Adaptive particle patterning in the presence of active synthetic nanomotors

Dazhi Xie,Shaoming Fu,Dongmei Fu,Bin Chen,Weidong He,Haiying Liang,Yingfeng Tu,Daniela A. Wilson,Fei Peng

For the maintenance of a biological system, spatial organization of material condensates within the cell through the dissipation of energy is crucial. Besides directed transport via microtubules, material arrangement can be achieved via motor protein facilitated adaptive active diffusiophoresis. For example, the distribution of membrane proteins during the cell division of Escherichia coli is affected by the MinD system. Synthetic active motors exhibit the ability to simulate natural motors. Here we propose an active Au–Zn nanomotor driven by water and discovered an interesting adaptive interaction mode of the diffusiophoretic Au–Zn nanomotors with passive condensate particles in different environments. It is found that the attraction/repulsion between the nanomotor and passive particles is adaptive, while an interesting hollow pattern is formed with a negatively charged substrate and a cluster pattern is favored with a positively charged substrate.

Self-propelling nanomotor made from halloysite and catalysis in Fenton-like reaction

Jian Wang,Yan Zhang,Weikun Ning,Peiping Zhang,Bo Jin,Yan Wang,Wei Zhang,Wenqing Li,Cundi Wei,Shiding Miao

A new class of catalytically self-propelled nanomotors were fabricated by modifying natural clay tubes, halloysite, with randomly distributed particles of MnO 2 and Fe 3 O 4 . The prepared MnO 2 –Fe 3 O 4 /HNTs composites were validated to be efficient Fenton catalysts in the degradation of rhodamine B (RhB). Compared to the previous preparation of rolled-up microtubes or other template-assisted syntheses, this strategy has its merits in utilizing clay minerals of abundance, cheap-price, and no complex instruments needed. The nanomotors were able to be prepared on a large scale. The MnO 2 –Fe 3 O 4 /HNTs motors displayed powerful autonomous movement, and a high velocity of up to 380 µm s −1 was achieved in 5.0 wt. % H 2 O 2 solution. For Fenton catalysis of RhB, the removal ratio of 94% dye molecules was obtained within 30 min, which was triply higher than other samples of the nonpropelling sample Fe 3 O 4 /HNTs. The autonomous movement provided adsorptive bubble separation, and the adsorption capacity was greatly enhanced by halloysite. These synergistic effects boosted the removal efficiency of dye molecules. The presence of magnetic MnO 2 –Fe 3 O 4 made these motors move directionally in external magnetic fields and provided a facile recovery for collecting heterogeneous catalysts.

An Endogenous Proton-Powered Adaptive Nanomotor for Treating Muscle Atrophy

Ming Liu,Zhicun Liu,Xiangkai Qiao,Cheng Chen,Hongtu Guo,Hao Gu,Junbo Li,Tiedong Sun

Nanomotors driven by endogenous enzymes are favored in biology and pharmacy due to their spontaneous driving and efficient biocatalytic activity, and have potential applications in the treatment of clinical diseases that are highly dependent on targeted effects. For diseases such as muscle atrophy, using energy molecules such as ATP to improve cellular metabolism is a relatively efficient treatment method. However, traditional adenosine triphosphate (ATP) therapies for muscle atrophy face limitations due to instability under physiological conditions and poor targeting efficiency. To address these challenges, we developed an endogenous proton-gradient-driven ATP transport motor (ATM), a nanomotor integrating chloroplast-derived FoF1-ATPase with a biocompatible flask-shaped organic shell (FOS). The ATM is synthesized by vacuum-injecting phospholipid-embedded FoF1-ATPase nanothylakoids into ribose-based FOS, enabling autonomous propulsion in acidic microenvironments through proton-driven negative chemotaxis (directional movement away from regions of higher proton concentration). This nanomotor converts proton gradients into ATP synthesis, directly replenishing cellular energy deficits in atrophic tissues. In vitro studies demonstrated high biocompatibility (>90% cell viability at 150 μg/mL) and pH-responsive motility, achieving speeds up to 4.32 μm/s under physiological gradients (ΔpH = 3). In vivo experiments using dexamethasone-induced muscle atrophy mice revealed that ATM treatment accelerated weight recovery and restored normal muscle morphology, with treated mice exhibiting cell sizes comparable to healthy controls (30–40 μm vs. 15–25 μm in untreated). These results highlight the ATM’s potential as a precision therapeutic platform for metabolic disorders, leveraging the natural enzyme functionality and synthetic material design to enhance efficacy while minimizing systemic toxicity.

Gastric Acid Powered Nanomotors Release Antibiotics for In Vivo Treatment of Helicobacter pylori Infection

Yang Wu,Zhiyong Song,Guiyun Deng,Kai Jiang,Huajuan Wang,Xueji Zhang,Heyou Han

Helicobacter pylori ( H. pylori ) infection has ≈75% probability of causing gastric cancer, so it is considered to be the strongest single risk factor for gastric malignancies. However, the harsh gastric acid environment has created obstacles to medical treatment. This work reports a nanomotor with a bottle-shaped container that can be loaded with small molecules of clarithromycin, nano calcium peroxide (CaO 2 ), and Pt nanoparticles (Pt NPs) by ultrasound. Nanomotors can quickly consume gastric acid through the chemical reaction of CaO 2 to temporarily neutralize gastric acid. The product hydrogen peroxide (H 2 O 2 ) is catalytically decomposed into a large amount of oxygen (O 2 ) by Pt NPs. The local concentration gradient of O 2 bubbles causes it to be expelled from the nanobottles through a narrow opening, and then push the nanobottles forward to provide maximum release and prodrug efficacy. Experiments in animal models show that 15 mg nanomotors can safely and quickly neutralize gastric acid in the stomach and simultaneously release prodrugs to achieve good therapeutic effects without causing acute toxicity. H. pylori burden in mice was 2.6 orders of magnitude lower than that in the control group. The stomach returns to normal pH within 1 d after administration.

Piezoelectric Amplification of Cascade Enzymatic Catalysis and Nanomotor Propulsion for Synergistic Electrodynamic-Starvation Tumor Therapy

Zhanlin Zhang,Kun Wei,Jie Meng,Junwu Wei,Yupeng Su,Huan Tan,Xiaohong Li

Piezoelectrodynamic therapy (PEDT) is compromised by hypoxia dilemma of tumors, while starvation therapy is constrained by insufficient enzyme activities. To address these challenges, Janus piezoelectric nanoparticles (NPs) are proposed to spatially immobilize glucose oxidase (GOx) and catalase (CAT), enabling piezoelectric potential-amplified enzyme activities and synergistic PEDT-starvation tumor therapy. Here hollow barium titanate (hBT) NPs are synthesized using SiO 2 templates, followed by partial Au deposition via the Pickering emulsion-masking method to create Janus hBT@Au NPs, which are then conjugated with GOx and CAT on opposing sides to yield C-hBT@Au-G NPs. The hollow structure of hBT enhances flexibility and deformation under ultrasonication, while Schottky heterojunctions with Au layers promote charge carrier transfer, amplifying piezoelectric effects and free electron transfer to boost GOx activities. Piezoelectric field-enhances selective tumor cell internalization of NPs and PEDT generation of reactive oxygen species (ROS), coupled with self-propagated GOx/CAT cascades, intensify tumor cytotoxicities and deplete intracellular adenosine triphosphate. The Janus architecture, ultrasonic cavitation, and O 2 generation collaboratively drive robust propulsion for efficient NP accumulation and deep ROS penetration into tumor tissues, thereby achieving full tumor suppression with negligible systemic toxicity. This design overcomes delivery barriers of tumor accumulation, intratumoral penetration, and cellular uptake and synergizes PEDT-starvation tumor therapy.

Cascade-driven nanomotors promote diabetic wound healing by eradicating MRSA biofilm infection

Jia Zheng,Yuanyuan Deng,Senfeng Zhao,Longhai Liu,Wei Wang,Jiawei Li,Wansong Chen,Liu Deng,Jianghua Li,You-Nian Liu

Bacterial infection, hypoxia, and inflammatory stress responses are the key obstacles that delay the healing of diabetic ulcers. Herein, DMSNs-Pt-LOX@Nisin (DPLN) nanomotors that exhibits cascade propulsion and can specifically target methicillin-resistant Staphylococcus aureus (MRSA) is developed. The DPLN can facilitate the conversion of the abundant lactic acid existed in diabetic wounds, resulting in the generation of hydrogen peroxide (H 2 O 2 ). Meanwhile, the unilateral Pt nanodendrites (Pt DNs) catalyze H 2 O 2 to produce O 2 , driving the nanomotors to rapidly penetrate into the biofilm. Moreover, by loading Nisin, which can bind to lipid II of Gram-positive bacteria, the nanomotors can target MRSA through a self-driven process. Both in vitro and in vivo experiments demonstrate that the nanomotors can eradicate MRSA biofilms. Furthermore, animal studies show that DPLN are able to expedite the process of wound healing in diabetic mice through the eradication of biofilms, facilitation of angiogenesis, and mitigation of inflammation. These novel nanomotors combining cascade propulsion and MRSA targeting represent an encouraging and potentially effective approach in diabetic ulcer treatment.

Single molecule–driven nanomotors reveal the dynamic-disordered chemomechanical transduction of active enzymes

Zhuodong Tang,Jingyu Wu,Shaojun Wu,Wenjing Tang,Jian-Rong Zhang,Wenlei Zhu,Jun-Jie Zhu,Zixuan Chen

Enzymes facilitate the conversion of chemical energy into mechanical work during biochemical reactions, thereby regulating the dynamic metabolic activity of living systems. However, directly observing the energy release facilitated by fluctuating individual enzymes remains a challenge, leading to a contentious debate regarding the underlying reasons for this phenomenon. Here, we aim to overcome this challenge by developing an oscillating nanomotor powered by a single-molecule enzyme, which allows real-time tracking of energy transduction in enzymatic reactions. Through analysis of the shifts in free energy profiles within the nanomotors, our results unveil not only the heterogeneous energy release patterns of individual enzyme molecules but also the dynamic disorder of a particular enzyme in energy release over extended monitoring periods. By exploring six distinct types of single-molecule enzymatic reactions, we provide the direct evidence supporting the argument that the reaction enthalpy governs the enzymatic energy release. This approach has implications for understanding the mechanism of enzymatic catalysis and developing highly efficient nanomotors.

A Programmable Oral Nanomotor Microcapsule for the Treatment of Inflammatory Bowel Disease

Jingwen Han,Jiamin Ye,Jiacheng Shi,Yueyue Fan,Xue Yuan,Ruiyan Li,Gaoli Niu,Muhammad Abubakar,Yong Kang,Xiaoyuan Ji

Given the unique anatomy and location of inflammatory bowel disease (IBD), oral pharmacological treatment is the mainstay for managing IBD because of its convenience and direct accessibility to the gastrointestinal tract. However, efficient delivery systems must be designed to navigate the harsh gastrointestinal environment during application. Here, an innovative oral drug delivery system was proposed using nanomotor (NM)-loaded soluble microcapsules for treating IBD. MnO 2 -Au-mSiO 2 NMs incorporate partially encapsulated MnO 2 antennas that convert reactive oxygen species into oxygen, autonomously propelling the NMs. Astaxanthin (AST), known for its anti-inflammatory properties, is loaded into the mesoporous silica (mSiO 2 ) of NMs (AST@NMs). AST effectively reduces inflammation and shifts macrophages from a proinflammatory (M1) phenotype to an anti-inflammatory (M2) phenotype. To protect the nanomotors from harsh digestive conditions and achieve intestinal-responsive release, using photocurable 3D printing, we fabricated enteric-coated oral microcapsules (MCs). Additionally, to prevent leakage of AST@NMs, a calcium alginate shell was cured in situ on the surface of the microcapsules (AST@NMs@MCs). In vitro and in vivo, AST@NMs@MCs effectively reduced inflammation, repaired the intestinal barrier, and modulated the gut microbiota. These findings underscore the protective effects of the microcapsules on AST@NM, highlighting their potential as a promising strategy for treating colitis.

Sulfur dioxide-releasing nanomotors improve the therapeutic effect of liver fibrosis by restoring the fenestrae of sinusoids

Lin Chen,Zhengwei Chen,Di Shi,Haifeng Ke,Chun Mao,Mimi Wan

The dual barriers formed by the capillarized liver sinusoids and excessive deposited extracellular matrix (ECM) significantly impede the retention of therapeutic agents in the fibrotic liver. Currently, there are limited reports on strategies capable of simultaneously overcoming these barriers. Here, we propose sulfur dioxide (SO 2 )-releasing nanomotors based on endogenous in vivo reactions to restore the fenestrae of sinusoids and degrade ECM by activating the endogenous signaling pathway to improve the retention of therapeutic agents in the damaged liver. These nanomotors leverage the specific enzyme concentration gradient in damaged liver tissue as a chemoattractant signal, guiding their targeted delivery. The nanomotors incorporate an l -cysteine-based substrate that, upon enzymatic catalysis, generates SO 2 . The released SO 2 can upregulate the cyclic guanosine monophosphate expression to restore the fenestrated phenotype of liver sinusoidal endothelial cells. Concurrently, SO 2 can stimulate the endogenous nitric oxide production to induce matrix metalloproteinase-1 activation to facilitate the collagen degradation. The animal experimental model also demonstrates the effective retention of nanomotors in damaged liver tissue and reversal of liver fibrosis.

Black Titania Janus Mesoporous Nanomotor for Enhanced Tumor Penetration and Near-Infrared Light-Triggered Photodynamic Therapy

Kexin Lv,Mengmeng Hou,Yufang Kou,Hongyue Yu,Mengli Liu,Tiancong Zhao,Jiacheng Shen,Xirui Huang,Jie Zhang,Mohamed F. Mady,Ahmed A. Elzatahry,Xiaomin Li,Dongyuan Zhao

Thanks to their excellent photoelectric characteristics to generate cytotoxic reactive oxygen species (ROS) under the light-activation process, TiO2 nanomaterials have shown significant potential in photodynamic therapy (PDT) for solid tumors. Nevertheless, the limited penetration depth of TiO2-based photosensitizers and excitation sources (UV/visible light) for PDT remains a formidable challenge when confronted with complex tumor microenvironments (TMEs). Here, we present a H2O2-driven black TiO2 mesoporous nanomotor with near-infrared (NIR) light absorption capability and autonomous navigation ability, which effectively enhances solid tumor penetration in NIR light-triggered PDT. The nanomotor was rationally designed and fabricated based on the Janus mesoporous nanostructure, which consists of a NIR light-responsive black TiO2 nanosphere and an enzyme-modified periodic mesoporous organosilica (PMO) nanorod that wraps around the TiO2 nanosphere. The overexpressed H2O2 can drive the nanomotor in the TME under catalysis of catalase in the PMO domain. By precisely controlling the ratio of TiO2 and PMO compartments in the Janus nanostructure, TiO2&PMO nanomotors can achieve optimal self-propulsive directionality and velocity, enhancing cellular uptake and facilitating deep tumor penetration. Additionally, by the decomposition of endogenous H2O2 within solid tumors, these nanomotors can continuously supply oxygen to enable highly efficient ROS production under the NIR photocatalysis of black TiO2, leading to intensified PDT effects and effective tumor inhibition.

Bioactive nanomotor enabling efficient intestinal barrier penetration for colorectal cancer therapy

Wang Zhi-Hao,Zeng Xuejiao,Huang Wanting,Yang Yanbo,Zhang Shuhao,Yang Mingzhu,Liu Hua,Zhao Fengqin,Li Airong,Zhang Zhenzhong,Liu Junjie,Shi Jinjin

Oral drug delivery systems had natural potential for colorectal cancer drug therapy. While the drug delivery efficiency is severely hindered by the complex intestinal barriers, especially mucus and epithelium barriers, resulting in unsatisfactory therapeutic effects and limited clinical translation. In this work, a bioactive self-thermophoretic and gas dual-driven nanomotor is developed for colorectal cancer therapy through efficient intestinal mucus and epithelial barrier penetration. The nanomotor shows intestinal mucus barrier penetration and the paracellular pathway reversibly opening properties of intestinal epithelium barrier, increasing the delivery efficiency of cisplatin by 3.5 folds. Owing to the targeted delivery of cisplatin and the reduced side effects on normal intestinal tissues, the therapeutic efficiency of the nanomotor for colorectal cancer in vivo is as high as 98.6%. With autonomous and reversible intestinal barriers penetration property, the nanoplatform may innovate the current oral drug delivery.

Liquid Metal Amplified Charge Separation in Photocatalytic Micro/Nanomotors for Antibacterial Therapy

Zichang Guo,Dongdong Jin,Haohui Li,Xinyu Zhu,Tianqi Zheng,Zirong Xu,Yi Chen,Xiaojia Liu,Yinuo Song,Dai Wang,Xiaohui Yan,Xing Ma

Photocatalytic micro/nanomotors (MNMs) driven by electrophoresis have attracted considerable attention by virtue of their active mobility and versatile functionality. However, the rapid recombination of photogenerated electron–hole pairs on light illumination severely compromises the involvement of charge species in the catalytic redox reactions of fuels, thus hindering both the propulsion and the application performance of photocatalytic MNMs. Herein, we report a facile strategy to amplify charge separation by incorporating liquid metal (LM) into the construction of photocatalytic MNMs, thereby strengthening the electrophoretic propulsion of MNMs and promoting the generation of reactive oxygen species (ROS) for antibacterial application. The MNMs are constructed with a gallium (Ga) LM core, coated with abundant graphite-phase carbon nitride (g-C3N4) nanosheets and half covered by a thin platinum layer. These MNMs exhibit self-propulsion in hydrogen peroxide (H2O2) solution, with their motion dynamics further enhanced by light irradiation. Theoretical calculations and simulations reveal that the composition between Ga and g-C3N4 forms an ohmic junction in the electronic energy band structure, which effectively improves the charge separation efficiency of electron–hole pairs. These results align well with the experimental electrochemical tests and consequently intensify the catalytic redox reactions of H2O2, as well as accelerate the charge migration across MNMs, contributing to the enhancement of their propulsion performance. Simultaneously, the amplified separation of electrons facilitates increased ROS generation, empowering the MNMs with motion-enhanced antibacterial activity against Escherichia coli. Finally, an in vivo wound healing experiment is conducted, verifying the superior antibacterial therapeutic performance of photocatalytic MNMs. This work not only provides insights into the role of charge species in phoretic motion of MNMs but also gives inspiration for developing photocatalytic MNMs with advanced biomedical applications.
