile method. The fabricated microgels display the ability of adapting to the surrounding medium of different glucose concentrations over a clinically relevant range (0–20 mM) and convert biochemical signals into optical signals. As nanobiosensors, the G1.0 PAMAM functionalized microgels exhibit high selectivity for glucose over various kinds of potential primary interferents, such as lactate, human serum albumin and metal ions, in the physiologically important glucose concentration range. Compared to traditional fluorescent dyes and quantum dots, which are limited by photobleaching and toxicity, this microgel with remarkable anti-photobleaching property and low toxicity makes it possible to be used for long-term continuous glucose monitoring. Through in vivo investigations, it can be observed that G1.0 PAMAM functionalized microgels can achieve wireless transdermal detection, indicating that the fabricated microgels have potential applications as a new generation of nanobiosensors for the highly sensitive and minimally invasive continuous glucose monitoring.

Antibiotic residues in foods pose a serious threat to human health. However, routine analysis techniques require bulky laboratory instruments and skilled personnel or give single-channel analysis results, exhibiting low practicality. Here, we explored a rapid and easy-to-use detection system combining a fluorescence nanobiosensor with a homemade fluorescence analyzer for the simultaneous identification and quantification of multiple antibiotics. The nanobiosensor assay worked based on the targeted antibiotics competing with signal labels of antigen-quantum dots (IQDs) to bind with recognition elements of antibody-magnetic beads (IMBs). The fluorescence signals of IMB-unbound IQDs in a magnetically separated supernatant, related to antibiotic concentration, were automatically collected and processed by our self-designed and homemade fluorescence analyzer which integrated mechanical control hardware (consisting of a mechanical arm, a ten-channel rotary bench, and an optical detection unit) and user control software (installed on a built-in laptop). The fluorescence analyzer enabled the analysis of 10 samples within 5 min in one round and permitted the real-time uploading of sample data to the cloud. By employing three QDs with emission wavelengths of 525 nm, 575 nm, and 625 nm, this multiplex fluorescence biosensing system demonstrated great sensitivity and accuracy for simultaneously analyzing enrofloxacin, tilmicosin, and florfenicol in chicken samples with detection limits of 0.34 μg kg−1, 0.7 μg kg−1, and 0.16 μg kg−1, respectively. Moreover, the biosensing platform performed well in a wealth of chicken samples covering various breeds from three Chinese cities. This study identifies a generic and user-friendly multiplex biosensor platform with significant potential for use in food safety and regulation.

The rapid and simple detection of circulating tumor cells (CTCs) can help with early diagnosis of tumor diseases and assist clinical judgment of tumor disease development status and prognosis. An important alteration in cancer progression is a significant increase in sialic acid (SA) levels, making sialic acid one of an interesting biomarker for cancer type diversity. Here, novel SERS nano-biosensors were fabricated employing molecularly imprinted polymers (MIPs) particles loaded with Ag nanoparticles (AgNPs) to form high affinity toward sialic acid on CTCs. In this work, the glycol-specific MIPs are produced using precipitation polymerization. The nanocomposite was characterized by Fourier transform infrared spectroscopy (FTIR), zeta potential, scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Consequently, CTC can be rapidly detected in mouse blood in a specific and sensitive manner, as low as 1 cells/ml. In addition, a good linear relationship was observed between CTC concentration and signal intensity ( R 2 = 0.9881). Moreover, the SERS nano-biosensors was used to detect CTC with excellent precision (relative standard deviation, RSD = 3.9%) and good repeatability (RSD = 6.8%). Therefore, this inexpensive, highly sensitive, and rapid method using SERS nano-biosensors can quickly detect CTCs and may prove to be a new platform to be an early diagnostic tool for cancer patient detection. Sepsis represents a life-threatening systemic inflammatory condition characterized by organ dysfunction, where the identification of specific biomarkers and development of effective diagnostic modalities are critical for timely clinical intervention to mitigate disease progression. Emerging evidence positions exosomal microRNAs (exomiRs) as promising novel biomarkers with multi-dimensional clinical utility spanning early diagnosis, therapeutic efficacy assessment, and longitudinal prognostic monitoring in sepsis management. However, the direct rapid detection of exomiRs in clinical specimens undergoes severe critical challenges due to the absence of robust detection methodologies with these characteristics of operational simplicity, high sensitivity and analytical robustness. Here, we designed and constructed a hairpin/loop-constrained three-dimensional topological self-assembling nanostructure, which was denoted as DNA molecular lock (DML), and developed a spatially allosteric-mediated electrochemical nanobiosensor for ultrasensitive and specific analysis of exomiR-125b. This independent, rapid, and portable detection method, combined with Ti 3 C 2 T x MXene/AuAgNPs nanozymes, achieved excellent sensitivity with a linear range from 2 fM to 200 nM and a low detection limit of 0.15 fM. It provided required low-volume plasma samples (2 mL) for analysis while supported Bluetooth-enabled wireless communication and real-time data processing. Furthermore, integration of machine learning algorithms with this developed nanobiosensor improved differential diagnostic accuracy to 96.6 % in the training cohort ( n = 83) and 89.2 % in the validation cohort ( n = 57), effectively distinguishing sepsis patients (SPs), general inflammation patients (GIPs), and healthy donors (HDs). Our study establishes the sensor's efficacy in improving sepsis detection through biomarker-guided analysis. These findings highlight the potential application of this developed nanobiosensors for portable, personalized bedside monitoring of exomiRs as the point-of-care testing (POCT) platforms.

Some veterinary drug residues in food products and environment have been widely regarded as severe threats to human health. Rapid and simultaneous detection methods are crucial to monitor and control veterinary drug usage. Here, we propose a fluorescence biosensor utilizing immunomagnetic beads (IMBs) and quantum dots (QDs) for the rapid and simultaneous detection of 1-adamantylamine (ADA), enrofloxacin (ENR) and tilmicosin (TIL) in raw chicken meat. A pretreatment method using sodium phosphotungstate–magnesium as extraction reagent was developed to simultaneously extract ADA, ENR and TIL from chicken meat with minor interference in background or response. By adding the IMBs modified with three types of antibodies and the QD-antigens modified with three types of BSA-antigens to sample, IMBs competitively conjugated to target antigens in a sample or QD-antigens. After magnetic separation, the residual QD-antigens were adopted to collect signals using fluorescence spectroscopy. Using QDs with well separated emission peaks, the detection of one type of targets was minorly interfered by the others. Under the optimum conditions, the biosensor exhibited the limit of detection of 0.96, 3.32, and 3.17 ng/mL for ADA, ENR and TIL in chicken samples, respectively, as well as good specificity. Due to the way of direct collection of signals in extracts, the tedious and complicated multiple magnetic separation and signal amplification procedures in conventional methods were avoided, thus the procedures were significantly simplified, and the reduction of the operation time of 30 min for sample pretreatment and 40 min for detection part was achieved. The biosensor might be promising in the rapid, in-field and sensitive screening of multiple veterinary drugs to ensure agriculture and food safety.

Engineered collaborative biochemical techniques and regulated nanomaterials (NMs) offer extraordinary opportunities for improving the analysis performance of lateral flow immunoassay (LFIA). Herein, inspired by the ability of macromolecules (e.g., proteins) to assemble into new functional units and the remarkable optical performance of engineered regulated NMs, goat anti-mouse immunoglobulin (GAMI) serves as the “crosslinker” integrate with gold‑manganese oxide (Au-MnO x ) to assemble the “signal tracers (STs)-crosslinker-antibody (mAb)” for elevating the mAb utilization efficiency. Notably, the “STs-crosslinker-mAb” assembly shows ~13.33-folds mAb utilization efficiency enhance, which perfectly response the challenge between limited sensitivity and sufficient signal intensity in competitive-type LFIA. The black color and rough structure of Au-MnO x offer higher colorimetric brightness (~2-folds than AuNPs) and enhanced mAb coupling efficiency (up to 92.47%), which further improves sensitivity under the premise of functional assembly to intensify the competitive immunoreaction. Additionally, the convenient synthesis conditions (~13 min at room temperature) even comparable to direct purchase commercial products indicate that using Au-MnO x undoubtedly increases the cost-effectiveness. Encouragingly, the Au-MnO x -GAMI-mAb based LFIA exhibited high sensitivity (LOD: 0.063 ng mL −1 for clenbuterol (CLE) monitoring) by elevating mAb utilization efficiency with the attendant enhancing immune competition response in a cost-effective manner, which provides an invigorating reference pathway in point-of-care immunoassay.

Herein, a sequentially responsive peptide DNA bilingual nanobiosensor is developed, which allows integrated quantification of amyloid signaling pathway. In this system, upstream beta-site amyloid precursor protein-cleaving enzyme 1 (BACE1) protease and downstream Aβ oligomer (AβO) are designed as two inputs for the AND DNA logic gate. In the existence of both inputs, peptide substrate with aptamer can be sequentially cleaved, reporting electrochemical and fluorescence dual-mode outputs. In comparison with conventional single protease activity assay based on peptide nanotechnology, this strategy permits accurate diagnosis of Alzheimer's disease (AD) from normal subjects. More importantly, it can achieve distinguished diagnosis between AD and type 2 diabetes mellitus patients. This bilingual nanobiosensor is successfully applied to detect BACE1 (1–100 U mL −1 ) and AβO (5–1000 pg mL −1 ) with limit of detections as low as 0.10 U mL −1 and 0.76 pg mL −1 , respectively. Furthermore, this strategy inspires advanced nanobiosensors to target the activation of other signaling pathways, which are potential tools for future biology and medicine investigation.

A Glypican-3 (GPC3) electrochemical aptamer nanobiosensor based on hemin/graphene nanohybrids (HGNs) peroxidase-like catalytic silver deposition and GPC3 aptamer has been constructed for the determination of GPC3. The HGNs were prepared by an one-step reduction method. Fourier transform infrared spectroscopy (FT-IR), ultraviolet spectroscopy (UV-vis), and transmission electron microscopy (TEM) were used to study the structure and morphological characteristics of the HGNs. The GPC3 electrochemical aptamer nanobiosensor was constructed using HGNs-aptamer (HGNs-Apt) as the signal probe and GPC3 aptamer as the capture probe. With the help of the catalytic action of peroxidase-like properties of HGNs, H 2 O 2 reduces the silver (Ag) ions in solution to metallic Ag, which deposit on the surface of the electrode. The amount of deposited Ag, which was derived from the amount of GPC3, was quantified by differential pulse voltammetry (DPV). Under optimal conditions, the current response of Ag had a good positive correlation with the GPC3 concentration in the range 10.0–100.0 μg mL −1 with a correlation coefficient of 0.9958. The detection limit was 3.16 μg mL −1 at a signal-to-noise ratio of 3, and the sensitivity was calculated to be 0.807 μA μM −1 cm −2 . The method is validated by analyzing spiked human serum samples with good recovery ranging from 101 to 122%. In addition, the GPC3 electrochemical aptamer nanobiosensor has acceptable selectivity, stability, and reproducibility.

: Bisphenol A (BPA), one of the most important endocrine disrupting chemicals , is a threat to human and wildlife health. Electrochemical enzyme biosensor has been regarded as ideal alternative analytical technique for ultrasensitive and rapid detection of BPA, while the unstable and easily deactivated nature of enzyme limits its development. In order to improve the stability of enzyme, tyrosinase was chosen as a model enzyme , and tyrosinase nanocapsules (nTyr) were prepared by encapsulating a single tyrosinase molecule into a thin network polymer shell through in-situ polymerization method in aqueous solution. The characterization of particle size distribution , TEM and FTIR indicated the successful formation of single tyrosinase molecule nanocapsule. And the porous network polymer shell of nTyr ensured the maintenance of tyrosinase activity and fast substrate transportation. The obtained nTyr was used to construct an electrochemical biosensor for BPA detection, exhibiting a low detection limit of 12 nmol L -1 and a wide linear range from 5 × 10 -8 to 2 × 10 -6 mol L -1 . Compared with native tyrosinase, the nTyr based biosensor displayed dramatically enhanced stability including thermal stability , organic solvent tolerance and acid/base tolerance. The excellent performance of nTyr based biosensor was not only attributed to the protection of biocompatible rigid polymer shells, but also the multipoint covalent attachments between tyrosinase cores and polymer shells. The robust biosensor was further used for rapid detection of BPA leached from plastic products with satisfactory results. The nTyr based nano-biosensor provides a prospective solution to resolve the stability problem of enzyme biosensors in different application scenarios.

MicroRNAs (miRNAs), a class of small, endogenous, noncoding RNA molecules, can serve as biomarkers for potential applications in cancer diagnosis, prognosis, and prediction due to its abnormal expression. As a result, a novel label-free biosensor with nanometer scale was prepared and employed in the detection of trace oligonucleotides based on the localized surface plasmon resonance (LSPR). The dielectric constant on the surface of DNA modified gold nanoparticle would change when probe single-strand DNA hybridized with target oligonucleotides, which resulted in the notable red shift of scattering peak position. The biosensor with excellent selectivity can be used in a real-time monitoring hybridization process. Notably, this method provided label-free detection of DNA and miRNA at single nanoparticle level with limit of detection up to 3 nM. Due to the advantages of LSPR scattering spectra, single nanoparticle biosensor can be designed for trace cancer-relevant miRNAs detection in the future.
