A self-assembled and H2O2-activatable hybrid nanoprodrug for lung infection and wound healing therapy

Rui Zhang,Danhua Mao,Yiyong Fu,Rong Ju,Guoqing Wei

Background: The pursuit of effective antibacterial strategies aimed at mitigating pathogenic bacterial infections while minimising drug resistance remains of paramount importance. A combinational therapeutic strategy that integrates distinct treatment components can enhance overall efficacy and mitigate undesired effects, thereby exhibiting considerable promise in combating bacterial infections. Methods: In this study, a meticulously engineered self-assembling hybrid nanoprodrug (CPBP NPs) has been devised, functioning as a hybrid prodrug of Ciprofloxacin (Cip) and hydroxybenzyl alcohol (HBA). Results: CPBP molecules can generate nanoassemblies via self-assembly and subsequently undergo decomposition to synchronously release Cip and HBA upon hydrogen peroxide (H 2 O 2 ) exposure. The CPBP NPs exert antibacterial and anti-inflammatory properties through the controlled release of Cip and HBA, while also facilitating the scavenging of reactive oxygen species. These CPBP NPs exhibit broad-spectrum antibacterial activity against both Gram-negative bacteria ( E. coli , 98.4%) and Gram-positive bacteria ( S. aureus , 98.5%). Notably, CPBP NPs not only accumulate in the lungs to facilitate organ-specific infection treatment but also expedite the healing process of infected wounds. Conclusions: Consequently, this H 2 O 2 -activatable hybrid nanoprodrug, possessing excellent biocompatibility, holds substantial promise for advancing clinical applications in managing bacterial infections.

Precision targeted melanoma therapy via cuproptosis/chemodynamic and chemotherapy: An engineering MCHS-CuMOF nanodelivery system

Yixun Zhang,Wenjie Xie,Jinchuang Li,Zhenguo Liang,Xin Zhou,Zeheng Tan,Zhenjie Wu,Jundong Lin,Yangjia Zhuo,Fen Zou,Biyan Wen,Jianwei Chen,Shanghua Cai,Jianheng Ye,Yingke Liang,Jianming Lu,Huikang Yang,Weide Zhong,Ronghua Yang

Melanoma, a highly aggressive skin cancer, continues to challenge current therapeutic modalities due to its resistance and high mortality rates. Recent advancements highlight cuproptosis, a copper-driven form of programmed cell death, as a promising target for melanoma treatment. This study integrated machine learning and large-scale genomic data to identify FDX1 as a pivotal gene in cuproptosis-related pathways for melanoma. We developed a novel nanomedicine, ACM@MCHS-CuMOF@Dox, combining Mesoporous Carbon Hollow Spheres (MCHS) loaded with Copper-based Metal-Organic Frameworks (CuMOFs) and Doxorubicin (Dox), to exploit this discovery. The nanomedicine leverages a biomimetic approach by incorporating A375 cell membranes, enhancing tumor-targeted delivery. Physicochemical characterization confirms optimal drug loading and pH/GSH-responsive release profiles. In vitro studies demonstrate that ACM@MCHS-CuMOF@Dox inhibits melanoma cell proliferation, migration, and invasion, outperforming other formulations. Mechanistic investigations revealed that ACM@MCHS-CuMOF@Dox induced robust apoptosis and cuproptosis through FDX1 downregulation, thereby enhancing oxidative stress and therapeutic efficacy. These findings underscore the potential of combining machine learning-driven target identification with advanced nanomedicine for precision melanoma therapy. This approach offers a promising strategy for overcoming current treatment limitations and advancing personalized cancer care.

Selenium-deposited tripterine phytosomes ameliorate the antiarthritic efficacy of the phytomedicine via a synergistic sensitization

Shiping Zhu,Chunhua Luo,Weifeng Feng,Yan Li,Mujuan Zhu,Shengyun Sun,Xingwang Zhang

Arthritis remains the notion of a hard-to-treat disease that raises an area of unmet clinical need. The phytomedicine tripterine (Tri) and trace element selenium (Se) have been shown to be of anti-inflammatory activity. This study was devoted to develop nanomedicine containing Tri and Se used for fighting against arthritis via a coordination mechanism. Se-deposited Tri phytosomes (Se@Tri-PTs) were prepared by a melting-hydration/ in situ reduction technique and characterized by particle size, ζ potential , morphology, and entrapment efficiency ( EE ). The resultant Se@Tri-PTs were 126 nm around in particle size with an EE of 98.85%. Se@Tri-PTs exhibited a sustained drug release both in 0.1 M HCl and pH 6.8 PBS compared with Se-free phytosomes (Tri-PTs). The in vivo antiarthritic test demonstrated that Se@Tri-PTs could result in significant resolution of arthritis and decline of inflammatory factors. Phytosomes primely facilitated the transepithelial transport of Tri, while Se enhanced the antiarthritic efficacy of the phytomedicine synergistically. The present work provides a proof-of-concept for the combined therapy of arthritis using Tri and Se in the form of nanoparticles .

Supramolecular PEGylation of camptothecin for cancer therapy

R. Sun,Q. Luo,X. Li,X. Huang,L. Teng,Z. Shen,W. Zhu

PEGylated drugs and their self-assembled nanoparticles have been used as a first-line technology in nanomedicines . However, premature disassembly before reaching the lesions under dilute conditions limits their bioavailability. This challenge can be overcome by covalent crosslinking. However, undefined molecular structures of the prodrugs are introduced, which hinder large-scale production and clinical translation. Herein, a supramolecular PEGylated camptothecin nanodrug with a crosslinked architecture and defined molecular drug structure was developed. PEGylated mono-tetraethylenepentamine β-cyclodextrin (mPEG-TEPA-CD) and adamantane modified camptothecin (AD-SS-CPT) were first synthesized. The host-guest coupling of β-cyclodextrin (β-CD) and adamantine (AD) moieties between mPEG-TEPA-CD and AD-SS-CPT induced the formation of supramolecular PEGylated camptothecin, which could self-assemble into micelles in the aqueous medium. They were further stabilized by the addition of a diacrylate crosslinker for reaction with the amine groups on mPEG-TEPA-CD, maintaining the unaltered AD-SS-CPT. The nanodrugs can internalize into tumor cells quickly and release free camptothecin in response to glutathione . They also have prolonged blood circulation time and can accumulate efficiently at tumor sites, thus showing effective antitumor activity . This supramolecular drug delivery system provides a new strategy for the design of nanodrugs.

Carrier-free nanodrugs for in vivo NIR bioimaging and chemo-photothermal synergistic therapy

Juanfang Lin,Chao Li,Yan Guo,Junjie Zou,Pengyu Wu,Yuqin Liao,Bingchen Zhang,Jingqing Le,Ruirui Zhao,Jing-Wei Shao

The combination of chemotherapy and photothermal therapy displays improved anti-cancer effects and lower systematic toxicity of a free drug compared with monotherapy. In this study, we designed innovative, carrier-free nanodrugs (PTX/ICG NDs) composed of the chemotherapeutic agent paclitaxel (PTX) and the photosensitizer indocyanine green (ICG) via self-assembly. The nanodrugs not only incorporated two different modalities into one delivery system for combined chemo-photothermal therapy but also enhanced the solubility of PTX without the need for any carrier. The as-prepared PTX/ICG NDs exhibited the merits of a relatively uniform size of 140 ± 1.4 nm, surface charge of −36 ± 2.2 mV, and high drug loading content of PTX. The combination strategy exerted a synergistic effect on the cytotoxicity of cancer cells in vitro, which could be attributed to the high cellular uptake and sustained release of PTX. Furthermore, an in vivo study indicated that PTX/ICG NDs showed higher accumulation in the tumor site than free ICG and possessed strong synergistic chemo-photothermal therapy efficacy against tumors in H22 tumor-bearing mice. Taken together, our study demonstrates that PTX/ICG NDs hold promise to become an alternative chemo-photothermal therapy agent to treat cancers.

Targeted delivery of a guanidine-pendant Pt(IV)-backboned poly-prodrug by an anisamide-functionalized polypeptide

Shao-Lu Li,Yaoyi Wang,Jingfang Zhang,Wei Wei,Hua Lu

We describe here a novel targeting polyion complex (Tg-PIC) system for the delivery and intracellular release of cisplatin. Briefly, a guanidinium-pendant Pt(IV)-backboned poly-prodrug termed P(DSP-Gu) is prepared with excellent aqueous solubility, high drug-loading and high potency. To enable prolonged circulation and selective cellular internalization, P(DSP-Gu) is complexed with anisamide-end-capped poly(ethylene glycol)-block-poly(L-phosphotyrosine)-block-poly(L-leucine) (AA-PEG-PpY-PLeu) to yield Tg-PIC via electrostatic coacervation. Tg-PIC is stabilized by hydrogen bonding between phosphate and guanidinium, the PEG corona, and the helical poly(L-leucine) segment forming the hydrophobic core. The anisamide group, a high affinity ligand recognizing the sigma (σ) receptors that are overexpressed on many human malignancies including prostate cancer, is incorporated at the surface of the Tg-PIC for active targeting and efficient internalization. In vitro, the Tg-PICs show targeted and efficient internalization into sigma receptor-positive PC3 cells, and can release toxic Pt(II) species due to the degradation of P(DSP-Gu) under the intracellular reducing conditions. In vivo, the Tg-PICs exhibit superior antitumor efficacy with reduced toxicity. Thus, the system holds considerable promise towards more effective and safe nanomedicine.

Hydrazided hyaluronan/cisplatin/indocyanine green coordination nanoprodrug for photodynamic chemotherapy in liver cancer

Guanghui Hou,Junmin Qian,Min Guo,Weijun Xu,Jinlei Wang,Yaping Wang,Aili Suo

It is still a huge challenge for concurrent highly efficient loading of chemotherapeutic agent and photosensitizer into single nanocarrier via stimuli-responsive linkages due to their different physicochemical properties and pharmacokinetics . Herein, based on the discovery of unique cisplatin-hydrazide and cisplatin-indocyanine green (ICG) coordination reactions, a multifunctional coordination nanoprodrug, cisplatin/ICG co-loaded hydrazided hyaluronan/bovine serum albumin (HBCI) nanoparticles , was developed by a desolvation-dual coordination process. The nanoprodrug exhibited ultrahigh drug loading efficiency and glutathione/NIR light dual-responsive drug release behavior. In vitro cellular studies demonstrated efficient internalization and apoptosis-inducing ability of the nanoprodrug in HepG2 cells. In vivo results confirmed the efficacious tumor accumulation and biosafety of HBCI nanoprodrug and synergistic effect of HBCI-based combined photodynamic chemotherapy on inhibiting tumor growth. Overall, this work not only provides a novel dual coordination approach for highly efficient loading of cisplatin and ICG but also verifies the therapeutic potential of HBCI nanoprodrug in combating hepatocellular carcinoma.

Vinpocetine-mediated multidimensional active targeted delivery across the blood-brain barrier

Zhongyi Ma,Xinlong Liu,Xinyu Xie,Yike Fang,Dingxi Jiang,Chen Zhao,Pingrong Li,Yu Fu,Lianghong Ye,Yuan Gao,Shuang Wu,Chong Li

Overcoming the challenges associated with brain-targeted drug delivery involves addressing both the limited biodistribution of drugs and microcirculation issues that hinder their accumulation in brain lesions. In this study, we propose a comprehensive strategy leveraging vinpocetine, a compound known to enhance cerebral blood flow (CBF), to improve drug delivery to the brain. Initially, the pre-administration of vinpocetine in the form of a microemulsion significantly enhances nanomedicine uptake into the brain by selectively boosting CBF. This creates a more favorable environment for drug distribution while also mitigating chemotherapy-induced cerebrovascular accidents. Building upon this, the use of vinpocetine as a targeting ligand for liposomes or lipid nanoparticles, compared to typical brain-targeting ligands (GSH, Apamin, and Angiopep-2), not only significantly improves drug distribution in the brain but also selectively targets glioma regions and maintains excellent immunocompatibility. Expanding on these findings, the direct conjugation of vinpocetine with therapeutic compounds leads to superior brain accumulation and enhanced antitumor efficacy, thereby extending its therapeutic potential. These results highlight the broad applicability of vinpocetine-based strategies for achieving efficient and precise brain-targeted drug delivery, offering promising prospects for treating a range of central nervous system disorders.

All-stage targeted therapy for invasive cryptococcosis through interaction between the secretory protein Cig1 and hemin

Liting Cheng,Zhongyi Ma,Xinlin Yang,Xue Wang,Yuqiong Wang,Xinlong Liu,Zhongjie Tang,Dingxi Jang,Guojian Liao,Tongbao Liu,Shuang Wu,Chong Li

Cryptococcosis, a serious systemic fungal infection caused by Cryptococcus neoformans ( C. neoformans ) and its variants, poses a significant clinical challenge due to its poor prognosis and severe health implications. The treatment of cryptococcal infections is complicated by several unique factors, stemming from both the pathogenic characteristics of the fungi and the biological barriers they exploit. These include the fungi's protective capsule, their ability to reside within host macrophages—thereby evading pharmacological intervention—and their involvement in multi-organ infections such as the lung and brain, in particular their strategic positioning within the brain, protected by the blood-brain barrier (BBB). To overcome these obstacles, precise active targeting emerges as a pivotal strategy. Identifying common targets is imperative to enhance therapeutic efficacy while ensuring the druggability of delivery systems. However, research on the methodology for selecting such shared targets remains sparse. In our investigation, we have pioneered the use of secreted proteins as shared target to trace the pathogens and their infection pathways. We identified the mannoprotein Cig1, prominently expressed on the surfaces of infected macrophages, lungs, and brains, as a viable shared target. On this basis, we utilized Hemin, a ligand for Cig1, to design liposomes (Hemin Lip) tailored for addressing complex fungal infections. By leveraging the interaction with the secreted protein Cig1, Hemin Lip specifically identifies and binds to organs and macrophages harboring cryptococcal infections, thereby facilitating targeted and efficacious clearance of both intracellular and extracellular fungus. Moreover, we have extended this targeting mechanism to other nanomedicinal platforms, including albumin nanoparticles. This study proposes an innovative drug delivery model that targets extracellular secretory proteins within the infection microenvironment, offering a streamlined formulation with the potential for effective therapy against complex infections.

Preparation and anti-tumor application of hyaluronic acid-based material for disulfide and copper ions co-delivery

Pang Long,Zhong Wei,Wang QiuBo,Feng HaoHui,Dong HaoNan,Wang Song,Cong HaiLin,Shen YouQing,Yu Bing

Disulfide (DSF) has been proved good anti-tumor effect and even better with coadministration of Cu 2+ . In this work, we report the use of hyaluronic acid (HA) based materials to construct vectors for the delivery of both DSF and Cu 2+ . HA was firstly modified with polyethylene glycol monomethyl ether (mPEG) and polycaprolactone (PCL) to synthesize an amphiphilic polymer (HA-PEG-PCL). DSF could be loaded in the hydrophobic core and Cu 2+ could be cooperated to the negative hydrophilic segment. The Cu 2+ also played a role as crosslinking agent, which prevented DSF leakage prematurely, avoiding the bad side effects to normal tissues. The interaction between HA and CD44 improved the distribution of nanodrugs in tumor cells. When the nanodrugs were delivered to the cancer cell, the acidic micro-environment would separate the Cu 2+ from the surface, leading to the disintegration of the micelles, promoting the release of DSF from the micelle core. The results of in vitro and in vivo experiments showed that the DSF and Cu 2+ co-delivery vector constructed in this work could enhance the antitumor effect and have low biological toxicity.

Plant-Derived Flavonoid and Antibiotic Self-Assembled Nanomedicines for Enhanced Efficacy Against Bacterial Infections

Mengying Zhang,Pingping Yuan,Yang Zhao,Shuo Yang,Xuan Ma,Qingjun Xu,Kezong Qi,Lin Li,Shaoqi Qu

Bacterial infections pose a serious challenge to public health and contribute to significant economic losses. The limited solubility of many antibiotics affects their absorption and therapeutic potential. Nanobased delivery systems offer a promising approach to improve solubility and therapeutic efficacy, though challenges such as nanocarrier toxicity and low drug loading rates remain. Here, we present a carrier-free drug delivery strategy to enhance the antibacterial efficacy. Two poorly water-soluble drugs, rifampicin (RIF) and isobavachalcone (IBC), were formulated into nanoparticles (RI@NP), with sodium lauryl sulfate and ammonium acetate added to improve the stability and polarity. RI@NP demonstrated superior efficacy at lower doses compared with RIF and IBC alone. Mechanistic studies revealed that RI@NP disrupts bacterial membrane integrity, leading to PMF collapse, impaired electron transport, ATP depletion, intracellular ROS accumulation, and ultimately bacterial cell death. Furthermore, RI@NP effectively eliminated 99% of Staphylococcus aureus within 24 h. Compared to the separate use of RIF and IBC, RI@NP showed enhanced efficacy in the mouse peritonitis-sepsis model, with a 100% survival rate and a 99% reduction in bacterial load in major organs. Our designed carrier-free RI@NP could achieve enhanced efficacy and shed light on the development of carrier-free nanoparticles against bacterial infections.

Limiting tumor cells comprehensively at micro and macro levels to improve the therapeutic effect of chemotherapy

Binghua Wang,Lulu Sun,Jing Zhao,Jingyi An,Yajie Jin,Xinwei Yang,Haixia Li,Hongling Zhang,Zhenzhong Zhang,A Youmei

Clinical data shows that antitumor treatments are often ineffective if tumor cells have metastasized. To gain an effective antitumor therapeutic effect, in this report, the tumor cell was limited to the primary site and simultaneously ablated by chemotherapy. Considering the extremely complicated process of cancer metastasis, we seek to comprehensively suppress tumor metastases at both micro and macro levels, which closely link to migration and interact with each other. At the micro level, the motility of the tumor cell was decreased via accelerating mitochondria fusion. At the macro level, the unfavorable hypoxia environment was improved. A liposome-based multifunctional nanomedicine was designed by coloading latrunculin B (LAT-B), an inhibitor of actin polymerization, and doxorubicin (DOX) into the hydrophobic bilayers and aqueous cavity, respectively. Meanwhile, an oxygen reservoir named perfluoropentane (PFP) was encapsulated into the liposome core to fulfill synergistic treatment of metastatic tumors. In this paper, we demonstrated that the metastasis of the tumor cell could be effectively inhibited by LAT-B through promoting mitochondria fusion without affecting its function, making it as an encouraging candidate for effective anti-metastasis therapy. Meanwhile, we found that the combination of LAT-B and DOX shows a synergistic effect against tumors because the combined effect of these two drugs cover the entire cell proliferation process. In a word, this report presents a potential improvement in the treatment of metastatic cancer.
